RESUMO

Defects in primary cilia lead to devastating disease because of their roles in sensation and developmental signaling but much is unknown about ciliary structure and mechanisms of their formation and maintenance. We used cryo-electron tomography to obtain 3D maps of the connecting cilium and adjacent cellular structures of a modified primary cilium, the rod outer segment, from wild-type and genetically defective mice. The results reveal the molecular architecture of the cilium and provide insights into protein functions. They suggest that the ciliary rootlet is involved in cellular transport and stabilizes the axoneme. A defect in the BBSome membrane coat caused defects in vesicle targeting near the base of the cilium. Loss of the proteins encoded by the Cngb1 gene disrupted links between the disk and plasma membranes. The structures of the outer segment membranes support a model for disk morphogenesis in which basal disks are enveloped by the plasma membrane.

While Slicer activity of Argonaute is central to RNAi, conserved roles of slicing in endogenous regulatory biology are less clear, especially in mammals. Biogenesis of erythroid Dicer-independent mir-451 involves Ago2 catalysis, but mir-451-KO mice do not phenocopy Ago2 catalytic-dead (Ago2-CD) mice, suggesting other needs for slicing. Here, we reveal mir-486 as another dominant erythroid miRNA with atypical biogenesis. While it is Dicer dependent, it requires slicing to eliminate its star strand. Thus, in Ago2-CD conditions, miR-486-5p is functionally inactive due to duplex arrest. Genome-wide analyses reveal miR-486 and miR-451 as the major slicing-dependent miRNAs in the hematopoietic system. Moreover, mir-486-KO mice exhibit erythroid defects, and double knockout of mir-486/451 phenocopies the cell-autonomous effects of Ago2-CD in the hematopoietic system. Finally, we observe that Ago2 is the dominant-expressed Argonaute in maturing erythroblasts, reflecting a specialized environment for processing slicing-dependent miRNAs. Overall, the mammalian hematopoietic system has evolved multiple conserved requirements for Slicer-dependent miRNA biogenesis.

A major feature of Saethre-Chotzen syndrome is coronal craniosynostosis, the fusion of the frontal and parietal bones at the coronal suture. It is caused by heterozygous loss-of-function mutations in either of the bHLH transcription factors TWIST1 and TCF12. Although compound heterozygous Tcf12; Twist1 mice display severe coronal synostosis, the individual role of Tcf12 had remained unexplored. Here, we show that Tcf12 controls several key processes in calvarial development, including the rate of frontal and parietal bone growth, and the boundary between sutural and osteogenic cells. Genetic analysis supports an embryonic requirement for Tcf12 in suture formation, as combined deletion of Tcf12 in embryonic neural crest and mesoderm, but not in postnatal suture mesenchyme, disrupts the coronal suture. We also detected asymmetric distribution of mesenchymal cells on opposing sides of the wild-type frontal and parietal bones, which prefigures later bone overlap at the sutures. In Tcf12 mutants, reduced asymmetry is associated with bones meeting end-on-end, possibly contributing to synostosis. Our results support embryonic requirements of Tcf12 in proper formation of the overlapping coronal suture.

Proper function of the vertebrate skeleton requires the development of distinct articulating embryonic cartilages. Irx transcription factors are arranged in co-regulated clusters that are expressed in the developing skeletons of the face and appendages. IrxB cluster genes are required for the separation of toes in mice and formation of the hyoid joint in zebrafish, yet whether Irx genes have broader roles in skeletal development remains unclear. Here, we perform a comprehensive loss-of-function analysis of all 11 Irx genes in zebrafish. We uncover conserved requirements for IrxB genes in formation of the fish and mouse scapula. In the face, we find a requirement for IrxAb genes and irx7 in formation of anterior neural crest precursors of the jaw, and for IrxBa genes in formation of endodermal pouches and gill cartilages. We also observe extensive joint loss and cartilage fusions in animals with combinatorial losses of Irx clusters, with in vivo imaging revealing that at least some of these fusions arise through inappropriate chondrogenesis. Our analysis reveals diverse roles for Irx genes in the formation and later segmentation of the facial skeleton.

Hallux amputations have long been performed for the definitive treatment of hallux osteomyelitis resulting from ulcerations. These amputations have been performed at various levels of the hallux. The aim of this study is to assess the long-term outcomes in patients with hallux amputations performed at these various levels and to determine whether there is an ideal anatomic level that would limit post-operative complications and need for revisional surgery. An Institutional Review Board (IRB)-approved retrospective chart review of 148 feet with hallux amputations performed at various levels from July 1, 2013 to July 16, 2020 at an academic medical center was conducted. A 2-year minimum follow up was required for inclusion in the study. Incidence of re-ulceration, need for further amputation, healing of index procedure, and revascularization status were evaluated. Statistical analysis utilizing chi square analysis was performed to calculate p-values where <.05 was statistically significant. In this retrospective study with a minimum of 2-year follow-up, there was a tendency for amputations performed at the level of the head of the proximal phalanx (21%) to have a lower rate of reulceration (24%) followed by amputations performed at the level of the metatarsophalangeal joint and interphalangeal joints (36%). However, neither proved to be statistically significant.

The Argentine shortfin squid, Illex argentinus, inhabits in the southwest Atlantic; it is a semelparous species which grows rapidly along its 1 year lifespan. The identification of its stocks is critical for sustainable fishery exploitation. Parasites have been used as biological indicators in a lower number of studies dealing with squids, therefore a validation of this methodology is necessary. The intra- and inter-cohort variability of parasite assemblages in the summer-spawning stock of I. argentinus was analysed to assess their value as indicators of stock structure. Four squid samples from the continental shelf of central Patagonia, corresponding to 3 consecutive cohorts, were examined for metazoan parasites. Results evidenced heterogeneity in terms of parasite assemblage composition and structure, dominated by short-lived gastrointestinal parasites, with a strong influence of host size, but no effect of squid sex. These changes are related to their recent habitats and diets, which change with ontogeny and migrations, clouding any interpretation of patterns when samples spatially or temporally separated are compared. Many squid species share these characteristics; therefore, it is recommended that the use of parasites as biological tags should be restricted to simultaneous sampling, while size or age must be considered for deriving proper conclusions.

A family of acylhydrazones have been prepared and characterized with the aim of investigating their potential as information storage systems. Their well-established synthetic methodologies allowed for the preparation of seven chemically stable acylhydrazones in excellent yields that have been photophysically and photochemically characterized. In addition, DFT and TD-DFT calculations have been performed to gain more insights into the structural, energetic and photophysical properties of the E/Z isomers. Our results reveal that E/Z configurational isomerization upon irradiation is highly dependent on the stabilization of the E or Z isomers due to the formation of intramolecular H bonds and the electronic/steric effects intrinsically related to their structures. In addition, Raman spectroscopy is also used to confirm the molecular structural changes after the formation of hydrogen bonds in the isomers.

In an urban wastewater treatment plant (WWTP), early knowledge of the pollutant load levels throughout the plant is key to optimize its processes and achieve better purification levels. Molecular spectrophotometry has begun to gain prominence in this wastewater characterization process, as it is a simple, fast, inexpensive and non-invasive technique. In this research work, different mathematical models based on genetic algorithms have been developed for the estimation of chemical oxygen demand (COD) and total suspended solids (TSS) from the spectral response of the samples, measured in the 380-700 nm range by means of a light-emitting diode (LED) spectrophotometer developed by the researchers. A field campaign was carried out in Mapocho-Trebal WWTP (Chile), where 550 samples were obtained in three different parts of the plant: at the inlet (raw wastewater), at the outlet (secondary treated wastewater) and at the outlet of the primary clarifier. A total of 18 estimation models have been calculated by mean of HeuristicLab software, which have presented a high accuracy, with a Pearson's coefficient between 80 and 90% in most cases. In order to achieve the most accurate models possible to characterize each part of the plant, specific models have also been developed, as well as combined models that are valid for all types of wastewater.

This work describes the synthesis and properties of a dicyanomethylene-substituted indolo[3,2-b]carbazole diradical ICz-CN. This quinoidal system dimerises almost completely to (ICz-CN)2 , which contains two long C(sp3 )-C(sp3 ) σ-bonds between the dicyanomethylene units. The minor open-shell ICz-CN component in the solid-state mixture was identified by EPR spectroscopy. Cyclic voltammetry and UV-visible spectroelectrochemical data, as well as comparison with reference monomer ICz-Br reveal that the nature of the one-electron oxidation of (ICz-CN)2 at ambient temperature and ICz-CN at elevated temperature is very similar in all these compounds due to the prevailing localization of their HOMO on the ICz backbone. The peculiar cathodic behaviour reflects the co-existence of (ICz-CN)2 and ICz-CN. The involvement of the dicyanomethylene groups stabilizes the close-lying LUMO and LUMO+1 of (ICz-CN)2 and especially ICz-CN compared to ICz-Br, resulting in a distinctive cathodic response at low overpotentials. Differently from neutral ICz-CN, its radical anion and dianion are remarkably stable under ambient conditions. The UV/Vis(-NIR) electronic transitions in parent (ICz-CN)2 and ICz-CN and their different redox forms have been assigned convincingly with the aid of TD-DFT calculations. The σ-bond in neutral (ICz-CN)2 is cleaved in solution and in the solid-state upon soft external stimuli (temperature, pressure), showing a strong chromism from light yellow to blue-green. Notably, in the solid state, the monomeric diradical species is predominantly formed under high hydrostatic pressure (>1 GPa).

The tear-producing lacrimal gland is a tubular organ that protects and lubricates the ocular surface. The lacrimal gland possesses many features that make it an excellent model in which to investigate tubulogenesis, but the cell types and lineage relationships that drive lacrimal gland formation are unclear. Using single-cell sequencing and other molecular tools, we reveal novel cell identities and epithelial lineage dynamics that underlie lacrimal gland development. We show that the lacrimal gland from its earliest developmental stages is composed of multiple subpopulations of immune, epithelial and mesenchymal cell lineages. The epithelial lineage exhibits the most substantial cellular changes, transitioning through a series of unique transcriptional states to become terminally differentiated acinar, ductal and myoepithelial cells. Furthermore, lineage tracing in postnatal and adult glands provides the first direct evidence of unipotent KRT5+ epithelial cells in the lacrimal gland. Finally, we show conservation of developmental markers between the developing mouse and human lacrimal gland, supporting the use of mice to understand human development. Together, our data reveal crucial features of lacrimal gland development that have broad implications for understanding epithelial organogenesis.

We report the synthesis and characterization of a novel series of push-pull chromophores bearing 1D linear and ß-branched thiophenes as π-conjugated spacers between a 2,2,4,7-tetramethyl-1,2,3,4-tetrahydroquinoline electron donor unit and dicyano- and tricyanovinylene electron acceptor groups. The effect of the introduction of ß-thiophenes on the linear and nonlinear (NLO) optical properties as well as electrochemical and thermal data is studied in detail by performing a comparative study between the branched and 1D linear systems. In addition, a parallel DFT computational study is used to evaluate structure-property relationships. The non-linear optical behavior of the molecules both in solution and in solid state as electro-optic (EO) films using a guest-host approach shows very promising performance for electro-optic applications with high molecular first hyperpolarizabilities (µß) of 4840 × 10-48 esu and electro-optic coefficients r33 reaching 650 pm V-1. One highlight is that the electro-optic films of the ß-branched chromophores are superior in terms of thermal stability in device operation as measured by a transmissive modified reflective Teng-Man method. This work provides guidelines for the design of improved electro-optic materials including ß-branched chromophores which could be useful for practical EO applications, where both enhanced ß and r33 values together with chemical and thermal stability are necessary.

Simple geometric shapes are associated with facial emotional expressions. According to previous research, a downward-pointing triangle conveys the threatening perception of an angry facial expression, and a circle conveys the pleasant perception of a happy facial expression. Some studies showed that downward-pointing triangles have the advantage to capture attention faster than circles. Other studies proposed that curvature enhances visual detection and guides attention. We tested a downward-pointing triangle and a circle as target stimuli for a speeded response task. The distractors were two stimuli that resulted from the mixture of both targets to control for low-level features' balanced presentation. We used 3 × 3, 4 × 4, and 5 × 5 matrices to test whether these shapes led attention to an efficient response. In Experiment 1, participants responded faster to the circle than to the downward-pointing triangle. They also responded slower to both targets as the number of distractors increased. In Experiment 2, we replicated the main findings of Experiment 1. Overall, the circle was detected faster than the downward-pointing triangle with small matrices, but this difference decreased as the matrix size increased. We suggest that circles capture attention faster because of the influence of low-level features, that is, curvature in this case.

During a parasitological survey of the olfactory sacs of 21 species of Rajiformes (Chondrichthyes) from the Southwestern Atlantic Ocean, copepods referable to Dendrapta Kabata (1964) (Siphonostomatoida: Lernaeopodidae) were found parasitizing the cuphead skate Bathyraja scaphiops (Rajiformes: Arhynchobatidae). Morphological analyses using both light and electron microscopy revealed that they belong to a new species. It can be easily distinguished from its congeners by the ratio between lengths of posterior process and trunk (1:0.8), the large to width ratio of trunk (1:0.7) and the armature of the antennule (1, 1, 5 + 1 aesthete). Dendrapta cameroni longiclavata is raised to full specific status, as Dendrapta longiclavata n. comb. Kabata & Gusev, 1966.

Local administrations are increasingly demanding real-time continuous monitoring of pollution in the sanitation system to improve and optimize its operation, to comply with EU environmental policies and to reach European Green Deal targets. The present work shows a full-scale Wastewater Treatment Plant field-sampling campaign to estimate COD, BOD5, TSS, P, TN and NO3-N in both influent and effluent, in the absence of pre-treatment or chemicals addition to the samples, resulting in a reduction of the duration and cost of analysis. Different regression models were developed to estimate the pollution load of sewage systems from the spectral response of wastewater samples measured at 380-700 nm through multivariate linear regressions and machine learning genetic algorithms. The tests carried out concluded that the models calculated by means of genetic algorithms can estimate the levels of five of the pollutants under study (COD, BOD5, TSS, TN and NO3-N), including both raw and treated wastewater, with an error rate below 4%. In the case of the multilinear regression models, these are limited to raw water and the estimate is limited to COD and TSS, with less than a 0.5% error rate.

Spectrophotometry has proven to be an effective non-invasive technique for the characterization of the pollution load of sewer systems, enabling compliance with new environmental protection regulations. This type of equipment has costs and an energy consumption which make it difficult to place it inside a sewer network for real-time and massive monitoring. These shortcomings are mainly due to the use of incandescent lamps to generate the working spectrum as they often require the use of optical elements, such as diffraction gratings, to work. The search for viable alternatives to incandescent lamps is key to the development of portable equipment that is cheaper and with a lower consumption that can be used in different points of the sewer network. This research work achieved the following results in terms of the measured samples: First, the development a calibration procedure that enables the use of RGB-LED technology as a viable alternative to incandescent lamps, within the range of 510 to 645 nm, with high accuracy. Secondly, demonstration of a simple method to model the transmittance value of a specific wavelength without the need for optical elements, achieving a cost-effective equipment. Thirdly, it provides a simple method to obtain the transmittance based on the combination of RGB colors. Finally its viability is demonstrated for the spectral analysis of wastewater.

A series of donor-acceptor-acceptor (D-A-A) and donor-acceptor-donor (D-A-D) systems based on a pyrimidine π-spacer with various substituents at the C-2 position has been successfully prepared. The synthesis involved site-selective palladium cross-coupling reactions of chloropyrimidines with triorganoindium reagents and proceed in good yields and with atom economy. 4-(N,N-Diphenylamino)phenyl was chosen as the donor group and thien-2-yl dicyanovinylene as the acceptor one. The optical, vibrational, electrochemical, and density functional theory (DFT) calculations of these molecular systems were analyzed, and experimental values show the important role of the substituents at the C-2 position of the pyrimidine with stronger electron accepting ability, absorption in a wide range of UV/vis, acceptable fluorescence lifetime, and effective intramolecular charge transfer (ICT) properties. The ICT was observed in both series by the bathochromic shift on increasing the polarity of the solvent. In addition, DFT calculations found lower lowest unoccupied molecular orbitals of D-A-A molecules that suggest good electron ejection and transportation, being good properties for their application in various organic optoelectronic devices.

The species of Anisakis constitute one of the most widespread groups of ascaridoid nematodes in the marine ecosystem. Three closely related taxa are recognised in the A. simplex (s. l.) complex, i.e. A. pegreffii, A. simplex (s. s.) and A. berlandi. They are distributed in populations of their intermediate/paratenic (fish and squids) and definitive (cetaceans) hosts. A panel of seven microsatellite loci (Anisl 05784, Anisl 08059, Anisl 00875, Anisl 07132, Anisl 00314, Anisl 10535 and Anisl 00185), were developed and validated on a total of N = 943 specimens of A. pegreffii and A. simplex (s. s.), collected in fish and cetacean hosts from allopatric areas within the range of distribution of these parasite species. In addition, the locus Anisl 7, previously detected in those Anisakis spp., was investigated. The parasites were first identified by sequence analysis of the EF1 α-1 nDNA. The panel of the microsatellites loci here developed have allowed to: (i) detect diagnostic microsatellite loci between the two species; (ii) identify specimens of the two species A. pegreffii, A. simplex (s. s.) in a multi-marker nuclear genotyping approach; (iii) discover two sex-linked loci in both Anisakis species and (iv) estimate levels of genetic differentiation at both the inter- and intra-specific level.

During an extensive research project involving 519 specimens of batoids, including 13 species of Rajiformes and Myliobatiformes (Chondrichthyes) from the Argentine Sea, three new species of Empruthotrema were found and are described using morphologic characteristics and two molecular markers: LSU rDNA and COI mtDNA. The new species can be distinguished from their congeners by the number and distribution of the marginal loculi, the length and morphology of male copulatory organ, and the presence of eyespots. Additionally, multivariate analysis identified the dimensions of the pharynx and ejaculatory bulb as diagnostic features. Host specificity and previous records of the genus in the region are discussed. This is the first description of new species in this genus for the Southwestern Atlantic Ocean, as well as for arhynchobatid hosts.

Corynosoma australe and C. cetaceum are the most frequently reported acanthocephalans in fish from the Argentine Sea, particularly in central and northern areas. Their definitive hosts are otariids and odontocete cetaceans, respectively. The low specificity of these larvae, in combination with high infective capability and long survival periods in fish, make them potentially good biological markers for stocks and other biological features of their fish hosts. In order to determine the distribution patterns of these species and their determining factors, a large dataset composed by newly collected fish samples, published and unpublished data from previous studies by the authors in the region were analysed in relation to host and environmental variables. The complete dataset comprised a total of 5084 fish, belonging to 29 species distributed in 21 families and 9 orders. Host size and trophic habits arose as the main determinants of abundance for both species of Corynosoma, showing higher abundances on larger fish and on higher trophic levels, as it is usual for trophically transmitted parasites. Biogeographic province and depth (indirectly representing the temperature of water) were the main drivers of the spatial distribution, displaying a latitudinal pattern associated to the temperature clines created by the interaction of Malvinas and Brazil currents, determining a decrease in abundance southwards and towards the deeper areas. No patterns were found regarding the distribution of definitive hosts. The knowledge of these distribution patterns of Corynosoma spp. in fish at regional scale, as well as of their causes, provides useful information to design management and conservation policies thus contributing to maintain the full and sustainable productivity of fisheries.

Three piranha species, Serrasalmus maculatus, S. marginatus and Pygocentrus nattereri, living sympatrically in the lower Paraná River (Argentina) were examined searching for nasal monogeneans to know its diversity and distribution. Four species of monogeneans belonging to Rhinoxenus were found parasitizing the nasal cavities. Two new species are described, and new morphological data of 2 previously described species is provided. Rhinoxenus argentinensis n. sp. is characterized by having the male copulatory organ (MCO) as a coiled tube with a reel shaped-base; an elongated accessory piece articulated to base of MCO; a sinistral vagina with a sclerotized vestibule, and a sclerotized cap of the ventral anchor laterally modified forming a triangular expansion. Rhinoxenus paranaensis n. sp. is characterized by having a MCO as a coiled tube with a reel-shaped base; an accessory piece with an elongate proximal portion, a dilated distal portion with digitiform projections articulated to base of MCO; a sinistral vagina with 5-6 cuticular spine-shaped processes, and a sclerotized cap of the ventral anchor modified laterally forming a bilobate expansion. Additionally, multivariate discriminant analyses showed significant morphometric differences in the ventral anchors among Rhinoxenus species parasitizing 'piranhas '.
