RESUMO

Superficial scald is a postharvest physiological disorder that occurs in pear during and after cold storage. In this study, the superficial scald index; α-farnesene and its oxidation products, conjugated trienols (CTols); phenolic content; and the expression of its related genes were investigated in two different pear cultivars, 'Wujiuxiang' (Pyrus communis L.) and 'Yali' (Pyrus bretschneideri R.), following 115 days of cold storage at 0 °C followed by 7 days of shelf life at 20 °C. The results indicated that the superficial scald occurred after 115 days of cold storage and became more severe during the shelf life of the 'Wujiuxiang' pear, whereas no scald was observed in 'Yali'. The α-farnesene levels increased rapidly at first and then decreased, while the CTols contents increased significantly in 'Wujiuxiang' as compared to 'Yali', and the expression levels of the genes involved in α-farnesene and CTols metabolism (HMGR1, HMGR2, GSTU7, GPX5, and GPX6), as well as the phenolic synthesis (PAL1, PAL2, C4H1, 4CL2, C3H, and ANR) of the peel, were significantly up-regulated at the onset of the superficial scald. In addition, the relative conductivity and contents of catechin and epicatechin were higher, and the expression level of the laccase gene (LAC7) significantly increased with the development of superficial scald, while lower contents of chlorogenic acid, arbutin, and isorhamnetin-3-3-glucoside, as well as the lower expression levels of a phenolic-synthesis-related gene (C4H3) and polyphenol oxidase genes (PPO1 and PPO5), were noticed in 'Wujiuxiang' as compared to 'Yali'. The results indicated that the onset and progression of superficial scald were associated with the accumulation of CTols, cell membrane breakdown, and higher catechin, epicatechin, and rutin contents, as well as the expression of associated genes of the peels of pear fruit.

Low concentration acetone gas detection is significantly important for diabetes diagnosis as 1.8⁻10 ppm of acetone exists in exhaled breath from diabetes patients. A new interlocking p+n field-effect transistor (FET) circuit has been proposed for Mn-doped ZnO nanoparticles (MZO) to detect the acetone gas at low concentration, especially close to 1.8 ppm. It is noteworthy that MZO in this interlocking amplification circuit shows a low voltage signal of <0.3 V to the acetone <2 ppm while it displays a transilient response with voltage signal >4.0 V to >2 ppm acetone. In other words, the response to acetone from 1 ppm to 2 ppm increases by ~1233%, which is competent to separate diabetic patients from healthy people. Moreover, the response to 2 ppm acetone is hardly influenced by high relative humidity of 85%. In the meanwhile, MZO in this interlocking circuit possesses a high acetone selectivity compared to formaldehyde, acetaldehyde, toluene and ethanol, suggesting a promising technology for the widespread qualitative screening of diabetes. Importantly, this interlocking circuit is also applicable to other types of metal oxide semiconductor gas sensors. The resistance jump of p- and n-FETs induced by the change of their gate voltages is deemed to make this interlocking circuit produce the transilient response.

Pure In2O3 nanoparticles are prepared by a facile precipitation method and are further modified by Ag. The synthesized samples are characterized by scanning electron microscopy, transmission electron microscopy, energy dispersive X-ray spectroscopy, X-ray diffraction, Raman and UV-Vis spectra. The results show the successful heterojunction formation between Ag and In2O3. Gas sensing property measurements show that the 5 mol % Ag-modified In2O3 sensor has the response of 67 to 50 ppm ethanol, and fast response and recovery time of 22.3 and 11.7 s. The response is over one magnitude higher than that of pure In2O3, which can be attributed to the enhanced catalytic activity of Ag-modified In2O3 as compared with the pure one. The mechanism of the gas sensor can be explained by the spillover effect of Ag, which enhances the oxygen adsorption onto the surface of In2O3 and thus give rise to the higher activity and larger surface barrier height.

This study investigated the soil physicochemical properties and vegetation community characteristics of the Baotou light rare earth tailings pond after 10 years of aggregate spray seeding ecological restoration (S1) and ordinary soil spray seeding ecological restoration (S2), and the naturally restored dam slope area without human intervention (S3). The results showed that the vegetation community of S1 was dominated by Caragana korshinskii Kom, and its importance and abundance values were 0.40 and 38.4, respectively, while the vegetation communities of S2 and S3 mainly comprised herbaceous plants. Additionally, the vegetation biomass of S1 was significantly higher than that of S2 and S3 by 215.20% and 1345.76%, respectively, and the vegetation diversity index of S1 was the highest among the three treatment groups. The soil porosity (SP), water content (W), electrical conductivity (EC), and available K were significantly improved in S1, while soil bulk density (BD) was significantly reduced compared with that of S2 and S3. In addition, redundancy analysis revealed that SP, EC, W, and K positively correlate with the biomass, Shannon, Pielou, Simpson, and Marglef indices. Principal component analysis further showed that the comprehensive score of S1 (0.983) was higher than that of S2 (- 0.261) and S3 (- 0.648). Collectively, these findings indicate that appropriate ecological restoration can improve soil structure and vegetation community characteristics, thereby accelerating vegetation restoration, ultimately increasing the stability of the ecosystem.

As a traditional folk medicine, pear paste has important nutritional and health effects. The physicochemical properties and antioxidant activities of pear pastes prepared from 23 different cultivars were investigated, including color parameters ( L*, a*, b* and h°), transmittance, pH, titratable acidity (TA), soluble sugar content, total phenolics content (TPC), total flavonoids content (TFC), DPPH and •OH radical scavenging activity (RSA), and ferric reducing antioxidant power (FRAP). It was demonstrated that the physicochemical properties and antioxidant activities of pear pastes from various cultivars differed significantly. Pear cultivars of "Mantianhong", "Xiangshui" and "Anli" possessing higher TPC and TFC exhibited excellent antioxidant activity determined by DPPH RSA, •OH RSA and FRAP, while the lowest TPC and TFC was observed for the cultivars of "Xueqing", "Nansui", "Hongxiangsu", and "Xinli No. 7", which also demonstrated the poor antioxidant activity. Multivariate analyses, including factor and cluster analysis, were used for the quality evaluation and separation of pear pastes based on their physicochemical and antioxidant properties. Factor analysis reduced the above thirteen parameters to final four effective ones, i.e. DPPH RSA, color b*, FRAP and TA, and subsequently these four parameters were used to construct the comprehensive evaluation prediction model for evaluating the quality of pear pastes. The pear pastes could be separated into three clusters and differentiated for the diverse of pear cultivars via cluster analysis. Consistently, "Mantianhong", "Xiangshui" and "Anli" pear with higher quality clustered into one group, in contrast, "Xueqing", "Nansui", "Hongxiangsu", and "Xinli No. 7" with lower quality clustered into the other group. It provided a theoretical method to evaluate the quality of pear paste and may help the fruit processing industry select the more suitable pear cultivars for pear paste making.

Rabies, caused by rabies virus (RABV), is an ancient zoonotic disease that severely threatens the public health throughout the world. Previous study indicated that interleukin-1ß (IL-1ß) plays an important role in RABV infection. However, the mechanism how IL-1ß affects RABV pathogenicity is still unknown yet. In this study, we confirmed that IL-1ß was able to reduce viral titers of RABV in different cells, and the recombinant RABV expressing IL-1ß, designated as rCVS-IL1ß, could be suppressed in different cells due to the expression of IL-1ß. Furthermore, the survival rates of mice infected with rCVS-IL1ß by intramuscular route was significantly higher than those of mice infected with parent virus rCVS, which is associated with the less viral loads for entry into the central nervous system (CNS). We further characterized that the cGAS-STING pathway was activated in rCVS-IL1ß infected bone marrow derived dendritic cells (BMDC), which could contribute to the decreased viral loads of RABV after intramuscular infection. Moreover, we also observed that the expression of IL-1ß by rCVS-IL1ß could compromise the blood-brain barrier (BBB) integrity by degrading the tight junction proteins, which allowing peripheral inflammatory cytokines, chemokines, and CD4+T cells to enter into the brain for the clearance of RABV in the CNS. Together, our study suggests that IL-1ß could attenuate RABV pathogenicity through activating cGAS-STING pathway in to decrease the viral entry into the CNS and enhance the BBB permeability to promote RABV clearance in the CNS as well, which provides new insight into developing effective therapeutics for rabies.

Artificial reefs have been widely deployed in the northern Yellow Sea. However, the differences in the ecological benefits on different types of artificial reef habitats are still poorly understood. In this study, the temporal and spatial differences on benthic fish communities were evaluated among concrete artificial reef habitat (CAR), rocky artificial reef habitat (RAR), ship artificial reef habitat (SAR) around Xiaoshi Island in northern Yellow Sea. The results indicated that all three types of artificial reef habitats can enhance the diversity variables of benthic fish communities, and fish abundance, species richness and Shannon-Wiener index of CAR were generally better than the other two. CAR and RAR hosted similar community composition of benthic fish, and all types of habitats showed significant differences in community composition between winter-spring and summer-autumn. Environmental factors, especially water temperature, can also affect the community composition by affecting the migration of temperature-preferred species. Overall, the enhancement effects of artificial habitats on fisheries productivity varied with fish species and reef types. This study will help to understand the ecological effects of different types of artificial reefs in northern Yellow Sea, and then could give an insight for scientific construction of artificial reefs in this region.

Flavonoids play important roles in regulating plant growth and development. In this study, three kaempferol 3-O-glycosides were identified and mainly accumulated in flowers but not in leaves or fruits of Malus. In Malus, flower petal color is normally white, but some genotypes have red flowers containing anthocyanin. Anthocyanin biosynthesis appears to be in competition with kaempferol 3-O-glycosides production and controlled by the biosynthetic genes. The white flower Malus genotypes had better-developed seeds than the red flower genotypes. In flowers, the overexpression of MYB10 in Malus domestica enhanced the accumulation of anthocyanin, but decreased that of kaempferol 3-O-glycosides. After pollination the transgenic plants showed slower pollen tube growth and fewer developed seeds. Exogenous application of different flavonoid compounds suggested that kaempferol 3-O-glycosides, especially kaempferol 3-O-rhamnoside, regulated pollen tube growth and seed set rather than cyanidin or quercetin 3-O-glycosides. It was found that kaempferol 3-O-rhamnoside might regulate pollen tube growth through effects on auxin, the Rho of plants (ROP) GTPases, calcium and the phosphoinositides signaling pathway. With the inhibition of auxin transport, the transcription levels of Heat Shock Proteins (HSPs) and ROP GTPases were downregulated while the levels were not changed or even enhanced when blocking calcium signaling, suggesting that HSPs and ROP GTPases were downstream of auxin signaling, but upstream of calcium signaling. In summary, kaempferol glycoside concentrations in pistils correlated with auxin transport, the transcription of HSPs and ROP GTPases, and calcium signaling in pollen tubes, culminating in changes to pollen tube growth and seed set.

Li2Si2O5 are generally obtained in form of granules with unavoidable impurities including Li2SiO3 and SiO2. Here, we demonstrated a facile hydrothermal route to synthesize novel 3D hierarchical Li2Si2O5 hydrate hollow flower-like microstructures assembled by rod subunits with high purity. The crystal growth was accomplished by complete transformation from poorly crystallized metastable phases formed in the initial stage including Li2SiO3, SiO2 and various Li2Si2O5 hydrate species to Li2Si2O5 hydrate rods. The transformation over many times gave a sustainable high chemical potential to direct the anisotropic growth of Li2Si2O5 hydrate rods with large aspect ratios. Besides, the variation of Li/Si molar ratios confirmed that Li2Si2O5 hydrate rods were obtained only at Li/Si = 1. The perfection and aspect ratio of the rods could be controlled very well by adjusting the hydrothermal temperatures and precursor concentrations. Some new points about obtaining pure phase and anisotropic morphology were discussed, including careful selection of precursors and synthetic method. The obtained novel 3D Li2Si2O5 hydrate structures exhibited a characteristic of mesoporous material and had an excellent adsorption capability of methylene blue with high adsorption amount of 49.42 mg·g-1 and color removal of 98.85%, indicating the potential use in wastewater treatment.

In recent years, increasing research has evaluated the use of natural products as antimicrobial food additives. In this study, antibacterial activity was evaluated for six dihydrochalcone compounds from Malus. Phloretin and 3-hydroxyphloretin exhibited antibacterial effects on both Gram-positive and Gram-negative bacteria, and the antibacterial capacity of these compounds was greater than that of their glycosylated derivatives. Within a certain range, dihydrochalcone hydrophobicity was positively correlated with antibacterial activity. Additionally, glycosylation at the 2'-position of the A-ring and hydroxyl group at the 3-position of the B-ring played a key role in the antibacterial activity of dihydrochalcones. Phloretin and 3-hydroxyphloretin caused damage to bacterial cells by significantly increasing protein and inorganic phosphate leakage. Compared to phloretin, 3-hydroxyphloretin exhibited a smaller effect on Gram-positive Micrococcus luteus and a greater effect on Gram-negative Klebsiella pneumoniae, suggesting different antibacterial mechanisms. At a low dihydrochalcone concentration, the respiration of M. luteus did not change, while membrane permeability increased significantly. These results indicate that the antibacterial mechanism of M. luteus was primarily damage to the cell membrane. However, damage to respiration and the cell membrane might occur simultaneously in K. pneumoniae, suggesting that the antibacterial mechanism of dihydrochalcones also depends on strain type. This study demonstrated the broad-spectrum antibacterial properties of dihydrochalcone compounds commonly found in the genus Malus to foodborne pathogens and elucidated the antibacterial mechanisms. It provides theoretical guidance for future research and application of dihydrochalcones in the food industry.

Nitrate pollution in rivers, lakes, shallow groundwater, and even deep groundwater occurs in many parts of the world. And, it's essential to assessing the relationship between nitrate pollution and human health, which is called human health risk assessment (HHRA). In this paper, groundwater samples were collected for their nitrate content in a typical karst hydrogeological unit in East China during the wet and dry seasons. Then, a human health risk assessment was conducted using the four-step risk assessment process developed by the US Environmental Protection Agency (USEPA), which aimed to determine the potential risk posed to human health by nitrate in the groundwater. To make the assessment more authentic and objective, the drinking water and dermal contact exposure pathways were considered, and the people were divided into four groups, including infants (0~6 months), children (7 months~17 years old), females (18 years and older), and males (18 years and older), in the wet and dry seasons to determine the impacts of the exposure pathway, age, sex, and precipitation period. The results indicated that more than half of the groundwater samples exceeded 10 mg/L (measured as nitrogen), which is the drinking water standard of China. The children and infants had greater health risks than the adults at the same groundwater nitrate concentration, and those two groups need to be paid more attention; the adult females had a greater health risk than the adult males in the two precipitation periods, which shows that the order of the health risk was infants ˃ children ˃ adult females ˃ adult males. In addition, the value of the hazard quotient (HQ) and the area of the adverse effects were both higher in the wet season than in the dry season, which explains that precipitation can affect the human health risk as well. The HQ caused by the drinking water exposure pathway was much higher than that caused by the dermal contact exposure pathway. This study can provide information for more effective and reasonable decisions to city managers for groundwater nitrate pollution prevention.

The antioxidant capacity (AC) of six dihydrochalcone compounds was evaluated using DPPH and ABTS assays. In water-based solution 3-hydroxyphlorizin exhibited the highest AC among all dihydrochalcones. In acetone and acidic solutions (pH = 2.5 or 2.0), presence of an o-dihydroxyl at the B-ring increased AC, whereas glycosylation at the A-ring decreased AC of dihydrochalcones. By comparing the AC of dihydrochalcones with similar structures, it was found that the o-dihydroxyl at the B-ring and 2'-hydroxyl group at the A-ring were critical for maintaining the AC of dihydrochalcones by promoting hydrogen atom transfer or single electron transfer mechanism. Sequential proton-loss electron transfer commonly occurred during free radical scavenging in water-based solution. Moreover, we report a unique phenomenon in which glycosylation at the 2'-position enhanced the dissociation ability of the 4'-hydroxyl group and increased the AC of dihydrochalcones containing o-dihydroxyl. We speculate that this increase in AC might occur through intramolecular electron transfer.

Although it is well known that human bone tissues have obvious orthotropic material properties, most works in the physical modeling field adopted oversimplified isotropic or approximated transversely isotropic elasticity due to the simplicity. This paper presents a convenient methodology based on harmonic fields, to construct volumetric finite element mesh integrated with complete orthotropic material. The basic idea is taking advantage of the fact that the longitudinal axis direction indicated by the shape configuration of most bone tissues is compatible with the trajectory of the maximum material stiffness. First, surface harmonic fields of the longitudinal axis direction for individual bone models were generated, whose scalar distribution pattern tends to conform very well to the object shape. The scalar iso-contours were extracted and sampled adaptively to construct volumetric meshes of high quality. Following, the surface harmonic fields were expanded over the whole volumetric domain to create longitudinal and radial volumetric harmonic fields, from which the gradient vector fields were calculated and employed as the orthotropic principal axes vector fields. Contrastive finite element analyses demonstrated that elastic orthotropy has significant effect on simulating stresses and strains, including the value as well as distribution pattern, which underlines the relevance of our orthotropic modeling scheme.
