RESUMO

In order to explore the role of heat shock proteins (Hsps) during thermal stress and development in the predatory mite Neoseiulus cucumeris (Oudemans), we cloned and characterized five full-length Hsp genes. We investigated the expression levels of these genes by quantitative real-time PCR. The five genes characterized here were NcHsp90, NcHsp75, NcHsp70, NcHsp60, and NcHsp40. These Hsps showed high sequence conservation and had greatest identity with heat shock proteins of Metaseiulus occidentalis and other mite and insect species. All five NcHsp genes showed changes in their levels of expression during development. Higher levels of expression were observed in adult females than in adult males, but there were no significant changes between pre-oviposition and post-oviposition stages in the females. NcHsp90, NcHsp75, and NcHsp70 expression levels were up-regulated after a heat shock, and the increases in NcHsp75 and NcHsp70 expression levels were maintained for at least 3 h. Up-regulation of NcHsp60 and NcHsp40 was not detected after 1 h at a high temperature (35-45 °C); however, a significant down-regulation was observed after 3 h heat exposure at 35 °C and 3 h recovery at 25 °C. Cold shock treatment (-5 to 15 °C) for 1 h did not acute elicit changes in the expression levels of any of the genes. At 5 °C, the expression levels of NcHsp90 significantly increased after 6 or 24 h exposure compared to the levels after 1 h exposure. Thus, expression of Hsp genes in N. cucumeris reflected developmental changes, sexual difference, and variable induced response to thermal stress. Increased expression of Hsps might protect N. cucumeris individuals under extreme temperature conditions. Therefore, it may be possible to enhance the thermal tolerance of commercially available N. cucumeris using temperature acclimation. Treatment at 35 °C should be suitable for such acclimation.

Neoseiulus cucumeris (Oudemans) (Acari: Phytoseiidae) is one of the most widely used and important biological control agents for thrips and other small pests worldwide. In the present study, we cloned two cDNAs of vitellogenins (Vgs, NcVg1 and NcVg2) and analyzed the effect of food source on the expression of both Vgs and fecundity in female adults. NcVgs showed higher sequence similarity to Vgs from Parasitiformes. Both neighbor-joining and maximum likelihood methods for phylogenetic analysis of NcVgs yielded similar topologies and showed that the Parasitiformes except Haemaphysalis longicornis segregated into a single clade that was separated into two subclades including one of both Vgs from N. cucumeris. Both transcripts, NcVg1 and NcVg2 revealed similar trends during developmental periods and reached the maximum level at the pre-oviposition period. When fed with different food sources, both NcVg1 and NcVg2 of female adults demonstrated a significant difference (P < 0.05) during the pre-oviposition period. Meanwhile, a positive correlation between the expression of Vgs and fecundity was observed. Therefore, the nutrients provided by the food sources affected fecundity resulting in differential expression of Vgs. Vitellogenin expression can be used as a molecular marker of fecundity of N. cucumeris.

Although the release of egg parasitoids has proven to be an effective strategy for the control of the fall armyworm (FAW), a single egg parasitoid, however, has exposed some deficiencies in practice, and it is worthwhile to explore whether the combination of multiple parasitoid species released can be used to control FAW by adopting an inter- or intra-specific relationship. In this study, we released compound parasitoid balls of Te. remus and Tr. chilonis in maize fields to explore the effects of combinations in different proportions of these two egg parasitoids on the control of the FAW. The results showed that the release of compound parasitoid balls improved the control effect on the FAW compared to the release of only Te. Remus (100%) and only Tr. Chilonis (100%). The treatments released with compound parasitoid balls significantly increased the egg parasitism rate against the FAW, reduced its populations, and alleviated its damage to maize as compared to the treatment with only Tr. chilonis (100%) released, whereas there was no significant difference in the egg parasitism rate and FAW populations between the treatments with the release of only Te. remus (100%) and that of compound parasitoid balls. Among the compound parasitoid balls with different proportions of egg parasitoids, the treatment with 80% Te. remus and 20% Tr. chilonis and that with 20% Te. remus and 80% Tr. chilonis could still retain more than 50% of the control effect against the FAW after 15 days of release and given the higher cost of production of Te. Remus, our results suggested that the combination of 20% Te. Remus and 80% Tr. Chilonis is a more cost-efficient ratio for the control of the FAW. Our findings may provide a new perspective for the sustainable control of the FAW.

BACKGROUND: The litchi fruit borer Conopomorpha sinensis Bradley is a major destructive pest of litchi and longan plants in China, India and South East Asia. Given its strong olfactory-based oviposition behaviour, interfering with the chemical communication between this insect pest and its host plant may serve as a potential control strategy. However, the chemical compounds associated with its egg-laying behaviour remain poorly understood. RESULTS: In this study, we investigated the olfactory preference of female C. sinensis for oviposition on intact mature fruits of the Feizixiao (FZX) and Guiwei (GW) varieties. Results showed that female C. sinensis preferred to lay eggs on FZX compared with GW fruits, and this preference was olfactory-induced. In addition, we identified differences in the chemical composition of the volatile blend and proportions between FZX and GW fruits, with terpenes being the main volatile components contributing to this divergence. Compounds that induced electrophysiological activity in female borers were subsequently screened from FZX. d-Limonene exhibited the strongest oviposition attraction among four candidates. Furthermore, this compound served as a volatile olfactory cue for recognition and orientation in female C. sinensis. CONCLUSION: The results of this study provide a deeper understanding of the olfactory preferences of female C. sinensis for oviposition on specific litchi varieties. © 2024 Society of Chemical Industry.

BACKGROUND: Anastatus japonicus Ashmead, a biological control agent utilized in China to control fruit bugs and forest caterpillars, is efficiently raised on large Chinese oak silkworm (Antheraea pernyi) eggs. Here, we investigated the biological parameters of non-diapaused and diapaused Anastatus japonicus after long-term storage within eggs of the host, Antheraea pernyi, under laboratory conditions. RESULTS: Diapaused mature larvae of Anastatus japonicus were more cold-tolerant than non-diapaused mature larvae, as reflected by a lower supercoiling point, lower freezing point, and higher survival rate at cold temperatures. Diapause induction enhanced the lifespan, fecundity and oviposition period of Anastatus japonicus than non-diapaused Anastatus japonicus when refrigerated for 6 months. However, after 12 months of refrigeration, the fecundity and oviposition period of Anastatus japonicus were significantly reduced with and without diapause. No difference in the progeny sex ratio of Anastatus japonicus was observed between diapause-induction treatment and those of non-diapaused. With the extension of refrigeration period from 6 months to 12 months, the lifespan, fecundity and oviposition period of Anastatus japonicus which were treated with diapause induction showed a sharp decrease. No significantly difference in the lifespan, fecundity and oviposition period of Anastatus japonicus was observed between diapause-induction treatment and those of non-diapaused when refrigerated for 12 months. CONCLUSION: These results suggest that the induction of diapause is an applicable technique to achieve mass production of Anastatus japonicus in long-term storage using eggs of the factitious host Antheraea pernyi, without compromising the quality of the parasitoid. The refrigeration period of diapaused Anastatus japonicus should not exceed 6 months. © 2023 Society of Chemical Industry.

RT-qPCR remains a vital approach for molecular biology studies aimed at quantifying gene expression in a range of physiological or pathological settings. However, the use of appropriate reference genes is essential to attain meaningful RT-qPCR results. Anastatus japonicus Ashmead (Hymenoptera: Helicopteridae) is an important egg parasitoid wasp and natural enemy of fruit bugs and forest caterpillars. While recent transcriptomic studies have analyzed gene expression profiles in A. japonicus specimens, offering a robust foundation for functional research focused on this parasitoid, no validated A. japonicus reference genes have yet been established, hampering further research efforts. Accordingly, this study sought to address this issue by screening for the most stable internal reference genes in A. japonicus samples to permit reliable RT-qPCR analyses. The utility of eight candidate reference genes (ACTIN, TATA, GAPDH, TUB, RPL13, RPS6, EF1α, RPS3a) was assessed under four different conditions by comparing developmental stages (larvae, pupae, adults), tissues (abdomen, chest, head), sex (male or female adults), or diapause states (diapause induction for 25, 35, 45, or 55 days, or diapause termination). RefFinder was used to calculate gene stability based on the integration of four algorithms (BestKeeper, Normfinder, geNorm, and ΔCt method) to determine the optimal RT-qPCR reference gene. Based on this approach, RPS6 and RPL13 were found to be the most reliable reference genes when assessing different stages of development, while ACTIN and EF1α were optimal when comparing adults of different sexes, RPL13 and EF1α were optimal when analyzing different tissues, and TATA and ACTIN were optimal for different diapause states. These results provide a valuable foundation for future RT-qPCR analyses of A. japonicus gene expression and function under a range of experimental conditions.

BACKGROUND: Anastatus japonicus Ashmead (Hymenoptera: Eupelmidae) is a solitary egg endoparasitoid that has been studied for inundative biological control of Halyomorpha halys Stål (Hemiptera: Pentatomidae) in China. In this study, we assessed the reproductive attributes and functional response of Anastatus japonicus on a factitious host, Antheraea pernyi (Guérin-Méneville) (Lepidoptera: Anthelidae) at 25 ± 1 °C, 70 ± 5% relative humidity and 16 h:8 h light/dark photoperiod. RESULTS: The mean lifetime fecundity of Anastatus japonicus females was 404.3 progeny produced over an average oviposition period of 42.3 days. The sex ratio of adult progeny was slightly male biased (51.2%), whereas more female progeny were produced before day 20 of a female's life. Single 1-day-old mated Anastatus japonicus females exhibited a type II functional response to increasing host densities (1-50 eggs), with an inverse host density-dependent pattern of percent parasitism. The upper limit to the daily attack rate was estimated as 7.6 Antheraea pernyi eggs. Furthermore, mutual interference among Anastatus japonicus females occurred when increasing densities of parasitoids (1, 2, 4, 8, 16) were exposed to 30 host eggs. CONCLUSION: Laboratory functional response result revealed that individual Anastatus japonicus might be unable to respond effectively to increasing host density in the field, which could be compensated by releasing larger numbers of wasps. Strong mutual interference among foraging Anastatus japonicus females should be considered in any future inundative biological control programs for the sustainable management of Halyomorpha halys or other host insect pests. © 2022 The Authors. The genus Anastatus comprises a large group of parasitoids, including several biological control agents in agricultural and forest systems. The taxonomy and phylogeny of these species remain controversial. In this study, the mitogenome of A. fulloi Sheng and Wang was sequenced and characterized. The nearly full-length mitogenome of A. fulloi was 15,692 bp, compromising 13 protein-coding genes (PCGs), 2 rRNA genes, 22 tRNA genes and a control region (CR). The total A + T contents were 83.83%, 82.18%, 87.58%, 87.27%, and 82.13% in the whole mitogenome, 13 PCGs, 22 tRNA genes, 2 rRNA genes, and CR, respectively. The mitogenome presented negative AT skews and positive GC skews, except for the CR. Most PCGs were encoded on the heavy strand, started with ATN codons, and ended with TAA codons. Among the 3736 amino acid-encoding codons, TTA (Leu1), CGA (Arg), TCA (Ser2), and TCT (Ser2) were predominant. Most tRNAs had cloverleaf secondary structures, except trnS1, with the absence of a dihydrouridine (DHU) arm. Compared with mitogenomes of the ancestral insect and another parasitoid within Eupelmidae, large-scale rearrangements were found in the mitogenome of A. fulloi, especially inversions and inverse transpositions of tRNA genes. The gene arrangements of parasitoid mitogenomes within Chalcidoidea were variable. A novel gene arrangement was presented in the mitogenome of A. fulloi. Phylogenetic analyses based on the 13 protein-coding genes of 20 parasitoids indicated that the phylogenetic relationship of 6 superfamilies could be presented as Mymaridae + (Eupelmidae + (Encyrtidae + (Trichogrammatidae + (Pteromalidae + Eulophidae)))). This study presents the first mitogenome of the Anastatus genus and offers insights into the identification, taxonomy, and phylogeny of these parasitoids.

Parasitoid wasps are rapidly developing as a model for evolutionary biology. Here we present chromosomal genomes of two Anastatus wasps, A. japonicus and A. fulloi, and leverage these genomes to study two fundamental questions-genome size evolution and venom evolution. Anastatus shows a much larger genome than is known among other wasps, with unexpectedly recent bursts of LTR retrotransposons. Importantly, several genomic innovations, including Piwi gene family expansion, ubiquitous Piwi expression profiles, as well as transposable element-piRNA coevolution, have likely emerged for transposable element silencing to maintain genomic stability. Additionally, we show that the co-option evolution arose by expression shifts in the venom gland plays a dominant role in venom turnover. We also highlight the potential importance of non-venom genes that are coexpressed with venom genes during venom evolution. Our findings greatly advance the current understanding of genome size evolution and venom evolution, and these genomic resources will facilitate comparative genomics studies of insects in the future.

The predatory stink bug Eocanthecona furcellata belongs to the subfamily Asopinae of Pentatomidae. In the current study, the complete mitochondrial genome of E. furcellata is determined. This mitogenome is 16,085 bp in size and comprises of 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and a control region. Gene order is identical to that of the putative ancestral arrangement of insects. Nucleotide composition is biased toward A and T, which together made up 75.5% of the entire genome. All tRNAs have the clover-leaf structure except for the tRNASer(AGN) and the length of them ranges from 61 to 73 bp. The monophyly of Pentatomidae is highly supported by the phylogenetic tree and E. furcellata is very close to other carnivorous species of the remaining Pentatomidae species.

Chinese silkworm (Antheraea pernyi) eggs are used as factitious hosts of Anastatus fulloi, and refrigeration of these eggs is essential for large-scale rearing of A. fulloi. We studied the effects of cold storage of A. pernyi eggs on egg quality and the fitness of A. fulloi reared on the eggs. Four cold storage treatments and two cold storage periods were assessed. The 0-3 °C refrigerator treatment was unsuitable for long-term (>70 days) storage. Cold storage at -5 °C and -18 °C increased the loss rate of A. pernyi eggs, but there was no significant difference between the control and 0-3 °C water bath treatment. The parasitism rate of A. fulloi was reduced when A. pernyi eggs were refrigerated for 6 or 12 months. There were no obvious differences in eclosion rate and percentage of females between control and eggs subjected to 6-month storage in 0-3 °C, -5 °C, and -18 °C water bath treatments. However, the eclosion rate and percentage of females decreased sharply when the storage period was 12 months. The overall eclosion rate of A. fulloi was reduced at the prolonged refrigeration time. Cold storage reduced host egg quality and their fitness suitability for A. fulloi. To minimize losses in the large-scale rearing of A. fulloi, A. pernyi eggs should be refrigerated in a 0-3 °C or -5 °C water bath treatment, and the storage period should not exceed 6 months.

We studied the parasitism capacity of Spalangia endius as a pupal parasite of Bactocera dorsalis after switching hosts. We used pupae of B. dorsalis and M. domestica as the hosts and studied parasitism by S. endius in the laboratory. The parasitism capacities were compared at different host densities and different parasitoid ages. The two functional responses of S. endius fitted a Holling Type II equation. More M. domestica were parasitized than B. dorsalis at all the densities. The ability of S. endius to control M. domestica was α/Th (parasitism capacity) = 32.1950, which was much stronger than that of control B. dorsalis, which was α/Th = 4.7380. The parasitism rate of wasps that had parasitized B. dorsalis had decreased by the emergence time of parasitoids. These results suggest that the parasitoid-pest ratio should be 1:25 to maintain a relatively stable parasitism rate for control of B. dorsalis. The S. endius colony reared on M. domestica successfully controlled a low-density population of B. dorsalis in the lab. We provide evidence suggesting that the parasitism capacity of S. endius needs to be improved.

BACKGROUND: Panonychus citri (McGregor) (Acari: Tetranychidae) population outbreaks after the citrus plantation's chemical application is a common observation. Dispersal behavior is an essential tool to understand the secondary outbreak of P. citri population. Therefore, in the current study, the dispersal activity of P. citri was observed on the leaf surfaces of Citrus reticulata (Rutaceae) treated with SYP-9625, abamectin, vegetable oil, and EnSpray 99. METHOD: Mites were released on the first (apex) leaf of the plant (adaxial surface) and data were recorded after 24 h. The treated, untreated, and half-treated data were analyzed by combining the leaf surfaces (adaxial right, adaxial left, abaxial right, and abaxial left). All experiments were performed in open-air environmental conditions. RESULTS: The maximum number of mites was captured on the un-treated or half-treated surfaces due to chemicals repellency. Chemical bioassays of the free-choice test showed that all treatments significantly increased the mortality of P. citri depending on application method and concentration. A significant number of mites repelled away from treated surfaces and within treated surfaces except adaxial left and abaxial right surfaces at LC30. In the no-choice test, SYP-9625 gave maximum mortality and dispersal by oils than others. No significant differences were observed within the adaxial and abaxial except abaxial surface at LC30. Therefore, the presence of tested acaricides interferes with P. citri dispersal within leaf surfaces of plantations depending on the mites released point and a preferred site for feeding.

Trichogramma is a useful species that is widely applied in biocontrol. Temperature profoundly affects the commercial application of T. chilonis. Different developmental transcriptomes of prepupae and pupae of T. chilonis under 10, 25, and 40°C were obtained from our previous study. In this study, transcriptomic analysis was further conducted to gain a clear understanding of the molecular changes in the prepupae of T. chilonis under different thermal conditions. A total of 37,295 unigenes were identified from 3 libraries of prepupae of T. chilonis, 17,293 of which were annotated. Differential expression analysis showed that 408 and 108 differentially expressed genes (DEGs) were identified after heat and cold treatment, respectively. Under heat stress, the pathway of protein processing in endoplasmic reticulum was found to be active. Most of the genes involved in this pathway were annotated as lethal (2) essential for life [l(2)efl] and heat shock protein genes (hsps), which were both highly upregulated. Nevertheless, most of the genes involved in another significantly enriched pathway of starch and sucrose metabolism were downregulated, including 1 alpha-glucosidase gene and 2 beta-glucuronidase genes. Under cold stress, no significantly enriched pathway was found, and the significantly enriched GO terms were related to the interaction with host and immune defenses. Together, these results provide us with a comprehensive view of the molecular mechanisms of T. chilonis in response to temperature stresses and will provide new insight into the mass rearing and utilization of T. chilonis.

Anastatus japonicus Ashmead is a widely used biological control agent against stink bugs that can be successfully reared using the large eggs of the Chinese silkworm. In this study, environmental factors responsible for the induction of diapause in A. japonicus were investigated on host eggs of the Chinese silkworm. A. japonicus exhibited a facultative, mature larval diapause within its host eggs. Second-third instar larva are the most sensitive stages to diapause stimuli. The accumulation of diapause stimuli during all the larval stages maximized the diapause response. A short photoperiod of 10L:14D and temperature of 17 °C led to the occurrence of the highest diapause response, while a long photoperiod (14L:10D) and low temperatures (11 and 14 °C) prevented the diapause. A specific exposure period was required to reach high diapause incidence. Diapausing mature larvae had a significantly higher survival rate after 180 days storage at 10 °C than that of nondiapausing mature larvae. Taken together, results suggest methods that could be exploited in the developmental regulation, field-release pretreatment technology, and long-term storage of A. japonicus.

BACKGROUND: Parasitoid venom is composed of a complex mixture of various active substances with different biological functions and is injected in the host during the parasitoid oviposition. Anastatus japonicus (Hymenoptera: Eupelmidae) is an egg parasite of Tessaratoma papillosa (Hemiptera: Tessaratomidae). Although the venom of this egg parasitoid plays an important role in the parasitic process, relatively little work has been done to address the mechanism. RESULTS: In the present study, proteomic analysis was performed to identify the proteins that play an important role in the parasitic process of A. japonicus. A total of 2084 proteins were identified, including 81 putative venom proteins, most of which were identified as Hexamerin, Chitinase 2, Calreticulin, Heat shock protein 83-like, Serine protease, Arginine kinase, Phosphoserine aminotransferase and Actin protein. Together the before (Be) and after (Af) parasitization venom contains 1628 proteins, including 212 DEPs with 181 and 31 significantly up-regulated and down-regulated respectively. In addition, 10 differentially expressed proteins (DEPs) with fold change ≥8.71 were subjected to RT-qPCR to validate the proteomic data. The differential expression analysis revealed that nine proteins were specifically present in the pre-parasitic venom, whereas 26 proteins were specific to the post-parasitic treatments. Results of RT-qPCR analysis showed high expression of the selected DEPs which further validated our proteomics data. CONCLUSION: These new proteomic data greatly enrich our current knowledge about key venom proteins associated with parasitic process in A. japonicus and contribute to better understanding of the parasitic mechanisms leading to the development of new biological control strategies. © 2020 Society of Chemical Industry.

Trichogramma is a kind of egg parasitoid wasp that is widely used to control lepidopterous pests. Temperature is one of the main factors that determines the various life activities of this species, including development, reproduction and parasitism efficiency. Heat shock proteins (HSPs) are highly conserved and ubiquitous proteins that are best known for their responsiveness to temperature and other stresses. To explore the potential role of HSPs in Trichogramma species, we obtained the full-length cDNAs of six HSP genes (Tchsp10, Tchsp21.6, Tchsp60, Tchsp70, Tchsc70-3, and Tchsp90) from T. chilonis and analyzed their expression patterns during development and exposure to temperature stress. The deduced amino acid sequences of these HSP genes contained the typical signatures of their corresponding protein family and showed high homology to their counterparts in other species. The expression levels of Tchsp10, Tchsp21.6 and Tchsp60 decreased during development. However, the expression of Tchsc70-3 increased from the pupal stage to the adult stage. Tchsp70 and Tchsp90 exhibited the highest expression levels in the adult stage. The expression of six Tchsps was dramatically upregulated after 1 h of exposure to 32 and 40°C but did not significantly change after 1 h of exposure to 10 and 17°C. This result indicated that heat stress, rather than cold stress, induced the expression of HSP genes. Furthermore, the expression of these genes was time dependent, and the expression of each gene reached its peak after 1 h of heat exposure (40°C). Tchsp10 and Tchsp70 exhibited a low-intensity cold response after 4 and 8 h of exposure to 10°C, respectively, but the other genes did not respond to cold at any time points. These results suggested that HSPs may play different roles in the development of this organism and in its response to temperature stress.

Chemoreception is critical for the survival of insects. Insects have a variety of behavioral responses, such as mating, host searching and ovipositing, in response to different odor signals detected in their living environment. Trichogramma chilonis, an egg parasitoid, acts as an efficient and effective biocontrol reagent for many agricultural and forestry insect pests in many parts of China. However, little is known about the molecular mechanism of the olfaction-evoked behavior in T. chilonis. In the present study, we conducted transcriptome profiling analysis of T. chilonis based on the Illumina high-throughput sequencing platform in order to explore differences of chemoreception between male and female T. chilonis. In this study, a total of 85 chemosensory genes were identified from transcriptomic data, including 45 odorant receptors (ORs), 22 odorant binding proteins (OBPs), 14 ionotropic receptors (IRs), 2 sensory neuron membrane proteins (SNMPs) and 2 chemosensory proteins (CSPs). From the analysis of the transcriptome, most of the candidate olfactory genes had similar expression levels in males and females, including a few OR and OBP genes (TchiOR38, TchiOR39, TchiOR40, TchiOR41, TchiOR42, TchiOR43, TchiOR44, TchiOR45, TchiOBP1, TchiOBP4, TchiOBP10, TchiOBP12, TchiOBP18 and TchiOBP19) which showed male-biased expression. Some annotated unigenes were chosen randomly to have qRT-PCR, which verified the correctness of analysis of transcriptome in T. chilonis. This is the first study to obtain and identify candidate genes related to chemoreception in T. chilonis. Our work lays a solid foundation for related future research on the chemosensory system of T. chilonis at the molecular level and helps advance the use of T. chilonis as biological control agents.

Asian corn borer, Ostrinia furnacalis (Guenée), is a key corn pest in the Asian-Western Pacific countries. It overwinters as full-grown larvae in plant stalks or in a spun-silk covering located in the plant debris in the temperate regions of China. Supercooling point (SCP) and survival rate after low sub-zero temperature treatment were assessed for field-collected populations in the laboratory using a cool bath with a 1°C/min cooling rate until -40°C. Mean SCPs were varied among geographical populations, with a significant decline from -22.7°C of Haikou, the multivoltine tropical population in the south, to -28.5°C of Gongzhuling, the univoltine temperate population in the northeast of China. In addition, there was more than 1°C difference in SCP between Gongzhuling univoltine and bivoltine populations that were from the same geographic origin. Mean SCPs of the Guangzhou population fluctuated over the year, with significantly lower SCPs in winter than in other seasons, which correlated with a significantly higher proportion of diapausing larvae in winter than in other seasons. Over 41% of overwintering larvae from the northeast population could withstand to be supercooled for a few minutes to the low sub-zero temperature of -40°C, but only 6.7% of their southern counterparts did so. The findings from this study suggest that O. furnacalis mostly takes advantage of freeze avoidance as diapausing larvae for overwintering in the southern region, whereas it exhibits freeze tolerance in diapause in the northeastern region.

BACKGROUND: The oriental fruit fly, Bactrocera dorsalis Hendel, causes serious losses to fruit production and is one of the most economically important pests in many countries, including China, Spalangia endius Walker is a pupal parasitoid of various dipteran hosts, and may be considered a potentially important ectoparasitic pupal parasitoid of B. dorsalis. However, lack of genetic information on this organism is an obstacle to understanding the mechanisms behind its interaction with this host. Analysis of the S. endius transcriptome is essential to extend the resources of genetic information on this species and, to support studies on S. endius on the host B. dorsalis. METHODOLOGY/PRINCIPAL FINDINGS: We performed de novo assembly RNA-seq of S. endius. We obtained nearly 10 Gbp of data using a HiSeq platform, and 36319 high-quality transcripts using Trinity software. A total of 22443 (61.79%) unigenes were aligned to homologous sequences in the jewel wasp and honeybee (Apis florae) protein set from public databases. A total of 10037 protein domains were identified in 7892 S. endius transcripts using HMMER3 software. We identified expression of six gustatory receptor and 21 odorant receptor genes in the sample, with only one gene having a high expression level in each family. The other genes had a low expression level, including two genes regulated by splicing. This result may be due to the wasps being kept under laboratory conditions. Additionally, a total of 3727 SSR markers were predicted, which could facilitate the identification of polymorphisms and functional genes within wasp populations. CONCLUSION/SIGNIFICANCE: This transcriptome greatly improves our genetic understanding of S. endius and provides a large number of gene sequences for further study.
