ABSTRACT

Studies on physiological responses to stimuli from physical factors are essential for understanding the dynamics of the microorganisms and higly important for the management of plant diseases. Besides, the development of an epidemiological model for pathogen populations requires studying their physiological responses to physical stimuli. The objective of this study was to evaluate the germination dynamics of spores from six isolates of Bipolaris bicolor under effects of light at 25 °C. Suspensions of 1.6 × 105 conidia mL- 1 from the B. bicolor isolates were inoculated onto Petri dishes containing agar-water culture medium and incubated in a BOD chamber under two physical conditions: (a) constant darkness and (b) constant light for five hours. The study was conducted in a completely randomized design, with a 6 × 2 factorial arrangement (six B. bicolor isolates and two physical conditions) and five replications. The length of the germ tube was measured hourly. The constant darkness resulted in higher mean germ tube growth for the pathogen; however, differences in the final germination percentage were found among the isolates. The isolate F-24-02 exhibited the highest germination adaptability to constant darkness, presenting the longest germ tube length.

ABSTRACT

A multitude of plants from the Brazilian savanna are known for their medicinal properties. Many plants contain endophytic fungi, which lead to the production of bioactive compounds by both the fungi and their hosts. This study investigated the bioprospecting of endophytic fungi recovered from the leaves of Palicourea rigida, a native medicinal plant of the Brazilian savanna. Four fungal taxa (Colletotrichum sp. SXS649, Pestalotiopsis sp. SXS650, the order Botryosphaeriales SXS651, and Diaporthe sp. SXS652) were recovered. The phenolic, flavonoid, extracellular degrading enzymes (amylase, cellulase, protease, and tannase) and antioxidant activity of these taxa were determined. Evaluation of the antimicrobial activity showed that the Botryosphaeriales SXS651 extract displays a minimum inhibitory concentration (MIC) of 23.20 mg mL-1 against Staphylococcus epidermidis and Pseudomonas aeruginosa, and the Diaporthe sp. SXS652 extract exhibited an MIC of 27.00 mg mL-1 against Escherichia coli. The Colletotrichum sp. SXS649 isolate inhibited tumors in potato discs by 69% at a concentration of 9.70 mg mL-1. All isolates had potential bioremediation criteria against soil contaminated with soybean oil, as proved by a high percentage of germination of Lactuca sativa and a reduction in phytotoxicity. Furthermore, the taxa under investigation demonstrated antagonistic action to phytopathogenic fungi, namely, Aspergillus niger, Inonotus rickii, Pestalotiopsis mangiferae, and Coniophora puteana, with an inhibition range between 34.2% and 76.9%. The preliminary toxicity assessment showed that all isolates possessed an LC50 of less than 100 mg mL-1 to the microcrustacean Artemia salina. These results indicate that the endophytic fungi of the Brazilian savanna are promising candidates for biotechnological and industrial applications and, in agricultural applications, for the biological control of phytopathogenic fungi.

ABSTRACT

This study evaluated the initial development of paricá (Schizolobium parahyba var. amazonicum) seedlings under different nitrogen rates with the application of Trichoderma spp., using a randomized complete block design in a 4x5 factorial scheme (strains and rates) with seven replications. The evaluated traits were plant height, stem diameter, leaf and stem fresh weights, leaf and stem dry weights, and aerial part dry and fresh weights. Trichoderma spp. strains did not satisfactorily promote paricá seedlings (Schizolobium parahyba var. amazonicum) under high nitrogen rates. However, the Trichoderma harzianum IBLF 006 WP strain was efficient only under low nitrogen availability.

ABSTRACT

The objective of this work was to accomplish the isolation, molecular identification and characterizing the physiology of the causal agent of the algal spot in mango trees. For this purpose, the pathogen growth was assessed in different culture media, with subsequent observation and measurements of the filamentous cells. The molecular identification was made using mycelium obtained from leaf lesions and pure algae colonies grown in culture medium. Descriptions based on DNA sequencing indicated that the algae is Cephaleuros virescens. The algae must be isolated primarily in liquid medium for further pricking into agar medium. The highest mycelial growth average in Petri dishes occurred when the algae were grown in Trebouxia and BBM. Trebouxia enabled larger cells in the filamentous cells when compared to other culture media.

Brazil has many important native species, which includes Schizolobium parahyba var. amazonicum (Huber ex Ducke) Barneby, however, the growth of this species is inhibited in soils with low boron contents, or excess boron, which causes phytotoxicity. Applications of strains of Trichoderma spp. increase the plants' tolerance to abiotic stresses, including tolerance to low or excess levels of nutrients such as boron (B). Thus, the objective of this work was to evaluate plant development and the effect of strains of Trichoderma spp. in Schizolobium parahyba var. amazonicum seedlings grown under different boron rates. A randomized block experimental design was used, with a 4×5 factorial scheme, consisting of 4 strains of Trichoderma spp. and 5 B rates (0, 0.5, 1.0, 1.5, and 2.0 mg dm-3), with seven replications, in greenhouse. Plant height, stem diameter, fresh and dry mass, leaf, stem and total were evaluated at 120 days after emergence. The stains of Trichoderma spp. and B rates presented significant interaction for all evaluated variables, decreasing phytotoxicity. The strain IBLF006WP (T. harzianum) showed a higher capacity of increasing the plants' tolerance to boron, followed by URM5911 (T. asperellum). However, the beneficial effect of increasing this tolerance with the application of these strains is only feasible for soils with high contents of this micronutrient in the soil.

ABSTRACT

This study aimed to evaluate six cauliflower genotypes regarding their resistance to black rot and their production performance. To do so, it was conducted two field experiments in Ipameri, Goiás, Brazil, in 2012 and 2013. It was used a randomized block design, with four replications (total of 24 plots). Each plot consisted of three planting lines 2.5 m long (six plants/line), spaced 1.0 m apart, for a total area of 7.5 m(2). Evaluations of black rot severity were performed at 45 days after transplanting, this is, 75 days after sowing (DAS), and yield evaluations at 90 to 105 DAS. The Verona 184 genotype was the most resistant to black rot, showing 1.87 and 2.25% of leaf area covered by black rot symptom (LACBRS) in 2012 and 2013. However, it was not among the most productive materials. The yield of the genotypes varied between 15.14 and 25.83 t/ha in both years, Lisvera F1 (21.78 and 24.60 t/ha) and Cindy (19.95 and 23.56 t/ha) being the most productive. However, Lisvera F1 showed 6.37 and 9.37% of LACBRS and Cindy showed 14.25 and 14.87% of LACBRS in 2012 and 2013, being both considered as tolerant to black rot.

ABSTRACT: The deficient development of fertile seeds of native forest plant species in Brazil limits the reproduction of these plants in various conditions. Among the limiting biotic factors in quality and quantity of the forest seeds, borer insects are quite prominent, before and after their dispersion. This study reports for the first time a host of the buprestid beetle Lius conicus (Gory & Laporte, 1840). The larval development of L. conicus takes place in the seed capsules of Vochysia haenkeana Mart. (Vochysiaceae), a typical tree species in the Brazilian cerrado biome. In two regions of the cerrado in Goiás State, Brazil, almost ripe fruits of V. haenkeana were collected directly from the plants. After natural drying, and fruit and seed processing in laboratory, damage caused by the L. conicus larvae was quantified and qualified. Bigger fruits were preferred as hosts. Fruits developing on the eastern side of the plant were most frequently occupied by L. conicus. Seed lots of bigger fruits showed damage up to 37.5% from the infestation by L. conicus larvae. There was only one larva per fruit, which damaged all the seeds of the capsule (three or four) and generally consumed around 26% of the seed dry mass.


RESUMO: O deficiente desenvolvimento de sementes férteis de espécies florestais nativas no Brasil limita a propagação dessas plantas em diversas condições. Entre os fatores bióticos limitantes da qualidade e quantidade das sementes florestais, destacam-se os insetos broqueadores, antes e depois da sua dispersão. Este estudo relata pela primeira vez um hospedeiro do besouro buprestídeo Lius conicus (Gory & Laporte, 1840). O desenvolvimento larval de L. conicus ocorre em cápsulas com sementes de Vochysia haenkeana Mart. (Vochysiaceae), uma típica árvore do bioma cerrado brasileiro. Em duas regiões do cerrado, no Estado de Goiás, no Brasil, frutos quase maduros de V. haenkeana foram coletados diretamente das plantas. Após secagem natural e beneficiamento de frutos e sementes em laboratório, os danos causados pelas larvas de L. conicus foram quantificados e qualificados. Frutos maiores foram os preferidas como hospedeiros desse inseto. Os frutos produzidos pela planta, voltados para ponto o cardeal leste, foram os mais infestados por L. conicus. Lotes de sementes provenientes de frutos maiores apresentaram infestação por L. conicus de até 37,5%. Observou-se que houve apenas uma larva desse inseto por fruto, que danificou todas as sementes da cápsula (três ou quatro) e consumiu geralmente cerca de 26% da massa seca da semente.

BACKGROUND: Alternaria alternata causes the Alternaria brown spot disease (ABS) in many tangerines and their hybrids worldwide. Plant extracts offer an alternative method for controlling this disease, which control is based on chemical fungicides. AIMS: To identify plant species with antifungal properties against A. alternata, the causal agent of the ABS. METHODS: Plant extracts prepared from leaves, barks, flowers, and stalks collected from 105 plant species in the State of Minas Gerais, Brazil, were tested for activity against the fungus A. alternata in vitro and in vivo. RESULTS: The most promising extract was obtained from Anadenanthera colubrina, which reduced the disease on Murcott tangor fruits to levels obtained with commercial fungicides. Artemisia annua, Cariniana estrelensis, Ficus carica, and Ruta graveolens presented moderate in vitro antifungal activity, but no effects were observed on the disease when the extracts were applied to fruits inoculated with the fungus. Besides, A. colubrina was the most active extract against A. alternata in the in vitro assay. CONCLUSIONS: The results obtained in the in vitro and in vivo assays suggested that the fungal growth test, which uses 96-well polypropylene plates, seems to be appropriate for selecting potential plant species for testing new methods to control ABS.

ABSTRACT

Alternaria brown spot (ABS), caused by the fungus Alternaria alternata pathotype tangerine, is one of the main phytosanitary problems for mandarin growers. About 15 applications per year of harmful fungicides are required for controlling ABS disease in citrus orchards. As chalcones seem to be less toxic to humans and environment than the commercial fungicides in use, this study initially aimed at synthesizing 137 chalcones through aldolic condensations between benzaldehydes and acetophenones. The resulting chalcones were screened for activity against A. alternata through a fungal growth assay that was carried out in 96-cell polypropylene plates, using the same concentration to all studied substances. The four active chalcones underwent conidia germination and mycelial growth, which confirmed the antifungal activity of the compounds. These chalcones were then poured onto Murcott tangor fruit that had been inoculated with conidia of the fungus. All four chalcones reduced the ABS progress to values significantly smaller (P0.05) than that observed for the control. Statistical calculations showed that the best results were afforded by two compounds, bearing a 2,4,5-trimethoxyphenyl group at position 3 of prop-2-enal and a 3-nitro- or 3-hydroxyphenyl group at position 1 of the aldehyde. Such compounds reduced the incidence of the disease in Murcott tangor fruit to values that did not differ statistically from those obtained with a commercial fungicide.


A mancha marron de Alternaria (MMA), causada pelo fungo Alternaria alternata patótipo tangerina, é um dos maiores problemas fitossanitários dos produtores de tangerina. Aproximadamente 15 aplicações de fungicidas de elevada periculosidade, por ano, são necessárias para o controle de MMA em várias plantações de citros. Como as chalconas parecem menos tóxicas para os seres humanos e para o ambiente que os fungicidas comerciais atualmente em uso, este estudo teve como objetivo inicial sintetizar 137 chalconas através da condensação aldólica de benzaldeídos com acetofenonas. As atividades das chalconas resultantes contra A. alternata foram avaliadas através do emprego de teste de crescimento fúngico em placas de polipropileno com 96 cavidades, empregando a mesma concentração para todas as substâncias estudadas. As quatro chalconas mais ativas foram submetidas a testes de germinação de conídios e de crescimento micelial, que confirmaram as atividades antifúngicas dos compostos selecionados. Estes foram então, aplicados em frutos de tangor Murcote que tinham sido inoculados com conídios do fungo. Todas as quatro chalconas reduziram o progresso de MMA a valores significativamente inferiores (P0.05) ao observado para o controle. Cálculos estatísticos mostraram que os melhores resultados foram obtidos para dois compostos, que tinham um grupo 2,4,5-trimetoxifenil na posição 3 do prop-2-enal e um grupo 3-nitro- ou 3-hidroxifenil na posição 1 do aldeído. Tais compostos reduziram a incidência da doença em frutos de tangor Murcote a valores que não diferiam estatisticamente do obtido com um fungicida comercial.

ABSTRACT

This study aimed to characterize the genetic variability in commercial accessions of passion fruit from the Federal District, Brazil, by RAPD markers. Genetic analyses were done with leaf samples of 30 accessions. DNA samples were amplified by RAPD technique, and respective markers converted into a binary matrix, from which the genetic distances between the accessions were estimated. Clustering analyis based on genetic distances allowed to detect a wide range of genetic variabillity among the accessions of sour passion fruit, and to separate them from the two sweet passion fruit. The graphical positioning of 'BRS Ouro Vermelho' confirms its potential to improve the genetic variability of commercial varieties of sour passion fruit. Dispersal of genetic distances among commercial accessions of sour passion fruit supports evidence for different genetic origins of the materials planted in the Federal District. The verified genetic variability indicates the potential success of future breeding programs for this region.


Este estudo teve como objetivo caracterizar a variabilidade genética de acessos de maracujá comerciais no Distrito Federal por meio de marcadores RAPD. Análises genéticas foram feitas com amostras foliares de 30 acessos. As amostras de DNA foram amplificadas pela técnica de RAPD e os respectivos marcadores convertidos em uma matriz binária, a partir da qual as distâncias genéticas entre os acessos foram estimadas. Análises de agrupamento baseadas em distâncias genéticas permitiram detectar uma ampla gama de variabilidade genética entre os acessos de maracujazeiro-azedo, bem como para separá-los dos dois de maracujazeiro-doce. O posicionamento gráfico de 'BRS Ouro Vermelho' confirma a sua importante contribuição para aumentar a variabilidade genética das atuais variedades comerciais de maracujazeiro-azedo. A dispersão das distâncias genéticas entre os acessos comerciais de maracujazeiro-azedo suportam as evidências de diferentes origens genéticas para os materiais plantados no Distrito Federal. A variabilidade genética verificada evidencia o potencial de sucesso de futuros programas de melhoramento para essa região.

ABSTRACT

A Talisia subalbens (Mart.) Radlk. (Sapindaceae) é uma espécie arbustiva apontada como endêmica do Mato Grosso. Objetivou-se neste estudo avaliar os efeitos da temperatura e condições de secagem sobre a germinação de sementes de T. subalbens. No primeiro experimento avaliou-se a germinação das sementes em diferentes regimes térmicos: 15ºC-25ºC e 25ºC-35ºC (alternadas), 25ºC e 35ºC (constante), e no segundo experimento, as sementes foram submetidas à diferentes condições de secagem: rápida (estufa à 35ºC) e lenta (temperatura ambiente à 28±2ºC). Os testes de germinação foram realizados em câmaras do tipo B.O.D., sob fotoperíodo de 12 horas, utilizando-se vermiculita como substrato. O delineamento foi o inteiramente casualizado, com quatro repetições de 25 sementes cada amostra. Os valores médios de porcentagem de germinação e índice de velocidade de emergência (IVE) foram comparados pelo teste de Tukey (5%). As sementes germinadas a 25ºC, 35ºC e 25ºC-35ºC apresentaram maiores porcentagens de germinação, não diferindo entre si, contudo, maior vigor foi observado à 35ºC, sendo esta considerada a temperatura ótima de germinação da espécie. Os menores valores de germinação e IVE ocorreram a 15ºC-25ºC. O grau de umidade inicial das sementes foi de 48%. A velocidade de secagem das sementes afeta negativamente a porcentagem de germinação, sendo o teor crítico de grau de umidade de 25%, e a secagem rápida mais favorável para a espécie.


Talisia subalbens (Mart.) Radlk. (Sapindaceae), a shrubby species, is considered endemic to Mato Grosso. The aim of this study was to evaluate the responses of its seeds to factors such as the optimum germination temperature and the effect of different drying conditions on the physiological quality of the seeds. In the first experiment, seed germination under different temperatures was evaluated: 15ºC-25ºC and 25ºC-35ºC (alternate), 25ºC and 35ºC (constant). In the second experiment, the seeds were submitted to different drying conditions: fast (oven at 35ºC) and slow (room temperature at 28±2ºC). The germination tests were performed in a B.O.D-type chamber, under a photoperiod of 12 hours, using vermiculite as the substrate. The experimental design was the completely randomized with four replications of 25 seeds for each sample, and the average values of germination at the different temperatures compared by the Tukey test (5%). Seeds germinated at 25ºC, 35ºC, and 25ºC-35ºC presented higher percentages of germination, not differing from each other. However, greater vigor was found at 35ºC, this being considered the optimum temperature. The lowest values of germination and germination index (GI) occurred at 15ºC-25ºC. The initial moisture of the seeds was 48%. The drying of the seeds negatively affected the germination, with the critic moisture content of 25%and the fast drying being more favorable to the germination.

ABSTRACT

A partir de 40 amostras de solo, provenientes dos estados do Rio Grande do Sul, Minas Gerais, São Paulo, Bahia, Goiás e Tocantins, uma coleção de 230 isolados monospóricos de Trichoderma spp. foi estabelecida, usando o meio seletivo TSM. Com o objetivo de selecionarem-se isolados com potencial para controle biológico de doenças, foram conduzidos testes de pareamento de culturas em meio BDA, a 20 ºC para Sclerotinia sclerotiorum e a 25 ºC para Fusarium solani f.sp. phaseoli. Antagonismo contra os dois patógenos foi observado em 10 por cento dos isolados. Avaliações ao microscópio eletrônico de varredura de sete isolados selecionados in vitro mostraram que nem todos promoveram o hiperparasitismo dos patógenos, sugerindo a existência de outros mecanismos de antagonismo, como antibiose ou competição.


From 40 soil samples collected in the Rio Grande do Sul, Minas Gerais, São Paulo, Bahia, Goiás and Tocantins states, Brazil, a collection of 230 monosporic isolates of Trichoderma spp. was established using TSM selective media. In order to select efficient isolates for biological control, dual culture tests were carried out on PDA media at 20 ºC for Sclerotinia sclerotiorum and at 25 ºC for Fusarium solani f.sp. phaseoli. From the whole collection, 50 isolates presented antagonism against F. solani and 111 isolates to S. sclerotiorum. The antagonism against both pathogens was found in only 10 percent of the isolates. Scanning electron microscopy assessments with seven in vitro selected isolates showed that not all promoted hyperparasitism on the pathogens, suggesting the existence of other mechanisms of antagonism, as antibiosis or competition.

Estudou-se a influência de extratos vegetais no crescimento, in vitro, de plântulas de Rosa x hybrida, objetivando o desenvolvimento de novos reguladores vegetais para a propagação dessa planta ornamental. Extratos metanólicos de Saintpaulia ionantha Wendl. (folhas), Hibiscus rosa-sinensis L. (folhas) e Bougainvillea spectabilis Wild. (folhas e flores) foram obtidos nas concentrações de 150 e 300 mg.L-1. Na ocasião de preparo do extrato todas as plantas se encontravam no período de floração. O experimento foi realizado em delineamento inteiramente casualizado com dez tratamentos e quatro repetições. Dois desses tratamentos foram utilizados como controle: meio de cultura com e sem BAP (6-benzilaminopurina). As variáveis analisadas foram: número e comprimento de brotos, número e comprimento de raízes, e número de folhas por broto. Quanto à variável número de brotos, todos os extratos testados se comportaram como o controle sem o BAP. No que diz respeito ao número e comprimento de raízes, apenas o extrato das folhas de H. rosa-sinensis na concentração de 300 mg.L-1 se mostrou diferente do controle sem o BAP, com valores estatisticamente iguais ao do controle com BAP. Excetuando-se o extrato de H. rosa-sinensis, todos reduziram o número de folhas por broto. Embora os efeitos observados não tenham sido tão pronunciados quanto os causados pelo BAP, os resultados indicam a presença de fitormônios nos extratos estudados.


This work aimed to study the influence of plant extracts on the in vitro growth of Rosa x hybrida plantlets, to contribute to the development of new plant growth regulators to propagate such ornamental plant. Methanolic extracts of Saintpaulia ionantha Wendl. (leaves), Hibiscus rosa-sinensis L. (leaves) and Bougainvillea spectabilis Wild. (leaves and flowers) were employed at 150 and 300 mg.L-1. Flowering plants were used to obtain the extracts. The experiment was carried out in a randomized design, with ten treatments and four repetitions. Culture medium with and without BAP (6-benzylaminopurine) were used as controls. The analyzed variables were shoot number and length, root number and length, and leaves per shoot. Regarding shoot number, all extracts were statistically identical to the control without BAP. When root number and length were taken into account, only the extract of H. rosa-sinensis leaves at 300 mg.L-1 differed from the control without BAP, with values statistically identical to the control with BAP. Except for the H. rosa-sinensis extract, all the others reduced the number of leaves per shoot. Although the observed effects were not as pronounced as those obtained with BAP, these results indicate the presence of plant growth regulators in the studied extracts.
