BACKGROUND: Lauric acid (LA), a major, natural, medium-chain fatty acid, is considered an efficient energy substrate for intense exercise and in patients with long-chain fatty acid ß-oxidation disorders. However, few studies have focused on the role of LA in exercise performance and related glucolipid metabolism in vivo. OBJECTIVES: We aimed to investigate the effect of dietary supplementation with LA on exercise performance and related metabolic mechanisms. METHODS: Male C57BL/6N mice (14 wk old) were fed a basal diet or a diet containing 1% LA, and a series of exercise tests, including a high-speed treadmill test, aerobic endurance exercises, a 4-limb hanging test, and acute aerobic exercises, were performed. RESULTS: Dietary supplementation with 1.0% LA accelerated the recovery from fatigue after explosive exercise (P < 0.05) and improved aerobic endurance and muscle strength in sedentary mice (P = 0.039). Lauric acid intake not only changed muscle fatty acid profiles, including increases in C12:0 and n-6/n-3 PUFAs (P < 0.001) and reductions in C18:0, C20:4n-6, C22:6n-3, and n-3 PUFAs (P < 0.05) but also enhanced fat mobilization from adipose tissue and fatty acid oxidation in the liver, at least partly via the AMP-activated protein kinase-acetyl CoA carboxylase pathway (P < 0.05). Likewise, LA supplementation promoted liver glyconeogenesis and conserved muscular glycogen during acute aerobic exercise (P < 0.05), which was accompanied by an increase in the mitochondrial DNA copy number and Krebs cycle activity in skeletal muscle (P < 0.05). CONCLUSIONS: Dietary supplemental LA serves as an efficient energy substrate for sedentary mice to improve aerobic exercise endurance and muscle strength through regulation of glucolipid metabolism. These findings imply that LA supplementation might be a promising nutritional strategy to improve aerobic exercise performance in sedentary people.

The new energy vehicle industry is booming, but the subsequent problem of vehicle power batteries' "scrap tide" is still severe. How to establish and improve the end-of-life power battery recycling system to avoid the "catastrophic" environmental consequences has become an urgent global problem needing a solution. This article constructs three recycling models for manufacturer recycling, retailer recycling, and mixed recycling. By using Stackelberg game and market real data, the influence of carbon trading policy outside the supply chain, power battery endurance capacity and advertising effects within the supply chain on the selection of recycling channels was studied. The results showed: (1) Different recycling channels did not affect the wholesale price, retail price, and market demand for raw material power batteries in the positive supply chain; (2) The total profit function of manufacturers and retailers had a "U-shaped" non-linear relationship with power battery endurance capacity and has a positive linear relationship with the advertising effect. Taking the R&D endurance capacity of 0.4 and the total endurance capacity of 62 kWh as the lowest dividing point, it will decrease first and then increase; (3) The increase in the recycling competition coefficient had a greater impact on the consumption of carbon emission rights in the mixed recycling model than on savings in carbon emission rights, and retailers were the indirect "victims" of rising carbon trading prices; (4) Endurance capacity, advertising effects, and carbon trading prices determined the economics of the recycling model and the carbon emission reduction potential. Manufacturers, retailers, and governments can refer to the value range of each variable to select the most appropriate recycling mode.

Objectives: The purpose was to investigate the effects of hot and humid environments on thermoregulation and aerobic endurance capacity and whether high skin temperature serves as a more important thermoregulatory factor affecting aerobic exercise capacity. Methods: A randomized cross-over design was applied to this study, in which nine Laser sailors performed the 6 km rowing test (6 km test) in both a warm (ambient temperature: 23 ± 1.4 °C; relative humidity: 60.5 ± 0.7%; wind speed: 0 km/h; WARM) and hot environment (ambient temperature: 31.8 ± 1.1 °C; relative humidity: 63.5 ± 4.9%; wind speed: 3.5 ± 0.7 km/h; HOT). Results: The time for completing 6 km test of HOT group was significantly longer than that of WARM group (P = 0.0014). Mean power of 3-4 km, 4-5 km and 5-6 km were significant lower in HOT group (P = 0.014, P = 0.02, P = 0.003). Gastrointestinal temperature and skin temperature were significantly higher in HOT group during the 6 km test (P = 0.016, P = 0.04). Heat storage at 5 min and 15 min of HOT group were significantly higher than that of WARM group (P = 0.0036; P = 0.0018). Heart rate and physiological strain index of HOT group were significantly higher than that of WARM group during the 6 km test (P = 0.01, P < 0.01). Conclusion: When skin temperature and core temperature both increased, high skin temperature may be the more important thermoregulatory factor that affected the aerobic endurance performance in hot and humid environments. The high skin temperature narrowed the core to skin temperature gradient and skin to ambient temperature gradient, which may result in greater accumulation of heat storage. The greater heat storage led to the lower muscle power output, which contributed to the reduction of the heat production.

Objective: Physical activity is a significant health determinant and is likely to be influenced by social-distancing rules imposed by authorities during the COVID-19 pandemic. This study explored gender-based differences in physical activity levels (PALs) and associated factors amid COVID-19 pandemic in adolescents. Methods: In this prospective analysis, 112 healthy adolescents (15.63 ± 1.21 years) participated. They were assessed at the baseline (before the announcement of COVID-19 as a global pandemic) for anthropometry, fitness status, and PALs (baseline-PALs), and next at the follow-up (three months of imposed social-distancing rules) for PALs (follow-up-PALs) over an internet-based platform through the Physical Activity Questionnaire for Adolescents. Results: A significant PALs decline was evidenced for the whole sample (P < .001) and such a decline was predominantly affected by significant PALs reduction among males (P < .001) rather than in females (P = .06). The fitness variables (explosive power, muscle endurance, flexibility, and aerobic endurance) were consistently positively correlated with the baseline- and follow-up-PALs in both genders with particularly prominent associations of PALs with explosive power and aerobic endurance. Conclusion: Physical literacy is necessary for PALs preservation, given the remarkable PALs decreases among adolescents during challenging circumstances such as the COVID-19 pandemic.


Objectif: L'activité physique est un déterminant important de la santé et est susceptible d'être influencée par les règles de distanciation sociale imposées par les autorités pendant la pandémie de COVID-19. Cette étude a exploré les différences basées sur le sexe dans les niveaux d'activité physique (PAL) et les facteurs associés au milieu de la pandémie de COVID-19 chez les adolescents. Méthodes: Dans cette analyse prospective, 112 adolescents en bonne santé (15,63 ± 1,21 ans) ont participé. Ils ont été évalués au départ (avant l'annonce du COVID-19 en tant que pandémie mondiale) pour l'anthropométrie, le statut de forme physique et les PAL (PAL de référence), et ensuite lors du suivi (trois mois de règles de distanciation sociale imposées) pour les PAL (suivi-PAL) sur une plate-forme internet via le questionnaire sur l'activité physique pour adolescents. Résultats: Une baisse significative des PAL a été mise en évidence pour l'ensemble de l'échantillon (p < ,001) et une telle baisse a été principalement affectée par une réduction significative des PAL chez les hommes (p < ,001) plutôt que chez les femmes (p = ,06). Les variables de forme physique (puissance explosive, endurance musculaire, flexibilité et endurance aérobie) étaient constamment corrélées positivement avec les PAL de référence et de suivi chez les deux sexes avec des associations particulièrement importantes de PAL avec la puissance explosive et l'endurance aérobie. Conclusion: La littératie physique est nécessaire pour la préservation des PAL, étant donné les diminutions remarquables des PAL chez les adolescents lors de circonstances difficiles telles que la pandémie de COVID-19.

BACKGROUND: Long-term non-invasive ventilation (NIV) is as an established treatment option for chronic hypercapnic COPD patients. Beneficial effects have also been shown during exercise, but this is restricted to rehabilitation programs. New portable NIV (pNIV) devices may now enable NIV application during walking at home. STUDY DESIGN AND METHODS: In two randomized crossover trials, the impact of pNIV on dyspnea and endurance capacity was investigated in patients with moderate to severe COPD. Participants performed a standardized 6-min walking test, with and without pNIV, using a pre-set inspiratory/expiratory positive airway pressure of 18/8 cmH2O. The first study was performed in NIV-naïve patients (Study I), while the second study was performed in those already established on long-term NIV (Study II). RESULTS: 38 patients (66.9 ± 7.4 years, mean FEV1: 30.3 ± 8%pred) and 23 patients (67.6 ± 8.7 years, mean FEV1: 29.8 ± 10.4%pred) participated in Study I and II, respectively. In Study I, the mean difference in the Borg Dyspnea Scale (BDS, primary outcome) score following walking was 3.2 (IQR 2-4) without pNIV, compared to 2.6 (IQR 1-4) with pNIV (ΔBDS 0.65, P = 0.04), while walking distance increased from 311.8 m (95%CI 276.9-346.6 m) to 326.3 m (95%CI 291.5-361.2 m) (P = 0.044) when pNIV was used. Accordingly, in Study II, the mean difference in BDS was 4.4 (IQR 3-6) without pNIV, compared to 4.5 (IQR 3-6) with pNIV (ΔBDS 0.09, P = 0.54), while walking distance decreased from 291.5 m (95%CI 246.1-336.9 m) to 258.4 m (95%CI 213-303.8 m) (P ≤ 0.001). INTERPRETATION: The use of a pNIV device during walking can improve dyspnea and walking distance in patients with moderate to severe COPD. Patients who do not already receive long-term NIV therapy are more likely to benefit compared to those undergoing long-term NIV. Careful patient selection is mandatory. BACKGROUND: Although evidence suggests that functional endurance capacity is the most important component associated with future health, little is known of how it is associated with multiple other physical fitness components. Since various physical fitness aspects do not change the same as functional endurance capacity during childhood, it is necessary to establish possible associations between functional endurance capacity and other physical fitness components in children. Therefore, the main purpose of the study was to test the associations between functional endurance capacity with other physical fitness components in 7-14-year-old children, stratified by gender. METHODS: In this cross-sectional study, we recruited 1612 children [mean age ± standard deviation (SD) = 9.72 ± 2.37 years; 52.5% girls). Health-related physical fitness components included: 1) body-mass index (kg/m2) calculated from height and weight (measure of body size), 2) sit-and-reach test (measure of flexibility), 3) standing broad jump (measure of explosive strength of lower extremities), 4) sit-ups in 30 s (measure of repetitive strength of the trunk), 5) 10 × 5 shuttle run test (measure of agility) and 6) 20-m shuttle run test (measure of functional endurance capacity). The associations were performed using generalized estimating equations with beta (ß) coefficients. RESULTS: After adjusting for age, functional endurance capacity was associated with sit-and-reach test (ß = 0.13, p < 0.001), standing broad jump (ß = 0.59, p < 0.001), sit-ups in 30 s (ß = 0.53, p < 0.001) and 10 × 5 shuttle run test (ß = - 0.56, p < 0.001) in boys. In girls, functional endurance capacity was associated with body-mass index (ß = - 0.12, p < 0.001), sit-and-reach test (ß = 0.21, p < 0.001), standing broad jump (ß = 0.25, p < 0.001), sit-ups in 30 s (ß = 0.36, p < 0.001) and 10 × 5 shuttle run test (ß = - 0.40, p < 0.001). No significant associations between functional endurance capacity and body-mass index in boys were observed. CONCLUSIONS: Although significant, functional endurance capacity is weakly to moderately associated with other physical fitness components, pointing out that such measure should be tested separately from other aspects of physical fitness in school-aged children.

We investigated the effects of caffeine mouth rinse on endurance performance, muscle recruitment (i.e., electromyographic activity of the vastus lateralis and rectus femoris), rating of perceived effort and heart rate. Twelve physically-active healthy men cycled at 80% of their respiratory compensation point until task failure. The participants rinsed their mouths for 10 seconds with placebo (PLA, 25 mL of a solution composed of non-caloric mint essence) or caffeine (CAF, 25 mL of 1.2% of anhydrous caffeine concentration with non-caloric mint essence) every 15 minutes of exercise. Time until exhaustion increased 17% (effect size = 0.70) in CAF compared to PLA (p = 0.04). The wavebands of low-frequency electromyographic activity (EMG) of the vastus lateralis and rectus femoris was lower in CAF group than PLA at 50% of the time until exhaustion (p = 0.04). The global EMG signal was lower in CAF group than PLA at 100% of the time until exhaustion (p = 0.001). The rating of perceived effort pooled was higher in CAF mouth rinse (p = 0.001) than PLA group. No effect was found on the heart rate between the groups (p > 0.05). Caffeine mouth rinse increases endurance performance, rating of perceived effort and decreases muscle activity during a moderate-intensity exercise.

Patients on maintenance hemodialysis (HD) spend significant sedentary time traveling to and receiving dialysis, which leaves little time for social or sport events. Also, chronic HD patients are not physically fit, too exhausted after HD, and are not necessarily motivated to perform sports. There is increasing evidence that endurance training during HD can both increase dialysis efficacy and improve the ability of patients to participate in social life. With limited spare time, how can we motivate patients to train? Would that training improve endurance and modify leisure activity choice? Since HD patients would be a captive audience, we would suggest that endurance training during HD would be a preferred solution, if pediatric and adolescent patients could be persuaded to participate. There are few data on how training should be organized, as well as which parameters are best markers of safety and efficacy. And, most importantly, motivational aspects for performing endurance training on a regular basis clearly have to be considered. It is therefore in any case important to assess whether training improves endurance and, thus, quality of life or also quality of care parameters.

Brain glycogen stored in astrocytes provides lactate as an energy source to neurons through monocarboxylate transporters (MCTs) to maintain neuronal functions such as hippocampus-regulated memory formation. Although prolonged exhaustive exercise decreases brain glycogen, the role of this decrease and lactate transport in the exercising brain remains less clear. Because muscle glycogen fuels exercising muscles, we hypothesized that astrocytic glycogen plays an energetic role in the prolonged-exercising brain to maintain endurance capacity through lactate transport. To test this hypothesis, we used a rat model of exhaustive exercise and capillary electrophoresis-mass spectrometry-based metabolomics to observe comprehensive energetics of the brain (cortex and hippocampus) and muscle (plantaris). At exhaustion, muscle glycogen was depleted but brain glycogen was only decreased. The levels of MCT2, which takes up lactate in neurons, increased in the brain, as did muscle MCTs. Metabolomics revealed that brain, but not muscle, ATP was maintained with lactate and other glycogenolytic/glycolytic sources. Intracerebroventricular injection of the glycogen phosphorylase inhibitor 1,4-dideoxy-1,4-imino-d-arabinitol did not affect peripheral glycemic conditions but suppressed brain lactate production and decreased hippocampal ATP levels at exhaustion. An MCT2 inhibitor, α-cyano-4-hydroxy-cinnamate, triggered a similar response that resulted in lower endurance capacity. These findings provide direct evidence for the energetic role of astrocytic glycogen-derived lactate in the exhaustive-exercising brain, implicating the significance of brain glycogen level in endurance capacity. Glycogen-maintained ATP in the brain is a possible defense mechanism for neurons in the exhausted brain.

It has been demonstrated that skeletal muscle adaptions, including muscle fibers transition, angiogenesis, and mitochondrial biogenesis are involved in the regular exercise-induced improvement of endurance capacity and metabolic status. Herein, we investigated the effects of pterostilbene (PST) supplementation on skeletal muscle adaptations to exercise training in rats. Six-week-old male Sprague Dawley rats were randomly divided into a sedentary control group (Sed), an exercise training group (Ex), and exercise training combined with 50 mg/kg PST (Ex + PST) treatment group. After 4 weeks of intervention, an exhaustive running test was performed, and muscle fiber type transformation, angiogenesis, and mitochondrial content in the soleus muscle were measured. Additionally, the effects of PST on muscle fiber transformation, paracrine regulation of angiogenesis, and mitochondrial function were tested in vitro using C2C12 myotubes. In vivo study showed that exercise training resulted in significant increases in time-to-exhaustion, the proportion of slow-twitch fibers, muscular angiogenesis, and mitochondrial biogenesis in rats, and these effects induced by exercise training could be augmented by PST supplementation. Moreover, the in vitro study showed that PST treatment remarkably promoted slow-twitch fibers formation, angiogenic factor expression, and mitochondrial function in C2C12 myotubes. Collectively, our results suggest that PST promotes skeletal muscle adaptations to exercise training thereby enhancing the endurance capacity.

An individual's diet affects numerous physiological functions and can play an important role in reducing the risk of cardiovascular disease. Epidemiological and clinical studies suggest that dietary flavanols can be an important modulator of vascular risk. Diets and plant extracts rich in flavanols have been reported to lower blood pressure, especially in prehypertensive and hypertensive individuals. Flavanols may act in part through signaling pathways that affect vascular function, nitric oxide availability, and the release of endothelial-derived relaxing and constricting factors. During exercise, flavanols have been reported to modulate metabolism and respiration (e.g., maximal oxygen uptake, O2 cost of exercise, and energy expenditure), and reduce oxidative stress and inflammation, resulting in increased skeletal muscle efficiency and endurance capacity. Flavanol-induced reductions in blood pressure during exercise may decrease the work of the heart. Collectively, these effects suggest that flavanols can act as an ergogenic aid to help delay the onset of fatigue. More research is needed to better clarify the effects of flavanols on vascular function, blood pressure regulation, and exercise performance and establish safe and effective levels of intake. Flavanol-rich foods and food products can be useful components of a healthy diet and lifestyle program for those seeking to better control their blood pressure or to enhance their physical activity. Key teaching points • Epidemiological and clinical studies indicate that dietary flavanols can reduce the risk of vascular disease. • Diets and plant extracts rich in flavanols have been reported to lower blood pressure and improve exercise performance in humans. • Mechanisms by which flavanols may reduce blood pressure function include alterations in signaling pathways that affect vascular function, nitric oxide availability, and the release of endothelial-derived relaxation and constriction factors. • Mechanisms by which flavanols may enhance exercise performance include modulation of metabolism and respiration (e.g., maximal oxygen uptake, O2 cost of exercise, and energy expenditure) and reduction of oxidative stress and inflammation. These effects can result in increased skeletal muscle efficiency and endurance capacity. • Further research is needed to clarify the amount, timing, and frequency of flavanol intake for blood pressure regulation and exercise performance.

PURPOSE: To test the reliability and validity of tethered swimming lactate minimum test in young swimmers. METHODS: Lactate minimum test was performed twice to test the reliability (experiment 1; n = 13). In addition, the validity was investigated through lactate minimum test relationships with tethered swimming lactate threshold and peak force obtained during graded exercise test (experiment 2; n = 11). Finally, the correlations with mean speeds observed during 200-m (s200m) and 30-minute continuous efforts (s30min) were also analyzed (experiment 3; n = 15). In all experiments, the lactate minimum test began with 3-minute all-out effort to induce lactatemia, followed by an exhaustive graded exercise test. RESULTS: The lactate minimum intensity and mean force during the entire 3-minute all-out effort (MF) showed high reliability (coefficient of variation < 8.9% and intraclass correlation coefficient > .93). The lactate minimum intensity was not different compared with lactate threshold (P = .22), presenting high correlations (r = .92) and agreement (95% limits of agreement = ±7.9 N). The mean force during the entire 3-minute all-out effort was similar to peak force obtained during graded exercise test (P = .41), presenting significant correlations (r = .88) and high indices of agreement (95% limits of agreement = ±11.3 N). In addition, lactate minimum test parameters correlated both with mean speeds observed during 200-m (r > .74) and 30-minute continuous efforts (r > .70). CONCLUSION: Thus, tethered swimming lactate minimum test can be used for training recommendations and to monitor aerobic adaptations in young swimmers.

Due to the controversy about the sensitive of lactate minimum intensity (LMI) to training and the need to develop other tool for aerobic fitness evaluation, the purpose of this study was to analyze the sensitivity of glucose minimum intensity (GMI) and LMI to endurance training. Eight trained male cyclists (21.4 ± 1.9 years, 67.6 ± 7.5 kg and 1.72 ± 0.10 m) were evaluated twice, before and after 12 weeks of training. GMI and LMI were calculated, respectively, by the lowest blood glucose and lactate values attained during an incremental test performed after a hyperlactemia induction, and VO2max was determined during standard incremental effort. The training was prescribed in three different zones and controlled by heart rate (HR). The training distribution was equivalent to 59.7%, 25.0% and 15.3% below, at and above anaerobic threshold HR respectively. The anaerobic threshold evaluated by GMI and LMI improvement 9.89 ± 4.35% and 10.28 ± 9.89 respectively, after training, but the VO2max 2.52 ± 1.81%. No differences were found between GMI and LMI in pre (218.2 ± 22.1 vs 215.0 ± 18.6 W) and post (240.6 ± 22.9 vs 237.5 ± 18.8 W) training situations. LMI and GMI were sensitive to 12-week aerobic training in cyclist; thus, both protocols can be used to assess aerobic adaptation, athletes diagnostic and prescribe training.

This study sought to determine the time course of training adaptations to two different sprint interval training programmes with the same sprint: rest ratio (1:8) but different sprint duration. Nine participants (M: 7; F: 2) were assigned to 15-second training group (15TG) consisting of 4-6 × 15-second sprints interspersed with 2-minute recovery, whereas eight participants (M: 5; F: 3) were assigned to 30-second training group (30TG) consisting of 4-6 × 30 second sprints interspersed with 4-minute recovery. Both groups performed their respective training twice per week over 9 weeks and changes in peak oxygen uptake (V˙O2peak) and time to exhaustion (TTE) were assessed every 3 weeks. Additional eight healthy active adults (M: 6; F: 2) completed the performance assessments 9 weeks apart without performing training (control group, CON). Following 9 weeks of training, both groups improved V˙O2peak (15TG: 12.1%; 30TG: 12.8%, P<.05) and TTE (15TG: 16.2%; 30TG: 12.8%, P<.01) to a similar extent. However, while both groups showed the greatest gains in V˙O2peak at 3 weeks (15TG: 16.6%; 30TG: 17.0%, P<.001), those in TTE were greatest at 9 weeks. CON did not change any of performance variables following 9 weeks. This study demonstrated that while the changes in cardiorespiratory function plateau within several weeks with sprint interval training, endurance capacity (TTE) is more sensitive to such training over a longer time frame in moderately-trained individuals. Furthermore, a 50% reduction in sprint duration does not diminish overall training adaptations over 9 weeks.

PURPOSE: To examine the effects of caffeine ingestion on physiological and perceptual responses in mentally fatigued individuals. METHODS: Eight male physically active subjects completed four cycling constant-workload tests in four experimental conditions at 80 % of maximal power output: control (C), mental fatigue (MF), mental fatigue plus caffeine ingestion (5 mg/kg) (MF-CAF), and mental fatigue plus placebo (MF-PLA). The mental fatigue was induced by a continuous performance task A-X version (AX-CPT). Before and after the AX-CPT, the profile of mood state (POMS) and blood samples for lactate measurement were collected. Oxygen consumption ([Formula: see text]), rating of perceived exertion (RPE), and electromyography (EMG) activity were measured during the cycling test. RESULTS: The time to exhaustion in C, MF, MF-PLA, and MF-CAF were 251 ± 30, 222 ± 23, 248 ± 28, and 285 ± 42 s, respectively. Delta values (corrected by C condition) were higher in MF-CAF than MF (P = 0.031). MF-CAF reported higher Vigor scores when compared with C (P = 0.046) and MF (P = 0.020). RPE at the first minute was significantly higher in MF-PLA than in C (P = 0.050); at the second minute, RPE was higher in MF-PLA than in C (P = 0.049) and MF-CAF (P = 0.048). EMG activity was not different between the conditions. CONCLUSIONS: Caffeine ingestion increased approximately 14 % endurance performance after the induction of mental fatigue. This effect was accompanied by a tendency to improvement in mood state (i.e., vigor). Therefore, caffeine ingestion can promote a beneficial effect on endurance performance in mentally fatigued individuals.

Molecular mechanisms of adaptation to exercise despite a large number of studies remain unclear. One of the crucial factors in this process is hypoxia inducible factor (HIF) that regulates transcription of many target genes encoding proteins that are implicated in molecular adaptation to hypoxia. Experiments were conducted on 24 adult male Fisher rats. Real-time PCR analysis was performed for quantitative evaluation of Hif3α, Igf1, Glut-4 and Pdk-1 in m. gastrocnemius, m. soleus, in lung and heart tissues. Mitochondrial respiratory function and electron microscopy were performed. Knockdown of Hif3α using siRNA increases time of swimming to exhaustion by 1.5 times. Level of mitochondrial NAD- and FAD-dependent oxidative pathways is decreased, however efficiency of phosphorylation is increased after Hif3α siRNA treatment. Expression of HIF target genes in muscles was not changed significantly, except for increasing of Pdk-1 expression in m. soleus by 2.1 times. More prominent changes were estimated in lung and heart: Igf1 gene expression was increased by 32.5 and 37.5 times correspondingly. Glut4 gene expression in lungs was increased from undetected level till 0.3 rel. units and by 84.2 times in heart. Level of Pdk1 gene expression was increased by 249.2 in lungs and by 35.1 times in hearts, correspondingly. Some destructive changes in muscle tissue were detected in animals with siRNA-inducing silencing of Hif3α.

Involuntary physical activity induced by the avoidance of electrical shock leads to improved endurance exercise capacity in animals. However, it remains unknown whether voluntary stand-up physical activity (SPA) without forced simulating factors improves endurance exercise capacity in animals. We examined the eff ects of SPA on body weight, cardiac function, and endurance exercise capacity for 12 weeks. Twelve male Sprague-Dawley rats (aged 8 weeks, n=6 per group) were randomly assigned to a control group (CON) or a voluntary SPA group. The rats were induced to perform voluntary SPA (lifting a load equal to their body weight), while the food height (18.0 cm) in cages was increased progressively by 3.5 every 4 weeks until it reached 28.5 cm for 12 weeks. The SPA group showed a lower body weight compared to the CON group, but voluntary SPA did not affect the skeletal muscle and heart weights, food intake, and echocardiography results. Although the SPA group showed higher grip strength, running time, and distance compared to the CON group, the level of irisin, corticosterone, genetic expression of mitochondrial biogenesis, and nuclei numbers were not affected. These findings show that voluntary SPA without any forced stimuli in rats can eff ectively reduce body weight and enhance endurance exercise capacity, suggesting that it may be an important alternative strategy to enhance endurance exercise capacity.

Endurance performance depends on the integration of several phenotypic traits influenced by multiple environmental and genetic factors. Objectives of the study were: (1) to examine the genotypic frequencies of the ACE I/D, ACTN3 R577X polymorphisms and endurance performance-related phenotypes, (2) to evaluate the dynamics of endurance performance parameters during a 5-year period in relation to ACE I/D and ACTN3 R577X genotypes in Estonian young skiers. Determination of VO2peak was performed in 58 skiers aged 15-19 years (41 males, 17 females) during a 5-year period. The control group consisted of 322 healthy non-athletic subjects (145 males, 177 females). The study groups were genotyped for the ACE I/D and ACTN3 R577X variants. Frequencies of the ACE ID and ACTN3 RR genotypes were significantly higher (p = 0.047 and p = 0.003, respectively) and the RX genotype was lower (p = 0.008) in young male skiers compared with controls. A significant relationship was found between change (Δ) of training volume and ΔVO2peak (mL·kg(-1)·min(-1)) (r = 0.475, p = 0.002). No significant main effect was detected between VO2peak (mL·kg(-1)·min(-1)) dynamics (comparison with the previous age group data) and ACE I/D and ACTN3 R577X genotypes interactions (F = 0.571, p = 0.770 and F = 0.650 and p = 0.705, respectively) in all young skiers. Study results indicated a significantly higher frequency of the ACE ID and ACTN3 RR genotypes among Estonian young male skiers compared with the male control group. Significant genotype-related differences in dynamics of VO2peak during a 5-year period were not found. In the future, longitudinal research including different gene variants may contribute to a better understanding of the nature of endurance performance. Key pointsSignificantly higher prevalence of the ACE ID and the ACTN3 RR genotypes were found among Estonian young male skiers compared with the male control group, which may be an advantage for the explosive speed and power capacity in race skiing.A more remarkable trend of increase in VO2peak (mL·kg(-1)·min(-1)) during the 5-year period was observed among male skiers with the ACTN3 XX genotype and among female skiers with the ACE ID genotype.No significant genotype-related associations in the dynamics of VO2peak were found during the 5-year period.

Based on the effects of n-3 polyunsaturated fatty acids (PUFA) such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) on reduction of blood viscosity, we theorized that PUFA could improve aerobic performance by increasing oxygen supply to tissues. Twenty male subjects were randomly divided into two groups (n = 10): a fish oil group (FG) and a control (CG). Maximal oxygen uptake and oxygen uptake during submaximal exercise were measured using a cycle ergometer. For 8 weeks, the FG then ingested capsules containing 3.6 g/day of EPA-rich fish oil, while the CG took 3.6 g/day of a medium-chain triglyceride. After supplementation, erythrocyte EPA and DHA in the FG were significantly increased. In the FG, a negative linear correlation was detected in the change between erythrocyte EPA and whole oxygen uptake during submaximal exercise pre- and post-supplementation. The present study showed that EPA-rich fish oil supplementation improves exercise economy in humans.

To investigate the activity of fermented deer antler on exercise endurance capacity, we evaluated endurance capacity in five-week-old male BALB/c mice by administering the fermented deer antler extract (FA) or the non-fermented deer antler extract (NFA) and then subjected the mice to exercise in the form of swimming. The mice administered 500 mg/kg/day of FA showed a significant increase in swimming time compared with mice administered placebo (16.55 min vs. 21.64 min, P<0.05). Serum lactate dehydrogenase (LDH), the marker of the liver and muscle damage, was significantly lower in FA groups. However, NFA groups did not show significantly different swimming time or serum LDH from that of the control group. Moreover, the FA-500 group had significantly higher hepatic superoxide dismutase (SOD) activity after forced swimming than the control and NFA groups (P<0.05). These findings suggest that fermentation may increase the exercise endurance capacity of the deer antler.
