RESUMO

Novel Supramolecular fluorescence receptor derived from calix-system i.e. calix[4]resorcinarene bearing dansylchloride as fluorophore was designed and synthesized. The compound was purified by column chromatography and characterized by elemental analysis, NMR and Mass spectroscopy. Tetradansylated calix[4] resorcinarene (TDCR) shows a boat conformation with C(2)v symmetry. The complexation behaviour of metal cations [Ag(I), Cd(II), Co(II), Fe(III), Hg(II), Cu(II), Pb(II), Zn(II), U(VI) (1 × 10(-4) M)] with tetra dansylated calix[4]resorcinarene (1 × 10(-6) M) was studied by spectophotometry and spectrofluorometry. Red shift in the absorption spectra led us to conclude that there is strong complexation Fe(III), Co(II) and Cu(II) with TDCR. These metal cations also produce quenching with red shifts in the emission spectra. The maximum quenching in emission intensity was observed in the case of Fe(III) and its binding constant was also found to be significantly higher than that of Co(II) and Cu(II). Quantum yield of metal complexes of Fe(III) was found to be lower in comparison with Co(II) and Cu(II) complexes. Stern Volmer analysis indicates that the mechanism of fluorescence quenching is either purely dynamic, or purely static.

In the present study, we report the synthesis of aqueous stable gold nanoparticles by using calix[4]arene polyhydrazide (CPH) as both reducing and capping agents. The calix[4]arene polyhydrazide reduced gold nanoparticles (CPH-AuNps) were characterized by UV/Vis, particle size analyzer (PSA) and transmission electron mictroscopy (TEM). The records confirmed high stability of CPH-AuNps in aqueous solution over a long period of time and even at varied pH. Additionally, CPH-AuNps have been investigated for its application as "Turn Off" fluorescent sensor for Hg[II]. A concentration of Hg[II] in the limit of 10 nM to 10 microM can be detected based on fluorescence quenching of the CPH-AuNPs and it was also concluded from the spectroscopic data that CPH-AuNPs possess excellent selectivity to Hg[II] over several metal ions like Pb[II], Cu[II], Cd[II], Mn[II], Zn[II] and Ni[II].

Globally 3 billion people are consuming water with moderately high concentrations of fluoride and arsenic. The development of a simple point of care (PoC) device or home device for the detection of fluoride/arsenic ensures safety before consuming water. Till date, lateral flow assay (LFA) based PoC devices can detect nucleic acids, viruses and diseases. An aluminium complex of rhodamine B functionalized oxacalix[4]arene (L) was designed to execute the LFA-based PoC device. Initially, Al3+ and Fe3+ ions were involved in complexation with the rhodamine B functionalized oxacalix[4]arene (L), resulting C1 (L-Al3+) and C2 (L-Fe3+) complexes respectively. The receptor L, as well as the probes (C1, C2), were characterized thoroughly using mass spectroscopy, FTIR, NMR, and EA. C1 and C2 were further utilized as recyclable probes for the detection of aqueous fluoride (21 ppb) and arsenate (1.92 ppb) respectively. The computational calculation indicates that upon complexation, the spirolactam ring opening at the rhodamine B site leads to optoelectronic changes. The consistency of LFA-based portable sensing device has been tested with water samples, synthetic fluoride standards and dental care products like toothpaste and mouthwash with concentrations ≥ 3 ppm. Moreover, fixed cell imaging experiments were performed to ascertain the in-vitro sensing phenomena.

An oxacalix[4]arene-Ce(iii) complex viz. L-Ce(III) has been introduced for the selective detection of As(v) and Cr(vi) oxyanions in aqueous medium. The binding mode of L-Ce(III) + AsO43-/CrO42- was completely investigated with fluorometric titration, time resolve fluorescent decay and FTIR analyses. Photoinduced electron transfer (PET) and chelation-enhanced fluorescence (CHEF) play an important role in the sensing of these oxyanions. The characteristic fluorescence of the L-Ce(III) complex has been quenched by AsO43- and CrO42- through cascading the ligating sites. Cyclic voltammetry (CV) experiments with various scan rates suggest that the electrochemical processes on the electrodes were controlled by diffusion. Both the analytes exhibit a lower limit of detection (LOD) below their standard EPA permissible limits. Moreover, the probe successfully detects the oxyanions in environmental real samples with excellent recovery ranging from 97 to 101%.

RESUMO

Kikuchi-Fujimoto disease (KFD) or histiocytic necrotizing lymphadenitis is a rare, benign, self-limiting disease with unknown etiology characterized by regional lymphadenopathy. A 30-year-old female presented with fever, weakness, multiple joint pain, oral ulcers, erythematous facial rashes, hemorrhagic crusting on both lips, and cervical lymphadenopathy of 2-month duration. Clinically, the disease was mimicking systemic lupus erythematosus, but immunofluorescence was negative for it. Lymph node biopsy suggested a diagnosis of KFD.

RESUMO

Calixarenes are considered as third generation supramolecules with hollow cavity-like architecture whereas nanoparticles are small entities with dimensions in the nanoscale. Many exciting achievements are seen when the calix system merges with nanoparticles which produces many fascinating facets in all fields of contemporary chemistry. The properties of nanoparticles which are tuned by calixarenes find applications in sensing, catalysis, molecular recognition, etc. Here, we have reviewed the chemistry of calix-based nanoparticles, and emphasis is laid on the modified, reducing, templated and stabilizing roles of calixarenes. This review covers the research being carried out in the domain of calix protected metal nanoparticles during last 18 years under the canopy of important 109 references. This article contains 58 figures which include 81 easy to understand structures. Calix-protected nanoparticles have enthralled researchers in the field of nanoscience with a tremendous growth in its applications, which heralds much promise to become in future a separate area of research.

A new calix[4]resorcinarene bearing eight hydroxamic acid groups (C4RAHA) has been synthesized and characterized by FT-IR, 1H-NMR and elemental analysis. Its analytical properties were investigated, and showed high affinity and selectivity toward thorium(IV) in the presence of large quantities of associated metal ions. Thorium(IV) was extracted from an ethyl acetate solution of C4RAHA at pH 4.5. The lambdamax and molar absorptivity (epsilon) for thorium(IV) were 341 nm and 7120 l mol(-1) cm(-1). The complexation of thorium(IV) with C4RAHA has a 4:1 metal:ligand stoichiometry, which is relatively rare. The extracts were directly aspirated in the plasma for ICP-AES measurements for thorium(IV) in the presence of a diversified matrix. The system obeyed Beer's law over the range 0.1 - 6.5 microg ml(-1) of thorium(IV) with a Sandell sensitivity of 0.0325 microg cm(-2). The preconcentration factor and overall stability constant evaluated at 25 degrees C for thorium(IV) were 133 and 15.86, respectively. The complexation was characterized by a favorable enthalpy change. A liquid-membrane transport study of thorium(IV) was carried out from the source to the receiving phase under controlled conditions, and a mechanism of transport proposed. To check the validity of the proposed method, thorium(IV) was determined in monazite sand, rare earth sand and water samples.

Calix[4]resorcinarene polyhydrazide (CPH) protected water dispersible fluorescent silver nanaoparticles (AgNps) were prepared by one-pot method using water soluble CPH and AgNO3. (CPH) bearing hydrazide group on its periphery acts as a reducing agent and its web type of structure as a stabilizing agent for the formation of calix protected silver nanoparticles (CPH-AgNps). CPH-AgNps were found to be highly stable over 120 days at room temperature and at varied pH. CPH-AgNps were characterized by UV/Vis-spectroscopy, particle size analyzer (PSA), transmission electron microscopy (TEM) and Energy dispersive X-ray analysis (EDX). Duly characterized nanoparticles were explored for their application as sensitive and selective fluorescent chemosensors for various metal ions. It was found that nanoparticles were selective and sensitive only for Fe(3+) ions with the linear range of detection from 0.1 µM to 10 µM. CPH-AgNps were also found to exhibit good antimicrobial activity when compared with standard Chloramphenicol. The selectivity and antimicrobial activity of CPH-AgNps suggests its potential use as a sensor for Fe(III) ions in ecosystems prone to industrial pollution and as an antimicrobial agent in biological applications.

Water dispersible stable gold nanoparticles (AuNps) have been synthesized by using calix[4]pyrrole octa-hydrazide (CPOH) as a reducing as well as stabilizing agent. CPOH-AuNps have been characterized by surface plasmon resonance, particle size analyzer and transmission electron microscopy. CPOH-AuNps are water dispersible, highly stable for more than 150 days at neutral pH with a size of less than 10nm and zeta potential of 15±2 MeV. Ion sensing property of CPOH-AuNps has been investigated for various metal ions Pb(II), Cd(II), Mn(II), Fe(III), Ni(II), Zn(II), Hg(II), Co(II) and Cu(II) by colorimetry and spectrofluorimetry. Among all the metal ions investigated, only Co(II) ions gives sharp colour change from ruby red to blue and is easily detectable by naked-eye. CPOH-AuNps being fluorescent in nature also shows great sensitivity and selectivity for Co(II) ions. Co(II) ions can be selectively detected at very low concentration level of 1 nM in a facile way of fluorescence quenching.

Two novel azocalix[4]pyrrole Amberlite XAD-2 polymeric chelating resins were synthesized by covalently linking diazotized Amberlite XAD-2 with calix[4]pyrrole macrocycles. The chelating resins were used for extraction, preconcentration and sequential separation of metal ions such as Cu(II), Zn(II) and Cd(II) by column chromatography prior to their determination by UV/vis spectrophotometry or flame atomic absorption spectrophotometry (FAAS) or inductively coupled plasma atomic emission spectroscopy (ICP-AES). Various parameters such as effect of pH on absorption, concentration of eluting agents, flow rate, total sorption capacity, exchange kinetics, preconcentration factor, distribution coefficient, breakthrough capacity and resin stability, were optimized for effective separation and preconcentration. The resin showed good ability for the separation of metal ions from binary and ternary mixture on the basis of pH of absorption and concentration of eluting agents. The newly synthesized resins showed good potential for trace enrichment of Cu(II), Zn(II) and Cd(II) metal ions, especially for Cu(II), as compared to the earlier reported resins. The synthesized resins were recycled at least 8-10 times without much affecting column sorption capacity. The presented method was successfully applied for determination of Cu(II), Zn(II) and Cd(II) in natural and ground water samples.

Congenital melanocytic nevus needs to be excised for aesthetic reasons and concerns regarding its malignant potential. Many methods like surgical resection, dermabrasion, curettage, chemical peeling, laser resurfacing, etc., are available for treatment. We hereby report the efficacy of excision of nevi by serial excision.

RESUMO

Resurfacing of facial rhytides (periorbital crow-feet wrinkles) was performed in three cases by carbon dioxide laser (Sharplan((R)) 1030 machine). Good to excellent results were observed. However, erythema and postinflammatory pigmentation were important side effects.

A rapid, specific and sensitive LC-MS/MS assay using solid-phase extraction (SPE) for the determination of pravastatin, in human plasma is described. The plasma filtrate obtained after SPE, using a polymer base, a hydrophilic-lipophilic balance (HLB) cartridge, was submitted directly to short-column liquid chromatography-tandem mass spectrometric (LC-MS/MS) assay, with negligible matrix effect on the analysis. For validation of the method, the recovery of the free analytes was compared with that from an optimized extraction method, and the analyte stability was examined under conditions mimicking the sample storage, handling, and analysis procedures. The extraction procedure yielded extremely clean extracts with a recovery of 107.44 and 98.93% for pravastatin and IS, respectively. The intra-assay and inter-assay precisions for the samples at the LLOQ were 3.30 and 7.31% respectively. The calibration curves were linear for the dynamic range 0.5-200 ng/mL with correlation coefficient r > or = 0.9988. The intra- and inter-assay accuracy ranged from 95.87 to 112.40%. The method is simple and reliable with a total run time of 3 min. This novel validated method was applied to the pharmacokinetic (PK) study in human volunteers receiving a single oral dose of 40 mg immediate release (IR) formulation.

A solvent extraction separation of uranium, in the presence of thorium, cerium and lanthanides with a new calix[4]resorcinarene bearing eight hydroxamic acid groups (C4RAHA) is described. Quantitative extraction of uranium is possible in ethyl acetate solution of C4RAHA at pH 8.0. The lambda(max) and molar absorptivity (varepsilon) for uranium is 356nm and 8352Lmol(-1)cm(-1). The Binding ratio of uranium with C4RAHA as evaluated by Job's method is 4:1. The system obeys Beer's law over the range 0.075-6.0mugml(-1) of uranium with Sandell sensitivity 0.0284mugcm(-2). A preconcentration factor of 142 was achieved by directly aspirating the extract for GF-AAS measurements. The two-phase stability constant evaluated at 25 degrees C for uranium is 15.91. The complexation is characterized by favorable enthalpy and entropy changes. A liquid membrane transport study of uranium was carried out from source to the receiving phase under controlled conditions and a mechanism of transport is proposed. Uranium has been determined in standard and environmental samples.
