ABSTRACT

BACKGROUND: Grandchild caring has positive as well as negative impact on the grandparents' psychological well-being and the findings are varied by culture and country. METHODS: Present study was intended to understand the relationship between caring for grandchildren and psychological well-being of grandparents living in skipped (SGH) and multi-generational households (MGH) in Indian demographical context. The present research involved In-depth Interviews (IDI) focusing on grandparents above 60 and grandchildren below 18, where the elder played a crucial role in caregiving. The study area was Malda, a district of West Bengal in India. Purposively 24 IDIs were selected. Psychological well-being was measured using open-ended questions. Thematic and content analyses were adopted to understand the perspective of grandparents. RESULTS: Most of the grandparents from SGH reported depression word frequently, while grandparents from MGH reported happy. In the content analysis, grandparents from SGH expressed tension, mental turmoil, and worry about grandchild's future. On the contrary, grandparents from MGH expressed happy, companionship, and worry about grandchild's future. Further, full time caring, compulsive reason behind grandchild caring, and working status were linked with living in SGH and grandchild caring, which were in turn connected with deteriorate psychological health. However, in MGH, a different scenario was observed, most grandparents were partially and non-compulsively engaged in grandchild caring and had expressed positive mental health. CONCLUSIONS: The Findings provide an intervention implication, particularly in the context of India's ageing population and their well-being by acknowledging the influence of household structure, caring intensity, motive behind grandchild caring, and working status on their psychological health. Understanding the importance of these key factors may help the policy maker and the individual to incorporate the most effective intervention to achieve sustainable development goal 3 and healthy ageing.

Two monophyletic Daphnia species (Daphnia magna and D. similis) were exposed to a sub-lethal concentration of Pb (50 µg/L) for nine generations under two food regimes (usual and restricted) and analyzed for acetylcholinesterase (AChE) activity, first reproduction delay, lifespan, and net reproductive rate (R0) at the subcellular, individual, and population levels, respectively. In the sixth generation, Pb-acclimated neonates were moved to clean media for three more generations to check for recovery. The net reproductive rate (R0) of D. magna was not affected by Pb. However, Pb stimulated reproduction, reduced lifespan, and decreased AChE activity. First reproduction delay and lifespan did not improve during the recovery process, suggesting a possible genetic adaptation. Food restriction reduced R0, lifespan, delayed hatching, and increased AChE activity; the opposite outcomes were observed for D. similis. The full recovery shown by R0 suggests the physiological acclimation of D. similis. Under food restriction, the animals exhibited a reduction of R0 and lifespan, delayed first reproduction, and increased AChE activity; however, there was no effect of Pb. The recovery process under food restriction showed that D. similis might not cope with Pb exposure, indicating a failed recovery. Such outcomes indicate that one model species' sensitivity may not represent another's sensitivity.

ABSTRACT

AIM: Linking information on family members in the Danish Civil Registration System (CRS) with information in Danish national registers provides unique possibilities for research on familial aggregation of diseases, health patterns, social factors and demography. However, the CRS is limited in the number of generations that it can identify. To allow more complete familial linkages, we introduce the lite Danish Multi-Generation Register (lite MGR) and the future full Danish MGR that is currently being developed. METHODS: We generated the lite MGR by linking the current version of the CRS with historical versions stored by the Danish National Archives in the early 1970s, which contain familial links not saved in the current CRS. We describe and compare the completeness of familial links in the lite MGR and the current version of the CRS. We also describe planned procedures for generating the full MGR by linking the current CRS with scanned archived records from Parish Registers. RESULTS: Among people born in Denmark in 1960 or later, the current CRS contains information on both parents. However, it has limited parental information for people born earlier. Among the 732,232 people born in Denmark during 1950-1959, 444,084 (60.65%) had information on both parents in the CRS. In the lite MGR, it was 560,594 (76.56%). CONCLUSIONS: The lite MGR offers more complete information on familial relationships than the current CRS. The lite and full MGR will offer an infrastructure tying together existing research infrastructures, registers and biobanks, raising their joint research value to an unparalleled level.

BACKGROUND: The occurrence of misattributed paternity has consequences throughout society with implications ranging from inheritance and royal succession to transplantation. However, its frequency in Sweden is unknown. OBJECTIVE: To estimate the contemporary frequency of misattributed paternity in Sweden. METHODS: The study was based on nationwide ABO blood group data and a nationwide register of familial relationships in Sweden. These data were analysed using both a frequentist Poisson model and the Bayesian Gibbs model. The conduct of the study was approved by the regional ethics committee in Stockholm, Sweden (reference numbers 2018/167-31 and 2019-04656). RESULTS: Nearly two million mother-father-offspring family units were included. Overall, the frequency of misattributed paternity was estimated at 1.7% in both models. Misattributed paternity was more common among parents with low educational levels, and has decreased over time to a current 1%. CONCLUSIONS: The misattributed paternity rate is similar to the rates in other West European populations. Apart from widespread societal implications, studies on heritability may consider misattributed paternity as a minor source of error.

ABSTRACT

OBJECTIVES: Adverse childhood experiences (ACEs) can detrimentally impact perinatal mental health, birth outcomes, and parenting behaviors. Proximal psychosocial stressors also increase risks to perinatal health and wellbeing. Additional research on effective perinatal mental health programs is needed, especially for individuals and families with historical and concurrent adversity, and those with moderate to severe symptoms. METHODS: The Mother-Baby Day Hospital at Hennepin County Medical Center provides trauma-informed, multi-generation treatment for perinatal women. Data were collected from patients between January 2016 and September 2019. Self-reported depression, anxiety, and maternal functioning assessments were administered pre- and post-treatment. Patients completed the ACE questionnaire and indicators of current psychosocial stressors (i.e., food insecurity, housing insecurity, and social support) at intake. A series of bivariate tests and hierarchical regression models examined relationships among variables, including whether distal and proximal adversity predicted post-treatment symptoms. RESULTS: 159 Perinatal patients consented to research and completed the ACEs questionnaire at first admission. High proportions of patients reported 4+ ACEs and current psychosocial stressors. Effect sizes for associations between ACEs, psychosocial stressors, and self-report symptoms were small to moderate. Individuals with food or housing insecurity entered treatment with higher anxiety. In regression models, the most robust predictors of post-treatment symptoms were pre-treatment symptoms. Effects of ACEs on post-treatment depression and food insecurity on post-treatment maternal functioning approached the adjusted significance cut-off (p < .01). CONCLUSIONS FOR PRACTICE: Current psychosocial stressors and ACEs did not substantially limit post-treatment depression, anxiety, and maternal functioning outcomes. High prevalence of ACEs and psychosocial stressors highlight the need for trauma-informed, multi-generation treatments to improve maternal mental health and parenting capacity.

BACKGROUND AND AIMS: It is widely accepted that changes in the environment affect mean trait expression, but little is known about how the environment shapes intra-individual and intra-population variance. Theory suggests that intra-individual variance might be plastic and under natural selection, rather than reflecting developmental noise, but evidence for this hypothesis is scarce. Here, we experimentally tested whether differences in intrinsic environmental predictability affect intra-individual and intra-population variability of different reproductive traits, and whether intra-individual variability is under selection. METHODS: Under field conditions, we subjected Onobrychis viciifolia to more and less predictable precipitation over 4 generations and 4 years. We analysed effects on the coefficient of intra-individual variation (CVi-i) and the coefficient of intra-population variation (CVi-p), assessed whether the coefficients of intra-individual variation (CsVi-i) are under natural selection and tested for transgenerational responses (ancestor environmental effects on offspring). KEY RESULTS: Less predictable precipitation led to higher CsVi-i and CsVi-p, consistent with plastic responses. The CsVi-i of all studied traits were under consistent stabilizing selection, and precipitation predictability affected the strength of selection and the location of the optimal CVi-i of a single trait. All CsVi-i differed from the optimal CVi-i and the maternal and offspring CsVi-i were positively correlated, showing that there was scope for change. Nevertheless, no consistent transgenerational effects were found in any of the three descendant generations, which contrasts with recent studies that detected rapid transgenerational responses in the trait means of different plant species. This suggests that changes in intra-individual variability take longer to evolve than changes in trait means, which may explain why high intra-individual variability is maintained, despite the stabilizing selection. CONCLUSIONS: The results indicate that plastic changes of intra-individual variability are an important determinant of whether plants will be able to cope with changes in environmental predictability induced by the currently observed climatic change.

Lead (Pb) stress affects hormone-mediated responses (e.g., reproduction) in insects. In this study, the effects of Pb stress (12.5-50 mg Pb/kg in larval artificial diets) on the reproduction of the common cutworm Spodoptera litura (Lepidoptera: Noctuidae) were investigated after 7 generations. The results showed that Pb stress did not reduce the longevity of adult females, but 50 mg Pb/kg significantly reduced the longevity of adult males, regardless of the generation. After 50 mg Pb/kg stress for one or 7 generations, the peak time of egg-laying was delayed, and egg production and hatchability were decreased significantly. The vitellin content in eggs was significantly inhibited by Pb stress. The S. litura vitellogenin (Vg) gene promoter was cloned and analyzed. Multiple putative transcription factors were predicted for the 2321 bp Vg promoter region, including the TATA box, GATA, basic helix-loop-helix (bHLH) transcription factor, Broad-Complex (BR-C) binding sites, etc. The fragment from -2222 to -211 bp of the Vg promoter was the activation domain for Vg, whereas the region from -211 to -55 bp repressed the activity of the Vg promoter. The construct promoter (-782/+76) in Trichoplusia ni (Hi5) cells significantly improved Vg expression, which was not affected by Pb stress (1 or 10 mg/ml). Therefore, Pb stress significantly inhibited the reproduction of S. litura but not by regulating the Vg promoter.

ABSTRACT

We evaluated the real effects of pollutants through a multi-generation study. We tested whether short-term exposure (48 h) of successive (first and second) generations of Chironomus yoshimatsui neonates (<24-h-old) to two acetylcholinesterase inhibitor insecticides, pyraclofos, and pirimicarb, would change insecticide sensitivity and life-cycle parameters over four generations. Additionally, we tested whether acetylcholinesterase (AChE) activity levels would be associated with this sensitivity change. Sensitivities (48 h EC50 value, using immobility as the endpoint) in chironomids (<24-h-old) and insect life-cycle parameters (the number of larvae per egg mass and adult size) were investigated. Parental chironomids produced larvae that were less sensitive than those in the control group following the two 48 h pirimicarb exposure events, whereas exposure to pyraclofos did not affect sensitivity. The AChE activity in larvae with low sensitivity to pirimicarb was significantly higher than that in the control. Thus, increased AChE activity might be associated with low sensitivity. The life-cycle parameters in chironomids recovered from the effects of pyraclofos and pirimicarb suggested they could adapt to the insecticides by changing biomass allocation. Our study suggested potential chemical risks of insecticide stress and how aquatic organisms adapt to it.

ABSTRACT

To better evaluate chemical damage in chronically contaminated habitats, a nine-generational exposure to Lead (Pb) was done with two monophyletic Daphnia species, from temperate (Daphnia magna) and tropical (Daphnia similis) environments. The multi-generational test consisted generally of a continuous Pb exposed set of organisms, plus an extra control set running simultaneously. To assess daphnids recovery after Pb exposure, some organisms from the sixth generation were transferred to clean media for three extra generations (recovery period; F6 to F9), while others were keep Pb exposed. All setups (control, Pb exposure and recovery period) were submitted to two different dietary regimes, the standard (3 × 105 cells/mL) and restricted food (1.5 × 105 cells/mL) regimes. To evaluate the effects of generational Pb exposure and food regimes, individual, functional and population related endpoints were assessed (number of offspring, body length and rate of population increase (r) and feeding rate (FR)). The tests were conducted on the first (F0) and last generations (F9). No differences were shown on number of offspring and feeding among F9 control and continuous Pb exposed D. magna, although a higher r was shown for F9 Pb exposed organisms. F9 Pb exposed D. similis also presented a higher r than F9 control, however, lethality was induced at high Pb exposure levels. At food restriction the patterns were opposite and D. magna died at high Pb exposure while Pb exposed D. similis was the only setup (compared to control and recovery period) to survive at high Pb exposure levels. Regarding the recovery period, D. magna (standard food) did not cope well with the Pb re-exposure and lethality was induced, while D. similis indicate a decreased Pb sensitivity (only setup that survived high Pb exposure levels). Under food restriction, both species presented a decreased Pb sensitivity and consequent failed recovery (possibly due to epigenetic changes). Both species presented similar patterns regarding generations. Organisms from F0 presented enhanced reproductive outputs in comparison to F9 and the contrary occurred to the FR (even in control organisms). Data show an acclimation under a generational Pb exposure, which could increase the population of adapted organisms in natural habitats. And, since there was not a full recovery after three generations in clean media, an indication of epigenetic changes for both species may also be considered.

BACKGROUND: Findings from animal and human studies indicate that anxiety and stress have a negative influence on the child and mother. The aim of this study was to explore the risk for having an anxiety diagnosis and the impact of the diagnosis in a three generational perspective. METHODS: The information was retrieved from Swedish population-based registries. All women who gave birth between 1973 and 1977 (n 169,782), their daughters (n 244,152), and subsequently also the offspring of the daughters (n 381,953) were followed until 2013. RESULTS: We found that 4% of the mothers and 6% of the grandmothers had been diagnosed with anxiety. Women who had mothers with an anxiety disorder were more than twice as likely to have an anxiety disorder themselves compared to all other women (OR = 2.20, 95% CI = 2.04-2.30). In the third generation, the children born to mothers with an anxiety disorder, the odds ratio of being diagnosed with anxiety was more than twice as high than for the rest of the population (OR = 2.54, 95% CI = 2.01-3.20). If both the mother and the grandmother had had an anxiety disorder the odds ratio for the child having a diagnosis of anxiety was three times higher (OR = 3.11, 95% CI = 2.04-4.75). Anxiety diagnosis in the two previous generations also increased the likelihood of the child having either more than two inpatient visits or more than 10 outpatient visits (OR = 2.64, 95% CI = 2.40-2.91 and OR = 2.21, 95% CI = 2.01-2.43, respectively). CONCLUSIONS: The intergenerational effect on anxiety is high. In order to minimize the risk for further transmission of anxiety disorders, increased awareness and generous use of effective treatment regimes might be of importance.

ABSTRACT

Collembola are soil dwelling organisms that provide important ecosystem services within soils. To increase realism in evaluating potential effects of plant protection products a Collembola two-generation study was developed. This test assesses the potential for recovery of Collembola when exposed to plant protection products. Juvenile individuals of Folsomia candida (Willem, Ann Soc Entomol Belg 46:275-283, 1902) which hatched under conditions of exposure to a test substance in a modified OECD 232 bioassay were introduced into a second consecutive bioassay containing the same test substance aged in soil. This test system determines whether a population which was initially impacted by a substance in a 1st bioassay shows normal reproduction or survival in a 2nd bioassay after aging of the test substance in soil. An intermediate period for juvenile growth is included between the 1st and 2nd bioassay in order to reduce the control treatment variability in reproduction and mortality to fulfill the validity criteria according to the OECD 232 guideline. The Collembola two-generation study is able to differentiate between substances showing either a potential long-term risk or comprising a low risk. Comparing the results of this two generation study with data from semi-field or field studies indicates a high degree of conservatism when this test is considered within a tiered risk assessment scheme. This approach represents a valuable tool which makes the risk assessment more efficient by providing an alternative refinement option for highly conservative tier 1 Collembola risk assessment.

ABSTRACT

5-α reductase (5αR) inhibitors have an anti-androgenic effect in mammals because they inhibit the conversion of testosterone to the potent androgen, dihydrotestosterone. Finasteride is a type-2 5αR inhibitor that is used as a human pharmaceutical for the treatment of prostate cancer, benign prostate hyperplasia and male pattern baldness. This study evaluated the impacts of finasteride (50, 500 and 5000µg/L) on the development and reproduction of medaka (Oryzias latipes) exposed continuously over multiple generations (F0, F1 and F2). The exposure was initiated with reproductively mature fish (F0 generation) and continued until the hatching of the F2 generation. There were no significant effects on survival, fecundity or fertility in the F0 (50, 500, 5000µg/L) and F1 (50, 500µg/L) generations. The F1 generation exposed to 5000µg/L exhibited significant mortality. Histopathology of the gonads demonstrated that medaka and pre-clinical species respond similarly to finasteride exposure. Intersex condition and maldeveloped gonads were observed in F0 generation males exposed to 5000µg/L and F1 generation males exposed to 500µg/L. F1 generation males exposed to 500µg/L displayed reduced gonadosomatic index with an increased incidence of testicular degeneration. Males in both generations exhibited an increased incidence of Leydig cell hyperplasia at concentrations ⩾500µg/L. F0 generation females exposed to 5000µg/L exhibited increased gonadosomatic index. An increased prevalence of accelerated post-ovulatory follicle involution was observed in females at concentrations ⩾500µg/L in both generations. The gonadal changes induced by finasteride support the idea that 5-α reductase inhibition impacts androgen signaling in fish. Results from this study are discussed in the context of differential expression of the androgen receptor between species of fish.

BACKGROUND: Shifting demographic trends in the United States (US) have resulted in increasing numbers of three-generation family households, where a child lives with a parent(s) and grandparent(s). Although similar demographic trends have been occurring in the United Kingdom (UK) and Australia, very little research has studied three-generation coresidence in these countries and no research has documented trends cross-nationally. OBJECTIVE: We investigate differences in the rates of three-generation coresidence in early childhood cross-nationally. METHOD: This study uses three longitudinal birth cohort studies: the Early Childhood Longitudinal Study - Birth Cohort for the US, the Millennium Cohort Study for the UK, and the Longitudinal Study of Australian Children - Birth Cohort to investigate cross-national differences three-generation coresidence in early childhood. RESULTS: We find that nearly one quarter of US children live in a three-generation household during early childhood, compared with 8% of children in the UK and 11% in Australia. Although there are large differences in the frequency of coresidence cross-nationally, we find that similar demographic groups live in three-generation households across contexts. In general, younger, less educated, lower income and minority mothers are more likely to live in three-generation households in all three countries.

Despite encouraging multi-generational cohabitation, the population of Japanese people living alone has increased. However, little is known about the association between health and multigenerational cohabitation. This study examined the relationship between self-rated health and living arrangements among Japanese adults using data from the Japan Multi-Institutional Collaborative Cohort Study (2013-2017). The analysis employed multivariate logistic regression to examine the associations. Our results showed no association between living arrangements and self-rated health when stratified by gender. Living alone was found to be associated with poor self-rated health among women aged 65 and above. A similar association may exist among men in the same age group. Among women aged < 65 years, two-generation cohabitation was associated with a good self-rated health, similar to those living alone. Among men aged < 65 years, neither living alone nor two-generation cohabitation was significantly associated with good self-rated health. We found no association between three- or plus-generation cohabitation and self-rated health. Therefore, our findings indicate associations between multigenerational cohabitation and self-rated health, but they vary by gender and age. Invested stakeholders in the public health field should consider the potential impact of living arrangements on health based on gender and age.

ABSTRACT

In this article, the optimal design of a novel multi-generation system for the production of electricity, cooling, heat and freshwater is discussed. In this system, a Proton exchange membrane fuel cell (PEM FC) is used to generate electricity, and the heat produced by it is absorbed by the Ejector Refrigeration Cycle (ERC) and used to provide cooling and heating capacity. A reverse osmosis (RO) desalination system is also used to supply freshwater. The esign variables in this research are operating temperature and pressure and current density of FC, as well as the operating pressure of the HRVG, evaporator, and condenser of the ERC system. In order to optimize the considered system, the exergy efficiency and total cost rate (TCR) of the system are considered as optimization objective functions. To this end, the genetic algorithm (GA) is used and the Pareto front is extracted. Also, three refrigerants R134a, R600 and R123 areused as ERC system refrigerant and their performance are evaluated. Finally, the optimal design point is selected. At the mentioned point, the exergy efficiency is 70.2% and the TCR of the system is 1.78 S/h.

ABSTRACT

BACKGROUND: Sweden and the UK invite all 65-year-old men to a population-based ultrasound-based screening program to detect abdominal aortic aneurysms (AAA). First-degree relatives of patients with AAA are reported to have an increased risk to develop AAA, both women and men, but are not invited to screening. The "Detecting AAA in First Degree Relatives to AAA patients" (DAAAD) was designed to detect the true prevalence in adult offspring to AAA patients and to evaluate if national registries could be used for identification of index persons and their adult children with a high risk for the disease. The aim of this study is to summarize the design and methodology for this registry-based study. METHODS: The study is based on a registry-based extraction and identification of a risk group in the population with a subsequent identification of their adult offspring. The targeted risk group suffers a heredity for a potentially lethal disease, AAA (n = 750) and matched control group without heredity for AAA is also identified and invited (n = 750). The participation rate in the population-based AAA screening program for men is 75% regionally. This population is younger and have a lower prevalence. A participation rate of 65% is considered clinically adequate. For the DAAAD study, a stratified analysis of the primary outcome, prevalence, will be performed for women and men separately. Two other planned projects are based on the material: firstly, evaluation of the anxiety for disease and health-related quality of life (HRQoL) and, secondly, the cost-effectiveness of the study. DISCUSSION: In conclusion, this feasibility study will be instrumental in supporting the development of a possible new model to invite persons with high risk to develop hereditary rare diseases. To our knowledge, this is a unique, safe, and most likely to be a cost-efficient model to invite targeted risk groups for selected screening. If the study design and the results are shown to be cost-effective at the detected participation rate and prevalence, it should be further evaluated and adopted to a national screening program. The model also invites both women and men, which is unique for this specific patient group, considering that all population-based screening programs only include men. ABSTRACT

In recent decades, nanotechnology has rapidly developed. Therefore, there is growing concern about the potential environmental risks of nanoparticles (NPs). Caenorhabditis elegans (C. elegans) has been used as a powerful tool for studying the potential ecotoxicological impacts of nanomaterials from the whole animal level to single cell level, especially in the area of reproduction. In this review, we discuss the reproductive toxicity of common nanomaterials in C. elegans, such as metal-based nanomaterial (silver nanoparticles (NPs), gold NPs, zinc oxide NPs, copper oxide NPs), carbon-based nanomaterial (graphene oxide, multi-walled carbon nanotubes, fullerene nanoparticles), polymeric NPs, silica NPs, quantum dots, and the potential mechanisms involved. This insights into the toxic effects of existing nanomaterials on the human reproductive system. In addition, we summarize how the physicochemical properties (e.g., size, charge, surface modification, shape) of nanomaterials influence their reproductive toxicity. Overall, using C. elegans as a platform to develop rapid detection techniques and prediction methods for nanomaterial reproductive toxicity is expected to reduce the gap between biosafety evaluation of nanomaterials and their application.

ABSTRACT

Hydrogen energy, as clean and efficient energy, is considered significant support for the construction of a sustainable society in the face of global climate change and the looming energy revolution. Hydrogen is one of the most important chemical substances on earth and can be obtained through various techniques using renewable and nonrenewable energy sources. However, the necessity for a gradual transition to renewable energy sources significantly hampers efforts to identify and implement green hydrogen production paths. Therefore, this paper's objective is to provide a technological review of the systems of hydrogen production from solar and wind energy utilizing several types of water electrolyzers. The current paper starts with a short brief about the different production techniques. A detailed comparison between water electrolyzer types and a complete illustration of hydrogen production techniques using solar and wind are presented with examples, after which an economic assessment of green hydrogen production by comparing the costs of the discussed renewable sources with other production methods. Finally, the challenges that face the mentioned production methods are illuminated in the current review.

ABSTRACT

It was reported that tris(1,3-dichloro-2-propyl) phosphate (TDCIPP) could inhibit the growth of F0-generation fish. However, multi-generation effects of TDCIPP on survival and growth of fish remain unknown. In this study, the effects of TDCIPP on survival and growth in F1 generation zebrafish were evaluated after two-generation exposure. Results demonstrated that TDCIPP inhibited the survival and growth of F1-generation zebrafish at 96 hpf and 30 dpf. Moreover, compared with the F0 generation, two-generation exposure resulted in a greater accumulation of TDCIPP in F1 generation zebrafish, and strongly down-regulated the expression of genes related to the GH/IGF axis (gh, igf1, igf2b) and HPT axis (tshß). Taken together, for the first time, this study revealed that exposure to TDCIPP for two generations at environmentally relevant concentrations aggravated the adverse effects on growth and survival in zebrafish.

ABSTRACT

BACKGROUND: Essential tremor (ET), one of the most common neurological disorders, has a phenotypically heterogeneous presentation characterized by bilateral kinetic tremor of the arms and, in some patients, tremor involving other body regions (e.g., head, voice). Genetic studies suggest that ET is genetically heterogeneous. METHODS: We analyzed whole genome sequence data (WGS) generated on 104 multi-generational white families with European ancestry affected by ET. Genome-wide parametric linkage and association scans were analyzed using adjusted logistic regression models through the application of the Pseudomarker software. To investigate the additional contribution of rare variants in familial ET, we also performed an aggregate variant non-parametric linkage (NPL) analysis using the collapsed haplotype method implemented in CHP-NPL software. FINDINGS: Parametric linkage analysis of common variants identified several loci with significant evidence of linkage (HLOD ≥3.6). Among the gene regions within the strongest ET linkage peaks were BTC (4q13.3, HLOD=4.53), N6AMT1 (21q21.3, HLOD=4.31), PCDH9 (13q21.32, HLOD=4.21), EYA1 (8q13.3, HLOD=4.04), RBFOX1 (16p13.3, HLOD=4.02), MAPT (17q21.31, HLOD=3.99) and SCARB2 (4q21.1, HLOD=3.65). CHP-NPL analysis identified fifteen additional genes with evidence of significant linkage (LOD ≥3.8). These genes include TUBB2A, VPS33B, STEAP1B, SPINK5, ZRANB1, TBC1D3C, PDPR, NPY4R, ETS2, ZNF736, SPATA21, ARL17A, PZP, BLK and CCDC94. In one ET family contributing to the linkage peak on chromosome 16p13.3, we identified a likely pathogenic heterozygous canonical splice acceptor variant in exon 2 of RBFOX1 (ENST00000547372; c.4-2A>G), that co-segregated with the ET phenotype in the family. INTERPRETATION: Linkage and association analyses of WGS identified several novel ET candidate genes, which are implicated in four major pathways that include 1) the epidermal growth factor receptor-phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit alpha-AKT serine/threonine kinase 1 (EGFR-PI3K-AKT) and Mitogen-activated protein Kinase 1 (ERK) pathways, 2) Reactive oxygen species (ROS) and DNA repair, 3) gamma-aminobutyric acid-ergic (GABAergic) system and 4) RNA binding and regulation of RNA processes. Our study provides evidence for a possible overlap in the genetic architecture of ET, neurological disease, cancer and aging.
