RESUMO

Prestes, J, Nascimento, DdC, Neto, IVS, Tibana, RA, Shiguemoto, GE, Perez, SEA, Botero, JP, Schoenfeld, BJ, and Pereira, GB. The effects of muscle strength responsiveness to periodized resistance training on resistin, leptin, and cytokine in elderly postmenopausal women. J Strength Cond Res 32(1): 113-120, 2018-The aim of this study was to evaluate muscle strength responsiveness and to determine whether interleukin 4 (IL-4), leptin, and resistin would be affected by the individual pattern of response to a resistance training (RT) periodization program. Twenty-six elderly postmenopausal women participated in the present study (mean age = 62.57 ± 6.69 years and body mass index = 28.09 ± 4.83 kg·m). Four-month longitudinal training program was performed consisting of 2 whole-body sessions per week with increasing intensity and decreasing volume by using 6-14 repetitions maximum (RM). Two acute whole-body RT sessions (before and after chronic training) were also performed comprising 3 sets of 12-14RM. The responsiveness was determined based on their relative muscle strength gains in 45° leg press. High responders were defined as relative muscle strength gains ≥32% and low responders <32% (n = 13 in each group). Muscle strength increased by a higher amount in the high-responsive group as compared with the low-responsive group in the 45° leg press and bench press, whereas muscle strength increased for both groups when compared with pretraining evaluation (p = 0.001). Low responders displayed higher plasma leptin levels when compared with high responders at baseline (p = 0.001), and after 16 weeks of training, leptin levels were significantly lower when compared with baseline (pretraining period). Both groups displayed a decrease in baseline resistin values after 16 weeks of RT, but only a statistically simple main effect was observed for low responders. In addition, there were no effect of time and no significant interaction between the responsiveness and time on IL-4 concentration. In conclusion, RT is effective in improving upper and lower limb muscle strength in elderly women, with higher magnitudes of increase seen for those classified as high responders. Resistin and leptin displayed a decrease over time, regardless of responsiveness classification.

RESUMO

Low-level laser therapy (LLLT) has shown efficacy in muscle bioenergetic activation and its effects could influence the mechanical performance of this tissue during physical exercise. This study tested whether endurance training associated with LLLT could increase human muscle performance in isokinetic dynamometry when compared to the same training without LLLT. The primary objective was to determine the fatigue index of the knee extensor muscles (FIext) and the secondary objective was to determine the total work of the knee extensor muscles (TWext). Included in the study were 45 clinically healthy women (21 ± 1.78 years old) who were randomly distributed into three groups: CG (control group), TG (training group) and TLG (training with LLLT group). The training for the TG and TLG groups involved cycle ergometer exercise with load applied to the ventilatory threshold (VT) for 9 consecutive weeks. Immediately after each training session, LLLT was applied to the femoral quadriceps muscle of both lower limbs of the TLG subjects using an infrared laser device (808 nm) with six 60-mW diodes with an energy of 0.6 J per diode and a total energy applied to each limb of 18 J. VT was determined by ergospirometry during an incremental exercise test and muscle performance was evaluated using an isokinetic dynamometer at 240°/s. Only the TLG showed a decrease in FIext in the nondominant lower limb (P = 0.016) and the dominant lower limb (P = 0.006). Both the TLG and the TG showed an increase in TWext in the nondominant lower limb (P < 0.001 and P = 0.011, respectively) and in the dominant lower limb (P < 0.000 and P < 0.000, respectively). The CG showed no reduction in FIext or TWext in either lower limb. The results suggest that an endurance training program combined with LLLT leads to a greater reduction in fatigue than an endurance training program without LLLT. This is relevant to everyone involved in sport and rehabilitation.

RESUMO

Tendon remodeling relies on extracellular matrix (ECM) restructuring by the matrix metallopeptidases (MMPs). The aim of this study was to investigate MMP-2 activity in different regions of the calcaneal tendon (CT) after resistance training (RT) in ovariectomized rats. Wistar adult female rats were grouped into sedentary (Sed-Intact), ovariectomized sedentary (Sed-Ovx), acute exercise (AcuteEx-Intact), ovariectomized acute exercise (AcuteEx-Ovx), resistance trained (ChronicEx-Intact), and ovariectomized resistance trained (ChronicEx-Ovx) (n = 10 each group). The RT protocol required the animals to climb a 1.1-m vertical ladder with weights attached to their tail. The sessions were performed once every 3 days with 4-9 climbs and 8-12 dynamic movements per scaling. The acute groups performed one session and the chronic groups underwent 12 weeks of RT. There was an increase in total MMP-2 activity in Sed-Ovx, AcuteEx-Intact, and ChronicEx-Intact compared with that in Sed-Intact in the proximal region of CT. AcuteEx-Ovx exhibited higher total MMP-2 than Sed-Ovx and AcuteEx-Intact in the distal region of CT. Chronic-Ovx presented lower total MMP-2 activity than Sed-Ovx and Chronic-Intact in the distal region of tendon. The active MMP-2 was higher for the AcuteEx-Ovx than Sed-Ovx and AcuteEx-Intact in proximal region of tendon. There was higher active MMP-2 in the distal region of tendon in the Acute-Ovx than in the Sed-Ovx and AcuteEx-Intact. Ovariectomy and resistance exercise modulate MMP-2 activity according to specific tendon region, indicating a differentiated tissue remodeling.

RESUMO

PURPOSES: There is evidence that the risk of colon cancer is reduced by appropriate levels of physical exercise. Nevertheless, the mechanisms involved in this protective effect of exercise remain largely unknown. Inflammation is emerging as a unifying link between a range of environment exposures and neoplastic risk. The carcinogen dimethyl-hydrazine (DMH) induces an increase in epithelial cell proliferation and in the expression of the inflammation-related enzyme cyclooxigenase-2 (COX-2) in the colon of rats. Our aim was to verify whether these events could be attenuated by exercise. METHODS: Four groups of eight Wistar rats were used in the experiment. The groups G1 and G3 were sedentary (controls), and the groups G2 and G4 were submitted to 8 wk of swimming training, 5 d.wk. The groups G3 and G4 were given subcutaneous injections of DMH immediately after the exercise protocols. Fifteen days after the neoplasic induction, the rats were sacrificed and the colon was processed for histological examination and immunohistochemistry staining of proliferating cell nuclear antigen (PCNA) and COX-2. RESULTS: We found a significant increase in the PCNA-labeling index in both DMH-treated groups of rats. However, this increase was significantly attenuated in the training group G4 (P < 0.01). Similar results were observed in relation to the COX-2 expression. CONCLUSIONS: From our findings, we conclude that exercise training exerts remarkable antiproliferative and antiinflammatory effects in the rat colonic mucosa, suggesting that this may be an important mechanism to explain how exercise protects against colonic cancer.

RESUMO

BACKGROUND/OBJECTIVES: Regular exercise training is effective to altering many markers of metabolic syndrome and its effects are strongly influenced by the type of consumed diet. Nowadays, resistance training (RT) has been frequently associated with low-carbohydrate high-fat diet (LCD). After long term these diets causes body weight (BW) regain with deleterious effects on body composition and metabolic risk factors. The effects of RT associated with long-term LCD on these parameters remain unexplored. We aimed to investigate the effects of RT when associated with long-term LCD on BW, feed efficiency, body composition, glucose homeostasis, liver parameters and serum biochemical parameters during BW regain period in rats. SUBJECTS/METHODS: Male Sprague-Dawley rats were fed with LCD (LC groups) or standard diet (STD) (ST groups). After 10 weeks-diet animals were separated into sedentary (Sed-LC and Sed-ST) and resistance-trained (RT-LC and RT-ST) groups (N = 8/group). RT groups performed an 11-week climbing program on a ladder with progressive load. Dual x-ray absorptiometry, glucose tolerance tests and insulin tolerance tests were performed at weeks 10 and 20. Liver and serum were collected at week 21. RESULTS: RT reduced feed efficiency, BW gain, liver fat and total and LDL cholesterol, and improved body composition and glucose clearance in animals fed on STD. In those fed with LCD, RT reduced caloric intake, BW regain, liver fat and serum triglycerides levels. However, improvement in body composition was inhibited and bone mineral density and glucose clearance was further impaired in this association. CONCLUSIONS: The LCD nullifies the beneficial effects of RT on body composition, glucose homeostasis and impairs some health parameters. Our results do not support the association of RT with LCD in a long term period.

RESUMO

In this study, we investigated the effects of resistance training (RT), caloric restriction (CR), and the association of both interventions in aortic vascular reactivity and morphological alterations, matrix metalloproteinase-2 (MMP-2) activity, insulin resistance and systolic blood pressure (SBP) in ovariectomized rats. Fifty female Holtzman rats were subjected to ovariectomy and Sham surgery and distributed into the following groups: Sham-sedentary, ovariectomized-sedentary, ovariectomized-resistance training, ovariectomized-caloric restriction, and ovariectomized-resistance training and caloric restriction groups. RT and 30% CR protocols were performed for 13 weeks. Analyses were conducted to evaluate the following: acetylcholine and sodium nitroprusside-induced relaxation of aortic rings, MMP-2 activity, insulin tolerance test, highlighting of the aorta wall cross-sectional area by hematoxylin-eosin stain, aorta vessel remodeling and SBP. We observed that ovariectomy decreased the potency of dependent and independent endothelium relaxation and MMP-2 activity, prevented insulin resistance, promoted aorta vessel remodeling in the cross-sectional area, and promoted the media-to-lumen ratio, the collagen content, and the alteration of the structure and elastic fibers of the vessel. The effects of the ovariectomy could contribute to SBP increases. However, the association of exercise and diet improved the relaxation potency in dependent and independent endothelium relaxation, elevated MMP-2 activity, ameliorate insulin sensitivity, increased the aorta cross-sectional area and media-to-lumen ratio, decreased collagen content and promoted histological parameters of the aorta vessel wall, preventing the increase of SBP. CONCLUSION: Our study revealed that the RT and CR separately, and even associatively, improved vascular function, activated MMP-2, and produced a beneficial hypertrophic remodeling, preventing the elevation of SBP in ovariectomized rats.

RESUMO

Estrogen deficiency is directly related to central obesity and low-grade inflammation. Hormonal replacement and exercise training are both able to decrease fat accumulation and inflammation in postmenopausal women. However, the efficiency of resistance training (RT) and estrogen replacement (ER) in minimizing adiposity and inflammation in the visceral adipose tissue (VAT) of ovariectomized (OVX) rats has not yet been elucidated. In this study, Sprague-Dawley rats were divided into the following 6 groups: sham-operated sedentary (Sham-Sed), OVX-Sed, Sham-RT, OVX-RT, OVX-Sed-ER, and OVX-RT-ER groups. ER was performed by implanting silastic capsules containing 17ß-estradiol. For RT, the animals were required to climb a 1.1-m vertical ladder with conical flasks containing weights attached to their tails for 12 weeks. Histological analyses were used to evaluate morphological changes. Gene expression levels were determined by quantitative real-time reverse transcriptase polymerase chain reaction, and protein concentrations were determined using Multiplex/Luminex assays. Ovariectomy increased the body mass (BM), adipocyte area, and inflammation in the VAT, the latter of which was indicated by reduced interleukin-10 (48%) and increased tumor necrosis factor (TNF)-α concentration (∼3%). RT efficiently decreased BM, adipocyte area, and inflammation in the OVX groups. The combination of RT and ER decreased BM (19%) and the TNF-α concentration (18%) and increased the gene and protein expression levels of adiponectin (173% and 18%). These results indicate that RT and the combination of RT and ER are efficient strategies for reducing the BM and improving the inflammatory status of OVX rats.

RESUMO

OBJECTIVE: The objective of this study was to assess the effects of resistance training on oxidative stress markers in the livers of ovariectomized rats. METHOD: Adult Sprague-Dawley rats were divided into the following four groups (n = 8 per group): sham-operated sedentary, ovariectomized sedentary, sham-operated resistance training, and ovariectomized resistance training. During the resistance training period, the animals climbed a 1.1-m vertical ladder with weights attached to their tails; the sessions were conducted 3 times per week, with 4-9 climbs and 8-12 dynamic movements per climb. The oxidative stress was assessed by measuring the levels of reduced glutathione and oxidized glutathione, the enzymatic activity of catalase and superoxide dismutase, lipid peroxidation, vitamin E concentrations, and the gene expression of glutathione peroxidase. RESULTS: The results showed significant reductions in the reduced glutathione/oxidized glutathione ratio (4.11±0.65 nmol/g tec), vitamin E concentration (55.36±11.11 nmol/g), and gene expression of glutathione peroxidase (0.49±0.16 arbitrary units) in the livers of ovariectomized rats compared with the livers of unovariectomized animals (5.71±0.71 nmol/g tec, 100.14±10.99 nmol/g, and 1.09±0.54 arbitrary units, respectively). Moreover, resistance training for 10 weeks was not able to reduce the oxidative stress in the livers of ovariectomized rats and induced negative changes in the hepatic anti-oxidative/oxidative balance. CONCLUSION: Our findings indicate that the resistance training program used in this study was not able to attenuate the hepatic oxidative damage caused by ovariectomy and increased the hepatic oxidative stress.

RESUMO

It is well established that atherogenic dyslipidemia, characterized by high levels of triglycerides (TG), total cholesterol (TC), and low-density lipoprotein (LDL) cholesterol and low levels of high-density lipoprotein (HDL) cholesterol, constitutes important risk factors for cardiovascular disease. Regular exercise has been associated with a reduced risk for metabolic diseases. However, studies supporting the concept that resistance exercise is a modifier of blood lipid parameters are often contradictory. The aim of this study was to investigate the effects of high-intensity resistance exercise on the serum levels of TG, TC, HDL and non-HDL cholesterol, glucose, and the liver function enzymes alanine aminotransferase (ALT, EC 2.6.1.2) and aspartate aminotransferase (AST, EC 2.6.1.1) in golden Syrian hamsters (Mesocricetus auratus (Waterhouse, 1839)) fed a hypercholesterolemic diet. Sedentary groups (S) and exercise groups (E) were fed a standard diet (SS and ES) or a cholesterol-enriched diet (standard plus 1% cholesterol, SC and EC). Resistance exercise was performed by jumps in the water, carrying a load strapped to the chest, representing 10 maximum repetitions (10 RM, 30 s rest, five days per week for five weeks). Mean blood sample comparisons were made by ANOVA + Tukey or ANOVA + Kruskal-Wallis tests (p < 0.05) to compare parametric and nonparametric samples, respectively. There were no differences in blood lipids between the standard diet groups (SS and ES) (p > 0.05). However, the EC group increased the glucose, non-HDL, and TC levels in comparison with the ES group. Moreover, the EC group increased the TG levels versus the SC group (p < 0.05). In addition, the ALT levels were increased only by diet treatment. These findings indicated that high-intensity resistance exercise contributed to dyslipidemia in hamsters fed a hypercholesterolemic diet, whereas liver function enzymes did not differ in regards to the exercise protocol.

RESUMO

Exercise can generate alterations in body composition and modulate the immune system. The objective of this study was to verify whether a circuit resistance training (CRT) protocol can increase lean body mass (LM), and reduce fat body mass (FM) and the percent of FM (%FM) of sedentary women, without inducing inflammatory responses, indicated by serum cytokine levels. The initial hypothesis was that CRT would improve body composition, without changing serum cytokine levels. The study consisted of 14 healthy, sedentary women, aged 33-45 years (mean +/- SD, 40.23 +/- 3.98 years), with a normal body mass index. They participated in 3 sessions per week of CRT, which included 2 rounds in 9 stations with 1 set of 8-12 repetition maximum at each station, for 10 weeks. During the 10-week CRT period, participants maintained their pretraining nutritional standard. Body composition was analysed with dual-energy X-ray absorptiometry both pre- and post-training. Blood samples were collected after 96 h of rest pre- and post-training, and 5 min, 24 h, and 48 h after the second and last training sessions to measure serum cytokine levels by flow cytometry. The nutritional standard was accompanied throughout the study period with 24-h dietary recall. Increases in LM (35.937 +/- 4.926 to 39.130 +/- 4.950 kg) and decreases in FM (21.911 +/- 8.150 to 17.824 +/- 4.235 kg) and %FM (37.10 +/- 10.84 to 31.19 +/- 6.06), without concurrent changes in serum cytokine levels, and in the nutritional standard (alpha = 0.05). The proposed CRT improved body composition and did not induce any changes in serum cytokine levels characteristic of the inflammatory response in women.

The position of lower limbs during lunge is different from squat without clear understanding on differences in muscle activation. The objective was to compare the electromyographic activity of the vastus lateralis (VL), vastus medialis (VM), biceps femoris (BF) and semitendinosus (ST) during the execution of the lunge to exhaustion of the lower limb, changing position of the legs frontally and posteriorly. Nine active women participated of this study with a mean (SD) age of 22 (3.4) years and body mass 60.3 (4.1) kg. The lunge was divided in two phases, changing the forward leg (random order). The electromyographic signals were captured by an eletromyograph, and the root mean square (RMS) values were analyzed during the concentric phase. There was a significant increase of the RMS in time for the LL positioned frontally and posterior (p< 0,001). For the forward position, the increase of RMS was 50 percent for VL, 54 percent for VM and 48 percent for BF. The backward position had an increase of 75 percent for VL, 113 percent for VM, 62 percent for BF and 48 percent for ST. The RMS was significantly higher for VM than for ST the forward position (p = 0,03) and ST and BF for the backward position (p = 0,02). No interaction was observed between time and muscle in the electromyigraphic activity. The normalized RMS was not statistically different comparing the forward and backward position of the dominant leg. Muscle activity was similar in both positions of the LL, showing increased activation of the VM and VL muscles in relation to the BF and ST.

The aim of this study was to investigate the influence of gender on Tthre and VO2 response during running exercise performed at vVO2max. Therefore, eighteen untrained individuals (9 male and 9 female) with normal weight and aged between 20 - 30 years (VO2max = 42.66 ± 4.50 vs 32.92 ± 6.03 mL.kg-1.min-1 and vVO2max = 13.2 ± 1.5 vs 10.3 ± 2.0 km.h-1, for male and female, respectively) were assessed. Subjects performed two exercise tests on treadmill. First one was an incremental test to determine VO2max, velocity at VO2max (vVO2max) and second test was performed at steady velocity - vVO2max - until exhaustion. The threshold time (Tthre) and VO2 kinetics response was determined. No significant differences were observed between men and women for time constant (τ) (35.76 ± 21.03 vs 36.5 ± 6.21s, respectively; P = 0.29); Tthre (308 ± 84.3 vs 282.11 ± 57.19s, respectively; P = 0.68), time to achieve VO2max (TAVO2max) (164.48 ± 96.73 vs 167.88 ± 28.59s, respectively; P = 0.29), time to achieve VO2max in Tthre percentage ( percentTthre) (50.24 ± 16.93 vs 62.63 ± 16.60 percent, respectively; P = 0.19); time maintained at VO2max (TMVO2max) (144.08 ± 42.55 vs 114.23 ± 76.96s, respectively; P = 0.13). These results suggest that the VO2 kinetics response and Tthre is similar between untrained men and women at the vVO2max.

The purpose of this study was to verify the effect of a sixteen weeks training program on sedentary individuals. Were included, 40 sedentary individuals, 20 male and 20 female, between 35-55 years old. The training program included 3 weekly one hour bouts, the sessions were divided in 30 minutes of aerobic exercise and 30 minutes of resistance exercise. For body fat percentage (%G), fat mass (Kg), lean body mass and the lean/fat mass ratio estimation, were used the JACKSON et al. (1980) equation for women and JACKSON; POLLOCK (1978) equation for men. The statistical analysis was done by test t of student with significance level of 5%. The sixteen weeks training program had positive effects on %G, reducing the inicial average value from 27,4% to 23,6% among males, and from 33,1% to 29,6% among females. The fat free mass was maintained in both sexes. There were significant reductions (p<0.05), in the abdominal and waist circunference, in male (abdominal= from 102cm to 98,5cm after training and waist= from 98cm to 94,5cm after training); and in women (abdominal= from 84cm to 80cm and waist= from 80cm to 76cm). In conclusion, the sixteen weeks training program could result in benefic effects in body composition of the participants in the study and is an important tool in the search of a health life quality and lifespan.

The purpose of this study was to verify the possibility of identifying the blood glucose threshold (GT) as well as to compare and correlate the GT with the lactate threshold (LT) during resistance exercises. Twelve healthy male volunteers aged 24.4 ± 1.2 years and adapted to resistance training were submitted to an incremental resistance exercise with graded intensities according to their maximal workload (kg) performed for 1 repetition (1RM) on both leg press (LP) and bench press (BP). The intensities applied for each 1 min stage were of 10 percent, 20 percent, 25 percent, 30 percent, 35 percent, 40 percent, 50 percent, 60 percent, 70 percent, 80 percent and 90 percent of 1RM, respectively, or until volitional exhaustion. Blood lactate and glucose measurements were done each 2 min rest between each stage (YSI 2300 S). The blood lactate and glucose responses were similar for both exercises. No differences were verified for the relative intensity ( percent 1RM) at which the inflection point of blood lactate and glucose curves were observed respectively for LP (36.6 ± 1.4 percent and 32.9 ± 1.5 percent) and BP (31.2 ± 1.2 percent and 31.2 ± 1.8 percent) (p > 0.05). Additionally, a high correlation was verified between LT and GT identified both on LP (r = 0.80) and SR (r = 0.73) (p < 0,05). It was concluded that it is possible to identify the LT and GT on resistance exercises. However, additional studies should investigate the meaning of these thresholds and their validity for exercise evaluation and training prescription.
