RESUMO

Poyang Lake is the largest fresh water lake in China. In this study, the objective was to examine the diversity of bacterial community in this environment. The phylogenetic composition of bacterioplankton communities from two sites and two dates (northern and southern sub-basins in October 2006 and in May 2007, respectively) in the water column of Poyang Lake were investigated by partially sequencing cloned 16SrRNA genes. Moreover, restriction fragment length polymorphism (RFLP) was applied in the 16SrRNA gene clones. In total, four clone libraries were constructed and 347 clones were screened by RFLP, yielding 153 operational taxonomic units, which mainly belonged to the proteobacteria, Actinobacteria, Bacteroidetes, Cyanobacteria, Verrucomicrobia and Planctomycetes. Our results showed that Beta-proteobacteria was the most significant lineage, with dominant numbers of operational taxonomic units in the northern October 2006, southern October 2006 and May 2007 libraries. The highest bacterial diversity occurred in the library from the southern sub-basin in May 2007 and the lowest in the library from the northern sub-basin in May 2007. Horizontal and temporal differences associated with the concentration of total phosphorus, water temperature and pH suggested that the trophic state and the physicochemical properties of lake play key roles in sustaining bacterial community composition structure.

A water-soluble 2',4',6'-trihydroxy phenylazo luminol dye (THPL) with a D-π-A conjugated structure was developed for the first time for the dual-channel spectral sensing of a wide-range of pH values in aqueous solutions, using phloroglucinol as the electron-donating group, heterocycle amide luminol as the electron-accepting one and -NN- as the π-conjugated bridge. Its UV-vis absorption spectrum, especially its spectral response to pH, was investigated in detail. The results confirmed that the proposed THPL exhibited superior UV-vis spectral properties, a strong molar absorption coefficient (εmax = 3.35 × 104 L mol-1 cm-1) and a small half-band width of ca. 126 nm. In particular, either the absorption intensity (Abs) or peak (λmax) changed linearly with pH values from 3.0 to 10.0 under the optimized conditions. Notably, THPL expressed an exclusive dual-channel spectral response to pH, i.e., Abs (a.u.) = 0.1316 + 0.0278 pH (R2 = 0.9921), λmax = 441.5 - 6.64 pH (R2 = 0.9968, pH = 3.0-6.0) and λmax = 359.6 + 7.38 pH (R2 = 0.9879, pH = 6.0-10.0). Under the optimized conditions, THPL was applied to detect pH values in some aqueous and fruit juice samples with satisfactory results. The reversible recognition mechanism was deduced by UV-vis titration, 1H NMR titration and theoretical calculation.

Heisenberg's uncertainty relations have played an essential role in quantum physics since its very beginning. The uncertainty relations in the modern quantum formalism have become a fundamental limitation on the joint measurements of general quantum mechanical observables, going much beyond the original discussion of the trade-off between knowing a particle's position and momentum. Recently, the uncertainty relations have generated a considerable amount of lively debate as a result of the new inequalities proposed as extensions of the original uncertainty relations. We report an experimental test of one of the new Heisenberg's uncertainty relations using a single 40Ca+ ion trapped in a harmonic potential. By performing unitary operations under carrier transitions, we verify the uncertainty relation proposed by Busch, Lahti, and Werner (BLW) based on a general error-trade-off relation for joint measurements on two compatible observables. The positive operator-valued measure, required by the compatible observables, is constructed by single-qubit operations, and the lower bound of the uncertainty, as observed, is satisfied in a state-independent manner. Our results provide the first evidence confirming the BLW-formulated uncertainty at a single-spin level and will stimulate broad interests in various fields associated with quantum mechanics.
