RESUMO

The plant hormone strigolactones (SLs) play crucial roles in regulating plant development and adaptations to abiotic stresses. Even though the functional roles of SLs have been identified in response to abiotic stresses, the function, and mechanism of SLs are not fully established under alkaline stress. In this study, we identified that exogenous SL could improve alkaline tolerance of soybean seedlings, especially when treated with 0.5 µM SL. The application of SL remarkably reduced the malondialdehyde content, hydrogen peroxide content, and increased the activity of antioxidant enzymes under alkaline stress, suggesting that SL improved the alkaline tolerance by regulating the antioxidant defense capacity. The RNA sequencing data showed 530 special differentially expressed genes under SL treatment and alkaline stress, mainly were associated with antioxidant processes and phenylpropanoid biosynthetic pathway. Some transcription factors were also induced by SL under alkaline stress as confirmed by quantitative real-time PCR (qRT-PCR). Furthermore, SL largely increased the Na content in leaves and decreased Na content in roots under alkaline stress, which suggested that SL might promote the transport of Na from the roots to the leaves of the soybean seedlings. Meanwhile, exogenous SL decreased the content of other elements such as K, Mg, Fe, and Cu in leaves or roots under alkaline stress. Collectively, our results suggested a role of SL in regulating antioxidant defense capacity, specific gene expression, and alterations in ionic contents to alleviate harmful effects of alkaline stress in soybean seedlings.

RESUMO

Some traditional Chinese medicine (TCM) has been applied in bone repair, however, hydroxy-safflower yellow A (HYSA), one composition of safflower of the typical invigorating the circulation of TCM, has little been studied in orthopedics field for osteogenesis and angiogenesis clinically. Herein, we hypothetically speculated that the synthetic bioactive glasses (BG, 1393) scaffolds carried HYSA by a 3D print technique could enhance osteogenic repair properties. Notably, scaffolds coating chitosan/sodium alginate endowed with excellent drug control release ability, and significantly improved the BG mechanical strength. HYSA was loaded into BG scaffolds by coating chitosan/sodium alginate film, and the osteogenesis and angiogenesis of the HYSA/scaffolds were evaluated in vitro and in vivo. In vitro the cell culture results exhibited that the high dose of HYSA (0.5 mg/ml) loaded scaffolds can promote the proliferation of bone marrow stromal cells (rBMSCs) and migration, tubule formation of human umbilical vein endothelial cells (HUVECs). The active alkaline phosphatase (ALP) of rBMSCs can also be improved by the high dose of HYSA/scaffolds. Results of qRT-PCR and Western blot indicated that the high dose of HYSA/scaffolds can up-regulate ALP, OCN, OPN and RUNX-2 expression and relative protein secretion of the HIF-1α and BMP-2. In the animal experiment, the high dose of HYSA/scaffolds has a significantly better capacity to promote new bone formation than the undoped scaffolds at 8 weeks post-surgery. Thus, our results claimed that the novel HYSA/scaffolds hold the substantial potential to be further developed as effective and safe bone tissue engineering biomaterials for bone regeneration by combining enhanced osteogenesis and angiogenesis.

RESUMO

Seasonally frozen ground regions occupy approximately 55% of the exposed land surface in the Northern Hemisphere, and frost heave is the common global problem in seasonally frozen soil areas. Frost heave induces uneven deformation of ground and damages railways, road paving, and buildings. How to mitigate frost heave is the most important technical issue in this field that has provoked great interest. Here, using freezing experiments, we investigate the effect of anionic polyacrylamide (APAM) polymer on frost susceptible soil. The results demonstrate a so-far undocumented inhibition of frost heave by APAM in freezing soil, namely APAM (tested at concentrations from 0.0 wt% to 0.60 wt%) slows down the frost heave by a factor of up to 2.1 (since 0.60 wt% APAM can decrease frost heave from 8.56 mm to 4.14 mm in comparison to the control experiment). Moreover, it can be observed that the maximum water content near the frozen fringe decreased from 53.4% to 31.4% as the APAM content increased from 0.0 wt% to 0.60 wt%, implying a mitigated ice lens growth. Hydrogen bonding between APAM and soil particles triggers an adsorption mechanism that accumulates soil particles, and thus can potentially inhibit the separation and growth of the ice lens. Moreover, the residue of APAM due to hydrogen bonding-induced adsorption in the pores of granular media may narrow seepage channels (capillary barriers) and provide an unfavourable condition for water migration. The use of APAM can also increase the viscosity of the solution, which causes a greater water migration resistance. This research provides new insights into APAM-influenced frost heave (introducing APAM into the soil can induce bridging adsorption between APAM polymer segments and a particle surface), can enable engineers and researchers to utilise chemical improvement design and to consider suitable actions (e.g., by injecting APAM solution into a frost susceptible soil or using APAM-modified soil to replace the frost susceptible soil) to prevent frost heave from having a negative impact on traffic roads and buildings in cold regions.

RESUMO

The macromorphic properties of carbon nanotubes perform poorly because of their size limitations: nanosize in diameters and microsize in length. In this work, to realize these dual purposes, we first used an electrochemical method to tear the surface of multiwalled carbon nanotubes (MWCNTs) to anchor photonic Eu3+-complexes there. Through the polar reactive groups endowed by the tearing, the Eu3+-complexes coordinate at the defected structures, obtaining the Eu3+-complex-anchored, unzipped, multiwalled carbon nanotubes (E-uMWCNTs). The controllable surface-breaking retains the MWCNTs' original, excellent mechanical properties. Then, to obtain the macromorphic structure with infinitely long fibers, a wet-spinning process was applied via the binding of a small quantity of polyvinyl alcohol (PVA). Thus, the wet-spun fibers with high contents of E-uMWCNTs (E-uMWCNT-Fs) were produced, in which the E-uMWCNTs took 33.3 wt%, a high ratio in E-uMWCNT-Fs. On the other hand, due to the reinforcing effect of E-uMWCNTs, the highest tensile strength can reach 228.2 MPa for E-uMWCNT-Fs. Meanwhile, the E-uMWCNT-Fs show high-efficiency photoluminescence and excellent media resistance performance due to the embedding effect of PVA on the E-uMWCNTs. Therefore, E-uMWCNT-Fs can exhibit excellent luminescence properties in aqueous solutions at pH 4~12 and in some high-concentration metal-ion solutions. Those distinguished performances promise outstanding innovations of this work.

RESUMO

Background: NDR1/HIN1-like (NHL) family genes are known to be involved in pathogen induced plant responses to biotic stress. Even though the NHL family genes have been identified and characterized in plant defense responses in some plants, the roles of these genes associated with the plant abiotic stress tolerance in wild soybean is not fully established yet, especially in response to alkaline stress. Methods: We identified the potential NHL family genes by using the Hidden Markov model and wild soybean genome. The maximum-likelihood phylogenetic tree and conserved motifs were generated by using the MEME online server and MEGA 7.0 software, respectively. Furthermore, the syntenic analysis was generated with Circos-0.69. Then we used the PlantCARE online software to predict and analyze the regulatory cis-acting elements in promoter regions. Hierarchical clustering trees was generated using TM4: MeV4.9 software. Additionally, the expression levels of NHL family genes under alkaline stress, ABA and MEJA treatment were identified by qRT-PCR. Results: In this study, we identified 59 potential NHL family genes in wild soybean. We identified that wild soybean NHL family genes could be mainly classified into five groups as well as exist with conserved motifs. Syntenic analysis of NHL family genes revealed genes location on 18 chromosomes and presence of 65 pairs of duplication genes. Moreover, NHL family genes consisted of a variety of putative hormone-related and abiotic stress responsive elements, where numbers of methyl jasmonate (MeJA) and abscisic acid (ABA) responsive elements were significantly larger than other elements. We confirmed the regulatory roles of NHL family genes in response to alkaline stress, ABA and MEJA treatment. In conclusion, we identified and provided valuable information on the wild soybean NHL family genes, and established a foundation to further explore the potential roles of NHL family genes in crosstalk with MeJA or ABA signal transduction mechanisms under alkaline stress.

RESUMO

Oxygen releasing biomaterials can facilitate the survival of living implants by creating environments with a viable oxygen level. Hydrophobic oxygen generating microparticles (HOGMPs) encapsulated calcium peroxide (CPO) have recently been used in tissue engineering to release physiologically relevant amounts of oxygen for several weeks. However, generating oxygen using CPO is mediated via the generation of toxic levels of hydrogen peroxide (H2 O2 ). The incorporation of antioxidants, such as catalases, can potentially reduce H2 O2 levels. However, the formulation in which catalases can most effectively scavenge H2 O2 within oxygen generating biomaterials has remained unexplored. In this study, three distinct catalase incorporation methods are compared based on their ability to decrease H2 O2 levels. Specifically, catalase is incorporated within HOGMPs, or absorbed onto HOGMPs, or freely laden into the hydrogel entrapping HOGMPs and compared with control without catalase. Supplementation of free catalase in an HOGMP-laden hydrogel significantly decreases H2 O2 levels reflecting a higher cellular viability and metabolic activity of all the groups. An HOGMP/catalase-laden hydrogel precursor solution containing cells is used as an oxygenating bioink allowing improved viability of printed constructs under severe hypoxic conditions. The combination of HOGMPs with a catalase-laden hydrogel has the potential to decrease peroxide toxicity of oxygen generating tissues.

RESUMO

Recapitulating inherent heterogeneity and complex microarchitectures within confined print volumes for developing implantable constructs that could maintain their structure in vivo has remained challenging. Here, we present a combinational multimaterial and embedded bioprinting approach to fabricate complex tissue constructs that can be implanted postprinting and retain their three-dimensional (3D) shape in vivo. The microfluidics-based single nozzle printhead with computer-controlled pneumatic pressure valves enables laminar flow-based voxelation of up to seven individual bioinks with rapid switching between various bioinks that can solve alignment issues generated during switching multiple nozzles. To improve the spatial organization of various bioinks, printing fidelity with the z-direction, and printing speed, self-healing and biodegradable colloidal gels as support baths are introduced to build complex geometries. Furthermore, the colloidal gels provide suitable microenvironments like native extracellular matrices (ECMs) for achieving cell growths and fast host cell invasion via interconnected microporous networks in vitro and in vivo. Multicompartment microfibers (i.e., solid, core-shell, or donut shape), composed of two different bioink fractions with various lengths or their intravolume space filled by two, four, and six bioink fractions, are successfully printed in the ECM-like support bath. We also print various acellular complex geometries such as pyramids, spirals, and perfusable branched/linear vessels. Successful fabrication of vascularized liver and skeletal muscle tissue constructs show albumin secretion and bundled muscle mimic fibers, respectively. The interconnected microporous networks of colloidal gels result in maintaining printed complex geometries while enabling rapid cell infiltration, in vivo.

RESUMO

A 3D printed flexible tactile sensor with graphene-polydimethylsiloxane (PDMS) microspheres for microstructure perception is presented. The structure of the tactile sensor is inspired by the texture of the human finger and is designed to enable the detection of various levels of surface roughness via the processing of tactile signals. The tactile sensor with a unique graphene-PDMS microsphere structure shows excellent comprehensive mechanical properties, including a robust stretching ability (elongation at break of the sensing layer is 70%), excellent sensing ability (short response time of 60 ms), high sensitivity (sensitivity up to 2.4 kPa-1), and cycle stability (over 2000 loading cycles). In addition, such versatility and sensitivity allow the electronic skin not only to accurately monitor pressure but also to distinguish various surface topographies with microscale differences, and to detect the action of an air fluid.

RESUMO

In the milestone of straggling to make water hydraulics more advantageous, the choice of coating polymer for water hydraulics valves plays an essential role in alleviating the impact of cavitation erosion and corrosion, and this is a critical task for designers. Fulfilling the appropriate selection, we conflicted properties that are vital for erosion and corrosion inhibitors, as well as the tribology in the sense of coefficient of friction. This article aimed to choose the best alternative polymer for coating on the selected substrate, that is, Cr2O3, Al2O3, Ti2O3. By applying PROMETHEE (Preference Ranking Organization Method for Enrichment Evaluations), the best polymer obtained with an analyzed performance attribute is Polytetrafluoroethylene (PTFE) that comes up with higher outranking (0.5932052). A Molecular Dynamics (MD) simulation was conducted to identify the stronger bonding with the regards of the better cleave plane between Polytetrafluoroethylene (PTFE) and the selected substrate. Polytetrafluoroethylene (PTFE)/Al2O3 cleaved in (010) plane was observed to be the strongest bond in terms of binding energy (3188 kJ/mol) suitable for further studies.

RESUMO

Animals and plants have numerous active protections for adapting to the complex and severe living environments, providing endless inspiration for extending the service life of materials and machines. Conch, a marine animal living near the coast and chronically suffering from the erosion of sand in water, has adapted to the condition through its anti-erosion conch shell. Romanesco broccoli, a plant whose inflorescence is self-similar in character, has a natural fractal bud's form. Coupling the convex domes on the conch shell and the fractal structure of Romanesco broccoli, a novel valve core structure of a water hydraulic valve was designed in this paper to improve the particle erosion resistance and valve core's service life. Three models were built to compare the effect among the normal structure, bionic structure, and multi-source coupling bionic structures, and were coined using 3D printing technology. A 3D printed water hydraulic valve was manufactured to simulate the working condition of a valve core under sand erosion in water flow, and capture the experimental videos of the two-phase flow. Furthermore, based on the water hydraulic platform and one-camera-six-mirror 3D imaging subsystem, the experiment system was established and used to compare the performance of the three different valve cores. As a result, the results showed that the coupling bionic structure could effectively improve the anti-erosion property of the valve core and protect the sealing face on the valve core from wear. This paper presents a novel way of combining advantages from both animal (function bionic) and plant (shape bionic) in one component design.

RESUMO

We discuss the application of the Agapito Curtarolo and Buongiorno Nardelli (ACBN0) pseudo-hybrid Hubbard density functional to several transition metal oxides. For simple binary metal oxides, ACBN0 is found to be a fast, reasonably accurate and parameter-free alternative to traditional DFT + U and hybrid exact exchange methods. In ACBN0, the Hubbard energy of DFT + U is calculated via the direct evaluation of the local Coulomb and exchange integrals in which the screening of the bare Coulomb potential is accounted for by a renormalization of the density matrix. We demonstrate the success of the ACBN0 approach for the electronic properties of a series technologically relevant mono-oxides (MnO, CoO, NiO, FeO, both at equilibrium and under pressure). We also present results on two mixed valence compounds, Co3O4 and Mn3O4. Our results for these binary oxides and all the materials we have investigated, obtained at the computational cost of a standard LDA/PBE calculation, are in excellent agreement with hybrid functionals, the GW approximation and experimental measurements.
