ABSTRACT
In previous studies it was found that the antimicrobial properties of pulcherrimin-producing Metschnikowia species are related to the formation of a red pigment-pulcherrimin and sequestration of free iron from their growth medium. For strains of Metschnikowia pulcherrima, M. sinensis, M. shaxiensis, and M. fructicola, at a high, ≈80 mg/kg, elemental Fe concentration in agar growth media we observed the essentially different (metal luster, non-glossy rust like, and colored) yeast biomass coatings. For the studied strains the optical and scanning electron microscopies showed the increased formation of chlamydospores that accumulate a red pigment-insoluble pulcherrimin rich in iron. The chlamydospore formation and decay depended on the iron concentration. In this study pulcherrimin in biomass of the selected Metschnikowia strains was detected by Mössbauer spectroscopy. At ≈80 mg/kg elemental Fe concentration the Mössbauer spectra of biomass of the studied strains were almost identical to these of purified pulcherrimin. Iron in pulcherrimin reached ≈1% of biomass by weight which is very high in comparison with elemental Fe percentage in growth medium and is not necessary for yeast growth. The pulcherrimin in biomass was also observed by Mössbauer spectroscopy at lower, ≈5 mg/kg, elemental Fe concentration. Through chemical binding of iron pulcherrimin sequestrates the soluble Fe in the growth media. However, at high Fe concentrations, the chemical and biochemical processes lead to the pulcherrimin accumulation in biomass chlamydospores. When soluble iron is sequestrated or removed from the growth media in this way, it becomes inaccessible for other microorganisms.

The pre-Alpine Marmarosh Massif is a tectonically complex unit of the crystalline basement within the Eastern Outer Carpathians. In the eastern (Ukrainian) segment of this massif, two units have been identified-the Bilyi Potok Nappe and the Dilove Nappe. Petrological investigations coupled with zircon U-Pb dating were performed on metavolcanic rocks (porphyroids) and their phyllite host rocks, sampled from three locations within the Dilove Nappe. The geochemical characteristics of the meta-rhyodacite porphyroids revealed a volcanic arc affinity of the protolith, with U-Pb zircon ages of 452.8 ± 1.5 Ma and 461.5 ± 1.6 Ma and zircon saturation temperatures in the range of 823-892 °C. The phyllite host rocks (meta-tuff) yield a U-Pb zircon maximum estimate for the eruption age at 584.7 ± 2.9 Ma. Peak amphibolite-facies metamorphism (M1) was estimated at the pressure of 600-900 MPa with a temperature range of 560-600 °C. Retrogression (M2), possibly related to Alpine nappe stacking and shearing, is assumed to have taken place at 200-250 MPa and 384-222 °C. The volcanic arc is interpreted as an early Caledonian arc that was subsequently accreted to the margin of Baltica during the closure of the easternmost Tornquist Ocean rather than Cenerian (early Paleozoic) orogenic events on the margin of East Gondwana. Cu accumulation in the internodal cell of charophyte Nitellopsis obtusa or its compartments was investigated after 3-h-exposure to lethal effective concentrations (8-day LC50) of CuO nanoparticle (nCuO) suspension or CuSO4 solution, i.e. 100 mg/L nCuO or 3.18 mg Cu/L as CuSO4. In both cases, the major part of Cu accumulated in the cell walls. The presence of CuO NPs in the cell wall and within the cell was visualized by scanning electron microscope images as well as confirmed by energy dispersive X-ray spectrum data. Although a threefold higher intracellular concentration of Cu was found after treatment with nCuO suspension, 3.18 mg Cu/L as CuSO4 induced fast and substantial depolarization of cell membrane potential contrary to that of 100 mg/L nCuO. A delayed effect of nCuO on the survival of the cells was also observed. This suggests that internally accumulated Cu was far less active and further supports the hypothesis of delayed toxicity of internalized nCuO NPs to charophyte cells.
