RESUMO

Some groups of invertebrates from intermittent wetlands produce dormant stages in response to environmental fluctuations. Dormancy is a strategy to survive such fluctuations and to persist in extreme aquatic habitats, such as temporary habitats. We investigated the hatching responses of invertebrate dormant stages across different depths of sediment in intermittent ponds. Our hypotheses were: (1) the richness and abundance of invertebrate hatchlings decrease as the depth of the sediment column increases, and (2) the composition of invertebrate hatchlings varies over the wetland sediment depth. Four intermittent ponds were sampled in southern Brazil. One sediment column of 30 cm depth was collected in each pond and stratified into 1 cm thick slices for analysis of the dormant stages. A total of 1,931 hatchlings distributed among 31 taxa were collected from the sediment columns over the experiment. The total richness and abundance of hatchlings (after bdelloid taxa exclusion) were negatively related with the sediment depth. The composition of aquatic invertebrates varied among the different strata over the sediment depth. As intermittent wetlands are ecosystems extremely susceptible to climate variations, the results help to understand the resilience of aquatic resistant communities from different sediment strata after drought events. The effect of in ovo threonine (Thr) supplementation on the ileal expression of glucose, peptide and amino acid transporters was assessed in Salmonella Enteritidis-challenged broiler chicks. At 17.5 days of incubation, fertile eggs were supplemented in the amniotic fluid with sterile saline or 3.5% threonine. Hatchlings were individually weighed, and Salmonella Enteritidis negative status was confirmed. At 2 days of age, half of the birds of each group were inoculated with sterile nutrient broth or Salmonella Enteritidis inoculum. Relative expression of sodium-dependent glucose transporter 1 (SGLT1), glucose transporter 2 (GLUT2), di- and tri-peptide transporter 1 (PepT1) and alanine, serine, cysteine, threonine transporter (ASCT1) was assessed at hatch, 2 and 9 days of age, i.e., before inoculation and 7 days post-inoculation (dpi). At 9 days of age (7dpi), threonine increased SGLT1 and GLUT2 expression, whereas GLUT2 expression decreased in Salmonella-challenged birds. There was a significant interaction between threonine and Salmonella for PepT1 and ASCT1. Threonine increased PepT1 expression only in non-challenged birds. In addition, in ovo supplementation increased expression of ASCT1 regardless of post-hatch inoculation; Salmonella inoculation resulted in decreased expression of ASCT1 only in supplemented birds. The results suggest that while intra-amniotic threonine administration in broiler embryos increases the expression of genes related to the absorption of monosaccharides and amino acids, Salmonella challenge may negatively affect the expression of protein related transporters in the ileum of broilers.

Viviparity (live-bearing) has independently evolved from oviparity (egg-laying) in more than 100 lineages of squamates (lizards and snakes). We might expect consequent shifts in selective forces to affect per-brood reproductive investment (RI = total mass of offspring relative to maternal mass) and in the way in which that output is partitioned (number vs. size of offspring per brood). Based on the assumption that newly born offspring are heavier than eggs, we predicted that live-bearing must entail either increased RI or a reduction in offspring size and/or fecundity. However, our phylogenetically controlled analysis of data on 1,259 squamate species revealed no significant differences in mean offspring size, clutch size or RI between oviparous and viviparous squamates. We attribute this paradoxical result to (1) strong selection on offspring sizes, unaffected by parity mode, (2) the lack of a larval stage in amniotes, favouring large eggs even in the ancestral oviparous mode and (3) the ability of viviparous females to decrease the mass of uterine embryos by reducing extra-embryonic water stores. Our analysis shows that squamate eggs (when laid) weigh about the same as the hatchlings that emerge from them (despite a many-fold increase in embryo mass during incubation). Most of the egg mass is due to components (such as water stores and the eggshell) not required for oviductal incubation. That repackaging enables live-born offspring to be accommodated within the mother's body without increasing total litter mass. The consequent stasis in reproductive burden during the evolutionary transition from oviparity to viviparity may have facilitated frequent shifts in parity modes.

Descriptions of egg cases and hatchlings of the Bali catshark Atelomycterus baliensis and the Australian marbled catshark Atelomycterus macleayi are provided. Comparisons are made with two of their congeners, banded sand catshark Atelomycterus fasciatus and coral catshark Atelomycterus marmoratus. The Atelomycterus egg cases have the same general morphology, i.e., elongate with anterior and posterior waists, a depressed and truncate anterior end sometimes with attachment fibres, a medially tapered posterior end with two horns sometimes terminating in short, tightly curled tendrils and four respiratory fissures (one at each left corner on opposing sides). Morphometric measurements taken of the egg cases of the four species differed significantly when subjected to multivariate analysis, with unique characters enabling distinction between them. The morphological characters that best distinguished the Atelomycterus species from each other were anterior border width, posterior waist width and posterior case width. Thus, egg case morphology could be used to distinguish the egg cases of the four Atelomycterus species examined, consistent with other studies on scyliorhinid and pentanchid egg cases. Colouration of A. baliensis and A. macleayi juveniles was similar to that of other Atelomycterus juveniles in being pale with a series of dark-brown distinct dorsal saddles along the body, blotches and sometimes with a low number of small dark spots.

This study investigated the effects of dietary supplementation with vitamin E (vit. E), selenium yeast (Se yeast), or both on egg incubation response, embryonic development, keet quality, and posthatch growth of helmeted guinea fowls. Two hundred and forty 24-week old helmeted guinea fowl hens (average weight 1.75 + 0.22 kg) and cocks (average weight 2.15 + 0.20 kg) were assigned into 24 pens; each pen housed 10 hens and 2 cocks. There were four dietary treatments consisting of a basal diet (control), basal diet supplemented with vit. E (30 IU/kg), Se yeast (0.3 mg/kg Se), or both. Six pens were assigned to each treatment. Egg incubation response were estimated using 504 settable eggs sampled from each treatment collected during 15 to 17 weeks in lay. A total of 72 fertile eggs sampled from each treatment were used for the estimation of embryonic development. Quality of day-old keets hatched was scored based on physical conditions, while posthatch growth was measured for 21 days. Guinea fowl breeders fed diet supplemented with both vit. E and Se yeast produced the highest (P < 0.05) number of fertile eggs, percentage fertility, number of hatchlings, hatchability of total eggs, and hatchability of fertile eggs. Supplementation with vit. E + Se yeast resulted in the heaviest (P < 0.05) embryo weight, relative embryo weight, least (P < 0.05) yolk sac weight, and relative yolk sac weight on 25 days of incubation. Hatchlings from breeders fed diet supplemented with Se yeast and vit. E + Se yeast showed normal swallowed yolk. Supplementation of maternal diet with vit. E, Se yeast, and vit. E + Se yeast resulted in improved (P < 0.05) feed conversion ratio of subsequent hatchlings during 1 to 7-day posthatch growth. It can be concluded that dietary supplementation of vit. E + Se yeast in guinea fowl breeders resulted in improved egg fertility, hatchability, heavier embryo weights, hatchlings of good quality, and improved posthatch growth during the first 7 days.

The antioxidant defense system protects turtles from oxidative stress during hibernation. The present study examined changes of the antioxidant enzymes both on mRNA level and enzyme activity level during hibernation of Chinese soft-shelled turtle Pelodiscus sinensis hatchlings. The upstream regulator NF-E2 related factor 2 (Nrf2) mRNA was also measured. Samples were taken at pre-hibernation (17.0°C, Mud temperature (MT)), hibernation (5.8°C, MT) and arousal (20.1°C, MT). Nrf2 exhibited a tissue-specific pattern of expression with a decrease in the brain, slight increase in the liver and heart during hibernation, and significant increase during arousal in all the three tissues. Superoxide dismutase (SOD) mRNA, catalase (CAT) mRNA, and glutathione peroxidase 3 (GPx3) mRNA exhibited a similar pattern as Nrf2 in the brain and liver during the entire hibernation period. Hepatic GPx4 mRNA level increased during hibernation and decreased during arousal, whereas it did not change in the heart. Cerebral SOD and CAT activities kept stable during the experimental period, but GPx activity decreased significantly during hibernation and arousal. Hepatic GPx enzyme activity did not change, whereas those of SOD and CAT exhibited a notable decrease during arousal. Malondialdehyde concentration did not increase during the hibernation process, indicating an effective protection of the antioxidant defense system.

As global warming has lengthened the active seasons of many species, we need a framework for predicting how advances in phenology shape the life history and the resulting fitness of organisms. Using an individual-based model, we show how warming differently affects annual cycles of development, growth, reproduction and activity in a group of North American lizards. Populations in cold regions can grow and reproduce more when warming lengthens their active season. However, future warming of currently warm regions advances the reproductive season but reduces the survival of embryos and juveniles. Hence, stressful temperatures during summer can offset predicted gains from extended growth seasons and select for lizards that reproduce after the warm summer months. Understanding these cascading effects of climate change may be crucial to predict shifts in the life history and demography of species.

A potential advantage of group movement in animals is increased locomotion efficiency. This implies a reduced energetic cost for individuals that occur in larger groups such as herds, flocks and schools. When chelonian hatchlings hatch in the underground nest with finite energy for their post-hatching dispersal phase, they face the challenge of minimizing energetic expenditure while escaping the nest. The term 'social facilitation' has been used to describe the combined digging effort of sea turtle hatchlings during nest escape. Given that in a normal clutch, a substantial part of the energy reserve within the residual yolk is used by hatchlings in the digging out process, a decreased cohort size may reduce the energy reserve available to cross the beach and sustain the initial swimming frenzy. This hypothesis was experimentally tested by varying cohort size in hatchling green turtles (Chelonia mydas) and measuring energy expenditure during the nest escape process using open-flow respirometry. The energetic cost of escaping through 40 cm of sand was calculated to vary between 4.4 and 28.3 kJ per individual, the cost decreasing as the number of individuals in the cohort increased. This represents 11-68% of the energy contained in a hatchling's residual yolk at hatching. The reduced energetic cost associated with large cohorts resulted from both a lower metabolic rate per individual and a shortened nest escape time. We conclude that synchronous digging activity of many hatchlings during nest escape evolved not only to facilitate rapid nest emergence but also to reduce the energetic cost to individuals.

The autonomic control of heart rate was studied throughout development in embryos of the green iguana, Iguana iguana by applying receptor agonists and antagonists of the parasympathetic and sympathetic systems. Acetylcholine (Ach) slowed or stopped the heart and atropine antagonized the response to Ach indicating the presence of muscarinic cholinoceptors on the heart of early embryos. However, atropine injections had no impact on heart rate until immediately before hatching, when it increased heart rate by 15%. This cholinergic tonus increased to 34% in hatchlings and dropped to 24% in adult iguanas. Although epinephrine was without effect, injection of propranolol slowed the heart throughout development, indicating the presence of ß-adrenergic receptors on the heart of early embryos, possibly stimulated by high levels of circulating catecholamines. The calculated excitatory tonus varied between 33% and 68% until immediately before hatching when it fell to 25% and 29%, a level retained in hatchlings and adults. Hypoxia caused a bradycardia in early embryos that was unaffected by injection of atropine indicating that hypoxia has a direct effect upon the heart. In later embryos and hatchlings hypoxia caused a tachycardia that was unaffected by injection of atropine. Subsequent injection of propranolol reduced heart rate both uncovering a hypoxic bradycardia in late embryos and abolishing tachycardia in hatchlings. Hypercapnia was without effect on heart rate in late stage embryos and in hatchlings.

In the last few decades, the cuttlefish market has grown to approximately 14% of the world's fisheries, and operators have begun to express concerns about the decline of this resource. In this context, the production of cuttlefish through aquaculture could offer a diversifying and valuable response to the increasing market demand and help alleviate the environmental pressure on this species. Therefore, the aim of this study is to identify a dry, cost-effective, and easy-to-administer diet that can successfully support the initial phases of cuttlefish growth and provide a similar performance to a krill-based diet, which closely mimics their natural diet. To achieve this objective, cuttlefish hatchlings were distributed among different experimental tanks, each receiving one of the five different diets (namely Diets A to E). Mortality and morphological parameters were monitored until day 10 post hatching, and the two most effective diets (Diets A and B) were chosen for further trials. The results indicated that Diet B had similar survival and growth rates to Diet A, which was based on frozen krill. Histological analysis revealed a comparable degree of gut maturity between the organisms fed the two diets. Likewise, levels of amylase and trypsin enzymes and hsp70, cat, and sod mRNA did not exhibit significant differences between the two groups. In conclusion, our findings provide preliminary evidence supporting the possibility of cultivating cuttlefish in captivity using a pelleted diet, representing a promising starting point for larger-scale breeding efforts.

Turtles are a potential sentinel species of aquatic ecosystem health as they inhabit aquatic ecosystems, are long lived, and potentially have high exposure to anthropogenic chemicals via food and water. This study investigated per- and polyfluoroalkyl substances (PFAS) tissue partitioning in female Emydura macquarii macquarii turtle, and the maternal offloading of (PFAS) into eggs and then hatchlings as well as the accumulation of PFAS in male and female Emydura macquarii macquarii serum. Significantly higher levels of perfluorosulfonic acids (PFSAs) and perfluorocarboxylic acids (PFCAs) were measured in the male serum compared to the female turtle serum, whereas perfluoroalkane sulfonamides (FASAs) were significantly higher in the female turtle serum. Perfluorooctane sulfonate (PFOS) was the predominant PFAS in the turtles whereas PFHxA was the predominant PFAS found in the surrounding water. PFHxA was not reported in any turtle tissue or the serum. The short-chain PFSAs and FASAs appeared to be highly associated with blood; long-chain PFSAs and PFCAs were more likely to be associated with tissue. Half of the PFHxS and all the long-chain PFSAs and PFCAs reported in the yolks were transferred into the hatchlings (by mass), suggesting a potential intergenerational effect.

It has been widely demonstrated that air and sand temperatures influence the anatomy of sea turtle hatchlings. We examined the impact of precipitation during the nesting season on the hatchling body size of loggerhead and green turtles from 37 beaches worldwide. Longitudinal data collected between 2012 and 2018 from Florida (US) and from a sample on Bõa Vista Island (Cabo Verde) carried out in 2019 showed that loggerhead body size at hatching was negatively correlated with precipitation, while precipitation was not correlated with hatchling body size in green turtles. A meta-analysis revealed that precipitation is positively correlated with hatchling mass in loggerhead turtles, while it is positively correlated with straight carapace length and width in green turtle hatchlings. The strongest influence of precipitation was found in the middle of the incubation period of loggerhead turtles in Cabo Verde, and we posit that this is due to an increase in the uptake of water for embryonic growth. These findings highlight the great importance of understanding the correlated effects of regional environmental variables, such as precipitation, on the development of sea turtle hatchlings and will have an impact on the evaluation of ongoing conservation and climate change discussions.

Invasive alien species control is recognized worldwide as a priority action to preserve global biodiversity. However, a lack of general life history knowledge for threatened species can impede the effectiveness of conservation actions. Galápagos pink land iguanas (Conolophus marthae) are endemic to Wolf Volcano, Galápagos, Ecuador. These iguanas are threatened by invasive alien species, particularly feral cats, that may affect their small population size. To guarantee the long-term survival of C. marthae, the Galápagos National Park Directorate is considering, along with an ongoing campaign of feral cat control, the implementation of a head-start program. However, the success of this management strategy necessarily relies on the identification of pink iguana nesting grounds, which were still unknown at the onset of this study. We modeled the movement patterns of male and female iguanas during the reproductive season, using location data collected from custom-made remote tracking devices installed on adult pink iguanas in April 2021. We first calculated for each individual the vector of distances from its starting location, which was defined as net displacement. We then used net displacement as the response variable in a generalized additive mixed model with day of the year as the predictor. Based on the hypothesis that males and females may behaviorally differ after mating, we looked for female-specific migratory behavior suggesting females were moving toward nesting areas. The results obtained confirmed our hypothesis, as females exhibited a distinct migratory behavior, reaching a small plateau area inside of Wolf Volcano's caldera and ca. 400 m below the volcano's northern rim. Moreover, once inside the caldera, females displayed a more aggregated distribution pattern. The movement data obtained allowed Galápagos National Park rangers to locate individual pink iguana nests and subsequently to sight and collect the first observed hatchlings of the species. This work constitutes a necessary baseline to perform dedicated studies of pink iguana nests and emerging hatchling iguanas, which is an essential step toward the development of an effective head-start program.

In November 2015, a tailings dam ruptured and affected the second largest nesting site of loggerhead sea turtles in Brazil. This study aimed to evaluate the reproductive success, and trace elements in female's plasma, freshly laid eggs, unhatched eggs, and dead hatchlings of loggerhead turtles that nest in the coastal area exposed to the mining waste (Povoação, Espírito Santo state) and compare them with animals from an area that was not affected by the tailings (Praia do Forte, Bahia state). Plasma concentrations of As, Cd, Cr, Fe, and Zn were significantly higher in samples from Povoação in comparison to turtles from Praia do Forte. In Povoação, unhatched eggs and dead hatchlings had higher As, Cu, Hg, Mn, and Zn concentrations than freshly laid eggs, and trace elements correlated with the hatching and emergence success. Our findings suggest that the higher concentrations of some metals may influence the incubation period and reproductive success of loggerheads in the affected area.

Suppression (p ≤ 0.05) of antioxidative/detoxification (except GPx and CYP3a) and cytoskeletal (except DHPR) genes but induction of metabolic (except for AST and TRY) and heat shock (except HSP60) genes of Labeo rohita hatchlings after 14 days of exposure to environmentally relevant concentrations of Triclosan (0.0063, 0.0126, 0.0252 and 0.06 mg/L) was followed by an increase (p ≤ 0.05) for most of the genes after 10 days recovery period. After recovery, LDH, ALT, CK, CHY, PA, HSP47 and DHPR declined, while SOD, CAT, GST, GR, GPx, CYP1a, CYP3a, AST, AChE, TRY, HSP60, HSP70, HSc71, HSP90 MLP-3, α-tropomyosin, desmin b and lamin b1 increased over exposure. Peak area of biomolecules (except 3290-3296, 2924-2925 and 2852-2855 cm-1) declined (p ≤ 0.01) more after recovery [except for an increase (p ≤ 0.01) at 1398-1401 cm-1]. CYP3a, CK, HSP90, MLP-3 and secondary structure of amide A are the most sensitive markers for the environmentally relevant concentrations of Triclosan.

We provide the raw data of 44 satellite-tracked loggerhead sea turtles from different life-stages collected between 2016 and 2018. Depending on life-stage and tag availability a different satellite tag was attached to the loggerhead carapace. Location data were collected using the Argos system. We made publically available for the first time in the Mediterranean: (i) the satellite-tracking data for 17 one-year-aged post-hatchlings of loggerhead sea turtle came from two nests laid on the Mediterranean Spanish coast; (ii) the satellite-tracking data for 4 loggerhead nesting females collected from nesting events occurred on the Spanish Mediterranean coast. Besides, another 23 juvenile and adult loggerhead sea turtles were monitored and their data were made also available. Our dataset provide the turtle identity name or number, the date and coordinates of the location data, and the Argos location class associated. Our data contribute to the knowledge about the spatial use of the loggerhead sea turtle in the Mediterranean Sea and could be used in further analysis regarding habitat use and dispersal of this species in the Mediterranean Sea. Specially, nesting females and post-hatchlings data contribute to shed light about these life-stages related to nesting events out of the usual nesting range in the western Mediterranean, for which data still remains scarce. Also, data could be compared to further similar research in satellite-tracking loggerhead sea turtles in the Mediterranean basin.

Ten nests were collected from Kerachut and Teluk Kampi, Penang Island between 2 August 2009 and 9 December 2009, and each one nest was split into three small clutch sizes for incubation at three nesting depths (45 cm, 55 cm and 65 cm), with a total of 30 modified nests for this experiment. Three important objectives were formulated; to observe on the survival hatchings among the three nesting depths, to study on the effects of sand temperature on incubation period among the three nesting depths, and to investigate the influence of sand temperature on hatchling's morphology. Main result shows that the mean survival of the hatchlings was 25.40% at 45 cm nesting depth, followed by mean 17.60% at 55 cm nesting depth, and lastly, the mean was 21.50% at 65 cm nesting depth. Overall, there are 56.63% survival hatchlings, 10.97% dead hatchlings and 32.40% unhatched eggs were produced. The incubation period was also found to be significantly correlated with sand temperature, p > 0.001, and nesting depth, p < 0.001. The hatchling's length and weight varies is sizes across the nesting depths, p < 0.001. However, the small difference in hatchling sizes per nesting depths are not strong enough to prove the significant correlation with sand temperature, p > 0.05. This article provides a basic knowledge from the splitting clutch design method. A sum of 50%-60% survivals hatchlings produced were incubating under small range of clutch sizes, 29 to 49 eggs. This article provides basic result on the survival hatchlings, eggs survivorship, incubation period, temperature, hatchling's morphology and discussion on implication of this method on conservation in Malaysia.


Terdapat 10 sarang penyu yang dikutip di pantai Kerachut dan Teluk Kampi, di antara 2 Ogos 2009 hingga 9 Disember 2009, dan setiap satu sarang dibahagikan kepada tiga bahagian kecil yang sama rata untuk proses pengeraman pada tiga kedalaman sarang yang berbeza (45 cm, 55 cm dan 65 cm). Jumlah keseluruhan sarang yang diubahsuai untuk menjalankan eksperimen ini ialah sebanyak 30 sarang. Tiga objektif utama telah diketengahkan; dimana penelitian ke atas anak penyu menetas di tiga kedalaman sarang yang berbeza, untuk mengkaji kesan suhu sarang ke atas masa pengeraman di antara tiga kedalaman sarang yang berbeza, dan kajian pengaruh suhu sarang ke atas morfologi anak penyu yang menetas. Keputusan utama menunjukkan min anak penyu menetas adalah sebanyak 25.40% di kedalaman sarang 45 cm, diikuti min 17.60% di kedalaman sarang 55 cm, dan yang terakhir, min 21.50% di kedalaman sarang 65 cm. Secara keseluruhannya, terdapat 56.63% anak penyu menetas, 10.97% anak penyu mati dan 32.40% telur yang tidak menetas telah dihasilkan. Didapati bahawa tempoh penetasan mempunyai signifikasi korelasi ke atas suhu sarang, p > 0.001 dan kedalaman sarang, p > 0.001. Sementara itu, panjang anak penyu dan berat anak penyu didapati berbeza saiz mengikut kedalaman sarang, p < 0.001. Namun begitu, perbezaan saiz anak penyu yang tidak begitu ketara yang dihasilkan mengikut kedalaman sarang tidak memberi kesan ke atas signifikasi korelasi dengan suhu sarang, p > 0.05. Kajian ini memberikan maklumat asas keputusan daripada kaedah pengasingan telur. Jumlah keseluruhan menunjukkan 50%­60% anak penyu menetas berjaya dihasilkan, walaupun dieramkan dalam jumlah kumpulan telur yang kecil, iaitu sebanyak 29­49 telur. Artikel ini adalah penting untuk memberikan maklumat asas mengenai anak penyu menetas, survival telur, tempoh penetasan, suhu, morfologi anak penyu dan perbincangan implikasi kaedah ini kepada konservasi di Malaysia.

Frequently, stranded sea turtles require rehabilitation under controlled conditions. Currently, few publications have described the conditions under which rehabilitation is to take place, particularly with respect to the hatchling life stage. To address this paucity of data, we conducted some experiments to assist rehabilitating facilities assess their handling of hatchlings. While in captivity, hatchlings are routinely handled, for example, for data collection and cleaning. Standardization of handling and housing protocols is necessary to define the most adequate rearing conditions to maintain hatchling welfare. Accordingly, the aim of this study was to assess plasma circulating corticosterone (Cort) concentration and growth, as a biomarker for the stress of hatchling loggerhead sea turtles (Caretta caretta) under controlled conditions. We performed two experiments to analyze handling frequency and stocking density. In both, Cort was measured and correlated with variations in animal weight and length. In handling experiments, Cort exhibited no significant increase when hatchlings were handled once a week, whereas Cort was significantly elevated when hatchlings were handled once every 2 weeks, suggesting that hatchlings have the ability to acclimate to frequent handling. However, hatchlings exhibited similar growth and mortality, regardless of handling regime. In stocking density experiments, hatchling isolation induced a significant elevation of Cort, in comparison with hatchlings placed with conspecifics at increasing densities. Growth increased in singly housed hatchlings, while mortality increased in tanks with three or more hatchlings. The results obtained suggest that Cort, growth, and mortality should be measured to assess hatchling welfare when kept under controlled conditions.

The efficient extraction and fixation of a tissue allows in preserving the cytoarchitecture, chemical composition and tissue organization, which is key in physiological and histopathological studies. The main goal of this study was to establish a microsurgery technique to obtain ocular tissue and provide an optimized immersion fixation protocol based on the 10 % formalin-intraocular injection on Olive ridley sea turtle hatchlings (Lepidochelys olivacea). To evaluate this optimized technique, a histological comparison between traditional immersion and intraocular/immersion protocols was done. The eyeball were processed into five protocols: Frozen eyes (Group 1), frozen eyes immersed in 10 % formalin (Group 2), fresh eyes immersed in 10 % formalin (Group 3), fresh eyes intraocularly injected with 0.1 M phosphate buffer solution (PBS) and then immersed in 10 % formalin (Group 4), and fresh eyes fixed by 10 % formalin-intraocular followed by 10 % formalin-immersion (Group 5). In comparison with all groups evaluated, the intraocular/immersion fixation protocol lead the conservation of eyeball shape, cell integrity and maintenance of the organization of the retina layers of sea turtle hatchlings. If this method will be the key in studying sea turtle, we suppose that this procedure, with minimal adjustments, could be useful in animals with similar eye anatomy.

Adenoviral infections may cause mild to severe morbidity or fatality in a large array of animal species. In crocodilians, hatchlings under 5 months of age are usually affected. However, there is a paucity of information on actual incidences in hatchlings originating from South Africa. Two cases of adenoviral hepatitis in crocodile hatchlings about 2 weeks old, bred on a commercial farm in South Africa, are described. Both hatchlings showed typical clinical signs of hepatitis. The identification of intranuclear inclusion bodies in the liver was used to differentiate between adenoviral hepatitis and chlamydial hepatitis. Although vertical transmission has never been proven in crocodiles, the young age of the affected hatchlings raises the possibility of vertical transmission. The lack of epidemiological information on adenoviral hepatitis in crocodiles highlights the need for further characterisation of the virus and targeted surveillance.
