RESUMO

The natural habitats of microorganisms in the human microbiome, ocean and soil ecosystems are full of colloids and macromolecules. Such environments exhibit non-Newtonian flow properties, drastically affecting the locomotion of microorganisms1-5. Although the low-Reynolds-number hydrodynamics of swimming flagellated bacteria in simple Newtonian fluids has been well developed6-9, our understanding of bacterial motility in complex non-Newtonian fluids is less mature10,11. Even after six decades of research, fundamental questions about the nature and origin of bacterial motility enhancement in polymer solutions are still under debate12-23. Here we show that flagellated bacteria in dilute colloidal suspensions display quantitatively similar motile behaviours to those in dilute polymer solutions, in particular a universal particle-size-dependent motility enhancement up to 80% accompanied by a strong suppression of bacterial wobbling18,24. By virtue of the hard-sphere nature of colloids, whose size and volume fraction we vary across experiments, our results shed light on the long-standing controversy over bacterial motility enhancement in complex fluids and suggest that polymer dynamics may not be essential for capturing the phenomenon12-23. A physical model that incorporates the colloidal nature of complex fluids quantitatively explains bacterial wobbling dynamics and mobility enhancement in both colloidal and polymeric fluids. Our findings contribute to the understanding of motile behaviours of bacteria in complex fluids, which are relevant for a wide range of microbiological processes25 and for engineering bacterial swimming in complex environments26,27.

RESUMO

Metamaterials are artificial materials that can achieve unusual properties through unique structures. In particular, their "invisibility" property has attracted enormous attention due to its little or negligible disturbance to the background field that avoids detection. This invisibility feature is not only useful for the optical field, but it is also important for any field manipulation that requires minimum disturbance to the background, such as the flow field manipulation inside the human body. There are several conventional invisible metamaterial designs: a cloak can isolate the influence between the internal and external fields, a concentrator can concentrate the external field to form an intensified internal field, and a rotator can rotate the internal field by a specific angle with respect to the external field. However, a multifunctional invisible device that can continuously tune across all these functions has never been realized due to its challenging requirements on material properties. Inside a porous medium flow, however, we overcome these challenges and realize such a multifunctional metamaterial. Our hydrodynamic device can manipulate both the magnitude and the direction of the internal flow and, at the same time, make negligible disturbance to the external flow. Thus, we integrate the functions of the cloak, concentrator, and rotator within one single hydrodynamic metamaterial, and such metamaterials may find potential applications in biomedical areas such as tissue engineering and drug release.

RESUMO

The entrapment of bacteria near boundary surfaces is of biological and practical importance, yet the underlying physics is not well understood. We demonstrate that it is crucial to include a commonly neglected thermodynamic effect related to the spatial variation of hydrodynamic interactions, through a model that provides analytic explanation of bacterial entrapment in two dimensionless parameters: α_{1} the ratio of thermal energy to self-propulsion, and α_{2} an intrinsic shape factor. For α_{1} and α_{2} that match an Escherichia coli at room temperature, our model quantitatively reproduces existing experimental observations, including two key features that have not been previously resolved: The bacterial "nose-down" configuration, and the anticorrelation between the pitch angle and the wobbling angle. Furthermore, our model analytically predicts the existence of an entrapment zone in the parameter space defined by {α_{1},α_{2}}.

RESUMO

Near-field hydrodynamic interactions in active fluids are essential to determine many important emergent behaviors observed, but have not been successfully modeled so far. In this work, we propose an effective model capturing the essence of the near-field hydrodynamic interactions through a tensorial coefficient of resistance, validated numerically by a pedagogic model system consisting of an Escherichia coli bacterium and a passive sphere. In a critical test case that studies the scattering angle of the bacterium-sphere pair dynamics, we prove that the near-field hydrodynamics can make a qualitative difference even for this simple two-body system: Calculations based on the proposed model reveal a region in parameter space where the bacterium is trapped by the passive sphere, a phenomenon that is regularly observed in experiments but cannot be explained by any existing model. In the end, we demonstrate that our model also leads to efficient simulation of active fluids with tens of thousands of bacteria, sufficiently large for investigations of many emergent behaviors.

RESUMO

Currently, natural products are one of the priceless options for finding novel chemical pharmaceutical entities. Ellipticine is a naturally occurring alkaloid isolated from the leaves of Ochrosia elliptica Labill. Ellipticine and its derivatives are characterized by multiple biological activities. The purpose of this review was to provide a critical and systematic assessment of ellipticine and its derivatives as bioactive molecules over the last 60 years. Publications focused mainly on the total synthesis of alkaloids of this type without any evaluation of bioactivity have been excluded. We have reviewed papers dealing with the synthesis, bioactivity evaluation and mechanism of action of ellipticine and its derivatives. It was found that ellipticine and its derivatives showed cytotoxicity, antimicrobial ability, and anti-inflammatory activity, among which cytotoxicity toward cancer cell lines was the most investigated aspect. The inhibition of DNA topoisomerase II was the most relevant mechanism for cytotoxicity. The PI3K/AKT pathway, p53 pathway, and MAPK pathway were also closely related to the antiproliferative ability of these compounds. In addition, the structure-activity relationship was deduced, and future prospects were outlined. We are confident that these findings will lay a scientific foundation for ellipticine-based drug development, especially for anticancer agents.

RESUMO

Cancer with low survival rates is the second main cause of death among all diseases in the world and consequently, effective antineoplastic agents are urgently needed. Allosecurinine is a plant-derived indolicidine securinega alkaloid shown bioactivity. The object of this study is to investigate synthetic allosecurinine derivatives with considerable anticancer capacity against nine human cancer cell lines as well as mechanism of action. We synthesized twenty-three novel allosecurinine derivatives and evaluated their antitumor activity against nine cancer cell lines for 72 h by MTT and CCK8 assays. FCM was applied to analyze the apoptosis, mitochondrial membrane potential, DNA content, ROS production, CD11b expression. Western blot was selected to analyze the protein expression. Structure-activity relationships were established and potential anticancer lead BA-3 which induced differentiation of leukemia cells towards granulocytosis at low concentration and apoptosis at high concentration was identified. Mechanism studies showed that mitochondrial pathway mediated apoptosis within cancer cells with cell cycle blocking was induced by BA-3. In addition, western blot assays revealed that BA-3 induced expression of the proapoptotic factor Bax, p21 and reduced the levels of antiapoptotic protein such as Bcl-2, XIAP, YAP1, PARP, STAT3, p-STAT3, and c-Myc. Collectively, BA-3 was a lead compound for oncotherapy at least in part, through the STAT3 pathway. These results were an important step in further studies on allosecurinine-based antitumor agent development.

RESUMO

For various engineering and industrial applications it is desirable to realize mechanical systems with broadly adjustable elasticity to respond flexibly to the external environment. Here we discover a topology-correlated transition between affine and non-affine regimes in elasticity in both two- and three-dimensional packing-derived networks. Based on this transition, we numerically design and experimentally realize multifunctional systems with adjustable elasticity. Within one system, we achieve solid-like affine response, liquid-like non-affine response and a continuous tunability in between. Moreover, the system also exhibits a broadly tunable Poisson's ratio from positive to negative values, which is of practical interest for energy absorption and for fracture-resistant materials. Our study reveals a fundamental connection between elasticity and network topology, and demonstrates its practical potential for designing mechanical systems and metamaterials.

RESUMO

Dimensionality often strongly affects material properties and phase transition behaviors, but its effects on crystal surfaces, such as roughening and premelting, have been poorly studied. Our simulation revealed that these surface behaviors are distinct in monolayer and multilayer Lennard-Jones (LJ) crystals. Solid surfaces fluctuate as capillary waves during the roughening process, but complete roughening is preempted by premelting. As the melting temperature is approached, the thickness of the premelted liquid layer approaches a constant (i.e., blocked premelting) for monolayer crystals, but diverges as a power law (i.e., complete premelting) for bilayer and trilayer crystals. The surface liquids of monolayer crystals contain crystalline patches and exhibits rough liquid-vapour and liquid-crystal interfaces, in contrast to the normal surface liquids of bilayer and trilayer crystals. Monolayer crystals melt heterogeneously from the surface without forming a hexatic phase and produce many vacancies.

RESUMO

Bacterial suspensions-a premier example of active fluids-show an unusual response to shear stresses. Instead of increasing the viscosity of the suspending fluid, the emergent collective motions of swimming bacteria can turn a suspension into a superfluid with zero apparent viscosity. Although the existence of active superfluids has been demonstrated in bulk rheological measurements, the microscopic origin and dynamics of such an exotic phase have not been experimentally probed. Here, using high-speed confocal rheometry, we study the dynamics of concentrated bacterial suspensions under simple planar shear. We find that bacterial superfluids under shear exhibit unusual symmetric shear bands, defying the conventional wisdom on shear banding of complex fluids, where the formation of steady shear bands necessarily breaks the symmetry of unsheared samples. We propose a simple hydrodynamic model based on the local stress balance and the ergodic sampling of nonequilibrium shear configurations, which quantitatively describes the observed symmetric shear-banding structure. The model also successfully predicts various interesting features of swarming vortices in stationary bacterial suspensions. Our study provides insights into the physical properties of collective swarming in active fluids and illustrates their profound influences on transport processes.

RESUMO

BACKGROUND: Virtual reality (VR) is an innovation that permits the individual to discover and operate within three-dimensional (3D) environment to gain practical understanding. This research aimed to examine the general efficiency of VR for teaching medical anatomy. METHODS: We executed a meta-analysis of randomized controlled studies of the performance of VR anatomy education. We browsed five databases from the year 1990 to 2019. Ultimately, 15 randomized controlled trials with a teaching outcome measure analysis were included. Two authors separately chose studies, extracted information, and examined the risk of bias. The primary outcomes were examination scores of the students. Secondary outcomes were the degrees of satisfaction of the students. Random-effects models were used for the pooled evaluations of scores and satisfaction degrees. Standardized mean difference (SMD) was applied to assess the systematic results. The heterogeneity was determined by I2 statistics, and then was investigated by meta-regression and subgroup analyses. RESULTS: In this review, we screened and included fifteen randomized controlled researches (816 students). The pooled analysis of primary outcomes showed that VR improves test scores moderately compared with other approaches (standardized mean difference [SMD] = 0.53; 95% Confidence Interval [CI] 0.09-0.97, p < 0.05; I2 = 87.8%). The high homogeneity indicated that the studies were different from each other. Therefore, we carried out meta-regression as well as subgroup analyses using seven variables (year, country, learners, course, intervention, comparator, and duration). We found that VR improves post-intervention test score of anatomy compared with other types of teaching methods. CONCLUSIONS: The finding confirms that VR may act as an efficient way to improve the learners' level of anatomy knowledge. Future research should assess other factors like degree of satisfaction, cost-effectiveness, and adverse reactions when evaluating the teaching effectiveness of VR in anatomy.

RESUMO

BACKGROUND: Knee osteoarthritis (KOA) is a degenerative joint disease leading to pain and disability for which no curative treatment exists. Intra-articular (IA) therapies are part of this multimodal approach and are approved by the Food and Drug Administration (FDA) and European Medicines Agency (EMA). Platelet-rich plasma (PRP), hyaluronic acid (HA), and corticosteroids (CS) have been increasingly used in recent years to treat KOA. PURPOSE: To determine whether IA-PRP was superior to IA-HA or IA-CS administration routes in these patients. MATERIAL AND METHODS: In this trial the patients were randomized to IA-HA (2 ml/week, for 3 weeks), IA-CS (1 ml) or IA-PRP (3 times, 4 ml, every 3 weeks) groups. The outcome was assessed using the Western Ontario and McMaster Universities (WOMAC) score prior to the first injection and then at 3, 6, 9 and 12 months. Pain was evaluated by a visual analogue scale (VAS) prior to treatment and after 12 months. RESULTS: In this study 120 patients were randomized into 3 groups. There was a significant improvement in all scores (WOMAC, VAS) in each group compared to the pretreatment values (P < 0.05). The mean WOMAC scores for the IA-HA group from pretreatment to 3, 6, 9, and 12 months were 47.23 ± 5.37, 25.02 ± 4.98, 26.38 ± 5.20, 27.86 ± 4.34, and 30.64 ± 8.36, respectively. Similar improvements were noted in the IA-CS and IA-PRP groups. There were no significant differences in the WOMAC scores between the 3 groups 3 months after treatment (P > 0.05) but IA-PRP showed significantly lower scores 6, 9 and 12 months after treatment (P < 0.05). CONCLUSION: Intra-articular PRP injections into the knee for symptomatic early stages of KOA are a valid treatment option. The clinical efficacy of IA-PRP is comparable to that of the IA-HA and IA-CS forms after 3 months and the long-term efficacy of IA PRP is superior to IA-HA and IA-CS.

RESUMO

The maintenance and improvement of ecosystem services on the Tibet Plateau are critical for national ecological security in China and are core objectives of ecological conservation in this region. In this paper, ecosystem service benefits of the Tibet Ecological Conservation Project were comprehensively assessed by estimating and mapping the spatiotemporal variation patterns of critical ecosystem services on the Tibet Plateau from 2000 to 2015. Furthermore, we linked the benefit assessment to the sustainable spatial planning of future ecological conservation strategies. Comparing the 8 years before and after the project, the water retention and carbon sink services of the forest, grassland and wetland ecosystems were slightly increased after the project, and the ecosystem sand fixation service has been steadily enhanced. The increasing forage supply service of grassland significantly reduced the grassland carrying pressure and eased the conflict between grassland and livestock. However, enhanced rainfall erosivity occurred due to increased rainfall, and root-layer soils could not recover in a short period of time, both factors have led to a decline in soil conservation service. The warm and humid climate is beneficial for the restoration of ecosystems on the Tibet Plateau, and the implementation of the Tibet Ecological Conservation Project has had a positive effect on the local improvement of ecosystem services. A new spatial planning strategy for ecological conservation was introduced and aims to establish a comprehensive, nationwide system to protect important natural ecosystems and wildlife, and to promote the sustainable use of natural resources.

RESUMO

Active fluids such as swarming bacteria and motile colloids exhibit exotic properties different from conventional equilibrium materials. As a peculiar example, a spherical tracer immersed inside active fluids shows an enhanced translational diffusion, orders of magnitude stronger than its intrinsic Brownian motion. Here, rather than spherical tracers, we investigate the diffusion of isolated ellipsoids in a quasi-two-dimensional bacterial bath. Our study shows a nonlinear enhancement of both translational and rotational diffusions of ellipsoids. More importantly, we uncover an anomalous coupling between particles' translation and rotation that is strictly prohibited in Brownian diffusion. The coupling reveals a counterintuitive anisotropic particle diffusion, where an ellipsoid diffuses fastest along its minor axis in its body frame. Combining experiments with theoretical modeling, we show that such an anomalous diffusive behavior arises from the generic straining flow of swimming bacteria. Our work illustrates an unexpected feature of active fluids and deepens our understanding of transport processes in microbiological systems.

RESUMO

The properties of a classical simple liquid are strongly affected by the application of an external potential that supports inhomogeneity. To understand the nature of these property changes, the equilibrium particle distribution functions of the liquid have, typically, been calculated directly using either integral equation or density functional based analyses. In this study, we develop a different approach with a focus on two distribution functions that characterize the inhomogeneous liquid: the pair direct correlation function c(r1,r2) and the pair correlation function g(r1,r2). With g(r1,r2) considered to be an experimental observable, we solve the Ornstein-Zernike equation for the inhomogeneous liquid to obtain c(r1,r2), using information about the well studied and resolved g(0)(r1,r2) and c(0)(r1,r2) for the parent homogeneous ((0)) system. In practical cases, where g(r1,r2) is available from experimental data in a discrete form, the resulting c(r1,r2) is expressed as an explicit function of g(r1,r2) in a discrete form. A weaker continuous form of solution is also obtained, in the form of an integral equation with finite integration limits. The result obtained with our formulation is tested against the exact solutions for the correlation and distribution functions of a one-dimensional inhomogeneous hard rod liquid. Following the success of that test, the formalism is extended to higher dimensional systems with explicit consideration of the two-dimensional liquid.

RESUMO

Colloidal suspensions exhibit shear thinning and shear thickening. The most common interpretation of these phenomena identifies layering of the fluid perpendicular to the shear gradient as the driver for the observed behavior. However, studies of the particle configurations associated with shear thinning and thickening cast doubt on that conclusion and leave unsettled whether these nonequilibrium phenomena are caused primarily by correlated particle motions or by changes in particle packing structure. We report the results of stokesian dynamics simulations of suspensions of hard spheres that illuminate the relation among the suspension viscosity, shear rate, and particle configuration. Using a recently introduced sampling technique for nonequilibrium systems, we show that shear thinning can be decoupled from layering, thereby eliminating layering as the driver for shear thinning. In contrast, we find that there is a strong correlation between shear thinning and a two-particle measure of the shear stress. Our results are consistent with a recent experimental study.

RESUMO

Colloidal suspensions self-assemble into equilibrium structures ranging from face- and body-centered cubic crystals to binary ionic crystals, and even kagome lattices. When driven out of equilibrium by hydrodynamic interactions, even more diverse structures can be accessed. However, mechanisms underlying out-of-equilibrium assembly are much less understood, though such processes are clearly relevant in many natural and industrial systems. Even in the simple case of hard-sphere colloidal particles under shear, there are conflicting predictions about whether particles link up into string-like structures along the shear flow direction. Here, using confocal microscopy, we measure the shear-induced suspension structure. Surprisingly, rather than flow-aligned strings, we observe log-rolling strings of particles normal to the plane of shear. By employing Stokesian dynamics simulations, we address the mechanism leading to this out-of-equilibrium structure and show that it emerges from a delicate balance between hydrodynamic and interparticle interactions. These results demonstrate a method for assembling large-scale particle structures using shear flows.
