RESUMO

Variations in cardioafferent traffic are relayed to the brain via arterial baroreceptors and have been shown to modulate perceptual processing. However, less is known about the cognitive-behavioral consequences of these effects and their role during stress. Here, we investigated in how far automatic responses during the Simon task were modulated by exposure to a laboratory stressor and the different phases of the cardiac cycle. In this study, 30 participants performed three blocks of a combined horizontal and vertical Simon task, which is characterized by either sensorimotor or cognitive response conflicts, respectively. Before each block, subjects were exposed to both the cold pressor test (CPT) and a control condition according to a within-subjects design. Target stimuli were presented during either systole or diastole. Behavioral and EEG-correlates of task processing were assessed along with subjective, cardiovascular, and endocrine measures of stress. The stress induction was successful yielding significant increases in all these measures compared to control. Moreover, we found the expected Simon effects: in incompatible compared to compatible trials performance was decreased and LRP latency as well as anterior N2 area increased. Importantly, accuracy was improved in compatible but reduced in incompatible trials during systole vs. diastole but only in the horizontal Simon condition. Stress dampened N2 area, however, no interactions with cardiac cycle were evident. These results indicate a faciliatory effect of cardioafferent traffic on automated sensorimotor processes.

EEG microstates represent functional brain networks observable in resting EEG recordings that remain stable for 40-120ms before rapidly switching into another network. It is assumed that microstate characteristics (i.e., durations, occurrences, percentage coverage, and transitions) may serve as neural markers of mental and neurological disorders and psychosocial traits. However, robust data on their retest-reliability are needed to provide the basis for this assumption. Furthermore, researchers currently use different methodological approaches that need to be compared regarding their consistency and suitability to produce reliable results. Based on an extensive dataset largely representative of western societies (2 days with two resting EEG measures each; day one: n = 583; day two: n = 542) we found good to excellent short-term retest-reliability of microstate durations, occurrences, and coverages (average ICCs = 0.874-0.920). There was good overall long-term retest-reliability of these microstate characteristics (average ICCs = 0.671-0.852), even when the interval between measures was longer than half a year, supporting the longstanding notion that microstate durations, occurrences, and coverages represent stable neural traits. Findings were robust across different EEG systems (64 vs. 30 electrodes), recording lengths (3 vs. 2 min), and cognitive states (before vs. after experiment). However, we found poor retest-reliability of transitions. There was good to excellent consistency of microstate characteristics across clustering procedures (except for transitions), and both procedures produced reliable results. Grand-mean fitting yielded more reliable results compared to individual fitting. Overall, these findings provide robust evidence for the reliability of the microstate approach.

EEG microstates are brief, recurring periods of stable brain activity that reflect the activation of large-scale neural networks. The temporal characteristics of these microstates, including their average duration, number of occurrences, and percentage contribution have been shown to serve as biomarkers of mental and neurological disorders. However, little is known about how microstate characteristics of prototypical network types relate to each other. Normative intercorrelations among these parameters are necessary to help researchers better understand the functions and interactions of underlying networks, interpret and relate results, and generate new hypotheses. Here, we present a systematic analysis of intercorrelations between EEG microstate characteristics in a large sample representative of western working populations (n = 583). Notably, we find that microstate duration is a general characteristic that varies across microstate types. Further, microstate A and B show mutual reinforcement, indicating a relationship between auditory and visual sensory processing at rest. Microstate C appears to play a special role, as it is associated with longer durations of all other microstate types and increased global field power, suggesting a relationship of these parameters with the anterior default mode network. All findings could be confirmed using independent EEG recordings from a retest-session (n = 542).

Early life events can have long-lasting effects that may impact the quality of life into adulthood. The link between childhood adversities and adult mental and physical health is well documented, however, the underlying mechanisms remain poorly understood. Executive functions are assumed to be a key factor in successfully dealing with cognitive-emotional challenges thereby contributing to stress resilience and mental health across the lifespan. Here, we examined whether cognitive control moderates the link between early life adversity and depression. Data was available from a sample of 424 participants aged 20-70 years (Clinicaltrials.gov: NCT05155397). They performed in the Stroop task and behavior as well as frontal theta power were recorded. Negative childhood experiences were assessed with the Childhood Trauma Questionnaire (CTQ), chronic stress was measured with the Trier Inventory for Chronic Stress (TICS) and depression symptoms with Beck's Depression Inventory (BDI). The CTQ predicted symptoms of chronic stress and depression. Regression models pointed to the TICS as a crucial mediator in the relationship between CTQ and BDI. However, parameters of cognitive control demonstrated a rather weak effect as moderators. These results indicate that chronic stress is an important mediator linking childhood trauma to depression but suggest only a limited role for cognitive control.

An increased prevalence of mixed-handedness has been reported in several neurodevelopmental and psychiatric disorders. Unfortunately, there is high between-study variability in the definition of mixed-handedness, leading to a major methodological problem in clinical laterality research and endangering replicability and comparability of research findings. Adding to this challenge is the fact that sometimes researchers use the concepts of mixed-handedness and ambidexterity interchangeably. Therefore, having a consensus on how to determine mixed-handedness and how to distinguish it from ambidexterity is crucial for clinical laterality research. To this end, hand preference and hand performance data from more than 600 participants from the Dortmund Vital Study (Trial registration: ClinicalTrials.gov NCT05155397), a population-based study in Germany, was analyzed to ascertain an optimal classification to determine mixed-handedness and ambidexterity. Using a combination of latent class analyses, effect size determination, and comparisons with the existing literature, we establish that an LQ cut-off criterion of +/-60 for mixed-handedness is optimal for future clinical laterality studies. Moreover, we show that mixed-handedness and ambidexterity are not identical and that the terms should not be used interchangeably. We further highlight the need for a consensus on how to mathematically determine ambidexterity as results of existing categorization schemes largely differ.Trial registration: ClinicalTrials.gov NCT05155397; https://clinicaltrials.gov/ct2/show/NCT05155397.

Theories of attention argue that objects are the units of attentional selection. In real-word environments such objects can contain visual and auditory features. To understand how mechanisms of selective attention operate in multisensory environments, in this pre-registered study, we created an audiovisual cocktail-party situation, in which two speakers (left and right of fixation) simultaneously articulated brief numerals. In three separate blocks, informative auditory speech was presented (a) alone or paired with (b) congruent or (c) uninformative visual speech. In all blocks, subjects localized a pre-defined numeral. While audiovisual-congruent and uninformative speech improved response times and speed of information uptake according to diffusion modeling, an ERP analysis revealed that this did not coincide with enhanced attentional engagement. Yet, consistent with object-based attentional selection, the deployment of auditory spatial attention (N2ac) was accompanied by visuo-spatial attentional orienting (N2pc) irrespective of the informational content of visual speech. Notably, an N2pc component was absent in the auditory-only condition, demonstrating that a sound-induced shift of visuo-spatial attention relies on the availability of audio-visual features evolving coherently in time. Additional exploratory analyses revealed cross-modal interactions in working memory and modulations of cognitive control.

Whereas neural representations of spatial information are commonly studied in vision, olfactory stimuli might also be able to create such representations via the trigeminal system. We explored in two independent multi-method electroencephalography-functional near-infrared spectroscopy (EEG+fNIRS) experiments (n1=18, n2=14) if monorhinal odor stimuli can evoke spatial representations in the brain. We tested whether this representation depends on trigeminal properties of the stimulus, and if the retention in short-term memory follows the "sensorimotor recruitment theory", using multivariate representational similarity analysis (RSA). We demonstrate that the delta frequency band up to 5 Hz across the scull entail spatial information of which nostril has been stimulated. Delta frequencies were localized in a network involving primary and secondary olfactory, motor-sensory and occipital regions. RSA on fNIRS data showed that monorhinal stimulations evoke neuronal representations in motor-sensory regions and that this representation is kept stable beyond the time of perception. These effects were no longer valid when the odor stimulus did not sufficiently stimulate the trigeminal nerve as well. Our results are first evidence that the trigeminal system can create spatial representations of bimodal odors in the brain and that these representations follow similar principles as the other sensory systems.

INTRODUCTION: Excessive stress is increasingly recognized as an important trigger of many diseases prevalent in modern societies, and monitoring such stress-related effects could aid prevention. The measurement of salivary markers of inflammation is emerging as a promising tool to non-invasively quantify stress' effects on immune processes in everyday life and thereby detect early aberrations before the manifestation of serious health problems. However, more laboratory-controlled research is needed in order to establish the timescale and determinants of salivary cytokine responses to acute stress. METHODS: We repeatedly exposed participants to Cold Pressor Stress Test (CPT) or a control procedure and measured a wide array of salivary cytokines as well as subjective, cardiovascular, and cortisol stress reactions. CPT exposure was repeated every 15 min, 3 times in total, with a duration of 3 min each. Saliva was sampled immediately after the first two exposures as well as in 15-min intervals until 60 min after the onset of the first intervention. RESULTS: We found that many cytokines were detectable in saliva. Specific stress effects were limited to IL-8 and IL-6, however, which decreased immediately or 15 min after stress onset, respectively. Moreover, IL-8 was negatively correlated to cortisol output in the stress but not in the control group. Significant increases were also observed in salivary TNFα and IFNγ; however, these effects were similar under both stress and control conditions. DISCUSSION: Our results show that particular salivary cytokines may be sensitive to immediate effects of acute CPT-induced stress and also highlight the importance of employing control procedures to discern stress effects from unrelated variations in salivary cytokines.

OBJECTIVE: Demographic changes encompass societies to maintain the work ability (WA) of aging workforces. The present study explored the relationship between modifiable lifestyle factors, cognitive functions, and their influence on WA, using a multi-group structural equation approach. METHOD: Cross-sectional data from 247 middle-aged and 236 older employees from the Dortmund Vital Study were included in this analysis. We proposed a model with three exogenous variables (Physical Fitness, Cognitive Functions, and Social Life), and with WA as the endogenous variable. WA was measured with the Work Ability Index (WAI), which considers job demands and individual physical and mental resources. Multi-group analyses were based on the principles of invariance testing and conducted using robust estimation methods. RESULTS: Results revealed that Social Life outside work had significant positive effects on WA in both, middle-aged and older adults. Physical Fitness had a significant effect on WA only in middle-aged adult, and Cognitive Functions had no significant influence on WA in either group. In older adults, Physical Fitness correlated with Cognitive Functions, whereas in middle-aged adults, Cognitive Functions marginally correlated with Social Life. CONCLUSIONS: Our results underline the importance of an active social life outside the workplace for WA, regardless of the employees' age. The influence of Physical Fitness on WA changes with increasing age, indicating the necessity to have a differentiated view of age effects and interacting influencing factors. ABSTRACTStress exposure and reactivity may show differential associations with handedness, but shallow phenotyping may influence the current knowledge. Importantly, different handedness measures do not necessarily show high correlations with each other and should not be used interchangeably as they may reflect different dimensions of laterality. Here, data on handedness from 599 participants in the population-based, longitudinal Dortmund Vital Study was used to determine various asymmetry indices. Hand preference was assessed with the Edinburgh Handedness Inventory (EHI) and the lateral preference inventory (LPI) measuring handedness, footedness, earedness, and eyedness. Hand performance was determined using the pegboard test. In addition, data on several dimensions of stress exposure and reactivity, including hair cortisol, and mental well-being was analysed to determine associations with handedness. All handedness measures correlated significantly with each other, with the strongest correlation between the EHI and the LPI handedness score. The EHI and LPI hand measures resulted in the highest effect sizes and most consistent correlations with stress or mental well-being. In contrast, the pegboard test only showed very little association with the stress and mental well-being measures. This highlights the importance of handedness phenotyping. Including preference measures is recommended to disentangle the link between handedness and mental health.

OBJECTIVE: We demonstrate and discuss the use of mobile electroencephalogram (EEG) for neuroergonomics. Both technical state of the art as well as measures and cognitive concepts are systematically addressed. BACKGROUND: Modern work is increasingly characterized by information processing. Therefore, the examination of mental states, mental load, or cognitive processing during work is becoming increasingly important for ergonomics. RESULTS: Mobile EEG allows to measure mental states and processes under real live conditions. It can be used for various research questions in cognitive neuroergonomics. Besides measures in the frequency domain that have a long tradition in the investigation of mental fatigue, task load, and task engagement, new approaches-like blink-evoked potentials-render event-related analyses of the EEG possible also during unrestricted behavior. CONCLUSION: Mobile EEG has become a valuable tool for evaluating mental states and mental processes on a highly objective level during work. The main advantage of this technique is that working environments don't have to be changed while systematically measuring brain functions at work. Moreover, the workflow is unaffected by such neuroergonomic approaches.

Immunological aging type definition requires establishing reference intervals from the distribution of immunosenescence biomarkers conditional on age. For 1605 individuals (18-97 years), we determined the comprehensive immune age index IMMAX from flow-cytometry-based blood cell sub-populations and identified age-specific centiles by fitting generalized additive models for location, scale, and shape. The centiles were uncorrelated with age and facilitated the categorization of individuals as immunologically slow or fast aging types. Using its 50th percentile as a reference, we rescaled the IMMAX to equivalent years of life (EYOL) and computed the immunological age gap as the difference between EYOL and chronological age. Applied to preliminary baseline and follow-up measurements from 53 participants of the Dortmund Vital Study (Clinical-Trials.gov Identifier: NCT05155397), the averaged changes in the IMMAX and EYOL conformed to the 5-year follow-up period, whereas no significant changes occurred concerning IMMAX centiles and age gap. This suggested that the participants immunologically adapted to aging and kept their relative positions within the cohort. Sex was non-significant. Methodical comparisons indicated that future confirmatory analyses with the completed follow-up examinations could rely on percentile curves estimated by simple linear quantile regression, while the selection of the immunosenescence biomarker will greatly influence the outcome, with IMMAX representing the preferable choice.

What mechanisms underlie the transfer of a working memory representation into a higher-level code for guiding future actions? Electrophysiological correlates of attentional selection and motor preparation processes within working memory were investigated in two retrospective cuing tasks. In the first experiment, participants stored the orientation and location of a grating. Subsequent feature cues (selective vs. neutral) indicated which feature would be the target for later report. The oscillatory response in the mu and beta frequency range with an estimated source in the sensorimotor cortex contralateral to the responding hand was used as correlate of motor preparation. Mu/beta suppression was stronger following the selective feature cues compared to the neutral cue, demonstrating that purely feature-based selection is sufficient to form a prospective motor plan. In the second experiment, another retrospective cue was included to study whether knowledge of the task at hand is necessary to initiate motor preparation. Following the feature cue, participants were cued to either compare the stored feature(s) to a probe stimulus (recognition task) or to adjust the memory probe to match the target feature (continuous report task). An analogous suppression of mu oscillations was observed following a selective feature cue, even ahead of task specification. Further, a subsequent selective task cue again elicited a mu/beta suppression, which was stronger after a continuous report task cue. This indicates that working memory is able to flexibly store different types of information in higher-level mental codes to provide optimal prerequisites for all required action possibilities.

Attentional models of time perception assume that the perceived duration of a stimulus depends on the extent to which attentional resources are allocated to its temporal information. Here, we studied the effects of auditory distraction on time perception, using a combined attentional-distraction duration-discrimination paradigm. Participants were confronted with a random sequence of long and short tone stimuli, most of which having a uniform (standard) pitch and only a few a different (deviant) pitch. As observed in previous studies, pitch-deviant tones impaired the discrimination of tone duration and triggered a sequence of event-related potentials (ERPs) reflecting a cycle of deviance detection, involuntary attentional distraction and reorientation (MMN, P3a, RON). Contrasting ERPs of short and long tone durations revealed that long tones elicited a more pronounced fronto-central contingent negative variation (CNV) in the time interval after the expected offset of the short tone as well as a more prominent centro-parietal late positive complex (LPC). Relative to standard-pitch tones, deviant-pitch tones especially impaired the correct discrimination of long tones, which was associated with a reduction of the CNV and LPC. These results are interpreted within the theoretical framework of resource-based models of time perception, in which involuntary distraction due to a deviant event led to a withdrawal of attentional resources from the processing of time information.

Age-related differences in the processing of audiovisual speech in a multi-talker environment were investigated analysing event-related spectral perturbations (ERSPs), focusing on theta, alpha and beta oscillations that are assumed to reflect conflict processing, multisensory integration and attentional mechanisms, respectively. Eighteen older and 21 younger healthy adults completed a two-alternative forced-choice word discrimination task, responding to audiovisual speech stimuli. In a cocktail-party scenario with two competing talkers (located at -15° and 15° azimuth), target words (/yes/or/no/) appeared at a pre-defined (attended) position, distractor words at the other position. In two audiovisual conditions, acoustic speech was combined either with informative or uninformative visual speech. While a behavioural benefit for informative visual speech occurred for both age groups, differences between audiovisual conditions in the theta and beta band were only present for older adults. A stronger increase in theta perturbations for stimuli containing uninformative visual speech could be associated with early conflict processing, while a stronger suppression in beta perturbations for informative visual speech could be associated to audiovisual integration. Compared to the younger group, the older group showed generally stronger beta perturbations. No condition differences in the alpha band were found. Overall, the findings suggest age-related differences in audiovisual speech integration in a multi-talker environment. While the behavioural benefit of informative visual speech was unaffected by age, older adults had a stronger need for cognitive control when processing conflicting audiovisual speech input. Furthermore, mechanisms of audiovisual integration are differently activated depending on the informational content of the visual information.

Stress is assumed to inhibit the top-down control of attention and to facilitate bottom-up processing. Evidence from human experiments, however, remains scarce. Previous studies have addressed how stress affects the interplay of bottom-up and top-down mechanisms of attention. A key open question is in how far such effects can actually be attributed to a stress-induced modulation of top-down attention control. We sought to isolate top-down from bottom-up effects by assessing stress effects on anticipatory changes in alpha oscillations that precede stimulus processing. Participants performed in a cued target detection task in which a cue prompted them to covertly shift their attention to left or right screen positions, 20 min after being exposed to the bilateral feet cold pressor test or a warm water control procedure. The stressor led to a substantial increase in cortisol, peaking 20 min post stressor, along with rises in heart rate, blood pressure, and subjective ratings of stress and arousal. As expected, cued attention deployment led to higher alpha power over posterior electrodes contralateral versus ipsilateral to the attended hemifield during the cue-target interval. Importantly, this purely endogenous effect was potentiated by stress, however, significant differences were restricted to the middle of the cue-target interval and thus temporally separated from the appearance of the target. These results indicate that stress does not impair top-down attentional control per se but may introduce a qualitative change modulating the way attention is deployed to meet action goals.

Our perception of time varies with the degree of cognitive engagement in tasks. The perceived passage of time accelerates while working on demanding tasks, whereas time appears to drag during boring situations. Our experiment aimed at investigating whether this relationship is mutual: Can manipulated announcements of elapsed time systematically affect the attentional resources applied to a cognitive task? We measured behavioral performance and the EEG in a whole report working memory paradigm with six items of different colors that each had to be reported after a short delay period. The 32 participants were informed about the current time after each 20 trials, while the clock was running at either 100% (normal), 120% (fast), or 80% (slow) of normal clock speed depending on the experimental block. The mean number of correctly reported colors per trial was significantly increased in the fast as compared to the slow and normal clock conditions. In the EEG, we focused on neural oscillations during working memory encoding and storage. As an electrophysiological correlate of task engagement, frontal theta power during the storage interval was increased in the fast clock condition. Also, the power of frontal theta oscillations predicted the number of correctly reported colors on a single-trial basis. This shows that a covert manipulation of clock speed can lead to an improvement in cognitive performance, presumably mediated by a higher allocation of attentional resources resulting from an adaptation of the subjective passage of time during an experiment.

Adaptively changing between different tasks while in locomotion is a fundamental prerequisite of modern daily life. The cognitive processes underlying dual tasking have been investigated extensively using EEG. Due to technological restrictions, however, this was not possible for dual-task scenarios including locomotion. With new technological opportunities, this became possible and cognitive-motor interference can be studied, even in outside-the-lab environments. In the present study, participants carried out a cognitive-motor interference task as they responded to cued, auditory task-switch stimuli while performing locomotive tasks with increasing complexity (standing, walking, traversing an obstacle course). We observed increased subjective workload ratings as well as decreased behavioural performance for increased movement complexity and cognitive task difficulty. A higher movement load went along with a decrease of parietal P2, N2 and P3 amplitudes and frontal Theta power. A higher cognitive load, on the other hand, was reflected by decreased frontal CNV amplitudes. Additionally, a connectivity analysis using inter-site phase coherence revealed that higher movement as well as cognitive task difficulty had an impairing effect on fronto-parietal connectivity. In conclusion, subjective ratings, behavioural performance and electrophysiological results indicate that less cognitive resources were available to be deployed towards the execution of the cognitive task when in locomotion compared to standing still. Connectivity results also show a scarcity of attentional resources when switching a task during the highest movement complexity condition. Summarized, all findings indicate a central role of attentional control regarding cognitive-motor dual tasking and an inherent limitation of cognitive resources.

Since the addition of fluoride to drinking water in the 1940s, there have been frequent and sometimes heated discussions regarding its benefits and risks. In a recently published review, we addressed the question if current exposure levels in Europe represent a risk to human health. This review was discussed in an editorial asking why we did not calculate benchmark doses (BMD) of fluoride neurotoxicity for humans. Here, we address the question, why it is problematic to calculate BMDs based on the currently available data. Briefly, the conclusions of the available studies are not homogeneous, reporting negative as well as positive results; moreover, the positive studies lack control of confounding factors such as the influence of well-known neurotoxicants. We also discuss the limitations of several further epidemiological studies that did not meet the inclusion criteria of our review. Finally, it is important to not only focus on epidemiological studies. Rather, risk analysis should consider all available data, including epidemiological, animal, as well as in vitro studies. Despite remaining uncertainties, the totality of evidence does not support the notion that fluoride should be considered a human developmental neurotoxicant at current exposure levels in European countries.

Understanding the contribution of cognitive processes and their underlying neurophysiological signals to behavioral phenomena has been a key objective in recent neuroscience research. Using a diffusion model framework, we investigated to what extent well-established correlates of spatial attention in the electroencephalogram contribute to behavioral performance in an auditory free-field sound localization task. Younger and older participants were instructed to indicate the horizontal position of a predefined target among three simultaneously presented distractors. The central question of interest was whether posterior alpha lateralization and amplitudes of the anterior contralateral N2 subcomponent (N2ac) predict sound localization performance (accuracy, mean RT) and/or diffusion model parameters (drift rate, boundary separation, non-decision time). Two age groups were compared to explore whether, in older adults (who struggle with multispeaker environments), the brain-behavior relationship would differ from younger adults. Regression analyses revealed that N2ac amplitudes predicted drift rate and accuracy, whereas alpha lateralization was not related to behavioral or diffusion modeling parameters. This was true irrespective of age. The results indicate that a more efficient attentional filtering and selection of information within an auditory scene, reflected by increased N2ac amplitudes, was associated with a higher speed of information uptake (drift rate) and better localization performance (accuracy), while the underlying response criteria (threshold separation), mean RTs, and non-decisional processes remained unaffected. The lack of a behavioral correlate of poststimulus alpha power lateralization constrasts with the well-established notion that prestimulus alpha power reflects a functionally relevant attentional mechanism. This highlights the importance of distinguishing anticipatory from poststimulus alpha power modulations.
