Abstract Purpose: To investigate the efficacy and mechanisms of root tuber of Polygonum ciliinerve (Nakai) ohwi (rPC) which has been used to treat bacterial infection in traditional Chinese medicine. Methods: With the mouse model of Staphylococcus aureus (S. aureus) pneumonia, the phenotype of rPC treated mice, including body weight, mortality, lung slices and bacterial burden were evaluated. Furthermore, inflammatory factors in bronchoalveolar lavage (BAL) were determined by ELISA and the distribution of T cells in lung was assessed by immunofluorescence assay. Results: rPC treatment could dose-dependently reduce weight loss and mortality in S. aureus-infected mice. Upon 10 mg/ml rPC treatment, S. aureus-infected mice showed about 8 grams increase in body weight (P<0.001) and 50% enhancement in mortality. The integrity of lung tissue and bacterial burden were also improved by rPC treatment. Moreover, rPC was found to modulate the immune response in infection. Conclusion: rPC has therapeutic potential for S. aureus infections and pneumonia with immunomodulatory functions.

RESUMO: A espécie Polygonum punctatum Elliott (Polygonaceae) é amplamente utilizada pela população como planta medicinal. O objetivo deste trabalho é o de avaliar o potencial genotóxico e mutagênico de P. punctatum utilizando raízes de bulbos e radículas em sementes germinadas de Allium cepa através do teste in vivo, e realizar comparações da extração do material vegetal por calor (infusões) e extração a frio (extrato). Para isso, foram preparadas dois tipos de soluções, infusões e extratos foliares de P. punctatum, em duas concentrações 0,4 g mL-1 e 2,4 g mL-1. A infusão foi preparada pela adição das folhas secas em água destilada fervente (100ºC), permanecendo por 10 minutos enquanto o extrato foi preparado através da maceração das folhas secas em água destilada fria. Para o teste em A. cepa foram utilizados, para cada tratamento, seis grupos de quatro bulbos e seis caixas gerbox com 50 sementes em cada caixa. Duas lâminas para cada tratamento foram obtidas através da técnica de esmagamento das raízes e coradas com orceína acética 2%. Foram contadas 2000 células por grupo de bulbos e 3000 células por grupo de sementes, observando-se a ocorrência de interrupções em metáfases, alterações cromossômicas estruturais, bem como a inibição ou aumento da divisão celular. Os valores do índice mitótico foram calculados e analisados estatisticamente pelo Teste χ2 (p≤0,05). Os resultados demonstraram que as infusões e os extratos de folhas apresentaram redução nos valores de índices mitóticos nas concentrações utilizadas em relação ao controle em água destilada. Foram identificadas alterações cromossômicas na divisão celular, tais como pontes anafásicas, em todas as concentrações de infusões e extratos indicando assim que P. punctatum possui atividade antiproliferativa e genotóxica.


ABSTRACT: The species Polygonum punctatum Elliott (Polygonaceae) is widely used by the Brazilian population as a medicinal plant. The aims of this study are to evaluate the genotoxic and mutagenic potential of P. punctatum, using its root bulbs and rootlets in germinated seeds of Allium cepa by in vivo testing, and to compare the extraction of plant material by heat (infusions) and cold (extract). Thus, two types of solutions - infusions and leaf extracts - of P. punctatum were prepared at the two concentrations of 0.4 g ml -1 and 2.4 g mL- 1. The infusion was prepared by addition of dry leaves in boiling distilled water (100ºC), remaining for 10 minutes, while the extract was prepared by maceration of dried leaves in cold distilled water. For the A. cepa, we used for each treatment six groups of six bulbs and six seedling boxes with 50 seeds each. Two slides for each treatment were obtained by the technique of crushing the roots, and they were stained with 2 % acetic orcein. For the analysis, 2000 cells per group of bulbs and 3000 cells per group of seeds were counted, and we noted the occurrence of interruptions in the metaphase, chromosomal aberrations, as well as inhibited or increased cell division. The values of the mitotic index were calculated and statistically analyzed by the χ2 test (p ≤ 0.05). The results showed that the infusions and extracts of leaves showed reduced values of mitotic indices in the concentrations used compared to the control in distilled water. Chromosomal alterations were identified in the cell division, in all concentrations of infusions and extracts, thus indicating that P. punctatum has an antiproliferative and genotoxic activity.

Polygonum lapathifolium is an invasive plant spread worldwide, which has been used in traditional medicine for its biological activities. We studied chemical profile of the aerial part using HPLC/DAD/ESI-MS. Eight flavonoids, two chalcones and gallic acid were isolated, identified nad tested for their activity in seven experimental models (for antioxidant, alpha/beta-glucosidase inhibitory, antimicrobial, anticholineserase activity). Polygonum lapathifolium es una planta invasora cuya presencia se extiende por todo el mundo y que ha sido utilizada en la medicina tradicional por sus actividades biológicas. Analizamos el perfil químico de la parte superficial utilizando HPLC/DAD/ESI-MS. Ocho flavonoides, dos chalconas y ácido gálico fueron aislados, identificados y analizados en relación a sus actividades en siete modelos experimentales (de actividad antioxidante, alfa/beta-glucosidasa inhibitoria, antimicrobiana, anticholineserasa). RESUMO

Pimenta pseudocaryophyllus (Gomes) Landrum é a única espécie representante brasileira deste gênero de Myrtaceae. Tem sido popularmente utilizada no tratamento de diversas enfermidades, assim como condimento e aromatizante de bebidas, por seu sabor muito similar ao cravo-da-índia. Em menor escala, é empregada em carpintaria e na arborização urbana. Diversos autores analisaram a composição química do óleo essencial da espécie em diferentes regiões do Brasil, encontrando eugenol, metileugenol, (E) metilisoeugenol, chavibetol, geranial e neral como composto predominante nos espécimes estudados. Visando ressaltar a importância de P. pseudocaryophyllus como potencial fonte de recursos assim como subsidiar ações de manejo adequado esta revisão apresenta os principais aspectos botânicos, ecológicos, etnobotânicos e farmacológicos da espécie.


Pimenta pseudocaryophyllus (Gomes) Landrum is the only Brazilian species representative of this Myrtaceae genus. It has being traditionally used in the treatment of many diseases as well for seasoning food and flavoring beverages due to its similarity with clove flavor. In a minor scale, it is used in carpentry and as urban tree. Many authors have analyzed the chemical composition of the leaf essential oil in different regions of Brazil, founding eugenol, metileugenol, (E) metilisoeugenol, chavibetol, geranial e neral as main component in the specimens studied. Aiming to highlight the importance of P. pseudocaryophyllus as potential source of natural products and to provide some base to the adequate management actions, a review on botanical, ecological, ethnobotanical and pharmacological aspects of this species is presented.

RESUMO

The flavonoids 5-hydroxy-7,8,4'-trimethoxy isoflavone and 5,3',4'-trihydroxy-7-methoxyflavanon ,the tannin derivate Ethyl gallate and lignang homoegonol 5-(3''-Hydroxypropyl)-7-methoxy-2-(3',4'-dimethoxyphenyl)benzofuran were isolated and purified from Polygonum segetum Kunth leaves using column chromatography and thin layer chromatography methods. These substances were identified on the basis of their physical properties (melting point and Rf value), qualitative chemical reactions and spectroscopic techniques UV and NMR (experiments 1H, 13C, 1H-1H COSY, HMQC) and spectrometric techniques GC-MS according to their complexity.


El presente estudio se encaminó a determinar los metabolitos secundarios presentes en las hojas de la especie vegetal: Polygonum segetum Kunth, colectada del humedal Juan Amarillo (Bogotá-Colombia). Como resultado se aislaron e identificaron a partir de propiedades físicas y técnicas espectroscópicas de UV, 1HRMN, COSY, DEPT 135, HMQC y APT, la 5-hidróxi-7,8,4´-trimétoxi isoflavona, el galato de etilo (derivado de taninos) y la 5,3´,4´-trihidróxi-7-metóxi flavanona. Además, por análisis GC-MS se identifico el lignano 5-(3,4-dimetóxifenil)-7-metóxibenzofuranpropanol homoegonol.

Over the last decades, the incidence of ultraviolet B (UVB)-related skin problems has been increasing. Damages induced by UVB radiation are related to mutations that occur as a result of direct DNA damage and/or the production of reactive oxygen species. We investigated the anti-oxidant effects of a Polygonum multiflorum thumb extract against skin damage induced by UVB irradiation. Female SKH-1 hairless mice were divided into three groups: control (N = 7), distilled water- (N = 10), and P. multiflorum extract-treated (PM, N = 10) groups. The PM (10 g) was extracted with 100 mL distilled water, cryo-dried and 9.8 g was obtained. The animals received a topical application of 500 æL distilled water or PM extract (1, 2, 4, 8, and 16 percent, w/v, dissolved in distilled water) for 30 min after UVB irradiation (wavelength 280-320 nm, 300 mJ/cm²; 3 min) of the dorsal kin for 14 days, and skin immunohistochemistry and Cu,Zn-superoxide dismutase (SOD1) activity were determined. SOD1 immunoreactivity, its protein levels and activities in the skin were significantly reduced by 70 percent in the distilled water-treated group after UVB irradiation compared to control. However, in the PM extract-treated groups, SOD1 immunoreactivity and its protein and activity levels increased in a dose-dependent manner (1-16 percent, w/v, PM extract) compared to the distilled water-treated group. SOD1 protein levels and activities in the groups treated with 8 and 16 percent, w/v, PM extract recovered to 80-90 percent of the control group levels after UVB. These results suggest that PM extract strongly inhibits the destruction of SOD1 by UV radiation and probably contains anti-skin photoaging agents.

Free radicals induce numerous diseases by lipid peroxidation, protein peroxidation, and DNA damage. It has been reported that numerous plant extracts have antioxidant activities to scavenge free radicals. Whether Polygonum aviculare L. (Polygonaceae) has antioxidant activity is unknown. In this study, dried Polygonum aviculare L. was extracted by ethanol, and the extract was lyophilized. The antioxidant activities of extract powder were examined by free radical scavenging assays, superoxide radical scavenging assays, lipid peroxidation assays and hydroxyl radical-induced DNA strand scission assays. The results show that the IC50 value of Polygonum aviculare L. extract is 50 µg/ml in free radical scavenging assays, 0.8 µg/ml in superoxide radical scavenging assays, and 15 µg/ml in lipid peroxidation assays, respectively. Furthermore, Polygonum aviculare L. extract has DNA protective effect in hydroxyl radical-induced DNA strand scission assays. The total phenolics and flavonoid content of extract is 677.4 ± 62.7 µg/g and 112.7 ± 13 µg/g. The results indicate that Polygonum aviculare L. extract clearly has antioxidant effects.

RESUMO

The aim of this paper is to present the structure of exocrine glands in P. glabrum, P. amphybium, P. minus, P. persicaria and P. punctatum, of them from section Persicaria, family Polygonaceae. The observations were performed with light, scanning and transmission electron microscope, using routine techniques for sample preparation. The structures observed at superficial view are composed of four cells flanking a small pore in the middle, and surrounded by a number of rather small cells, followed by normal epidermic cells. In the cross section the structure is composed of a great cavity filled with secretion, and communicated with the external side of the pore. The cavity is internally surrounded by a flat cell epithelium separated from the mesophyll by a layer of parenchymatic cells.
