RESUMO

Sensors, the underlying technology that supports the Internet of Things, are undergoing multi-disciplinary integration development to constantly improve the efficiency of human production and life. Simultaneously, the application scenarios in emerging fields such as medical diagnosis, environmental monitoring and industrial safety put forward higher requirements for sensing capabilities. Over the last decade, single-atom catalysts (SACs) have attracted tremendous attention in fields such as environment and energy due to their high atom utilization efficiencies, controllable active sites, tailorable coordination environments and structural/chemical stability. These extraordinary characteristics extend the sensitivity and selectivity of sensors beyond their current limitations. Here, we start with the working principles of SAC-based sensors, and summarize the relationship between sensor performance and intrinsic properties of SACs, followed by an overview of the design strategy development. We then review the recent advances in SAC-based sensors in different fields and highlight the future opportunities and challenges in their exciting applications.

Anaerobic co-fermentation is a favorable way to convert agricultural waste, such as swine manure (SM) and apple waste (AW), into lactic acid (LA) through microbial action. However, the limited hydrolysis of organic matter remains a main challenge in the anaerobic co-fermentation process. Therefore, this work aims to deeply understand the impact of cellulase (C) and protease (P) ratios on LA production during the anaerobic co-fermentation of SM with AW. Results showed that the combined use of cellulase and protease significantly improved the hydrolysis during the enzymatic pretreatment, thus enhancing the LA production in anaerobic acidification. The highest LA reached 41.02 ± 2.09 g/L within 12 days at the ratio of C/P = 1:3, which was approximately 1.26-fold of that in the control. After a C/P = 1:3 pretreatment, a significant SCOD release of 45.34 ± 2.87 g/L was achieved, which was 1.13 times the amount in the control. Moreover, improved LA production was also attributed to the release of large amounts of soluble carbohydrates and proteins with enzymatic pretreated SM and AW. The bacterial community analysis revealed that the hydrolytic bacteria Romboutsia and Clostridium_sensu_stricto_1 were enriched after enzyme pretreatment, and Lactobacillus was the dominant bacteria for LA production. This study provides an eco-friendly technology to enhance hydrolysis by enzymatic pretreatment and improve LA production during anaerobic fermentation.

Perilla as herb, ornamental, oil and edible plant is widely used in East Asia. Until now, the mechanism of regulated leaf coloration is still unclear. In this study, four different kinds of leaf colors were used to measure pigment contents and do transcriptome sequence to postulate the mechanism of leaf coloration. The measurements of chlorophyll, carotenoid, flavonoid, and anthocyanin showed that higher contents of all the aforementioned four pigments were in full purple leaf 'M357', and they may be determined front and back leaf color formation with purple. Meanwhile, the content of anthocyanin was controlled back leaf coloration. The chromatic aberration analysis and correlative analysis between different pigments and L*a*b* values analysis also suggested front and back leaf color change was correlated with the above four pigments. The genes involved in leaf coloration were identified through transcriptome sequence. The expression levels of chlorophyll synthesis and degradation related genes, carotenoid synthesis related genes and anthocyanin synthesis genes showed up-/down-regulated expression in different color leaves and were consistent of accumulation of these pigments. It was suggested that they were the candidate genes regulated perilla leaf color formation, and genes including F3'H, F3H, F3',5'H, DFR, and ANS are probably important for regulating both front and back leaf purple formation. Transcription factors involved in anthocyanin accumulation, and regulating leaf coloration were also identified. Finally, the probable scheme of regulated both full green and full purple leaf coloration and back leaf coloration was postulated.

Tumor metastasis is one of the main reasons for the high mortality rate associated with colorectal cancer (CRC). However, its underlying mechanisms have not been fully understood. Here, we reported that the expression of discoidin domain receptor 2 (DDR2) was significantly upregulated in CRC tissues compared to that in normal adjacent tissues. The expression level of DDR2 was negatively associated with prognosis of CRC patients. Therefore, DDR2 may play an oncogenic role in CRC development. Furthermore, DDR2 induced epithelial mesenchymal transition in CRC cells and regulated their invasive and metastatic capacity in vitro and in vivo. Mechanistically, increased DDR2 expression level activated the AKT/GSK-3ß/Slug signaling pathway. In conclusion, these findings showed that DDR2 promoted CRC metastasis and DDR2 inhibition might represent an effective therapeutic strategy for local advanced and metastatic CRC treatment.

Orientation of local cardiomyocyte aggregates in the human left ventricle free wall experiences an oscillation in the laminar structure regions, besides its gradual change trend. We described this oscillation using five transmural samples imaged at the European Synchrotron Radiation Facility with an isotropic voxel size of 3.5 × 3.5 × 3.5 µm3 . In the reconstructed volume of each sample, we manually selected a region containing a regular laminar structure as the region of interest and measured the distribution of the orientation of local cardiomyocyte aggregates inside using a Fourier-based method. Then, we extracted the gradual change part of the orientation of cardiomyocyte aggregates with a three-dimensional centered Gaussian filter and measured the angle between the original orientation vector of local cardiomyocyte aggregates and its gradual change part. Further, we assessed the measured angles in different local coordinates. The results indicate that the oscillation amplitude of the orientation of cardiomyocyte aggregates is regional in the left ventricle wall, which may promote our understanding of the rearrangement mechanism of the cardiomyocyte aggregates and provide a new biomarker to study the heart physiological status.

Micro-computed tomography (Micro-CT) is inevitably required to inspect long large objects with high resolution. It is well known that helical CT solves the so-called "long object" problem, but it requires that the measured object be strictly located in the lateral field of view (FOV). Therefore, developing a novel scanning method to extend the FOV in both the lateral and axial directions (i.e., the large helical FOV) is necessary. Recently, due to the application of linearly distributed source arrays and the characteristics of easy extension of the FOV and engineering implementation, straight-line scanning systems have attracted much attention. In this paper, we propose a segmented helical computed tomography (SHCT) based on multiple slant source-translation. SHCT can readily extend the helical FOV by adjusting the source slant translation (SST) length, pitch (or elevation of the SST trajectory), and number of scanning circles. In SHCT, each projection view is truncated laterally and axially, but the projection data set within the cylindrical FOV region is complete. To ensure reconstruction efficiency and avoid the lateral truncation, we propose a generalized backprojection-filtration (G-BPF) algorithm for SHCT approximate reconstruction. Experimental results verify the effectiveness of the proposed SHCT methods for imaging large and long objects. As the pitch decreases, the proposed SHCT methods can reconstruct competitive, high-quality volumes.

Recently, to easily extend the helical field-of-view (FOV), the segmented helical computed tomography (SHCT) method was proposed, as well as the corresponding generalized backprojection filtration (G-BPF) type algorithm. Similar to the geometric relationship between helical and circular CT, SHCT just becomes full-scan multiple source-translation CT (F-mSTCT) when the pitch is zero and the number of scan cycles is one. The strategy of G-BPF follows the idea of the generalized Feldkamp approximate cone-beam algorithm for helical CT, i.e., using the F-mSTCT cone-beam BPF algorithm to approximately perform reconstruction for SHCT. The image quality is limited by the pitch size, which implies that satisfactory quality could only be obtained under the conditions of small pitches. To extend the analytical reconstruction for SHCT, an effective single-slice rebinning (SSRB) method for SHCT is investigated here. Transforming the SHCT cone-beam reconstruction into the virtual F-mSTCT fan-beam stack reconstruction task with low computational complexity, and then some techniques are developed to address the challenges involved. By using the basic BPF reconstruction with derivating along the detector (D-BPF), our experiments demonstrate that SSRB has fewer interlayer artifacts, higher z-resolution, more uniform in-plane resolution, and higher reconstruction efficiency compared to G-BPF. SSRB could promote the effective application of deep learning in SHCT reconstruction.

The human RecQ helicase BLM is involved in the DNA damage response, DNA metabolism, and genetic stability. Loss of function mutations in BLM cause the genetic instability/cancer predisposition syndrome Bloom syndrome. However, the molecular mechanism underlying the regulation of BLM in cancers remains largely elusive. Here, we demonstrate that the deubiquitinating enzyme USP37 interacts with BLM and that USP37 deubiquitinates and stabilizes BLM, thereby sustaining the DNA damage response (DDR). Mechanistically, DNA double-strand breaks (DSB) promotes ATM phosphorylation of USP37 and enhances the binding between USP37 and BLM. Moreover, knockdown of USP37 increases BLM polyubiquitination, accelerates its proteolysis, and impairs its function in DNA damage response. This leads to enhanced DNA damage and sensitizes breast cancer cells to DNA-damaging agents in both cell culture and in vivo mouse models. Collectively, our results establish a novel molecular mechanism for the USP37-BLM axis in regulating DSB repair with an important role in chemotherapy and radiotherapy response in human cancers.

University campuses of China accommodate over 30 million students and consume a large amount of fossil fuel energy, leading to high carbon emission. Implementation of bioenergy (e.g. biomethane) is one of promising ways to mitigate emission and foster low-carbon emitting campus. Biomethane potential from anaerobic digestion (AD) of food waste (FW) in 2344 universities of 353 cities of mainland China have been estimated herein. Results have shown that 1.74 million tons of FW are discharged from campus canteens annually, that can generate 195.8 million m3 biomethane and reduce 0.77 million ton CO2-eq. Wuhan, Zhengzhou, and Guangzhou are the top three cities having the most biomethane potential from campus FW, accounting up to 8.92, 7.89, and 7.28 million m3 year-1, respectively. Technical challenges and solutions have been summarized and discussed such as FW purity, accumulation of ammonia and fatty acid, foaming, and plant site selection. Low-carbon campuses are supposed to be achieved by using bioenergy, like biomethane, in appropriate ways after resolving technical and management challenges.

This paper is to investigate the high-quality analytical reconstructions of multiple source-translation computed tomography (mSTCT) under an extended field of view (FOV). Under the larger FOVs, the previously proposed backprojection filtration (BPF) algorithms for mSTCT, including D-BPF and S-BPF (their differences are different derivate directions along the detector and source, respectively), make some errors and artifacts in the reconstructed images due to a backprojection weighting factor and the half-scan mode, which deviates from the intention of mSTCT imaging. In this paper, to achieve reconstruction with as little error as possible under the extremely extended FOV, we combine the full-scan mSTCT (F-mSTCT) geometry with the previous BPF algorithms to study the performance and derive a suitable redundancy-weighted function for F-mSTCT. The experimental results indicate FS-BPF can get high-quality, stable images under the extremely extended FOV of imaging a large object, though it requires more projections than FD-BPF. Finally, for different practical requirements in extending FOV imaging, we give suggestions on algorithm selection.

The rapid development of information technology has made the amount of information in massive texts far exceed human intuitive cognition, and dependency parsing can effectively deal with information overload. In the background of domain specialization, the migration and application of syntactic treebanks and the speed improvement in syntactic analysis models become the key to the efficiency of syntactic analysis. To realize domain migration of syntactic tree library and improve the speed of text parsing, this paper proposes a novel approach-the Double-Array Trie and Multi-threading (DAT-MT) accelerated graph fusion dependency parsing model. It effectively combines the specialized syntactic features from small-scale professional field corpus with the generalized syntactic features from large-scale news corpus, which improves the accuracy of syntactic relation recognition. Aiming at the problem of high space and time complexity brought by the graph fusion model, the DAT-MT method is proposed. It realizes the rapid mapping of massive Chinese character features to the model's prior parameters and the parallel processing of calculation, thereby improving the parsing speed. The experimental results show that the unlabeled attachment score (UAS) and the labeled attachment score (LAS) of the model are improved by 13.34% and 14.82% compared with the model with only the professional field corpus and improved by 3.14% and 3.40% compared with the model only with news corpus; both indicators are better than DDParser and LTP 4 methods based on deep learning. Additionally, the method in this paper achieves a speedup of about 3.7 times compared to the method with a red-black tree index and a single thread. Efficient and accurate syntactic analysis methods will benefit the real-time processing of massive texts in professional fields, such as multi-dimensional semantic correlation, professional feature extraction, and domain knowledge graph construction.

BACKGROUND: Emerging evidence has identified miR-138 as a tumor suppressor that can suppress the proliferation of various cancers. Meanwhile, the cause of abnormal miR-138 expression in cervical cancer remains uncertain. This study clarified the mechanism by which miR-138 regulates proliferation, invasion, metastasis, and EMT in cervical cancer cells. RESULTS: miR-138 expression in human cervical cancer and adjacent normal tissue was measured using qPCR. SiHa and C33A cells were used to determine the function of miR-138 via miR-138 mimic or inhibitor transfection, followed by wound healing, Cell Counting Kit-8, flow cytometry, and Transwell assays. Epithelial and mesenchymal marker expression was analyzed using Western blotting. DNA methylation in the miR-138 promoter was examined using bisulfite sequencing PCR. The downstream target genes of miR-138 were identified via bioinformatics analysis and luciferase reporter assays. A tumor xenograft model was employed to validate DNA methylation-induced miR-138 downregulation and tumor growth inhibition in cervical cancer in vivo. miR-138 levels were significantly lower in cervical cancer tissues than in adjacent control tissues. Furthermore, lower miR-138 expression and higher CpG methylation in the miR-138 promoter were identified in lymph node-positive metastatic cervical cancer tumors versus that in non-metastatic tumor tissues. Upon miR-138 overexpression, cell proliferation, metastasis, invasion, and EMT were suppressed. miR-138 agomir transfection and demethylating drug treatment significantly inhibited cervical tumor growth and EMT in tumor xenograft models. DNA methylation inhibited miR-138 transcription, and enhancer of zeste homolog 2 (EZH2) downregulation mediated the tumor suppressor function of miR-138 in cervical cancer. CONCLUSION: We demonstrated that miR-138 suppresses tumor progression by targeting EZH2 in cervical cancer and uncovered the role of DNA methylation in the miR-138 promoter in its downregulation. These findings demonstrated the potential of miR-138 to predict disease metastasis and/or function as a therapeutic target in cervical cancer.

Pork accounts for an important proportion of livestock products. For pig farming, a lot of manpower, material resources and time are required to monitor pig health and welfare. As the number of pigs in farming increases, the continued use of traditional monitoring methods may cause stress and harm to pigs and farmers and affect pig health and welfare as well as farming economic output. In addition, the application of artificial intelligence has become a core part of smart pig farming. The precision pig farming system uses sensors such as cameras and radio frequency identification to monitor biometric information such as pig sound and pig behavior in real-time and convert them into key indicators of pig health and welfare. By analyzing the key indicators, problems in pig health and welfare can be detected early, and timely intervention and treatment can be provided, which helps to improve the production and economic efficiency of pig farming. This paper studies more than 150 papers on precision pig farming and summarizes and evaluates the application of artificial intelligence technologies to pig detection, tracking, behavior recognition and sound recognition. Finally, we summarize and discuss the opportunities and challenges of precision pig farming.

Exposure to acidic and alkaline conditions were found to cause the excess accumulation of reactive oxygen species in tree peony, thereby causing damage and inhibiting plant growth and development. The activities of antioxidant enzymes were also found to be significantly up-regulated, especially under alkaline conditions; this explained why tree peony is better adapted to alkaline than to acidic conditions. Through pairwise comparisons, 144 differentially expressed genes (DEGs) associated with plant growth, photosynthesis, and stress were identified. The DEGs related to stress were up-regulated, whereas the remaining DEGs were almost all down-regulated after acid and alkaline treatments. The nutrient assimilation was greatly inhibited. Chlorophyll synthesis genes were suppressed, and chlorophyll content was reduced. The development and structures of stomata and chloroplasts and the transcription of related genes were also influenced. Among photosynthesis-related DEGs, electron transport chains were the most sensitive. The suppressed expression of photosynthesis genes and the reduced light-harvesting capacity, together with the impairment of chloroplasts and stomata, finally led to a sharp decrease in the net photosynthetic rate. Carbohydrate accumulation and plant biomass were also reduced. The present study provides a theoretical basis for the response mechanisms of tree peony to adverse pH conditions and enriches knowledge of plant adaptation to alkaline conditions.

Itoh hybrids are intersectional hybrids in Paeonia L. with sect. Moutan and sect. Paeonia as paternal and maternal parents, respectively. Therefore, these hybrids have herbaceous stems with improved ornamental value introduced by the paternal parent. Although both of their parents are diploids, Itoh hybrids are triploids. Moreover, the parental origin of their chromosomes has not been extensively studied. This study systematically analyzed the genome size, ploidy, and karyotype of Itoh hybrids and compared them with their parental taxa. Although the monoploid genome size of Itoh hybrids was different, it was not significantly different from that of the parents. However, the size of varieties in the two parental taxa was significantly different from the wild species, probably due to genome rearrangements caused by artificial selection. Further karyotype analysis, correlation analysis, and hierarchical clustering could not identify the parental origin of chromosomes in Itoh hybrids. Verification through genomic and fluorescence in situ hybridization (GISH and FISH) suggested that for the three sets of chromosomes in Itoh hybrids, two were from the paternal parent, and one was from the maternal parent. One of the first two sets was from wild species, and the other from a cultivated variety. GISH could not label the chromosomes of cultivated peonies from the sect. Moutan, probably due to the huge and complex genomes compared with the wild species. Meanwhile, 5S rDNA-based FISH was first applied in Paeonia, which may be used for ploidy assessment. This work may give insights into the utilization of Itoh hybrid resources.

Low propagation rate is the primary problem that limits industry development of tree peony. In this study, a highly efficient regeneration system for tree peony using somatic embryogenesis (SE) was established. The transcriptomes of zygotic embryo explants (S0), non-embryonic callus (S1), embryonic callus (S2), somatic embryos (S3), and regenerated shoots (S4) were analyzed to determine the regulatory mechanisms that underlie SE in tree peony. The differentially expressed genes (DEGs) were identified in the pairwise comparisons of S1-vs-S2 and S1-vs-S3, respectively. The enriched DEGs were primarily involved in hormone signal transduction, stress response and the nucleus (epigenetic modifications). The results indicated that cell division, particularly asymmetric cell division, was enhanced in S3. Moreover, the genes implicated in cell fate determination played central roles in S3. Hormone signal pathways work in concert with epigenetic modifications and stress responses to regulate SE. SERK, WOX9, BBM, FUS3, CUC, and WUS were characterized as the molecular markers for tree peony SE. To our knowledge, this is the first study of the SE of tree peony using transcriptome sequencing. These results will improve our understanding of the molecular mechanisms that underly SE in tree peony and will benefit the propagation and genetic engineering of this plant.

RESUMO

Gibberellin (GA) is frequently used in tree peony forcing culture, but inappropriate application often causes flower deformity. Here, 5-azacytidine (5-azaC), an efficient DNA demethylating reagent, induced tree peony flowering with a low deformity rate by rapidly inducing PsFT expression, whereas GA treatment affected various flowering pathway genes with strong pleiotropy. The 5-azaC treatment, but not GA, significantly reduced the methylation level in the PsFT promoter with the demethylation of five CG contexts in a 369 bp CG-rich region, and eight light-responsive related cis-elements were also predicted in this region, accompanied by enhanced leaf photosynthetic efficiency. Through GO analysis, all methylation-closer differentially expressed genes (DEGs) were located in the thylakoid, the main site for photosynthesis, and were mainly involved in response to stimulus and single-organism process, whereas GA-closer DEGs had a wider distribution inside and outside of cells, associated with 12 categories of processes and regulations. We further mapped five candidate DEGs with potential flowering regulation, including three kinases (SnRK1, WAK2, and 5PTase7) and two bioactive enzymes (cytochrome P450 and SBH1). In summary, 5-azaC and GA may have individual roles in inducing tree peony flowering, and 5-azaC could be a preferable regulation approach; DNA demethylation is suggested to be more focused on flowering regulation with PsFT playing a core role through promoter demethylation. In addition, 5-azaC may partially undertake or replace the light-signal function, combined with other factors, such as SnRK1, in regulating flowering. This work provides new ideas for improving tree peony forcing culture technology.

From April to June 2019, poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) (P3(HA)) microbead samples were exposed to an operational wastewater reclamation facility (WWRF) in an aerobic aeration basin in Athens, Georgia. Samples were withdrawn from the facility over a 13-week timeframe, and the particles were examined by Raman microscopy and thermogravimetric analysis/mass spectroscopy (TGA/MS) coupled with differential scanning calorimetry (DSC). The activated sludge from this facility was also used as an inoculum to examine carbon mineralization under controlled respirometry experiments to corroborate biological degradation rates determined from both the environmental and laboratory approach. Respirometry, Raman microscopy, and TGA/MS-DSC methods all measured similar biodegradation timelines for microbeads bound to an epoxy substrate, indicating that the three methods are temporally comparable and may be used to measure material biological degradation. Samples of epoxy-bound P3(HA) microbeads, free microbeads, the P3(HA) film, and poly(lactic acid) (PLA) film demonstrated carbon mineralization of 90.0, 89.4, 95.0, and 8.15%, respectively, relative to the cellulose positive control. Using a modified Gompertz growth model, the biological degradation rate coefficients (Rm) were determined for cellulose, P3(HA) film, epoxy-bound P3(HA) microbeads, and free P3(HA) microbeads and found to be 31.6, 30.2, 17.5, and 18.7 mL CO2·g-1·day-1, respectively. Moreover, P3(HA) microbeads can efficiently mineralize in WWRF infrastructure at a rate comparable to cellulose.

BACKGROUND: Feed accounts for about 70% of the total cost of poultry meat production. Residual feed intake (RFI) has become the preferred measure of feed efficiency because it is phenotypically independent of growth rate and body weight. In this study, our aim was to estimate genetic parameters and identify quantitative trait loci (QTL) for feed efficiency in 3314 purebred broilers using a genome-wide association study. Broilers were genotyped using a custom 55 K single nucleotide polymorphism (SNP) array. RESULTS: Estimates of genomic heritability for seven growth and feed efficiency traits, including body weight at 28 days of age (BW28), BW42, average daily feed intake (ADFI), RFI, and RFI adjusted for weight of abdominal fat (RFIa), ranged from 0.12 to 0.26. Eleven genome-wide significant SNPs and 15 suggestively significant SNPs were detected, of which 19 clustered around two genomic regions. A region on chromosome 16 (2.34-2.66 Mb) was associated with both BW28 and BW42, and the most significant SNP in this region, AX_101003762, accounted for 7.6% of the genetic variance of BW28. The other region, on chromosome 1 (91.27-92.43 Mb) was associated with RFI and ADFI, and contains the NSUN3 and EPHA6 as candidate genes. The most significant SNP in this region, AX_172588157, accounted for 4.4% of the genetic variance of RFI. In addition, a genomic region containing the gene AGK on chromosome 1 was found to be associated with RFIa. The NSUN3 and AGK genes were found to be differentially expressed in breast muscle, thigh muscle, and abdominal fat between male broilers with high and low RFI. CONCLUSIONS: We identified QTL regions for BW28 and BW42 (spanning 0.32 Mb) and RFI (spanning 1.16 Mb). The NSUN3, EPHA6, and AGK were identified as the most likely candidate genes for these QTL. These genes are involved in mitochondrial function and behavioral regulation. These results contribute to the identification of candidate genes and variants for growth and feed efficiency in poultry.

A circular grating angle encoder is a key component in the dynamic torque calibration system. To improve the accuracy of an angle measurement, in this paper, the source of the angle measurement error of the circular grating is analyzed; an eccentricity error model and an inclination error model are proposed, respectively; further, these two models are combined to establish a total error model. Through the simulation study with the models, the conditions, in which the eccentricity error or inclination error can be ignored, are discussed. The calibration and compensation methods of the angle measurement error are given, and a progressive error compensation function which integrates the first harmonic fitting and the second harmonic fitting is obtained. An experiment is performed to verify the proposed calibration and compensation methods. The peak-to-peak value of the compensated angle measurement error of the single reading head can be reduced by about 93.76%, which approximates to the error of the mean value of the double reading heads. The experimental results show that the error calibration and compensation method based on the proposed error model can effectively compensate the angle measurement error of the circular grating with a single reading head, and obtain a high-precision measurement angle.
