ABSTRACT

Several colubrid snakes produce venomous oral secretions. In this work, the venom collected from Venezuelan opisthoglyphous (rear-fanged) Philodryas olfersii snake was studied. Different proteins were present in its venom and they were characterized by 20% SDS-PAGE protein electrophoresis. The secretion exhibited proteolytic (gelatinase) activity, which was partially purified on a chromatography ionic exchange mono Q2 column. Additionally, the haemorrhagic activity of Philodryas olfersii venom on chicken embryos, mouse skin and peritoneum was demonstrated. Neurotoxic symptoms were demonstrated in mice inoculated with Philodryas olfersii venom. In conclusion, Philodryas olfersii venom showed proteolytic, haemorrhagic, and neurotoxic activities, thus increasing the interest in the high toxic action of Philodryas venom.

ABSTRACT

OBJECTIVE: The main goal was to explore the different toxin properties (proteolytic, hemorrhagic, and neurotoxic) of Leptodeira annulata ashmeadii Duvernoy's gland secretion (DGS). METHODS: To separate and characterize the different proteins present in L. annulata ashmeadii DGS, 20% sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) was run. To partially purify the proteolytic activity, Mono Q2 column ion-exchange chromatography was used. Hemorrhagic activity was analyzed in chicken embryos and mice (skin and peritoneum). Neurotoxic disorders were analyzed in mice inoculated with L. annulata ashmeadii DGS. RESULTS: The approximate relative mass of 21 protein bands was determined using the Multi-Analyst PC version 1.1 (Bio-Rad) program. L. annulata ashmeadii DGS proved to have proteolytic, hemorrhagic, and neurotoxic activities. CONCLUSIONS: Given the properties of the secretions of its Duvernoy's gland, L. annulata ashmeadii should be added to the list of venomous colubrids.

ABSTRACT

Várias serpentes da família Colubridae produzem secrecões orais venenosas. Neste trabalho, foi estudado o veneno coletado da presa posterior da serpente opistóglifa venezuelana Philodryas olfersii. Deferentes proteínas estavam presentes no veneno, sendo caracterizadas pela eletroforese de proteínas (SDS-PAGE) a 20%. A secrecão mostrou atividade proteolítica (gelatinase) a qual foi parcialmente purificada em uma coluna de intercâmbio iônico (mono Q2). Adicionalmente, a atividade hemorrágica do veneno de Philodryas olfersii foi demonstrada em embriões de galinha, pele e peritônio de rato. Os sintomas neurológicos foram demonstrados em camundongos inoculados com veneno de Philodryas olfersii. Em conclusão, o veneno da Philodryas olfersii mostrou atividade proteolítica, hemorrágica, e neurotóxica, assim aumentando o interesse na elevada acão tóxica do veneno da Philodryas olfersii.

ABSTRACT

Muchas serpientes Colubridae producen secreciones orales tóxicas. En este trabajo se ha estudiado el veneno de la secreción de la glándula de Duvernoy colectado de especies opistoglifas (colmillos posteriores) de Colubridae Venezolanas. Se corrieron electroforesis de poliacrilamida al 20 por ciento (SDS-PAGE), a fin de separar y caracterizar las proteínas presentes en la secreción de la glándula de Duvernoy de la falsa coral Erythrolamprus bizona. El veneno mostró actividad proteolítica (gelatinasa). Para purificar parcialmente esta última actividad, se empleó una columna Mono Q (Bio-Rad, USA) de cromatografía de intercambio iónico. Para probar la actividad hemorrágica, de esta secreción de Duvernoy, se utilizaron embriones de pollo, piel y peritoneo de ratones, donde se demostró fuerte actividad hemorrágica. Al observar los síntomas neurológicos en ratones, ocasionados por la secreción de Duvernoy de E. bizona, se pudo constatar la intensa actividad neurotóxica de este veneno. En conclusión, la secreción de la glándula de Duvernoy de E. bizona mostró tener actividades proteolíticas, neurotóxicas y hemorrágicas
