RESUMO

Many Vicia species (Fabaceae) were proven to possess bioactive compounds with potential health beneficial properties. The present study was designed to determine the phenolic constituents, antioxidant and enzyme inhibition activities of aerial parts and seed of V. peregrina. Hexane, ethyl acetate and methanol extracts were prepared by maceration and aqueous extract by infusion. The chemical compositions of the extracts were determined using HPLC-MS/MS technology. The antioxidant activities were examined using various assays including free radical scavenging (ABTS and DPPH), reducing ability (CUPRAC and FRAP), metal chelation, and phosphomolybdenum. The enzyme inhibitory effects were investigated against cholinesterase, tyrosinase, amylase and glucosidase. The highest total phenolics and flavonoids contents were recorded in the methanol extracts of the seed (45.42 mg GAE/g) and aerial parts (40.33 mg RE/g) respectively. The aerial parts were characterized by higher accumulation of chlorogenic acid (9893.86 µg g-1 ), isoquercitrin (9400.33 µg g-1 ), delphindin 3,5 diglucoside (9113.28 µg g-1 ), hyperoside (6337.09 µg g-1 ), rutin (3489.83 µg g-1 ) and kaempferol-3-glucoside (2872.84 µg g-1 ). Generally, the methanol and aqueous extracts of the two studied parts exerted the best antioxidant activity with highest anti-DPPH (61.99 mg TE/g), anti-ABTS (101.80 mg TE/g) and Cu++ (16169 mg TE/g) and Fe+++ (172,36 mg TE/g) reducing capacity were recorded from the seed methanol extract. Methanol extract of the seed showed the best anti-tyrosinase activity (75.86 mg KAE/g). These results indicated that V. peregrina is rich with bioactive phenolics suggesting their use in different health promoting applications.

MAIN CONCLUSION: Species have plasticity across altitude gradients in leaf morphology and function, and their response to high altitude conditions was mainly reflected in leaf cell metabolism and gas exchange. Leaf morphological and functional adaptation to altitude has received research attention in recent years, but there are no studies for forage legumes. Here we report differences in 39 leaf morphology and functional traits of three leguminous forages (alfalfa, sainfoin and perennial vetch) at three sites in Gansu Province, China, ranging from 1768 to 3074 m altitude to provide information for potential use in breeding programmes. With increasing altitude, plant water status increased, reflecting increase in soil water content and decreased average temperature, which lead to leaf intercellular CO2 concentration. Stomatal conductance and evapotranspiration increased significantly but water-use efficiency decreased. At high altitude, ΦPSII decreased but non-photochemical quenching and chlorophyll a:b ratio increased while spongy mesophyll tissue and leaf thickness increased. These changes may be due to UV or low-temperature damage of leaf protein and metabolic cost of plant protection or defence responses. Contrary to many other studies, leaf mass per area decreased significantly at higher altitude. This was consistent with predictions under the worldwide leaf economic spectrum on the basis that soil nutrients increased with increasing altitude. The key species differences were more irregularly shaped epidermal cells and larger stomatal size in perennial vetch compared to alfalfa or sainfoin that enhanced gas exchange and photosynthesis by generating mechanical force, increasing guard cell turgor, and promoting stomatal operation. The lower adaxial stomatal density also enhanced water-use efficiency. These adaptations might confer perennial vetch an advantage in environments with extreme diurnal temperature fluctuation or in frigid conditions.

In the present study, in order to improve the growth performance of hairy vetch (Vicia villosa Roth., Local landrace from Ardabil, Iran) seedlings grown in the soil contaminated with heavy metals Pb and Zn, our attention was directed toward the application of biochar, inoculation with conidial suspension of Trichoderma harzianum Rifai-T22 and management of phosphorus (P) nutrition. Heavy metal toxicity reduced leaf greenness, membrane stability index, maximum quantum yield of PSΙΙ (Fv/Fm), P concentration and uptake in plant tissues and root and shoot biomass, but increased Pb and Zn concentration and uptake in root and leaf, H2O2 and malondialdehyde content and CAT and POX activity in the leaves. The application of biochar, inoculation with Trichoderma fungus and P supplementation increased the shoot P content, which might contribute to the alleviation of P insufficiency and a subsequent elevation in P transfer to aboveground biomass, and eliminated the toxicity of heavy metal on hairy vetch plants, which was revealed in reducing oxidative stress and enhancing plant growth performance. The biochar considerably increased Zn immobilization, while being able to slightly stabilize Pb. Co-application of Trichoderma and 22 mg P/kg soil (22P) increased the concentration and uptake of Zn in the roots and decreased the translocation of this element to the shoots, especially when biochar was not amended. Although the biochar and P inputs could compensate the negative Trichoderma effects, the results suggested that biochar application in combination with fungal inoculation and 22-P supplementation could not only increase hairy vetch growth performance but also decline heavy metal uptake to ensure the production of a forage crop in soils polluted with heavy metals based on the nutritional standards of livestock.

Long-chain polyprenyl diphosphate synthases play a critical role in the formation of the prenyl side-chain of ubiquinones, but up to date, their functions have scarcely been characterized in insects. Here, we first cloned the complementary DNAs encoding the subunits of decaprenyl diphosphate synthase (DPPS) in the vetch aphid Megoura viciae, an important agricultural pest insect. The results showed that there existed three DPPS subunits, designated as MvDPPS1, MvDPPS2a, and MvDPPS2b, with an open reading frame of 1218, 1275, and 1290 bp, and a theoretical isoelectric point of 7.91, 6.63, and 9.62, respectively. The sequences of MvDPPS1s from different aphid species were nearly identical, while the sequences of MvDPPS2a and MvDPPS2b shared only moderate sequence similarity. Phylogenetic analysis clearly separated MvDPPS2a and MvDPPS2b, indicating a functional differentiation between them. Functional coexpression analysis in Escherichia coli showed that MvDPPS1 plus MvDPPS2a and MvDPPS1 plus MvDPPS2b, respectively, catalyzed the formation of the prenyl side-chain of the ubiquinone coenzyme Q10 (CoQ10). Interestingly, MvDPPS1 plus MvDPPS2b catalyzed the formation of the prenyl side-chain of a ubiquinone other than CoQ10. RNA interference-mediated knockdown of MvDPPS2a imposed no significant effect on MvDPPS2b, and vice versa, suggesting no compensatory action between them. In the end, we detected the product CoQ10 in the aphid, the first identification of CoQ10 in an insect species. Taken together, we characterized two functional DPPSs in M. viciae, one of which might be multifunctional. Our study helps to understand the functional plasticity of the terpenoid backbone biosynthesis pathway in insects.

Vicia amoena is a high-nutritional quality forage similar to alfalfa. However, studies on the genetic background of V. amoena are scarce. In the present study, the genetic variation of 24 V. amoena populations was assessed with newly developed simple sequence repeat (SSR) markers. A total of 8799 SSRs were identified in the V. amoena genomic-enriched sequences, and the most abundant repeat number was four. A total of 569 sampled individuals were assayed to evaluate the genetic diversity of the V. amoena populations based on 21 polymorphic SSR primers. The polymorphism information content (PIC) ranged from 0.896 to 0.968, with an average of 0.931, which indicated that the markers were highly informative. Based on analysis of molecular variance, 88% of the variance occurred within populations, and the remaining 12% of the variance occurred among populations. The high degree of gene flow (Nm= 4.958) also showed slight differentiation among the V. amoena populations. The V. amoena populations were mainly clustered by steppe and mountain habitats based on principal coordinate analysis (PCoA) and STRUCTURE analysis. This indicated that the elevation and special habitat of geographical origins may be important factors affecting the clustered pattern of V. amoena populations. Neighbour-joining (NJ) analysis did not separate the populations well by geographical origin, which indicated that the genetic structure of V. amoena was complex and needs further study. Overall, our results showed that the newly developed SSR markers could benefit the V. amoena research community by providing genetic background information to help establish a foundation for breeding improvement and germplasm resource conservation.

PREMISE: Annual and perennial life history transitions are abundant among angiosperms, and understanding the phenotypic variation underlying life span shifts is a key endeavor of plant evolutionary biology. Comparative analyses of trait variation and correlation networks among annual and perennial plants is increasingly important as new herbaceous perennial crops are being developed for edible seed. However, it remains unclear how seed to vegetative growth trait relationships correlate with life span. METHODS: To assess the relative roles of genus and life span in predicting phenotypic variation and trait correlations, we measured seed size and shape, germination proportion, and early-life-stage plant height and leaf growth over 3 mo in 29 annual and perennial, herbaceous congeneric species from three legume genera (Lathyrus, Phaseolus, and Vicia). RESULTS: Genus was the strongest predictor of seed size and shape variation, and life span consistently predicted plant height and leaf number at single time points. Correlation networks revealed that annual species had significant associations between seed traits and vegetative traits, whereas perennials had no significant seed-vegetative associations. Each genus also differed in the extent of integration between seed and vegetative traits, as well as within-vegetative-trait correlation patterns. CONCLUSIONS: Genus and life span were important for predicting aspects of early-life-stage phenotypic variation and trait relationships. Differences in phenotypic correlation may indicate that selection on seed size traits will impact vegetative growth differently depending on life span, which has important implications for nascent perennial breeding programs.

The plants belonging to the genus Vicia are of great interest as a source of many bioactive compounds and micronutrients. A snapshot of their cultivation, habitat, main components, from which essential oils can be obtained, is given. The traditional medicinal uses of Vicia plants are also reported, as well as the wide spectrum of the main biological activities attributed to Vicia plants is discussed regarding potential health beneficial properties, in particular anti-Parkinson, anticholinesterase, antidepressant, anticonvulsant, antimicrobial, cytotoxic, antioxidant, antiinflammatory and antinociceptive, antidiabetic, antihemolytic, anticoagulant, estrogenic, diuretic, antihypoxic activities.

Vicia palaestina Boiss. is an annual herb that grows in dry areas of eastern Mediterranean countries. It belongs to section Cracca subgenus Vicilla, which is characterized by having a high content in the non-protein amino acid canavanine. The seeds from some of these vetches are also rich in lectins. The purification and characterization of a single-chain lectin from the seeds of V. palaestina is described here. This lectin was the most abundant protein in albumin extracts. It has affinity for the glycoconjugate N-acetylgalactosamine and inhibits proliferation of the cancerous Caco-2 and THP-1 cell lines. In addition to their high nutritional value, the seeds from V. palaestina represent a source of lectins with health promoting and pharmacological potential because of their antiproliferative activity.

This report describes an outbreak of hairy vetch toxicosis afflicting a herd of cattle with a fatal cutaneous and systemic granulomatous disease. It highlights how this condition remains poorly recognized by cattle production professionals in Europe and the need for communication about vetch-associated diseases.


Cet article décrit une épidémie de toxicose due à la vesce velue touchant un troupeau de bovins se manifestant par une maladie granulomateuse systémique et cutanée fatale. Ceci illustre comment cette atteinte reste peu connue par les professionnels de l'élevage en Europe et le besoin de communiquer sur les maladies associées à la vesce velue.


Este artículo describe un brote de toxicosis de veza vellosa (Vicia villosa) que afectó a un rebaño de ganado con una enfermedad granulomatosa cutánea y sistémica mortal. Destaca cómo esta condición sigue siendo poco reconocida por los profesionales de la producción ganadera en Europa y la necesidad de comunicación sobre las enfermedades asociadas al consumo de veza.


Este relato descreve um surto de intoxicação por ervilhaca peluda em um rebanho de gado com uma doença granulomatosa cutânea e sistêmica. Destaca-se como essa enfermidade continua pouco reconhecida pelos profissionais de bovinocultura na Europa, e a necessidade de comunicação sobre doenças associadas à ervilhaca.

This study aimed to investigate the replacement value of half time grazing of wheat stubbles by vetch, which had been cropped under the context of conservation agriculture (CA). Three grazing treatments were evaluated on Barbarine lambs (initial BW 18 ± 1.42 kg). Treatment 1 consists of 6-hr grazing on dried vetch only (V). For treatment 2, the sheep were grazing 3 hr on wheat stubbles in morning and 3 hr on dried vetch in the afternoon (VWS). Treatment 3 consists of 6-hr grazing of wheat stubbles only (WS). At grain maturity stage, biomass yield of vetch averaged 7 tons DM/ha allowing a grazing period of 2 consecutive months. Along this period, vetch conserved its pods indehiscent. Biomass and nutritive value of vetch and wheat stubbles were decreasing from the start to the end of the grazing period. Residual biomass was higher in vetch and wheat stubble assigned to treatment VWS. WS lambs spent more time on walking and standing, while V and VWS lambs allocated more time on biomass uptake. The DM, OM and CP intakes were higher in animal grazing vetch alone or combined to wheat stubble. Rumen fermentation parameters (pH, ammonia nitrogen concentration and protozoa count) were not affected (p > .05) by any of the three treatments. The average daily gain of lambs on V and VWS lambs was three times greater (p < .05) than that of WS lambs (164, 152 and 49.5 g respectively). Cold carcass yield averaged 444, 428 and 388 g/ kg for lambs assigned to V, VWS and WS treatments respectively. It is concluded that grazing vetch alone or combined with WS increased substantially the growth performance and carcass yield of lambs compared with WS grazing only. Therefore, dried vetch grazing could be a solution to make possible mulching and biomass uptake by sheep under the context of CA.

Symbiotic viruses exist in many insects; however, their functions in host insects are not well understood. In this study, we explored the role of acyrthosiphon pisum virus (APV) in the interaction of its host aphid Acyrthosiphon pisum with plants. APV is primarily located in aphid salivary glands and gut and propagated in the insect. APV is horizontally transmitted to host plants during aphid feeding, but the virus does not replicate in the host plant. When the pea host race of aphids colonized two low-fitness plants, Medicago truncatula and Vicia villosa, the virus titers in both the aphids and plants significantly increased. Furthermore, APV infection strongly promoted the survival rate of the pea host race on V. villosa. Transcriptomic analysis showed that only 0.85% of aphid genes responded to APV infection when aphids fed on V. villosa, with a fold change in transcript levels of no more than fourfold. The improved survival due to APV infection was apparently related to the inhibitory effect of the virus on levels of phytohormone jasmonic acid (JA) and JA-isoleucine. Our data suggest a benefit of the symbiotic virus to its aphid host and demonstrate a novel case of symbiotic virus-mediated three-species interaction.

Vicia species, commonly known as vetches, include legume plants which nowadays can be found in many countries around the world. Their use to improve soil health and productivity is crucial in management schemes that make sustainable agriculture possible, but they can also play a part in the phytoremediation of polluted environments. Furthermore, they harbor a large community of rhizospheric microorganisms, such as biodegradative bacteria and plant growth-promoting rhizobacteria, which can help to increase phytoremediation efficiency. Their mutualistic association with Rhizobium sp. has also been proposed as an attractive bioremediation tool. Thus, Vicia species could make a remarkable difference in the ecological restoration of polluted soils, thanks to their dual role as cover crops and phytoremediator plants. This mini-review discusses recent advances in the use of Vicia. Challenges and opportunities connect with the application of these species will also be revised, as well as aspects that remain to be explored.

The contents of photosynthetic pigments are an important indicator of many processes taking place in the plant body. Still, however, our knowledge of the effects of polyploidization, a major driver of speciation in vascular plants, on the contents of photosynthetic pigments is very sparse. We compared the contents of photosynthetic pigments among natural diploids, natural tetraploids, and synthetic tetraploids. The material originated from four natural mixed-cytotype populations of diploid and autotetraploid Vicia cracca (Fabaceae) occurring in the contact zone between the cytotypes in Central Europe and was cultivated under uniform conditions. We explored whether the contents of pigments are primarily driven by polyploidization or by subsequent evolution of the polyploid lineage and whether the patterns differ between populations. We also explored the relationship between pigment contents and plant performance. We found very few significant effects of the cytotype on the individual pigments but many significant interactions between the cytotype and the population. In pair-wise comparisons, many comparisons were not significant. The prevailing pattern among the significant once was that the contents of pigments were determined by polyploidization rather than by subsequent evolution of the polyploid lineage. The contents of the pigments turned out to be a useful predictor of plant performance not only at the time of material collection, but also at the end of the growing season. Further studies exploring differences in the contents of photosynthetic pigments in different cytotypes using replicated populations and assessing their relationship to plant performance are needed to assess the generality of our findings.

This study proposed the General Health Index of Cheese (GHIC) as an indicator for the presence of health-promoting compounds in cheese and compared the antioxidant capacity and phenolic and fatty acid contents of cheeses from goats consuming 7 forage species. Ninety-one homogeneous Red Syrian goats were randomly assigned to 1 of 7 feeding treatments (Festuca arundinacea, Hordeum vulgare, Triticosecale, Pisum sativum, Trifolium alexandrinum, Vicia sativa, and Vicia faba minor). The housed goat groups received the scheduled forage ad libitum. Forage species affected the antioxidant capacity, the phenolic and fatty acid contents, the Health Promoting Index, and the GHIC. Trifolium alexandrinum, Triticosecale, and Hordeum vulgare showed a clear advantage in terms of beneficial fatty acids content in goat cheese. Cheese from the Triticosecale group also showed a high antioxidant capacity value even if its polyphenol content was intermediate compared with others. Trifolium alexandrinum and Triticosecale had the highest value of the new index GHIC. This comparison suggests that there are important differences in fatty acid profile and polyphenol content among cheeses from goats fed grasses and legumes commonly used in the Mediterranean area. In this first approach, GHIC index, which combines the positive components found in cheese, seems to be a useful tool to provide an indication concerning the general health value of the product.

Neonicotinoid seed treatments are extensively used to systemically protect corn from invertebrate herbivory. Interseeding cover crops can promote beneficial insect communities and their ecosystem services such as predation on pests, and this practice is gaining interest from farmers. In this study, cereal rye (Secale cereale) and hairy vetch (Vicia villosa) were planted between rows of early vegetative corn that had been seed-treated with thiamethoxam. Thiamethoxam and its insecticidal metabolite, clothianidin were quantified in cover crop leaves throughout the growing season. Thiamethoxam was present in cereal rye at concentrations ranging from 0 to 0.33 ± 0.09 ng/g of leaf tissue and was detected on six out of seven collection dates. Cereal rye leaves contained clothianidin at concentrations from 1.05 ± 0.22 to 2.61 ± 0.24 ng/g and was present on all sampling dates. Both thiamethoxam and clothianidin were detected in hairy vetch on all sampling dates at rates ranging from 0.10 ± 0.05 to 0.51 ± 0.11 ng/g and 0.56 ± 0.15 to 9.73 ± 5.04 ng/g of leaf tissue, respectively. Clothianidin was measured at a higher concentration than its precursor, thiamethoxam, in both plant species on every sampling date. Neonicotinoids entering interseeded cover crops from adjacent treated plants is a newly discovered route of exposure and potential hazard for non-target beneficial invertebrates. Future research efforts should examine the effects of systemic insecticides on biological communities in agroecosystems whose goal is to diversify plant communities using methods such as cover cropping.

In recent decades, some bumble bee species have declined, including in North America. Declines have been reported in species of bumble bees historically present in Ontario, including: yellow bumble bee (Bombus fervidus) (Fabricus, 1798), American bumble bee (Bombus pensylvanicus) (DeGeer, 1773), and yellow-banded bumble bee (Bombus terricola) (Kirby, 1837). Threats contributing to bumble bee population declines include: land-use changes, habitat loss, climate change, pathogen spillover, and pesticide use. A response to the need for action on pollinator preservation in North America has been to encourage 'bee-friendly' plantings. Previous studies show differences in common and at-risk bumble bee foraging; however, similar data are unavailable for Ontario. Our research question is whether there is a difference in co-occurring at-risk and common bumble bee (Bombus spp.) floral use (including nectar and pollen collection) in protected areas in southern Ontario. We hypothesize that common and at-risk species forage differently, predicting that at-risk species forage on a limited selection of host plants. We conducted a field survey of sites in southern Ontario, using observational methods to determine bumble bee foraging by species. The results of a redundancy analysis show a difference in foraging between common and at-risk bumblebee species. At-risk bumble bee species show a preference for foraging on invasive, naturalized Vicia cracca (tufted vetch). This finding raises the question of how to preserve or provide forage for at-risk bumble bees, when they show an association with an invasive species often subject to control in protected areas.

This study evaluated the varietal variability and interrelationship of the grain and stover yields, and stover quality characteristics of four common vetch varieties (three improved varieties and one local variety) used on the Tibetan Plateau. Stover quality attributes determined included chemical composition, in vitro true digestibility of dry matter (IVTD) and neutral detergent fibre (dNDF). The improved varieties were superior to the local variety in grain yield, harvest index and potential utility index (PUI). The wide ranges in the proportions of leaf (24.3%-34.6%), pod (22.0%-45.6%) and stem (29.5%-44.7%) fractions were observed. Significant varietal differences (p < .001) were observed for the whole-stover content of crude protein [CP; 78.1-101 g/kg dry matter (DM)], neutral detergent fibre (aNDF; 520-564 g/kg DM), acid detergent fibre (ADF; 520-564 g/kg DM), IVTD (597-658 g/kg DM) and dNDF (305-384 g/kg aNDF). The leaf fraction had the highest levels of CP, IVTD and dNDF, while the highest contents of aNDF and ADF were observed in the stem fraction. Canonical correlation analysis showed that significant (p < .001) correlations between the quality attributes of whole stover and proportions and quality attributes of stover morphological fractions. Pearson correlation between the grain and stover yield was negative and significant (r = -.295; p = .001), while the correlations between the grain/stover yield and stover quality traits were positive or non-significant. Ranking of the varieties differed when grain yield, PUI and stover quality scores were investigated. Of the varieties tested, Lanjian No.2 has the best potential for use as a ruminant feed, as indicated by PUI and stover quality.

Plants engineer the rhizosphere to their advantage by secreting various nutrients and secondary metabolites. Coupling transcriptomic and metabolomic analyses of the pea (Pisum sativum) rhizosphere, a suite of bioreporters has been developed in Rhizobium leguminosarum bv viciae strain 3841, and these detect metabolites secreted by roots in space and time. Fourteen bacterial lux fusion bioreporters, specific for sugars, polyols, amino acids, organic acids, or flavonoids, have been validated in vitro and in vivo. Using different bacterial mutants (nodC and nifH), the process of colonization and symbiosis has been analyzed, revealing compounds important in the different steps of the rhizobium-legume association. Dicarboxylates and sucrose are the main carbon sources within the nodules; in ineffective (nifH) nodules, particularly low levels of sucrose were observed, suggesting that plant sanctions affect carbon supply to nodules. In contrast, high myo-inositol levels were observed prior to nodule formation and also in nifH senescent nodules. Amino acid biosensors showed different patterns: a γ-aminobutyrate biosensor was active only inside nodules, whereas the phenylalanine bioreporter showed a high signal also in the rhizosphere. The bioreporters were further validated in vetch (Vicia hirsuta), producing similar results. In addition, vetch exhibited a local increase of nod gene-inducing flavonoids at sites where nodules developed subsequently. These bioreporters will be particularly helpful in understanding the dynamics of root exudation and the role of different molecules secreted into the rhizosphere.

Nanoviruses are multi-component plant-infecting single-stranded DNA viruses. Using a viral metagenomics-informed approach, a new nanovirus and two associated alphasatellite molecules have been identified in an uncultivated asymptomatic Vicia cracca plant in the Rhône region of France. This novel nanovirus genome includes eight genomic components (named DNA-R, DNA-S, DNA-M, DNA-C, DNA-N, DNA-U1, DNA-U2 and DNA-U4) and, across all components, shares < 66% pairwise sequence identity with other nanovirus genomes. The two associated alphasatellites share 62% identity with each other and < 81% identity will all other nanovirus-associated alphasatellites.

Despite the significant livestock contribution to households' nutrition and incomes in many African smallholder farms, milk productivity remains low. Inadequate feeding is the main reason for the underperformance. To contribute towards addressing this, an on-farm feeding trial was undertaken in Ol-joro-Orok Central Kenya. A feed basket using oat (Avena sativa) cv Conway and vetch (Vicia villosa) was compared to farmers practice. Milk production (kg) and quality parameters, including butterfat, protein, lactose, and density, were monitored, and cost-benefit analysis (CBA) undertaken. Feeding both oat and vetch increased milk production by 21% (morning) and 18%, (evening), equivalent to 1.4 kg/day. Increases (%) in quality were butter fat (18.2), solid-non-fat (16.5), lactose (16.2), and protein (16.1). Concomitantly, the CBA returned positive results, supporting the hypothesis of economic advantage in using oat and vetch in milk production in the area, and possibly in other similar areas.
