Anthropogenic causes are overtaking natural factors to reshape patterns of biodiversity and ecosystem functioning. Mangrove reforestation aimed at reversing losses of mangroves has been conducted worldwide for several decades. However, how reforestation influences the link between ecological processes that shape community diversity and the consequent effects on ecosystem functions such as biomass production is less well known. Here we used data collected before and after mangrove planting to examine the effects of reforestation on molluscan species richness and biomass production by testing the changes in species richness, compositional similarities, distance-decay effects (community similarity decreases with increasing geographical distance) in metacommunity across a regional scale of 480 km (23-27 °N) in southeast Chinese coasts. Additionally, we further detected the impact of landscape configuration caused by different intensities of reforestation on the mollusc community. After the mangrove reforestation, mollusc species richness and biomass increased significantly. The increases in species richness and biomass of mollusc community were mediated by reducing distance-decay effect, indicating an increase in relationship strength between species richness and biomass might be associated with a decrease in distance-decay effect with rising mangrove habitat. We highlight the importance of considering the effects of anthropogenic changes on the relationship between biodiversity and ecosystem functioning. Quantifying the distance-decay effect of these influences enables management decisions about coastal restoration to be based upon ecological mechanisms rather than wishful thinking or superficial appearance.

Pulmonary fibrosis (PF) is a chronic interstitial lung disease with no effective therapies. Galectin-3 (Gal-3), a marker of oxidative stress, plays a key role in the pathogenesis of PF. Fibroblast-myofibroblast differentiation (FMD) is an important source of fibrotic cells in PF. Previous studies showed that melatonin (MT) exerted anti-fibrotic effect in many diseases including PF through its antioxidant activity. In the present study we investigated the relationships among Gal-3, NRF2, ROS in FMD and their regulation by MT. We established an in vitro model of FMD in TGF-ß1-treated human fetal lung fibroblast1 (HFL1) cells and a PF mouse model via bleomycin (BLM) intratracheal instillation. We found that Gal-3 expression was significantly increased both in vitro and in vivo. Knockdown of Gal-3 in HFL1 cells markedly attenuated TGF-ß1-induced FMD process and ROS accumulation. In TGF-ß1-treated HFL1 cells, pretreatment with NRF2-specific inhibitor ML385 (5 µM) significantly increased the levels of Gal-3, α-SMA and ROS, suggesting that the expression of Gal-3 was regulated by NRF2. Treatment with NRF2-activator MT (250 µM) blocked α-SMA and ROS accumulation accompanied by reduced Gal-3 expression. In BLM-induced PF model, administration of MT (5 mg·kg-1·d-1, ip for 14 or 28 days) significantly attenuated the progression of lung fibrosis through up-regulating NRF2 and down-regulating Gal-3 expression in lung tissues. These results suggest that Gal-3 regulates TGF-ß1-induced pro-fibrogenic responses and ROS production in FMD, and MT activates NRF2 to block FMD process by down-regulating Gal-3 expression. This study provides a useful clue for a clinical strategy to prevent PF. Graphic abstract of the mechanisms. MT attenuated BLM-induced PF via activating NRF2 and inhibiting Gal-3 expression.

BACKGROUND: Premenstrual dysphoric disorder (PMDD) is a mood disorder characterized by psychological and physical symptoms. Differences in white matter have been associated with affective and anxiety disorders, which share some symptoms with PMDD. However, whether white matter structure differs between the brains of individuals with PMDD and healthy controls is not known, nor is its relation to symptom severity. METHODS: We performed tract-based spatial statistics and voxel-based morphometry analyses of diffusion tensor imaging metrics and white matter volume, using 2 neuroimaging data sets (n = 67 and n = 131) and a combined whole-brain and region-of-interest approach. We performed correlation analyses to investigate the relationship between regions with different white matter microstructure and volume and PMDD symptom severity. RESULTS: We found greater fractional anisotropy in the left uncinate fasciculus (d = 0.69) in individuals with PMDD compared to controls. Moreover, the volume of the right uncinate fasciculus was higher in individuals with PMDD compared to controls (d = 0.40). As well, the severity of premenstrual depression was positively correlated with fractional anisotropy in the right superior longitudinal fasciculus (r = 0.35). LIMITATIONS: It is challenging to interpret group differences in diffusion tensor imaging metrics in terms of their underlying biophysical properties. The small size of the control group in the diffusion tensor imaging study may have prevented effects of interest from being detected. CONCLUSION: The findings of the present study provide evidence of differential cerebral white matter structure associated with PMDD and its symptoms.

In this study, the protein bioaccessibility of soymilk gels produced by the addition of glu-cono-δ-lactone (GDL) and fermentation with lactic acid bacteria (LAB) was examined using an in vitro gastrointestinal simulated digestion model. The in vitro protein digestibility, soluble protein content, free amino acids contents, degree of hydrolysis, electrophoretic patterns, and peptide content were measured. The results suggested that acid-induced soymilk gel generated by GDL (SG) showed considerably reduced in vitro protein digestibility of 75.33 ± 1.00% compared to the soymilk gel induced by LAB (SL) of 80.57 ± 1.53% (p < 0.05). During the gastric digestion stage, dramatically higher (p < 0.05) soluble protein contents were observed in the SG (4.79−5.05 mg/mL) than that of SL (4.31−4.35 mg/mL). However, during the later intestinal digestion phase, the results were the opposite. At the end of the gastrointestinal digestion phase, the content of small peptides was not significantly different (p > 0.05) between the SL (2.15 ± 0.03 mg/mL) and SG (2.17 ± 0.01 mg/mL), but SL showed higher content of free amino acids (20.637 g/L) than that of SG (19.851 g/L). In general, soymilk gel induced by LAB had a higher protein bioaccessibility than the soymilk gel coagulated by GDL.

Fibroblast-myofibroblast differentiation (FMD) is a critical cellular phenotype during the occurrence and deterioration of pulmonary fibrosis (PF). FMD can increase with an elevated level of reactive oxygen species (ROS) on fibroblasts under oxidative stress. Thioredoxin-interacting protein (TXNIP) is an α-arrestin family protein that regulates the level of intracellular ROS. Nuclear factor erythroid 2-related factor 2 (Nrf2) can protect against FMD in PF. However, the relationship between Nrf2 and TXNIP in FMD remains elusive. Therefore, we established TGF-ß1-induced FMD in vitro and bleomycin (BLM)-induced mouse PF model in vivo to explore whether the activation of Nrf2 can inhibit TXNIP-mediated FMD in PF. Dimethyl itaconate (DMI) was selected to activate Nrf2. Our results showed that TXNIP was elevated and FMD was aggravated in mice lung tissues after BLM administration compared with the saline group. Inversely, Nrf2 decreased TXNIP expression and alleviated FMD in PF. In vitro, TXNIP overexpression enhanced FMD and increased the level of ROS. In contrast, TXNIP deficiency by small interfering RNA (siRNA) attenuated TGF-ß1-induced FMD and reduced ROS. An increase in ROS by H2 O2 can upregulate TXNIP expression. Moreover, Nrf2 also inhibited TGF-ß1-induced FMD and the increase of ROS, with reducing expression of TXNIP, and the inhibitory effect was better than TXNIP siRNA. These results suggest that activation of Nrf2 by DMI can protect against PF via inhibiting TXNIP expression. Our study may provide new therapeutic targets and treatment approaches for PF.

Acetylcholine (ACh), the major neurotransmitter secreted by cholinergic neurons, is widely found in the peripheral and central nervous systems, and its main function is to complete the transmission of neural signals. When cholinergic neurons are impaired, the synthesis and decomposition of ACh are abnormal and the neural signalling transition is blocked. To some extent, the concentration changes of ACh reflects the occurrence and development of many kinds of nervous system diseases, such as Alzheimer's disease, Parkinson's disease, Myasthenia gravis and so on. Thus, researches of the physiological and pathological roles and the tracking of the concentration changes of ACh in vivo are significant to the prevention and treatment of these diseases. In the paper, the pathophysiological functions and the comprehensive research progress on detection methods of ACh are summarized. Specifically, the latest research and related applications of the optical and electrochemical biosensors are described, and the future development directions and challenges are prospected, which provides a reference for the detection and applications of ACh.

Pulmonary fibrosis (PF) is a progressive and devastating lung disease of unknown etiology, excessive fibroblast proliferation serves as a key event to promote PF. Transcription factor forkhead box M1 (FOXM1) is not only a well-known proto-oncogene, but also an essential driver of cell proliferation. Recently, 5'-AMP-activated protein kinase (AMPK) is reported to reduce the incidence of PF. However, it remains elusive whether have an underlying relationship between AMPK and FOXM1 in fibroblast proliferation-mediated PF. Here, the progression of lung fibroblast proliferation and the expression levels of AMPK and FOXM1 were observed by intratracheally instilled of bleomycin (BLM) and intraperitoneal injection of metformin in C57BL/6 J mice. Meanwhile, human fetal lung fibroblast1 (HFL1) cells were respectively treated with AMPK activator metformin or AMPK inhibitor Compound C, or FOXM1 depletion by transfected small interfering RNA (siRNA) to unveil roles of AMPK, FOXM1 and the link between them on platelet-derived growth factor (PDGF)-induced fibroblast proliferation. Our results demonstrated that AMPK activated by metformin could down-regulate FOXM1 and alleviate BLM-induced mouse PF model. In vitro, activation of AMPK attenuated PDGF-induced fibroblast proliferation accompanied by the down-regulation of FOXM1. In contrast, inhibition of AMPK enhanced PDGF-induced fibroblast proliferation along with activating FOXM1. These findings suggest that AMPK can ameliorate the progression of fibroblast proliferation during PF via suppressing the expression of FOXM1 and provide new insight into seek PF treatment approaches.

Inhibiting myocardial fibrosis can help prevent cardiovascular diseases, including heart failure. Magnolol (Mag), a natural component of Magnoliae officinalis, has been reported to inhibit fibrosis. However, the mechanism of Mag activity and its effects on myocardial fibrosis remain unclear. Here, we investigated the involvement of ALDH2, an endogenous protective agent against myocardial fibrosis, in the Mag-mediated inhibition of cardiac fibroblast proliferation and collagen synthesis. We found that Mag significantly inhibited cardiac fibroblast proliferation and collagen synthesis, based on the results of MTT, EdU and western blot assays. Moreover, molecular docking, molecular dynamics simulation and surface plasmon resonance (SPR) assays showed that Mag could bind directly and stably to ALDH2. Further analysis of the mechanism of these effects indicated that treatment with Mag dose-dependently enhanced ALDH2 activity without altering protein expression. Mag could enhance the activity of recombinant human ALDH2 proteins with a half-maximal effective concentration of 5.79 × 10-5 M. In addition, ALDH2 activation via Alda-1 inhibited cardiac fibroblast proliferation and collagen synthesis, while ALDH2 inhibition via daidzin partially blocked the suppressive effects of Mag. In summary, Mag may act as a natural ALDH2 agonist and inhibit cardiac fibroblast proliferation and collagen synthesis.

BACKGROUND: Desmoglein2 (DSG2) is a transmembrane protein that helps regulate intercellular connections and contributes to desmosome assembly. Desmosome are associated with cell adhesion junctions, which play an important role in cancer progression specially cancer cell migration and invasion. However, DSG2 expression in colon cancer (CC) and its association with CC patients' overall survival (OS) are still unclear. METHODS: We collected 587 CC samples, 41 colitis tissues and 114 pericarcinomatous tissues, as well as corresponding clinicopathological data about the patients who contributed them. All samples were tested immunohistochemically in tissue microarrays. Kaplan-Meier method was used for calculating patient survival. Univariate and multivariate analyses was used for investigating DGS2 link with CC patient's clinicopathological factors. Bioinformatics analysis was also used in study. RESULTS: The results showed that DSG2 expression was lower in CC tissues than in pericarcinomatous tissues (P < 0.001). DSG2 expression was associated with differentiation (P = 0.033), lymph node metastasis (P = 0.045), distant metastasis (P = 0.006) and AJCC stage (P < 0.001). Univariate analysis indicated that poor OS in patients with CC was associated with low DSG2 expression (P < 0.001), tumor size (P < 0.001), lymph node metastasis (P < 0.001), distant metastasis (P < 0.001), AJCC stage (P < 0.001) and venous invasion (P < 0.001). In multivariate analysis, low DSG2 expression (P < 0.001), distant metastasis (P < 0.001), AJCC stage (P = 0.002), venous invasion (P < 0.001) were independent prognostic factors for CC patients. Bioinformatics analysis indicated that low DSG2 expression affects protein activation, regulates the P53-related pathway in CC, and activates the EGFR pathway. CONCLUSIONS: The results suggest that low DSG2 expression is associated with poor survival for CC patients. DSG2 could be a prognostic biomarker for CC.

An excellent aptasensor for electrochemical detection of amyloid-ß oligomers (AßOs) at trace levels was fabricated based on a triple-helix aptamer switch (THAS) via target-triggered signal transduction DNA displacement events. Specifically, a single-stranded anti-AßO aptamer (Apt) carrying two symmetrical arm segments was first attached via Au-S binding to an Au electrode. Gold nanoparticle (GNP)-tagged signal transduction probes (GNP-STPs) were simultaneously hybridized with the two arm segments of the Apt, and a rigid THAS was formed on the Au electrode. Compared to the conventional hybrid, the number of GNPs on the Au electrode increased significantly with the THAS, effectively improving the stability of the Apt to avoid lodging. Trithiocyanuric acid (TA) was utilized to further gather the GNPs and form network-like TA/GNPs. As a result, the differential pulse voltammetry (DPV) response of GNPs was clearly enhanced. When AßOs were present, target-triggered signal transduction DNA displacement events were carried out from THAS via the reaction of the Apt with the AßOs, which caused the GNP-STP to dissociate from the Au electrode, and thus a significant reduction in the DPV response was observed. The assay was able to sensitively detect trace AßOs by monitoring the AßO-controlled DPV response change. It exhibited a wide linear range from 1 fM to 10 pM with a low detection limit of 0.5 fM, and was successfully employed for the determination of AßOs in 20 serum samples, with good recovery. Moreover, the developed assay can provide a sensitive and selective platform for many studies or investigations related to Alzheimer's disease (AD) monitoring and treatment.

BACKGROUND: Symptom phrase recognition is essential to improve the use of unstructured medical consultation corpora for the development of automated question answering systems. A majority of previous works typically require enough manually annotated training data or as complete a symptom dictionary as possible. However, when applied to real scenarios, they will face a dilemma due to the scarcity of the annotated textual resources and the diversity of the spoken language expressions. METHODS: In this paper, we propose a composition-driven method to recognize the symptom phrases from Chinese medical consultation corpora without any annotations. The basic idea is to directly learn models that capture the composition, i.e., the arrangement of the symptom components (semantic units of words). We introduce an automatic annotation strategy for the standard symptom phrases which are collected from multiple data sources. In particular, we combine the position information and the interaction scores between symptom components to characterize the symptom phrases. Equipped with such models, we are allowed to robustly extract symptom phrases that are not seen before. RESULTS: Without any manual annotations, our method achieves strong positive results on symptom phrase recognition tasks. Experiments also show that our method enjoys great potential with access to plenty of corpora. CONCLUSIONS: Compositionality offers a feasible solution for extracting information from unstructured free text with scarce labels.

We prepared Y2 Mg2 Al2 Si2 O12 :xTb3+ (x = 0.02, 0.04, 0.06, 0.08, 0.10, 0.12, 0.14 and 0.16) luminescent materials using a nodulizing procedure. The phase and luminescence properties of these materials were investigated. X-ray diffraction results demonstrated that Tb3+ ions doped into Y2 Mg2 Al2 Si2 O12 hosts successfully and the Y2 Mg2 Al2 Si2 O12 :xTb3+ materials showed a pure cubic phase. Y2 Mg2 Al2 Si2 O12 :xTb3+ materials had the characteristic Tb3+ emission bands derived from 5 D4 →7 F6 , 7 F5 , 7 F4 , and 7 F3 transitions when excited at 365 nm. The green emission band that derived from the 5 D4 →7 F5 transition was highest due to the high possibility of both electric-dipole and magnetic-dipole transitions. Emission spectra indicated that the critical concentration of Tb3+ in the Y2 Mg2 Al2 Si2 O12 host was 0.14. The concentration quenching of Y2 Mg2 Al2 Si2 O12 :Tb3+ was derived from a dipole-dipole interaction.

The aggregation of amyloid-ß oligomers (AßOs) with extremely strong neurotoxicity has been proved to be the main pathogenesis of Alzheimer's disease (AD). For sensitive quantification of AßOs, a switchable electrochemical aptasensor is proposed. Metal organic framework carrying Au nanoparticles (AuNPs@CuMOF) has been used to label signaling displaced-probe (SD), which formed triple helix switch (THS) by hybridizing with label-free anti-AßOs aptamer (Apt) on the electrodeposited palladium electrode (EPd). Thus, a relatively strong response of differential pulse voltammetry (DPV) was produced (switch on). With the specific binding between AßOs and Apt, the DPV response obviously decreased, owing to destroyed structure of THS and the separation of AuNPs@CuMOF/SD from the EPd (switch off). The mode of "switch on-off" can dramatically enhance the AßOs-dependent DPV intensity change. As a result, the switchable EA exhibited excellent selectivity and sensitivity with the linear range from 0.5 fM to 500 fM and the detection limit of 0.25 fM. When evaluating the AßOs of artificial cerebrospinal fluid (aCSF) samples, the switchable EA exhibited desirable feasibility, and the results are basically consistent with the enzyme linked immunosorbent assay (ELISA). The pollution control of local government to industrial enterprises is actually a long-term, complex and dynamic game process. This study develops an evolutionary game model to analyze the operation mechanism of local governments' different expenditure preferences on the production behavior of industrial polluting enterprises, so as to specify the behavioral characteristics and optimal strategy of local environmental governance. The results indicated that whether the relationship between local governments and polluting enterprises in environmental governance becomes cooperative or collusive depends on their game sequence and initial endowment. Under the condition of realizing a steady state of cooperation, polluting enterprises would advance faster toward clean production if local governments distribute more environmental expenditures on cost subsidies for enterprises to implement clean production. The findings of this study provide decision-making basis for local governments to control industrial pollution.

Construal level refers to the level of abstractness that events and objects are represented, with high construal level promoting relational processing, whereas low construal level, item-specific processing. Three experiments (total N = 432) were conducted to investigate how priming construal level affects encoding of Deese/Roediger-McDermott (DRM) lists (e.g., bed, dream … converged to a critical item sleep), and then modulates free recall and recognition memory performance. Before and during list encoding, participants were primed with high/low construal level by identifying global/local letter in composite letters in a Navon task, choosing category (e.g., body)/exemplar (e.g., liver) to common objects (e.g., internal organ) in a two-alternative-forced-choice task, or by narrating on a particular incident they had power over other people or someone else had power over them. Half of critical items (and yoked associates) were studied and half were not, so their hit rates, false alarm rates, and memory sensitivity were measured. Results showed the typical false memory effect and lower memory sensitivity for critical items (vs. yoked associates), but there was no significant effect of primed construal level on memory performance in all experiments, despite our sample sizes ensuring strong statistical power. Hence, construal-level priming does not modulate memory performance in the DRM task.

The present study examined the intergenerational factors in transmitting autobiographical memories from one generation to the next. Older adults from Beijing, China reported a collection of personally important autobiographical memories and their middle-aged children recalled important parental memories. The parent-child dyads independently recalled and provided ratings of mnemonic characteristics for the memories. Across generations, consensus memories, which refer to the memories that both parents and children considered as important in the parent's life, were characterised by the substantial material change that the events brought about in their lives. While parent-child interaction affected the number of events passed onto children, it only affected the number of script-divergent events that were not recorded in the life script of a culture, but not the number of script-consistent events. In addition, children whose parents were rusticated and relocated to rural areas during the Cultural Revolution remembered more historical memories than children whose parents were not rusticated. The findings shed light on the process that one generation gains the biographical knowledge and historical experience of a prior generation. Theoretical implications are discussed.

Jinlianhua Decoction (JD), composed of Flos Trollii, Herba Taraxaci, Folium Isatidis, Radix Puerariae Lobatae, and Folium Perillae in a ratio of 6:15:10:10:6, is a prescription for Fengwen which is a group of febrile diseases due to wind in Chinese medicine. It was originally used for the prevention and treatment of severe acute respiratory syndrome (SARS), and could also be used to treat influenza due to their common pathomechanism. To elucidate the unclear pharmacodynamic basis of JD, the LC-QExactive-MS system was used to qualitatively analyze its main components in this study. As a result, 89 compounds were identified and 24 important ones were selected thereby to further perform the simultaneous quantification in 8 batches of JD samples using LC-QTrap-MS with multiple reaction monitoring (MRM). Based on the qualitative and quantitative results in combination with the bioactivities reported, 16 compounds including orientin, 2″-O-ß-l-galactopyranosylorientin, puerarin, trollisin I, rosmarinic acid, 2″-O-(2'″-methylbutanoyl) isoswertisin, daidzin, scutellarin, 3'-methoxy puerarin, vitexin, 3'-hydroxy puerarin, 2″-O-(2'″-methylbutanoyl) vitexin, kaempferol, caffeic acid, 3,4-dimethoxybenzoic acid, and cynaroside were determined as the major components of JD. This study provides a useful combinational method for analyzing the major pharmacodynamic substances of JD and lays a foundation for the quality control research of the decoction.

Avulsion fracture of the ischial tuberosity (AFIT) is a rare adolescent sports injury. At present, there is no consensus on its therapeutic paradigm, but conservative treatment appears to be the predominate choice. Furthermore, the degree of fracture displacement (DFD) remains as an important factor in determining whether AFIT needs internal fixation. The aim of the present study was to review and update the injury mechanism, clinical manifestations, imaging examination, diagnosis and differential diagnosis, and treatment of AFIT. A literature search was performed on a variety of databases using text words, and the results were limited to the English language. This review provides an important reference for the diagnosis and treatment of AFIT. AFIT can be easily misdiagnosed. Therefore, a detailed medical history and imaging examination are crucial for a correct diagnosis and differential diagnosis. For the choice of treatment of AFIT, it is necessary to consider not only the size of the fracture and DFD, but also the long-term functional needs of the patient.

Life transitions like war, marriage, and immigration presumably organize autobiographical memory. Yet little is known about how the magnitude of a given transition affects this mnemonic impact. To examine this issue, we collected (a) word-cued events, (b) event-dating protocols, (c) personally important events, and (d) transitional impact scores of the Cultural Revolution (1966-1976) and important events from Chinese adults who had been adolescents during the revolution. There were three main findings. First, rusticated participants, who moved from cities to rural areas during the Cultural Revolution, dated autobiographical memories in relation to this collective transition more frequently than nonrusticated participants, with the former group reporting a greater material (but not psychological) change in their lives due to this collective transition than the latter group. Second, material change predicted the degree to which the self-nominated important events served as temporal landmarks in event dating. Third, we observed that the events that people typically considered important and those that typically served as temporal landmarks changed as a function of age but displayed the similar temporal distributions. We conclude by considering the theoretical implications of these findings.

The present study manipulated participants' narrative perspectives (1st-personal pronoun "I" and 3rd-personal pronoun "He/She") to vary their field and observer visual perspectives that they took to retrieve autobiographical events and examine how the shifts in narrative perspective could influence the self-rated emotional intensity of autobiographical memory. Results showed that when narrative perspectives effectively shifted participants' visual perspectives from field to observer, they felt attenuated emotional intensities of positive and negative autobiographical memories. However, this did not occur when narrative perspectives effectively shifted the visual perspectives from observer to field. Multiple mediator models further showed that the changes in psychological distance and imagery vividness (a distance-related construct) of autobiographical memory mediated the relationship between the narrative perspective shift from the 1st- to 3rd-person and the reduction in the intensities of negative and positive emotion. This provides support for the role of psychological distancing in reducing the emotional intensity of autobiographical memory.
