RESUMO

Discrete organometallic complexes with defined structures are proceeding rapidly in combating malignant tumors due to their multipronged treatment modalities. Many innovative superiorities, such as high antitumor activity, extremely low systemic toxicity, active targeting ability, and enhanced cellular uptake, make them more competent for clinical applications than individual precursors. In particular, coordination-induced regulation of luminescence and photophysical properties of organic light-emitting ligands has demonstrated significant potential in the timely evaluation of therapeutic efficacy by bioimaging and enabled synergistic photodynamic therapy (PDT) or photothermal therapy (PTT). This review highlights instructive examples of multimodal radiochemotherapy platforms for cancer ablation based on self-assembled metallacycles/metallacages, which would be classified by functions in a progressive manner. Finally, the essential demands and some plausible prospects in this field for cancer therapy are also presented.

Toad venom, a traditional Chinese medicine, exhibits remarkable medicinal properties of significant therapeutic value. The peptides present within toad venom possess a wide range of biological functions, yet the neuropeptide B (NPB) and it modification requires further exploration to comprehensively understand its mechanisms of action and potential applications. In this study, a fusion peptide, ANTP-BgNPB, was designed to possess better analgesic properties through the transdermal modification of BgNPB. After optimizing the conditions, the expression of ANTP-BgNPB was successfully induced. The molecular dynamics simulations suggested that the modified protein exhibited improved stability and receptor binding affinity compared to its unmodified form. The analysis of the active site of ANTP-BgNPB and the verification of mutants revealed that GLN3, SER38, and ARG42 were crucial for the protein's recognition and binding with G protein-coupled receptor 7 (GPR7). Moreover, experiments conducted on mice using the hot plate and acetic acid twist body models demonstrated that ANTP-BgNPB was effective in transdermal analgesia. These findings represent significant progress in the development of transdermal delivery medications and could have a significant impact on pain management.

Toad venom is a traditional Chinese medicine with high medicinal value. The existing quality evaluation standards of toad venom have obvious limitations because of the lack of research on proteins. Thus, it is necessary to screen suitable quality markers and establish appropriate quality evaluation methods for toad venom proteins to guarantee their safety and efficacy in clinical applications. SDS-PAGE, HPLC, and cytotoxicity assays were used to analyze differences in protein components of toad venom from different areas. Functional proteins were screened as potential quality markers by proteomic and bioinformatic analyses. The protein components and small molecular components of toad venom were not correlated in content. Additionally, the protein component had strong cytotoxicity. Proteomics analysis showed that 13 antimicrobial proteins, four anti-inflammatory and analgesic proteins, and 20 antitumor proteins were differentially expressed extracellular proteins. A candidate list of functional proteins was coded as potential quality markers. Moreover, Lysozyme C-1, which has antimicrobial activity, and Neuropeptide B (NPB), which has anti-inflammatory and analgesic activity, were identified as potential quality markers for toad venom proteins. Quality markers can be used as the basis of quality studies of toad venom proteins and help to construct and improve safe, scientific, and comprehensive quality evaluation methods.

The development of highly specific biomimetic recognition material is a challenge for rapid detection of harmful residues in foodstuff. In this study, a paper-based boronate affinity metal-organic framework/molecularly imprinted polymer microfluidic chip (FZS-BA@MIP) was constructed based on the in situ construction strategy, which was also designed as a highly specific biomimetic recognition module. Here, the homogeneous zeolitic imidazole framework-8 (ZIF-8) membrane served as a great scaffold and enrichment layer. Besides, the recognition layer of MIP was prepared based on a highly oriented boronate affinity surface imprinting strategy. With the aid of the liquid flow channel, the highly specific enrichment and visual detection for antibiotic residues like kanamycin in actual products were achieved on the paper chip module of an integrated lateral flow platform. The whole analysis process could be accomplished within 30 min. In brief, this study offered a new integrated biomimetic recognition platform for visually detecting harmful veterinary residues containing cis-diols, which demonstrated promising commercial value in point-of-care testing of foodborne hazardous compounds.

A three-dimensional structured porous graphene oxide-polyethylenimine bead (pGP) is synthesized for immobilizing gold nanoparticles and modifying glutathione molecules (denoted as pGP/AuG). The pGP/AuG has open pore structure, honeycomb-like channels, and excellent hydrophilicity. By taking advantages of the porous structure, abundant binding sites, and multivalent interactions between glycopeptides and both glutathione molecules and free amino groups, the pGP/AuG is adopted to the selective enrichment of N-linked glycopeptides with low limit of detection (2 fmol), high enrichment selectivity (1:500), binding capacity (333.3 mg/g), recovery yield (91.3 ± 2.1%), and repeatability (< 6.0% RSD) using matrix-assisted laser desorption/ionization time of flight mass spectrometry detection method. Furthermore, the practical applicability of pGP/AuG is evaluated, in which 209 N-glycosylated peptides corresponding to 128 N-glycosylated proteins are identified from 1 µL human serum in three independent analysis procedures, suggesting the great potential for application in glycoproteome fields.Graphical abstract Schematic presentation of preparation for porous graphene oxide-based hydrophilic beads (pGP/AuG) with honeycomb-like microstructure. Introduction: This trial was conducted to compare the effect of diets supplemented with plant essential oil (PEO) and coated plant essential oil (CEO) on growth performance, immunity, antioxidant activity, and fecal microbiota of weaned piglets. Methods: A total of 360 21-day-old weaned piglets were randomly allocated into three groups, namely, CON, PEO, and CEO (basal diets supplemented with 0, 500 mg/kg PEO, and 500 mg/kg CEO, respectively) for a 4-week feeding trial. Results and discussion: The results showed that dietary supplementation with CEO improved the average final weight and average daily gain, decreased the diarrhea rate, increased antioxidant enzyme activities, enhanced immunoglobulin concentrations, and decreased concentrations of pro-inflammatory cytokines in the serum of weaned piglets (p < 0.05). In addition, CEO addition increased the fecal concentrations of propionic acid and isovaleric acid of piglets (p < 0.05). Spearman correlation analysis showed that fecal microorganisms at the genus level were closely correlated with the volatile fatty acid concentrations. The present study indicated that PEO and CEO could improve growth performance, enhance immunity, and increase antioxidant capacity by modulating the microbial flora in weaned piglets. Moreover, CEO addition seemed to offer more positive results than of PEO addition.

The development of a green, safe, and accurate sample preparation method for the determination of trace metal elements in environmental samples is of great importance. Choline chloride-based deep eutectic solvents (DESs) were used to extract heavy metal elements from litterfall and the target analytes were measured using inductively coupled plasma optical emission spectrometry. Factors such as the type, ratio, dosage, and extraction time and temperature of the DESs were studied. A DES system based on choline chloride and maleic acid had the highest extraction efficiency of 98.5%, 88.4%, 90.2%, and 93.7% for Cd, Cu, Zn, and Fe. Under the optimized conditions, the limits of detection and limits of quantification were in the range of 0.04-0.70 and 0.13-2.30 mg kg-1. The repeatability (n = 3), estimated in terms of the relative standard deviation, ranged from 1.14% to 3.40%. The proposed method was validated for accuracy using GBW10087. Notably, the energy consumption of the newly developed method was only one-fifth that of a traditional acid digestion method. This work not only presents an environmentally friendly method for the determination of trace element concentrations in environmental samples but also deepens our understanding of DES systems.

The surface functional groups of hydrochar are crucial to its surface properties, and their contents are strongly positively correlated with the adsorption performance. In this study, acrylate-functionalized hydrochar (AHC) with varying contents of O-containing functional groups (OFGs) was synthesized via hydrothermal carbonization (HTC) of bamboo, acrylic acid and an initiator, and then deprotonated with NaOH. The AHCs were analyzed by various characterization techniques. During HTC, the higher amount of acrylic acid added led to higher carbon, oxygen and carboxyl contents, and to the larger specific surface area and pore volume of AHC. The adsorption kinetics, isotherms, thermodynamic, ionic strength and pH effects of Pb(II) on AHC were studied. Adsorption isotherms and kinetics obeyed Langmuir and pseudo-second-order models, respectively, indicating adsorption is monolayer chemical process. The adsorptive ability was well linearly related to the OFG contents of AHC. When acrylic acid was added to 25 mL during HTC, the adsorbing ability of AHC over Pb(II) reached 193.90 mg g-1. Hence, direct HTC of acrylic acid, biomass and an initiator can prepare hydrochar with controllable OFG contents, which is a prospective adsorbent for treating metal cations.

Soil is the basis of agricultural production and accessing accurate information on soil nutrients is essential. Traditional methods of soil composition detection, which are based on chemical analysis, are characterized by being costly and polluting. Spectroscopic analysis has proven to be a rapid, non-destructive and effective technique for predicting soil properties in general and potassium, phosphorus and organic matter in particular. However, previous research on soils has rarely combined optimization algorithms with machine learning techniques, which has led to suboptimal model accuracy and convergence speed. In this study, a total of 184 soil samples were collected from three cities of Linhai, Yueqing and Longyou County, Zhejiang Province, China. After measuring pH values, alkali-hydrolyzable nitrogen (SAN), available phosphorus (SAP), available potassium (SAK) and soil organic matter (SOM) contents, along with their corresponding spectroscopic measurements, nine pretreatment methods and their combinations are adopted. A novel assessment model, integrating support vector machine and dung beetle optimization algorithm (DBO-SVR), is proposed to predict pH values and SAN, SAP, SAK, SOM content. Meanwhile, the DBO algorithm is compared with three mainstream optimization algorithms (particle swarm optimization (PSO), whale optimization algorithm (WOA) and grey wolf optimizer (GWO)). Results showed that the DBO-SVR model was shown best performance with Rp, RMSEP and RPD of 0.9842, 0.1306, 5.6485 respectively for prediction of pH value, with Rp, RMSEP and RPD of 0.8802, 15.0574 mg/kg and 2.0508, respectively for assessment of SAN content, with Rp, RMSEP and RPD of 0.9790, 12.8298 mg/kg, and 4.5132, respectively for assessment of SAP content, with Rp, RMSEP and RPD of 0.8677, 22.5107 mg/kg, and 1.9546, respectively for assessment of SAK content, and with Rp, RMSEP and RPD of 0.9273, 2.6427g/kg , and 2.1821, respectively for assessment of SOM content. This study demonstrates that the combination of near-infrared (NIR) spectroscopy and the DBO-SVR algorithm is capable of predicting soil nutrient composition with greater accuracy and efficiency.

The adsorption behaviors of biochar are largely impacted by biomassfeedstock. In this study, two biochars were prepared from torrefaction of ammonium persulfate- and potassium persulfate-pretreated bamboo and then activated by cold alkali, which are named as ASBC and KSBC, respectively. The two biochars were characterized by different instruments, and their adsorption properties over cationic methylene blue (MB) were compared. The type of persulfates little affected the specific surface areas, but significantly impacted O (29.54 % vs. 35.113 %) and N (12.13 % vs. 3.74 %) contents, functional groups, and zeta potentials of biochars. MB adsorption onto ASBC/KSBC is a single-layer chemical endothermic process and ASBC/KSBC exhibit high adsorption capacity over MB (475/881 mg·g-1) at 303 K. Obviously, the sorption capacity of MB onto KSBC much surpasses that of MB onto ASBC. These results indicate biomass pre-treatment is a cheap and convenient method to prepare biochars with unique physicochemical and adsorptive properties.

antibiotic resistance and drug-resistant bacterial infections pose significant threats to public health. Antimicrobial peptides (AMPs) are a promising candidate for related-infection therapy, but their clinical application is limited by their high synthesis cost and susceptibility to protease degradation. To address these issues, cationic poly(α-amino acid)s based on lysine have been developed as synthetic mimics of AMPs. In this study, we introduce a new class of cationic AMP synthetic mimics based on functional poly(methionine)s. We synthesized a series of sulfonium cationic poly(d,l-methionine)s with varying chain lengths via a convenient polymerization on α-amino acid thiocarboxyanhydride (α-NTA) using tert-butyl-benzylamine as the initiator, followed by alkylation with iodomethane. Our optimal methionine polymer demonstrated potent and broad-spectrum antibacterial activity against antibiotic-resistant bacteria, as well as excellent biocompatibility with mammalian cells and rapid bactericidal performance. Our findings suggest that sulfonium poly(methionine)s have the potential to address the challenge of drug-resistant bacterial infections.

PEGylated black phosphorus nanosheets (PEG-BPNSs) have shown promising applications in biomedicine and potentially interact with the vasculature following iatrogenic exposures. Whether the exposure to PEG-BPNSs could induce toxic effects on endothelial cells that line the blood vessels remains largely unknown. Herein, we investigate the cellular response and transcriptional profiling of human umbilical vein endothelial cells (HUVECs) after the exposure to BPNSs and PEG-BPNSs. BPNSs and PEG-BPNSs induce cellular elongation and cause significant cytotoxicity to HUVECs at 0.8 µg/mL, with viabilities of 87.8% and 87.7% respectively. The transcriptome analysis indicates that BPNSs and PEG-BPNSs at 0.4 µg/mL cause marked alterations in the expression of genes associated with detection of stimulus, ion transmembrane transport and components of plasma membrane. BPNSs and PEG-BPNSs at 0.4 µg/mL decrease the transendothelial electrical resistance (TEER) across monolayers of HUVECs by 22.8% and 20.3% compared to the control, respectively. The disturbance of tight junctions (TJs) after 24 h exposure to 0.4 µg/mL BPNSs and PEG-BPNSs is indicated with the downregulated mRNA expression of zona occluden-1 (ZO-1) by respective 16.5% and 29.9%, which may be involved in the impairment of endothelial barrier integrity. Overall, the response of HUVECs to PEG-BPNSs and BPNSs has no statistical difference, suggesting that PEGylation does not attenuate the BPNSs-induced endothelial injury. This study demonstrates the detrimental effects of BPNSs and PEG-BPNSs on barrier integrity of HUVECs, contributing to our understanding on the potential toxicological mechanisms.

In this study, carbon dots (CDs), cellulose nanofibers (CNF) and essential oil nanoemulsion (EON) were extracted from the aril waste of Torreya grandis following nuts production. These three nanomaterials were formulated for the preparation of a composite film to be employed for postharvest tomato storage. Visual, microscopical and physicochemical properties of the prepared nanocomposite films were analyzed at different levels of CDs and CNF for optimization purposes. The UV absorption and antioxidant capacity of gelatin film with 10 % CDs (G/10CD) were enhanced compared with gelatin (G) film, concurrent with a reduction in water barrier capacity, water contact angle (WCA) and tensile strength (TS). Compared with G/10CD film, the WCA of G film after incorporation of 10 % CDs and 3 wt% CNF (G/10CD/3CNF) was significantly increased by 14.5°at 55 s. In contrast, TS increased by 1.26 MPa, as well as the significant enhancement in water barrier capacity. The above composite film mixed with EON (G/10CD/3CNF/EON) exerted further antimicrobial effects against Escherichia coli. G/10CD/3CNF/EON coating effectively extended tomato shelf life compared with the control group. Therefore, this new eco-friendly film presents several advantages of biodegradability, sustainability as well as multifunctional properties posing it as potential packaging material for food applications.

To explore the effect and its mechanism of alkali-assisted phosphorylation on the functional properties of Camellia Oleifera seeds cake glutelin (CSCG), CSCG was treated with different concentration of sodium trimetaphosphate (STMP, 1.0, 2.0, 3.0, 4.0, and 5%, w/v) in different pH environment (3.0, 5.0, 7.0, 9.0, and 11.0). The results showed that alkali assist improved the phosphorylation degree of CSCG, and the optimum pH value is 9.0. FT-IR and XPS confirmed the successful modification of phosphate groups on CSCG through covalent interaction. Alkali-assisted phosphorylation decreased the particle size and increased electronegativity of CSCG, as well as changed in its surface hydrophobicity, crystallinity, and intrinsic fluorescence. All these changes of protein structure triggered by alkali-assisted phosphorylation led to the improvement of water solubility, water/oil absorption capacity, emulsifying ability, foamability, and in vitro digestibility of CSCG. This work could provide a theoretical basis for industrial production of CSCG with excellent functional properties.

Improving nitrogen use efficiency (NUE) without compromising yield remains a crucial agroecological challenge in theory and practice. Some meta-analyses conducted in recent years investigated the impact of nitrogen (N) fertilizer on crop yield and gaseous emissions, but most are region-specific and focused on N sources and application methods. However, various factors affecting yield and N fertilizer efficiency in wheat crops on a global scale are not extensively studied, thus highlighting the need for a comprehensive meta-analysis. Using 109 peer-reviewed research studies (published between 2000 and 2022) from 156 experimental sites (covering 36.8, 38.6 and 24.6% of coarse, medium, and fine texture soils, respectively), we conducted a global meta-analysis to elucidate suitable N management practices and the key factors influencing N fertilization efficiency in wheat as a function of yield and recovery efficiency and also explained future perspectives for efficient N management in wheat crop. Overall, N fertilization had a significant impact on wheat yield. A curvilinear relationship was found between N rates and grain yield, whereas maximum yield improvement was illustrated at 150-300 kg N ha-1. In addition, N increased yield by 92.18% under direct soil incorporation, 87.55% under combined chemical and organic fertilizers application, and 72.86% under split application. Site-specific covariates (climatic conditions and soil properties) had a pronounced impact on N fertilization efficiency. A significantly higher yield response was observed in regions with MAP > 800 mm, and where MAT remained < 15 °C. Additionally, the highest yield response was observed with initial AN, AP and AK concentrations at < 20, < 10 and 100-150 mg kg-1, respectively, and yield response considerably declined with increasing these threshold values. Nevertheless, regression analysis revealed a declining trend in N recovery efficiency (REN) and the addition of N in already fertile soils may affect plant uptake and RE. Global REN in wheat remained at 49.78% and followed a negative trend with the further increase of N supply and improvement in soil properties. Finally, an advanced N management approach such as "root zone targeted fertilization" is suggested to reduce fertilizer application rate and save time and labor costs while achieving high yield and NUE.

Astringency removal is important for the quality of Torreya grandis nut and occurs after harvest. Here, we evaluated the effect of NaHCO3 treatment on astringency removal and compared the differential metabolites of the seed coat and kernel using a UHPLC QQQ-MS-based metabolomics approach. The result revealed the nut astringency was primarily enriched in the seed coat with more soluble tannins. The NaHCO3 treatment greatly shortened the de-astringency process, as indicated by a faster conversion of soluble tannins to insoluble tannins and more acetaldehyde production. Besides, a total of 293 metabolites, including 92 phenolic acids and 37 flavonoids, were tentatively characterized in the seed coat. A further comparative analysis of the metabolomics indicated epigallocatechin, gallocatechin, catechin, procyanidin B1, B2, B3 and C1 to be the major metabolites influenced by the NaHCO3 treatment. This study provides new insights regarding the metabolite differences of Torreya grandis nuts processed with different de-astringent treatments.

In this paper, Eu(n+), Sm3+ doped Fe3O4 nanoparticles were prepared via solvothermal method, in which Ferric chloride is used as the iron source, and anhydrous EuCl3, SmCl3 as doping source. Eu, Sm valence in doped Fe3O4 nanoparticles, and effects of Eu, Sm doping amount on their structure, morphology, magnetic properties and PL properties were discussed. The results show, the Eu ions had doped Fe3O4 nanoparticles in the mixed-valence state, when the Eu and Sm doping amount were increased, the doped Fe3O4 nanoparticles changed from hollow nanospheres into spherical particles, and finally changed into uniform cube-shaped particles with 13 nm in diameter. Moreover, the doping sites for doping ions in doped Fe3O4 nanoparticles were discussed from Rietveld analysis of XRD pattern of the doped Fe3O4 nanoparticles. And the changes of the magnetic and PL properties with the doping amount were further discussed. It was found that higher Sm(3+)-doping amount led to stronger magnetic dipole transitions, while the Eu(n+)-doping amount had little effect on the magnetic dipole transitions, thus resulting in different changes in their saturation magnetization with doping amount.

Background: Recent experimental studies have shown that vegetable oil supplementation ameliorates high-fat diet- (HFD-) induced hyperlipidemia and oxidative stress in mice via modulating hepatic lipid metabolism and the composition of the gut microbiota. The aim of this study was to investigate the efficacy of the Torreya grandis kernel oil (TKO) rich in unpolysaturated fatty acid against hyperlipidemia and gain a deep insight into its potential mechanisms. Methods: Normal mice were randomly divided into three groups: ND (normal diet), LO (normal diet supplement with 4% TKO), and HO (normal diet supplement with 8% TKO). Hyperlipidemia mice were randomly divided into two groups: HFN (normal diet) and HFO (normal diet supplement with 8% TKO). Blood biochemistry and histomorphology were observed; liver RNA-seq, metabolomics, and gut 16S rRNA were analyzed. Results: Continuous supplementation of TKO in normal mice significantly ameliorated serum total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), and free fatty acid (FFA) accumulation, decreased blood glucose and malondialdehyde (MDA), and enhanced superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) levels. According to GO and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis, most differentially expressed genes (DEGs) were significantly enriched in the biosynthesis of unsaturated fatty acid pathways, and significantly changed metabolites (SCMs) might be involved in the metabolism of lipids. High-dose TKO improved gut alpha diversity and beta diversity showing that the microbial community compositions of the five groups were different. Conclusion: Supplementation of TKO functions in the prevention of hyperlipidemia via regulating hepatic lipid metabolism and enhancing microbiota richness in normal mice. Our study is the first to reveal the mechanism of TKO regulating blood lipid levels by using multiomics and promote further studies on TKO for their biological activity.

The potential utilization value of Camellia seed cake was explored by extracting albumin (CSCA) to develop nanoparticles for lutein delivery. First, thermal property and amphiphilicity of CSCA were evaluated to guide nanoparticle preparation. Next, CSCA nanoparticles modified with chitosan (CS) were prepared through a thermally induced self-assembly method derived by electrostatic attraction and hydrophobic interaction. The optimized nanoparticles were prepared from CSCA:CS at a mass ratio of 2:1 with pH of 4.5, and an incubation temperature and time of 80 ℃ and 10 min, respectively. The nanoparticles had the highest effective loading capacity for lutein at 5.89 ± 0.78%, and the corresponding encapsulation efficiency was 43.82 ± 5.69%. The storage stability of lutein was improved by nanoparticle loading, and the bioaccessibility of lutein in simulated intestinal digestion increased from 26.8 ± 4.4% to 57.3 ± 9.6% after encapsulation into nanoparticles. These findings may facilitate the development of new and sustainable proteins from plant waste for delivery system applications.
