RESUMO

Pagiophloeus tsushimanus is a new, destructive, and monophagous weevil pest that thrives on Cinnamomum camphora, found in Shanghai. The functions of chemosensory genes involved in the host location and intraspecific communication of P. tsushimanus remain unknown. The male-female transcriptomes of P. tsushimanus adults were assembled using Illumina sequencing, and we focused on all chemosensory genes in transcriptomes. In general, 58,088 unigenes with a mean length of 1018.19 bp were obtained. In total, 39 odorant binding proteins (OBPs), 10 chemosensory proteins (CSPs), 22 olfactory receptors (ORs), 16 gustatory receptors (GRs), eight ionotropic receptors (IRs), and five sensory neuron membrane proteins (SNMPs) were identified. PtsuOBPs comprised four subfamilies (20 Minus-C, one Plus-C, two Dimer, and 15 Classic). Both PtsuOBPs and PtsuCSPs contained a highly conserved sequence motif of cysteine residues. PtsuORs including one olfactory receptor co-receptors (Ptsu/Orco) comprised seven predicted transmembrane domains. Phylogenetic analysis revealed that PtsuOBPs, PtsuCSPs, and PtsuORs in P. tsushimanus exhibited low homology compared to other insect species. The results of tissue- and sex-specific expression patterns indicated that PtsuOBPs and PtsuORs were highly abundant in the antennae; whereas, PtsuCSPs were not only highly abundant in antennae, but also abdominal apexes, wings, and legs. In conclusion, these results enrich the gene database of P. tsushimanus, which may serve as a basis for identifying novel targets to disrupt olfactory key genes and may provide a reverse validation method to identify attractants for formulating potential eco-friendly control strategies for this pest.

Orthaga olivacea Warre (Lepidoptera: Pyralidae) is an important agricultural pest of camphor trees (Cinnamomum camphora). To further supplement the known genome-level features of related species, the complete mitochondrial genome of Orthaga olivacea is amplified, sequenced, annotated, analyzed, and compared with 58 other species of Lepidopteran. The complete sequence is 15,174 bp, containing 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, 2 ribosomal RNA (rRNA) genes, and a putative control region. Base composition is biased toward adenine and thymine (79.02% A+T) and A+T skew are slightly negative. Twelve of the 13 PCGs use typical ATN start codons. The exception is cytochrome oxidase 1 (cox1) that utilizes a CGA initiation codon. Nine PCGs have standard termination codon (TAA); others have incomplete stop codons, a single T or TA nucleotide. All the tRNA genes have the typical clover-leaf secondary structure, except for trnS(AGN), in which dihydrouridine (DHU) arm fails to form a stable stem-loop structure. The A+T-rich region (293 bp) contains a typical Lepidopter motifs 'ATAGA' followed by a 17 bp poly-T stretch, and a microsatellite-like (AT)13 repeat. Codon usage analysis revealed that Asn, Ile, Leu2, Lys, Tyr and Phe were the most frequently used amino acids, while Cys was the least utilized. Phylogenetic analysis suggested that among sequenced lepidopteran mitochondrial genomes, Orthaga olivacea Warre was most closely related to Hypsopygia regina, and confirmed that Orthaga olivacea Warre belongs to the Pyralidae family.

A laboratory study was made on the calling and mating behaviors of adult Orthaga achatina at (27 +/- 1) degrees C, (60 +/- 10)% RH, and 14 L: 10 D photoperiod. The female O. achatina did not call until the start of scotophase, with the calling percentage increased sharply at the 5th hour and reached the peak at the 6th-7th hour of scotophase. The calling percentage varied with moth age, being the highest (> 70%) at the age of 2-3 d. The mating behavior could be divided into two successive periods, i.e., calling period and mating period. The mating between female and male O. achatina occurred during the 5th-9th hour of scotophase, with the peak at the 6th-7th hour into scotophase, which was in accordance with the peak time of female calling behavior. Females mated only one time during their whole life. Treatment 1 female : 2 male showed a significantly higher mating percentage, but a significantly shorter mating duration time, compared with treatment 1 female : 1 male.

Fruits of Nectandra megapotamica Mez and Cinnamomum camphora (L.) (Lauraceae) were collected in Rio Grande do Sul, Brazil, in 2004. Species of five families of insects were found inside the fruits: two fly species (Diptera: Drosophilidae: Drosophila spp.), six beetle species, Heilipus sp., Conotrachelus sp. (Curculionidae), Hypothenemus sp. (Scolytidae) and three species of Carpophilus (Nitidulidae), and moths (Lepidoptera: Elachistidae). The moth especimes were identified as Stenoma catenifer Walsingham, the avocado borer. The occurrence of the moth was predominant from early April until middle May. The natural larval infestation level was low. Two new host plant of the pest were identified.
