RESUMO

The objective of this study was to describe the nutritional strategies used on Canadian dairy farms with automated milking systems (AMS), both at the feed bunk and the concentrate offered at the AMS, as well as to determine what dietary components and nutrients, as formulated, were associated with milk production and milking behaviors on those farms. Formulated diets (including ingredients and nutrient content) and AMS data were collected from April 1, 2019, until September 30, 2020, on 160 AMS farms (eastern Canada [East] = 8, Ontario [ON] = 76, Quebec [QC] = 22, and western Canada [West] = 54). Both partial mixed ration (PMR) and AMS concentrate samples were collected from May 1 to September 30, 2019, on 169 farms (East = 12, ON = 63, QC = 42, West = 52). We collected AMS milking data for 154 herds. For each farm (n = 161), milk recording data were collected and summarized by farm to calculate average milk yield and components. Multivariable regression models were used to associate herd-level formulated nutrient composition and feeding management practices with milk production and milking behavior. Milk yield (mean ± SD = 37.0 ± 0.3 kg/d) was positively associated with the PMR ether extract (EE) concentration (+0.97 kg/d per percentage point [p.p.] increase) and with farms that fed barley silage as their major forage source (n = 16; +2.18 kg/d) as compared with haylage (n = 42), whereas farms that fed corn silage (n = 96; +1.23 kg/d) tended to produce more milk than farms that fed haylage. Greater milk fat content (4.09 ± 0.28%) was associated with a greater PMR-to-AMS concentrate ratio (+0.02 p.p. per unit increase) and total diet net energy for lactation (+0.046 p.p. per 0.1 Mcal/kg increase), but a lesser percentage of NFC of the PMR (-0.016 p.p. per p.p. increase of NFC percentage). Milk protein content (3.38 ± 0.14%) was positively associated with the forage percentage of the PMR (+0.003 p.p. per p.p. increase of forage percentage) and the total diet starch percentage (+0.009 p.p. per p.p. increase of starch percentage), but was negatively associated with farms feeding corn silage (-0.1 p.p. compared with haylage) as their major forage. Greater milking frequency (2.77 ± 0.40 milkings/d) was observed on farms with free-flow cow traffic systems (+0.62 milkings/d) and was positively associated with feed push-up frequency (+0.013 milkings/d per additional feed push-up), but negatively associated with PMR NFC content and forage percentage of the total ration (-0.017 milkings/d per p.p. increase of forage percentage). Lastly, greater milking refusal frequency (1.49 ± 0.82 refusals/d) was observed on farms with free-flow cow traffic systems (+0.84 refusals/d) and farms feeding barley silage (+0.58 refusals/d) than with guided flow and farms feeding either corn silage or haylage, respectively. These data give insight into the ingredients, nutrient formulations and type of diets fed on AMS dairy farms across Canada and the association of those factors with milk production and milking behaviors.

The objectives of this study were to determine the farm-level hyperketolactia (HKL) prevalence, as diagnosed from milk ß-hydroxybutyrate (BHB) concentration, on dairy farms milking with an automatic milking system (AMS) and to describe the farm-level housing, management, and nutritional risk factors associated with increased farm-average milk BHB and the within-herd HKL prevalence in the first 45 DIM. Canadian AMS farms (n = 162; eastern Canada n = 8, Quebec n = 23, Ontario n = 75, western Canada n = 55) were visited once between April to September 2019 to record housing and herd management practices. The first test milk data for each cow under 45 DIM were collected, along with the final test of the previous lactations for all multiparous cows, from April 1, 2019 to September 30, 2020. The first test milk BHB was then used to classify each individual cow as having HKL (milk BHB ≥ 0.15 mmol/L) at the time of testing. Milk fat and protein content, milk BHB, and HKL prevalence were summarized by farm and lactation group (all, primiparous, and multiparous). During this same time period, formulated diets for dry and lactating cows, including ingredients and nutrient composition, and AMS milking data were collected. Data from the AMS were used to determine milking behaviors and milk production of each herd during the first 45 DIM. Multivariable regression models were used to associate herd-level housing, feeding management practices, and formulated nutrient composition with first test milk BHB concentrations and within-herd HKL levels separately for primiparous and multiparous cows. The within-herd HKL prevalence for all cows was 21.8%, with primiparous cows having a lower mean prevalence (12.2 ± 9.2%) than multiparous cows (26.6 ± 11.3%). Milk BHB concentration (0.095 ± 0.018 mmol/L) and HKL prevalence for primiparous cows were positively associated with formulated prepartum DMI and forage content of the dry cow diet while being negatively associated with formulated postpartum DMI, the major ingredient in the concentrate supplemented through the AMS, and the PMR-to-AMS concentrate ratio. However, multiparous cows' milk BHB concentration (0.12 ± 0.023 mmol/L) and HKL prevalence were positively associated with the length of the previous lactation, milk BHB at dry off, prepartum diet nonfiber carbohydrate content, and the major forage fed on farm, while tending to be negatively associated with feed bunk space during lactation. This is the first study to determine the farm-level risk factors associated with herd-level prevalence of HKL in AMS dairy herds, thus helping optimize management and guide diet formulation to promote the reduction of HKL prevalence.

This study was conducted to investigate the role of Na+ on ruminal short-chain fatty acid (SCFA) absorption and barrier function when isolated ruminal epithelium was exposed to high and low pH ex vivo. Nine Holstein steer calves (322 ± 50.9 kg of body weight) consuming 7.05 ± 1.5 kg dry matter of a total mixed ration were euthanized and ruminal tissue was collected from the caudal-dorsal blind sac. Tissues were mounted between 2 halves of Ussing chambers (3.14 cm2) and exposed to buffers that contained low (10 mM) or high (140 mM) Na+ with low (6.2) or high (7.4) mucosal pH. The same buffer solutions were used on the serosal side except that pH was maintained at 7.4. Buffers used to evaluate SCFA uptake contained bicarbonate to determine total uptake or excluded bicarbonate and included nitrate to determine noninhibitable uptake. Bicarbonate-dependent uptake was calculated as the difference between the total and noninhibitable uptake. Acetate (25 mM) and butyrate (25 mM) were spiked with 2-3H-acetate and 1-14C-butyrate, respectively, and were then added to the mucosal side, incubated for 1 min, and tissues were analyzed to evaluate rates of SCFA uptake. Tissue conductance (Gt) and the mucosal-to-serosal flux of 1-3H-mannitol were used to assess barrier function. There were no Na+ × pH interactions for butyrate or acetate uptake. Decreasing mucosal pH from 7.4 to 6.2 increased total acetate and butyrate uptake, and bicarbonate-dependent acetate uptake. Flux of 1-3H-mannitol was not affected by treatment. However, high Na+ concentration reduced Gt and prevented an increase in Gt from flux period 1 to flux period 2. The results of this study indicate that although providing more Na+ to the ruminal epithelium does not affect SCFA uptake or mannitol flux, it may help stabilize tissue integrity.

Impaired locomotion (lameness) may negatively affect the ability and desire of cows to milk voluntarily, which is a key factor in success of automated milking systems (AMS). The objective of this study was to identify factors associated with herd-level lameness prevalence and associations of lameness and other farm-level factors with milking activity, milk yield, and milk quality in herds with AMS. From April to September 2019, 75 herds with AMS in Ontario, Canada, were visited, and data on barn design and farm management practices were collected. Data from AMS were collected, along with milk recording data, for the 6-mo period before farm visits. Farms averaged 98 ± 71 lactating cows, 2.3 ± 1.5 robot units/farm, 43.6 ± 9.4 cows/robot, 36.4 ± 4.9 kg/d of milk, a milking frequency of 3.01 ± 0.33 milkings/d, and a herd average geometric mean SCC of 179.3 ± 74.6 (× 1,000) cells/mL. Thirty percent of cows/farm (minimum of 30 cows/farm) were scored for body condition (1 = underconditioned to 5 = over conditioned) and locomotion (1 = sound to 5 = lame; clinically lame ≥3 out of 5 = 28.3 ± 11.7%, and severely lame ≥4 out of 5 = 3.0 ± 3.2%). Clinical lameness (locomotion score ≥3) was less prevalent on farms with sand bedding, with increased feed bunk space per cow, and on farms with non-Holstein breeds versus Holsteins, and tended to be less prevalent with lesser proportion of underconditioned cows (with body condition score ≤2.5). Severe lameness occurrence (farms with any cows with locomotion score ≥4) was associated with a greater proportion of underconditioned cows and in farms with stalls with greater curb heights. Herd average milk yield/cow per day increased with lesser prevalence of clinical lameness (each 10-percentage-point decrease in clinical lameness prevalence was associated with 2.0 kg/cow per day greater milk yield) and greater milking visit frequency per day, and tended to be greater with increased feed push-up frequency. Lesser herd average somatic cell count was associated with lesser clinical lameness prevalence, herd average days in milk, and proportion of overconditioned cows, and somatic cell count tended to be lesser for farms with sand bedding versus those with organic bedding substrates. The results highlight the importance of minimizing lameness prevalence, using of sand bedding, ensuring adequate feed access and feed bunk space, and maintaining proper cow body condition to optimize herd-level productivity and milk quality in AMS herds.

The objective of this study was to benchmark the herd-level housing and management strategies of automated milking system (AMS) farms across Canada and assess the associations of these herd-level housing factors and management practices with milk production and quality. Canadian AMS farms (n = 197; Western Canada: n = 50, Ontario: n = 77, Quebec: n = 59, Atlantic Canada: n = 11) were each visited once from April to September 2019, and details were collected related to barn design and herd management practices. Milk-recording data for the 6 mo before farm visits were collected. Farms averaged (± standard deviation) 110 ± 102 lactating cows, 2.4 ± 1.9 AMS units/farm, 47.5 ± 14.9 cows/AMS, 36.7 ± 5.0 kg/d of milk, 4.13 ± 0.34% fat, 3.40 ± 0.16% protein, and a herd-average somatic cell count of 186,400 ± 80,800 cells/mL. Farms mainly used freestall housing systems (92.5%), organic bedding substrates (73.6%), and free flow cow traffic systems (87.8%); farms predominantly milked Holsteins (90.4%). Multivariable regression models were used to associate herd-level housing factors and management practices with milk production and quality. At the herd level, feed push-up frequency (mean = 12.8 ± 8.3 times per day) and feed bunk space (mean = 64 ± 21.5 cm/cow) were positively associated with milk yield. Greater milk yield was associated with herds using inorganic (sand) versus organic bedding, milking Holsteins versus non-Holsteins, and using a form of mechanical ventilation versus natural ventilation alone. Milk fat and milk protein content were only associated with breed. Herds with lower somatic cell counts had more frequent alley cleaning (mean = 12.1 ± 7.5 times per day), wider lying alleys (mean = 304.5 ± 40.0 cm), and sand bedding. The results highlight the importance of using sand bedding, using mechanical ventilation, keeping feed pushed up, ensuring alleys are clean, and ensuring adequate space at the feed bunk for maintaining herd-level productivity and milk quality in farms with AMS.

The objective of this study was to determine whether the partial replacement of barley starch with lactose (fed as dried whey permeate; DWP) affects N utilization, whole-body urea kinetics, and production in dairy cows. Eight lactating Holstein cows were used in a replicated 4 × 4 Latin square design with 28-d periods. Four cows in one Latin square were ruminally cannulated and used to determine dietary effects on whole-body urea kinetics and N utilization. Cows were fed a barley-based diet that contained 3.6% (dry matter basis) total sugar (TSG; designated control), or diets that contained 6.6, 9.6, or 12.6% TSG. Dietary TSG content was increased by the replacement of barley grain with DWP (83% lactose). Diets were isonitrogenous (∼17.3% crude protein), and starch contents of the control, 6.6, 9.6, and 12.6% TSG diets were 24.3, 22.2, 21.2, and 19.1%, respectively. Whole-body urea kinetics were measured using 4-d infusions of [15N15N]-urea with concurrent total collections of feces and urine. Dry matter intake (mean = 26.7 kg/d), milk yield (mean = 34.9 kg/d), and milk protein and fat contents were unaffected by diet. Ruminal ammonia-N concentration decreased linearly as TSG content increased, whereas ruminal butyrate concentration increased linearly as TSG content increased. Urinary excretion of total N and urea-N changed quadratically, whereas urinary excretion of total N (% of N intake) tended to change quadratically as TSG content increased. Fecal N excretion linearly increased as TSG content increased. A quadratic response was observed for total N excretion as TSG content increased. Milk N and retained N were not affected by diet. As TSG content increased, we observed quadratic responses in the omasal flow of fluid-associated and total bacterial nonammonia N, endogenous production of urea-N, urea-N recycled to the gastrointestinal tract, and urea-N returned to the ornithine cycle. Dietary TSG content did not affect the anabolic utilization of recycled urea-N or the proportion of recycled urea-N that was used for bacterial growth. Our results indicate that feeding DWP did not influence dry matter intake, milk yield, or milk composition. Feeding DWP decreased ruminal ammonia-N concentration, but this did not result in positive responses in milk protein secretion or N balance. The quadratic response in omasal flow of total bacterial nonammonia N indicated that including TSG beyond 9.6% of diet dry matter might depress ruminal microbial protein synthesis.

AIMS: The effects of partial replacement of a barley-based concentrate with flaxseed-based products on the rumen bacterial population of lactating Holstein dairy cows were evaluated. METHODS AND RESULTS: Treatments fed were CONT, a normal diet that included barley silage, alfalfa hay and a barley-based concentrate that contained no flaxseed or faba beans; FLAX, inclusion of a nonextruded flaxseed-based product containing 55·0% flaxseed, 37·8% field peas and 6·9% alfalfa; EXT, similar to FLAX, but the product was extruded and EXTT, similar to FLAX, but product was extruded and field peas were replaced by high-tannin faba beans. The rumen bacterial population was evaluated by utilizing 16S rRNA gene sequencing. Most abundant phyla, families and genera were unaffected. However, some taxa were affected; for example, unsaturated fatty acid content was negatively correlated with Clostridiaceae, and tannin content was negatively correlated with BS11 and Paraprevotellaceae. CONCLUSIONS: Predominant rumen bacterial taxa were not affected, but the abundance of some taxa found in lower proportions shifted, possibly due to sensitivity to unsaturated fatty acids or tannins. SIGNIFICANCE AND IMPACT OF THE STUDY: Flaxseed-based products were effective for partially replacing barley-based concentrate in rations of lactating dairy cows. No negative effects of these products were observed on the abundance of predominant rumen bacterial taxa, with only minor shifts in less abundant bacteria.

The objective of this study was to determine the effects of feeding canola meal (CM) or wheat dried distillers grains with solubles (W-DDGS) as the major source of protein in diets varying in crude protein (CP) content on ruminal fermentation, microbial protein production, omasal nutrient flow, and production performance in lactating dairy cows. Eight lactating dairy cows were used in a replicated 4×4 Latin square design with 29-d periods (21 d of dietary adaptation and 8 d of measurements) and a 2×2 factorial arrangement of dietary treatments. Four cows in 1 Latin square were ruminally cannulated to allow ruminal and omasal sampling. The treatment factors were (1) source of supplemental protein (CM vs. W-DDGS) and (2) dietary CP content (15 vs. 17%; DM basis). Diets contained 50% forage and 50% concentrate, and were fed twice daily at 0900 and 1600 h as total mixed rations for ad libitum intake. Dry matter intake and milk yield were unaffected by dietary treatments; however, milk yield in cows that were fed CM was numerically greater (+1.1 kg/d) when compared with cows fed W-DDGS. Feeding CM increased milk lactose content compared with feeding W-DDGS. Milk urea nitrogen and ruminal NH3-N concentrations were greater in cows fed the high-CP compared with those fed the low-CP diet. The rumen-degradable protein supply was greater in cows fed the high-CP when compared with those fed the low-CP diet when diets contained CM, whereas rumen-degradable protein supply was lower in cows fed the high-CP when compared with those fed the low-CP diet when diets contained W-DDGS. Total N flow at the omasal canal was not affected by diet; however, omasal flow of NH3-N was greater in cows fed CM when compared with those fed W-DDGS. The rumen-undegradable protein supply was greater in cows fed the low-CP when compared with those fed the high-CP diet when diets contained CM, whereas rumen-undegradable protein supply was lower in cows fed the low-CP when compared with those fed the high-CP diet when diets contained W-DDGS. Omasal flow of fluid-associated bacteria was greater and that of particle-associated bacteria tended to be greater in cows fed CM when compared with those fed W-DDGS; however, omasal flow of total microbial nonammonia N was unaffected by dietary treatment. Omasal flows of threonine and tryptophan were greater, whereas that of histidine and lysine tended to be greater in cows fed CM when compared with those fed W-DDGS. Our results show that when dairy diets are formulated to contain 15 or 17% CP, CM or W-DDGS can be used as the major source of protein and achieve similar levels of milk production.

Our objective was to examine the effects of feeding coextruded and nonextruded supplements consisting of wheat dried distillers grains with solubles with peas (WDDGS-peas) or canola meal (WDDGS-CM) on ruminal fermentation, omasal flow, and production performance in Holstein cows. Eight cows (4 ruminally cannulated) were used in a replicated 4×4 Latin square with 28-d periods and a 2×2 factorial arrangement of dietary treatments. Dietary treatments were coextruded or nonextruded mixtures of WDDGS-peas and WDDGS-CM that were included in total mixed rations at 15.1% [dry matter (DM) basis]. Diet had no effect on DM intake. Milk yield was greater in cows fed coextruded diets compared with those fed nonextruded diets. Milk fat content was greater in cows fed nonextruded diets compared with those fed coextruded diets, but milk fat yield was greater in cows fed coextruded diets compared with those fed nonextruded diets. Milk yield tended to be greater and milk protein yield was greater in cows fed WDDGS-peas compared with those fed WDDGS-CM. Cows fed nonextruded diets had a greater milk urea-N concentration compared with those fed coextruded diets. Cows fed coextruded diets had greater ruminal digestion of DM and tended to have greater ruminal digestion of organic matter compared with those fed nonextruded diets. Total-tract digestibilities of organic matter, crude protein, ether extract, and starch were greater, whereas that of acid detergent fiber and neutral detergent fiber tended to be greater in cows fed coextruded compared with those fed nonextruded diets. Total-tract digestibility of ether extract was lower whereas that of starch was greater and that of crude protein tended to be greater in cows fed WDDGS-peas compared with those fed WDDGS-CM. Total N excretion and milk N efficiency were unaffected by diet. Ruminal NH3-N concentration tended to be greater in cows fed WDDGS-CM compared with those fed WDDGS-peas. Ruminal propionate concentration was greater whereas plasma urea-N concentration tended to be lower in cows fed coextruded compared with those fed nonextruded diets. Plasma glucose concentration was greater in cows fed diets containing WDDGS-CM compared with those fed diets containing WDDGS-peas, but the difference in plasma glucose concentration between WDDGS-CM and WDDGS-peas was greater in cows fed coextruded diets compared with those fed nonextruded diets. In summary, feeding coextruded compared with nonextruded supplements or WDDGS-peas compared WDDGS-CM increased yields of milk, fat, and protein.

The objective of this study was to determine how interactions between dietary crude protein (CP) and rumen-degradable protein (RDP) concentrations alter urea-nitrogen recycling, nitrogen (N) balance, omasal nutrient flow, and milk production in lactating Holstein cows. Eight multiparous Holstein cows (711±21kg of body weight; 91±17d in milk at the start of the experiment) were used in a replicated 4×4 Latin square design with a 2×2 factorial arrangement of dietary treatments and 29-d experimental periods. Four cows in one Latin square were fitted with ruminal cannulas to allow ruminal and omasal sampling. The dietary treatment factors were CP (14.9 vs. 17.5%; dry matter basis) and RDP (63 vs. 69% of CP) contents. Dietary RDP concentration was manipulated by including unprocessed or micronized canola meal. Diet adaptation (d 1-20) was followed by 8d (d 21-29) of sample and data collection. Continuous intrajugular infusions of [(15)N(15)N]-urea (220mg/d) were conducted for 4d (d 25-29) with concurrent total collections of urine and feces to estimate N balance and whole-body urea kinetics. Proportions of [(15)N(15)N]- and [(14)N(15)N]-urea in urinary urea, and (15)N enrichment in feces were used to calculate urea kinetics. For the low-CP diets, cows fed the high-RDP diet had a greater DM intake compared with those fed the low-RDP diet, but the opposite trend was observed for cows fed the high-CP diets. Dietary treatment had no effect on milk yield. Milk composition and milk component yields were largely unaffected by dietary treatment; however, on the low-CP diets, milk fat yield was greater for cows fed the low-RDP diet compared with those fed the high-RDP diet, but it was unaffected by RDP concentration on the high-CP diets. On the high-CP diets, milk urea nitrogen concentration was greater in cows fed the high-RDP diet compared with those fed the low-RDP diet, but it was unaffected by RDP concentration on the low-CP diets. Ruminal NH3-N concentration tended to be greater in cows fed the high-CP diet compared with those fed the low-CP diet, and it was greater in cows fed the high-RDP diet as compared with those fed the low-RDP diet. Nitrogen intake and both total N and urea-N excretion in urine were greater for cows fed the high-CP diet compared with those fed the low-CP diet. However, N balance and urinary excretion of purine derivatives were unaffected by dietary treatment. Urea-N entry rate (UER) was greater in cows fed the high-CP diet compared with those fed the low-CP diet; however, UER was unaffected by dietary RDP concentration. The proportion of urea-N recycled to the gastrointestinal tract (as a percentage of UER) was greater in cows fed the low-CP diet compared with those fed the high-CP diet. In summary, reducing dietary CP concentration decreased urinary N excretion but had no effect on milk yield, thus resulting in an overall improvement in milk N efficiency.

In cows fed diets based on corn-alfalfa silage, replacing starch with sugar improves milk production. Although the rate of ruminal fermentation of sugar is more rapid than that of starch, evidence has been found that feeding sugar as a partial replacement for starch does not negatively affect ruminal pH despite increasing diet fermentability. The mechanism(s) for this desirable response are unknown. Our objective was to determine the effects of replacing barley or corn starch with lactose (as dried whey permeate; DWP) on ruminal function, short-chain fatty acid (SCFA) absorption, and nitrogen (N) utilization in dairy cows. Eight lactating cows were used in a replicated 4 × 4 Latin square design with 28-d periods and source of starch (barley vs. corn) and level of DWP (0 vs. 6%, DM basis) as treatment factors. Four cows in 1 Latin square were ruminally cannulated for the measurement of ruminal function, SCFA absorption, and N utilization. Dry matter intake and milk and milk component yields did not differ with diet. The dietary addition of DWP tended to increase ruminal butyrate concentration (13.6 vs. 12.2 mmol/L), and increased the Cl(-)-competitive absorption rates for acetate and propionate. There was no sugar effect on minimum ruminal pH, and the duration and area when ruminal pH was below 5.8. Minimum ruminal pH tended to be lower in cows fed barley compared with those fed corn (5.47 vs. 5.61). The duration when ruminal pH was below pH 5.8 tended to be shorter (186 vs. 235 min/d), whereas the area (pH × min/d) that pH was below 5.8 was smaller (47 vs. 111) on the corn than barley diets. Cows fed the high- compared with the low-sugar diet had lower ruminal NH3-N concentration. Feeding the high-sugar diet tended to increase apparent total-tract digestibility of dry matter and organic matters and increased apparent total-tract digestibility of fat. Apparent total-tract digestibility of N tended to be greater in cows fed barley compared with those fed corn, whereas apparent total-tract digestibility of acid-digestible fiber was greater in cows fed corn compared with those fed barley. In conclusion, partially replacing dietary corn or barley starch with sugar upregulated ruminal acetate and propionate absorption, suggesting that the mechanisms for the attenuation of ruminal acidosis when sugar is fed is partly mediated via increased SCFA absorption.

Urea transport (UT-B) proteins are known to facilitate urea movement across the ruminal epithelium; however, other mechanisms may be involved as well because inhibiting UT-B does not completely abolish urea transport. Of the aquaporins (AQP), which are a family of membrane-spanning proteins that are predominantly involved in the movement of water, AQP-3, AQP-7, and AQP-10 are also permeable to urea, but it is not clear if they contribute to urea transport across the ruminal epithelium. The objectives of this study were to determine (1) the functional roles of AQP and UT-B in the serosal-to-mucosal urea flux (Jsm-urea) across rumen epithelium; and (2) whether functional adaptation occurs in response to increased diet fermentability. Twenty-five Holstein steer calves (n=5) were assigned to a control diet (CON; 91.5% hay and 8.5% vitamin and mineral supplement) or a medium grain diet (MGD; 41.5% barley grain, 50% hay, and 8.5% vitamin and mineral) that was fed for 3, 7, 14, or 21 d. Calves were killed and ruminal epithelium was collected for mounting in Ussing chambers under short-circuit conditions and for analysis of mRNA abundance of UT-B and AQP-3, AQP-7, and AQP-10. To mimic physiologic conditions, the mucosal buffer (pH 6.2) contained no urea, whereas the serosal buffer (pH 7.4) contained 1 mM urea. The fluxes of (14)C-urea (Jsm-urea; 26 kBq/10 mL) and (3)H-mannitol (Jsm-mannitol; 37 kBq/10 mL) were measured, with Jsm-mannitol being used as an indicator of paracellular or hydrophilic movement. Serosal addition of phloretin (1 mM) was used to inhibit UT-B-mediated urea transport, whereas NiCl2 (1 mM) was used to inhibit AQP-mediated urea transport. Across treatments, the addition of phloretin or NiCl2 reduced the Jsm-urea from 116.5 to 54.0 and 89.5 nmol/(cm(2) × h), respectively. When both inhibitors were added simultaneously, Jsm-urea was further reduced to 36.8 nmol/(cm(2) × h). Phloretin-sensitive and NiCl2-sensitive Jsm-urea were not affected by diet. The Jsm-urea tended to increase linearly as the duration of adaptation to MGD increased, with the lowest Jsm-urea being observed in animals fed CON [107.7 nmol/(cm(2) × h)] and the highest for those fed the MGD for 21 d [144.2 nmol/(cm(2) × h)]. Phloretin-insensitive Jsm-urea tended to increase linearly as the duration of adaptation to MGD increased, whereas NiCl2-insensitive Jsm-urea tended to be affected by diet. Gene transcript abundance for AQP-3 and UT-B in ruminal epithelium increased linearly as the duration of MGD adaptation increased. For AQP-7 and AQP-10, gene transcript abundance in animals that were fed the MGD was greater compared with that of CON animals. These results demonstrate that both AQP and UT-B play significant functional roles in urea transport, and they may play a role in urea transport during dietary adaptation to fermentable carbohydrates.

A study was conducted to determine the effects of including either wheat-based (W-DDGS) or corn-wheat blend (B-DDGS) dried distillers grains with solubles as the major protein source in low- or high-crude protein (CP) diets fed to dairy cows on ruminal function, microbial protein synthesis, omasal nutrient flows, urea-N recycling, and milk production. Eight lactating Holstein cows (768.5 ± 57.7 kg of body weight; 109.5 ± 40.0 d in milk) were used in a replicated 4 × 4 Latin square design with 28-d periods (18d of dietary adaptation and 10d of measurements) and a 2 × 2 factorial arrangement of dietary treatments. Four cows in one Latin square were ruminally cannulated for the measurement of ruminal fermentation characteristics, microbial protein synthesis, urea-N recycling kinetics, and omasal nutrient flow. The treatment factors were type of distillers co-product (W-DDGS vs. B-DDGS) and dietary CP content [15.2 vs. 17.3%; dry matter (DM) basis]. The B-DDGS was produced from a mixture of 15% wheat and 85% corn grain. All diets were formulated to contain 10% W-DDGS or B-DDGS on a DM basis. No diet effect was observed on DM intake. Yields of milk, fat, protein, and lactose, and plasma urea-N and milk urea-N concentrations were lower in cows fed the low-CP compared with those fed the high-CP diet. Although feeding B-DDGS tended to reduce ruminal ammonia-N (NH3-N) concentration compared with feeding W-DDGS (9.3 vs. 10.5mg/dL), no differences were observed in plasma urea-N and milk urea-N concentrations. Additionally, dietary inclusion of B-DDGS compared with W-DDGS did not affect rumen-degradable protein supply, omasal flows of total N, microbial nonammonia N (NAN), rumen-undegradable protein, and total NAN, or urea-N recycling kinetics and milk production. However, cows fed the low-CP diet had lower N intake, rumen-degradable protein supply, ruminal NH3-N concentration, and omasal flows of N, microbial NAN, and total NAN compared with those fed the high-CP diet. Feeding the low-CP compared with the high-CP diet also resulted in lower endogenous urea-N production, urea-N recycled to the gastrointestinal tract, and urea-N excretion in urine. In summary, our results indicate that both W-DDGS and B-DDGS can be included as the major protein sources in dairy cow diets without compromising nutrient supply and production performance. However, feeding the low-CP diet lowered omasal flows of microbial protein and metabolizable protein, which, in turn, resulted in lower milk production compared with feeding the high-CP diet.

The aim of this study was to determine whether dietary Na-butyrate supplementation affects butyrate and glucose oxidation by ruminal epithelial preparations and whether this effect can be acutely modulated by substrate (glucose and butyrate) supply. Eighteen Suffolk wether lambs (6 lambs/treatment) were blocked by body weight and, within block, randomly assigned to the control treatment (CON) or to diets containing differing Na-butyrate inclusion rates (1.58 or 3.16%) equating to 1.25 (B1.25), and 2.50% (B2.50) butyrate on a dry matter basis, respectively. All lambs received their diet for a period of 14 d. After dietary adaptation, lambs were killed and the ruminal epithelium was harvested from the ventral sac, minced finely, and used for in vitro incubations. Incubation medium contained either a constant concentration of glucose (4 mM) with increasing butyrate concentrations (0, 5, 15, 25, or 40 mM) or a constant butyrate concentration (15 mM) with increasing glucose concentrations (0, 1, 2, 4, or 8 mM) to allow for the evaluation of whether acute changes in the concentration of metabolic substrates affect the oxidation of glucose and butyrate. We observed no interactions between the in vivo and in vitro treatments. Increasing dietary butyrate supplementation linearly decreased glucose oxidation by ruminal epithelial preparations, but had no effect on butyrate oxidation. Increasing butyrate concentration in vitro decreased (cubic effect) glucose oxidation when butyrate concentration ranged between 5 and 15 mM; however, glucose oxidation was increased with a butyrate concentration of 40 mM. Butyrate oxidation decreased (cubic effect) as glucose concentration increased from 1 to 4 mM; however, butyrate oxidation increased when glucose was included at 8mM. The results of this study demonstrate that dietary butyrate supplementation can decrease glucose oxidation by the ruminal epithelium, but the relative supply of glucose and butyrate has a pronounced effect on substrate oxidation.

A study was conducted to determine the effects of replacing canola meal (CM) as the major protein source with wheat-based dried distillers grains with solubles (W-DDGS) on ruminal fermentation, microbial protein production, omasal nutrient flow and animal performance. Eight lactating dairy cows were fed in a replicated 4 × 4 Latin square design with 28-d periods (20 d of dietary adaptation and 8 d of measurements). Four cows in one Latin square were ruminally cannulated for measurements of ruminal fermentation characteristics and flow of nutrients at the omasal canal. Cows were fed either a standard barley silage-based total mixed ration containing CM as the major protein supplement (0% W-DDGS, control) or diets formulated to contain 10, 15, and 20% W-DDGS (dry matter basis), with W-DDGS replacing primarily CM. Diets were isonitrogenous (18.9% crude protein) and contained 3.0, 3.2, 3.5, and 3.7% ether extract for 0, 10, 15, and 20% W-DDGS, respectively. Diets contained 50% forage and 50% concentrate. Inclusion of W-DDGS linearly increased dry matter intake (29.5, 31.2, 30.2, and 31.9 kg/d for 0, 10, 15, and 20% W-DDGS, respectively). The addition of W-DDGS in place of CM resulted in a 1.2- to 1.8-kg increase in milk yield (42.9, 44.7, 44.1, and 44.5 kg/d for 0, 10, 15, and 20% W-DDGS); however, a quadratic change in feed efficiency (i.e., milk yield/DM intake) occurred as the dietary level of W-DDGS increased. Treatments did not differ for milk fat, protein, and lactose concentrations; however, quadratic changes were observed in milk yields of fat (1.48, 1.56, 1.62, and 1.55 kg/d for 0, 10, 15, and 20% W-DDGS, respectively), protein (1.44, 1.46, 1.49, and 1.42 kg/d) and lactose (1.96, 2.02, 2.09, and 1.93 kg/d). Ruminal fermentation characteristics did not change except that the inclusion of 20% W-DDGS resulted in a decrease and a tendency for a decrease in molar concentrations of isobutyrate and total volatile fatty acids, respectively. Omasal flow of total bacterial nonammonia N (NAN) and bacterial efficiency (g of total bacterial NAN flow/kg of organic matter truly digested in the rumen) were not different among diets; however, feeding W-DDGS resulted in a quadratic increase in nonammonia nonbacterial N flow at the omasal canal (271, 318, 336, and 311 g/d for 0, 10, 15, and 20% W-DDGS, respectively). These data indicate that W-DDGS can substitute for CM as the major protein source in dairy cow diets without negatively affecting ruminal fermentation, microbial protein production, and omasal nutrient flow, and can potentially increase dry matter intake and milk yield.

The objective of this study was to determine the interactive effects of barley grain processing and source of supplemental fat on ruminal nutrient digestion, duodenal nutrient flow, microbial protein synthesis, and milk fatty acid (FA) profiles in dairy cows. Four Holstein cows (656 ± 28 kg of BW; 80 ± 12 d in milk) fitted with permanent ruminal and duodenal cannulas were used in a 4×4 Latin square design with a 2×2 factorial arrangement of experimental treatments, in which barley was either dry-rolled (DRB) or pelleted (PB) and supplemented with full-fat canola seed (canola) or full-fat flaxseed (flaxseed). Cows fed DRB consumed 1.8 to 3.5 kg/d more dry matter compared with those fed PB. Mean ruminal pH was lower and the duration (min/d) and area (pH × min) of total ruminal acidosis (RA; ruminal pH <5.8), mild RA (5.8 >ruminal pH >5.5), and severe RA (5.5 >ruminal pH >5.2), and the duration of acute RA (ruminal pH <5.2) were greater in cows fed PB compared with those fed DRB. Milk yield was unaffected by diet. Milk fat content and yield were greater in cows fed DRB than in cows fed PB. Cows fed PB had greater duodenal flows of C18:1 trans, C18:1 cis, C18:2 trans-9,trans-12, C18:2 trans-9,cis-12, C18:2 trans-11,cis-15, and C18:2 trans-10,cis-12 compared with those fed DRB. Duodenal flows of C18:1 cis, C18:1 trans, total C18:1, C18:2n6, C18:2 cis-9,trans-11, C18:2 trans-10,cis-12, C18:2 trans-11,trans-13, total conjugated linoleic acid (CLA), and C18:3n3 were greater in cows fed flaxseed compared with those fed canola. Milk concentrations of C18:3n3 and total CLA tended to be greater in cows fed PB compared with those fed DRB. When compared with cows fed canola, milk concentrations of C18:2 trans-11,cis-15, C18:2 cis-9,trans-11, C18:2 trans-11,trans-13, total CLA, and C18:3n3 were greater in cows fed flaxseed. Ruminal digestion of neutral detergent fiber and acid detergent fiber was greater in cows fed DRB compared with those fed PB. Duodenal flow of ammonia-N tended to be greater in cows fed PB compared with those fed DRB. Duodenal flow of nonammonia nonmicrobial N was greater in cows fed flaxseed compared with those fed canola; however, barley grain processing did not affect duodenal nonammonia nonmicrobial N flow. Duodenal flow of microbial nonammonia N and microbial efficiency were not affected by diet. In summary, barley grain processing and source of fat altered ruminal FA biohydrogenation, and this was reflected in changes in duodenal FA flow and milk FA profiles; however, ruminal microbial protein synthesis was unaltered.

The risk for ischiofemoral impingement has been mainly related to a reduced ischiofemoral distance and morphological variance of the femur. From an evolutionary perspective, however, there are strong arguments that the condition may also be related to sexual dimorphism of the pelvis. We, therefore, investigated the impact of gender-specific differences in anatomy of the ischiofemoral space on the ischiofemoral clearance, during static and dynamic conditions. A random sampling Monte-Carlo experiment was performed to investigate ischiofemoral clearance during stance and gait in a large (n = 40 000) virtual study population, while using gender-specific kinematics. Subsequently, a validated gender-specific geometric morphometric analysis of the hip was performed and correlations between overall hip morphology (statistical shape analysis) and standard discrete measures (conventional metric approach) with the ischiofemoral distance were evaluated. The available ischiofemoral space is indeed highly sexually dimorphic and related primarily to differences in the pelvic anatomy. The mean ischiofemoral distance was 22.2 ± 4.3 mm in the females and 29.1 ± 4.1 mm in the males and this difference was statistically significant (P < 0.001). Additionally, the ischiofemoral distance was observed to be a dynamic measure, and smallest during femoral extension, and this in turn explains the clinical sign of pain in extension during long stride walking. In conclusion, the presence of a reduced ischiofemroal distance and related risk to develop a clinical syndrome of ischiofemoral impingement is strongly dominated by evolutionary effects in sexual dimorphism of the pelvis. This should be considered when female patients present with posterior thigh/buttock pain, particularly if worsened by extension. Controlled laboratory study.

Eight multiparous Holstein cows (676 +/- 57 kg of body weight; 121 +/- 17 d-in-milk) were used in a replicated 4 x 4 Latin square design to determine the effects of 4 sources of carbohydrate on milk yield and composition, ruminal fermentation, and microbial N flow to the duodenum. Four cows in one of the Latin squares were fitted with permanent ruminal cannulae. Diets contained (DM basis) 50% forage in combinations of alfalfa hay and barley silage, and 50% concentrate. The concentrate portion of the diets contained barley, corn, wheat, or oats grain as the primary source of carbohydrate. Intake of DM ranged from 24.0 to 26.2 kg/d, and it tended to be lower in cows fed the wheat-based diet compared with those fed the barley-based diet; consequently, milk yield tended to be lower in cows fed the wheat-based diet compared with those fed the barley-based diet. Cows fed the barley- or wheat-based diets had a lower milk fat content compared with those fed the corn-based diet. Ruminal fermentation characteristics were largely unaffected by the source of dietary carbohydrate, with similar ruminal pH and volatile fatty acid and ammonia concentrations for the first 6 h after the morning feeding. Dietary treatment did not affect total tract apparent digestibility of DM, organic matter, and neutral detergent fiber; however, total tract apparent digestibility of starch in cows fed the oats-based diet was higher compared with those fed the corn-and wheat-based diets. Nitrogen that was used for productive purposes (i.e., N secreted in milk + N apparently retained by the cow) tended to be lower in cows fed the wheat-based diet compared with cows fed the barley-, corn-, or oats-based diets. Urinary purine derivative (PD) excretion was similar in cows fed the barley-, corn-, and wheat-based diets; however, purine derivative excretion was higher in cows fed the barley-based diet compared with those fed the oats-based diet. Consequently, estimated microbial N flow to the duodenum was 49 g/d higher in cows fed the barley-based diet compared with those fed the oats-based diet. Improved production performance with corn and barley diets appeared to be due to greater nutrient absorption than in cows fed oats and wheat diets, rather than improved nutrient utilization efficiency.

The objective of this study was to determine the effects of methods of barley grain processing and source of supplemental fat on urea-N transfer to the gastrointestinal tract (GIT) and the utilization of this recycled urea-N in lactating dairy cows. Four ruminally cannulated Holstein cows (656.3 +/- 27.7 kg of BW; 79.8 +/- 12.3 d in milk) were used in a 4 x 4 Latin square design with 28-d periods and a 2 x 2 factorial arrangement of dietary treatments. Experimental diets contained dry-rolled barley or pelleted barley in combination with whole canola or whole flaxseed as supplemental fat sources. Nitrogen balance was measured from d 15 to 19, with concurrent measurements of urea-N kinetics using continuous intrajugular infusions of [15N 15N]-urea. Dry matter intake and N intake were higher in cows fed dry-rolled barley compared with those fed pelleted barley. Nitrogen retention was not affected by diet, but fecal N excretion was higher in cows fed dry-rolled barley than in those fed pelleted barley. Actual and energy-corrected milk yield were not affected by diet. Milk fat content and milk fat yield were higher in cows fed dry-rolled barley compared with those fed pelleted barley. Source of supplemental fat did not affect urea-N kinetics. Urea-N production was higher (442.2 vs. 334.3 g of N/d), and urea-N entering the GIT tended to be higher (272.9 vs. 202.0 g of N/d), in cows fed dry-rolled barley compared with those fed pelleted barley. The amount of urea-N entry into the GIT that was returned to the ornithine cycle was higher (204.1 vs. 159.5 g of N/d) in cows fed dry-rolled barley than in pelleted barley-fed cows. The amount of urea-N recycled to the GIT and used for anabolic purposes, and the amounts lost in the urine or feces were not affected by dietary treatment. Microbial nonammonia N supply, estimated using total urinary excretion of purine derivatives, was not affected by diet. These results show that even though barley grain processing altered urea-N entry into the GIT, the utilization of this recycled urea-N for microbial production was unaffected as the additional urea-N, which entered the GIT was returned to ureagenesis.

Early-lactating dairy cows mobilize body protein to provide amino acids that are directed toward gluconeogenesis and milk protein synthesis. Propylene glycol (PG) is a precursor of ruminal propionate, and feeding PG has been reported to improve energy supply by increasing blood glucose. Our hypothesis was that feeding PG could spare body protein by providing an alternative source of carbon for gluconeogenesis. The major objectives of this study were 1) to delineate the effects of pre- and postpartum PG supplementation in transition dairy cows on whole-body nitrogen balance, urinary 3-methylhistidine (3-MH) excretion, body composition, and gene expression profiles for the major protein degradation pathways in skeletal muscle; and 2) to characterize the changes in body protein metabolism during the periparturient period. Sixteen pregnant cows (7 primiparous and 9 multiparous) were paired based on expected calving dates and then randomly assigned within each pair to either a basal diet (control) or basal diet plus 600 mL/d of PG. Diets were fed twice daily for ad libitum intake, and PG was fed in equal amounts as a top dress from d -7 to d 45. All measurements were conducted at 3 time intervals starting at d -14 +/- 5, d 15, and d 38 relative to calving. Propylene glycol had no effect on whole-body N balance, urinary 3-MH excretion, or body composition. However, N balance was lower at d 15 and 38, compared with d -14. Urinary excretion of 3-MH was lower at d -14 than at d 15 and 38. Supplemental PG had no effect on body weight (BW) and all components of empty BW. On average, cows fed both diets mobilized 19 kg of body fat and 14 kg of body protein between d -14 and d 38. Supplemental PG had no effect on mRNA abundance in skeletal muscle for m-calpain, and the 14-kDa ubiquitin-carrier protein E2 (14-kDa E2) and proteasome 26S subunit-ATPase components of the ubiquitin-mediated proteolytic pathway; however, PG supplementation downregulated mRNA expression for mu-calpain at d 15, and tended to downregulate mRNA expression for ubiquitin at d 15 and 38. Relative to calving, mRNA abundance for m- and mu-calpain, ubiquitin, and 14-kDa E2 were greater at d 15 compared with d -14 and d 38. In summary, these results indicate that transitional effects on whole-body metabolism and gene expression for the Ca(2+)-dependent and ubiquitin-mediated proteolytic pathways in skeletal muscle were more pronounced than those elicited by PG supplementation.
