RESUMO

Engineered male and female biomimetic reproductive tissues are being developed as autonomous in vitro units or as integrated multi-organ in vitro systems to support germ cell and embryo function, and to display characteristic endocrine phenotypic patterns, such as the 28-day human ovulatory cycle. In this Review, we summarize how engineered reproductive tissues facilitate research in reproductive biology, and overview strategies for making engineered reproductive tissues that might eventually allow the restoration of reproductive capacity in patients.

Polybrominated diphenyl ethers (PBDEs) are a group of environmental endocrine-disrupting chemicals (EDCs). According to recent studies, the associations between PBDEs and rodent male reproductive system are controversial. Therefore, we performed this systematic review and meta-analysis to clarify the effects of PBDEs on rodent male reproductive system. Data were extracted from articles collected from PubMed, Web of Science, and other databases. The pooled standard mean deviation (SMD) with 95% confidence interval (95% CI) was used to evaluate the association between the male rodent reproductive system impairment and PBDE exposure. Ten articles were included in the present meta-analysis and systematic review. The summary SMD of reproductive toxicity associated with PBDEs was -0.46 (95% CI: -0.69, -0.22), and the overall association between DE-71 along with BDE-209 and male reproductive system damage was statistically significant (SMD = -0.7 95% CI: -1.18, -0.21, p = .041; SMD = -0.41, 95% CI: -0.77, -0.05, p = .000). The adverse impact of PBDEs on rodent male reproductive system, especially seminal vesicle (SMD = -1.09, 95% CI: -1.49, -0.49, p = .523) and ventral prostate (SMD = -1.27, 95% CI: -1.88, -0.65, p = .821), were clearly demonstrated. Moderate (SMD = -0.81, 95% CI: -1.182, -0.437, p = .197) and high (SMD = -0.41, 95% CI: -0.76, -0.05, p = .000) dosage may have exerted effect than relatively low (SMD = -0.29, 95% CI: -0.17, 0.12, p = .136) dosage. In conclusion, our meta-analysis and systematic review suggested PBDE toxicity on the male reproductive system.

The aim of this paper was to observe the toxic effect of Tripterygium Glycosides Tablets on the reproductive system of Ⅱ type collagen induced arthritis(CIA) male rats, and to explore the toxic mechanism preliminarily. Fifty SD rats were randomly divided into normal control group(Con), model group(CIA), Tripterygium Glycosides Tablets clinical equivalent dose groups of 1, 2, 4 times(9, 18, 36 mg·kg~(-1)), 10 rats in each group, and were given by gavage once a day for 42 days after the first immunization. The organ index of testis and epididymis were calculated on days 21 and 42. Histopathological and morphological changes of testis and epididymis were observed under optical microscope. Sperm count, sperm malformation rate and sperm kinetic parameters in epididymal tissues were observed by computer assisted sperm analysis(CASA). The concentration of testosterone(T), nitric oxide synthase(NOS) and aromatase(CYP19 A1) in serum were detected by ELISA. Immunohistochemistry was used to observe the expression of Bax and Bcl-2 related proteins in the apoptosis pathway of testis and epididymis. The results showed that, compared with Con group, CIA group significantly increased the rate of testicular spermatogenic tubule lesion and sperm malformation, decreased the average path speed, and no significant changes were observed in other groups. Tripterygium Glycosides Tablets at 4 times clinical equivalent dose can significantly reduce the testis index(P<0.01), each dose group can reduce the epididymis index(P<0.05). Each dose group of Tripterygium Glycosides Tablets could cause different degrees of damage to the testis and epididymis, the proportion of testicular histopathology lesions increased, the number of spermatogenic cells in the seminiferous tubules decreased, and so on. It could reduce the number of sperm, increase the rate of sperm deformity, make the parameters of sperm dynamics abnormal, and so on. Tripterygium Glycosides Tablets at 4 times dose could significantly reduce the content of serum sex hormone T and key enzyme of androgen synthesis(P<0.05 or P<0.01), but had no effect on CYP19 A1. The expression of Bax and Bcl-2 in testis and epididymis were increased by 2 and 4 times doses of Tripterygium Glycosides Tablets(P<0.05, P<0.01 or P<0.01). The results showed that 21 d administration of Tripterygium Glycosides Tablets at equal or higher doses could induce obvious toxic effect to the reproductive organs of CIA male rats, and lower the level of serum sex hormone T and the key enzyme of androgen synthesis, NOS. The mechanism of abnormal changes of Bax and Bcl-2 in Testis and epididymis is still to be elucidated.

RESUMO

The new species Oxycheila alenatiki sp. nov. (Cicindelidae: Megacephalini) is described from central Bolivia and compared to related species. Colour photographs of habitus and male genitalia of O. alenatiki sp.nov. and related species O. gracillima Bates, 1872 and O. oberthueri Horn, 1897 are compared. A modified taxonomic key to the gracillima species group is provided.

Kodaianellissus gibbusis sp. nov. is described from Rawlakot, Azad Jammu Kashmir (Pakistan), which is first record of the genus Kodaianellissus Wang, Bourgoin Zhang, 2017 from the country. Morphological characters including male genitalia structure are described and illustrated. A checklist and key to all species of Kodaianellissus are provided. A molecular analysis with morphological characters indicates its placement into the Kodaianellissus.

RESUMO

The Bittacidae of South China consist of two species of Bicaubittacus and eight species of Bittacus, among which three species are described here as new to science. Bittacus acutus sp. n. from Guangxi is recognized mainly by the hat-shaped epandrial lobe with an acute ventro-distal process in the male genitalia. Bittacus shaoguanensis sp. n. from Guangdong is distinguished by the triangular epandrial lobe in the male genitalia, and a V-shaped streak in the central part of wings. Bittacus longilobus sp. n. from Guangdong is recognizable by the main body of the epandrial lobe trapezoid, with an elongate ventro-distal lobe twice as long. The number of described species of the Chinese Bittacus is now raised to 41. A key to the species of Bittacidae in South China is provided.

RESUMO

A new species of the genus Sovia Evans, 1949, viz. S. liuzihaoi Huang Fan sp. nov. is described from Mt. Ailao, Xinping Yi and Dai Autonomous County, representing a remarkable geographically southward extension of this genus in Yunnan Province, Southwestern China. In general appearance S. liuzihaoi Huang Fan sp. nov. resembles S. separata magna (Evans, 1932), but in our molecular phylogenetic analyses, this species is clustered with S. lii Xue, 2015. The adults and male genitalia of the new species and related species are illustrated.

RESUMO

A new species of Enosis Mabille, 1889 from southern Brazil, E. ester Lemes, Mielke Casagrande sp. nov, is described and illustrated. The new species is closely related to Enosis angularis (Möschler, 1877) but can be distinguishable by a set of characters on wings and male genitalia. An updated geographic distribution for E. angularis is given and illustrations of the female genitalia are provided for the first time.

A prevalence study was conducted on German sheep flocks including goats if they cohabitated with sheep. In addition, a novel approach was applied to identify an infection at the herd-level before lambing season with preputial swabs, suspecting venereal transmission and ensuing colonisation of preputial mucosa with Coxiella (C.) burnetii. Blood samples and genital swabs were collected from breeding males and females after the mating season and were analysed by enzyme-linked immunosorbent assay (ELISA) and quantitative polymerase chain reaction (qPCR) respectively. In total, 3367 animals were sampled across 71 flocks. The true herd-level prevalence adjusted for misclassification probabilities of the applied diagnostic tests using the Rogan-Gladen estimator for the prevalence estimate and a formula by Lang and Reiczigel (2014) for the confidence limits, ranged between 31.3% and 33% (95% confidence interval [95% CI] 17.3-45.5) detected by the ELISA and/or qPCR. Overall 26-36.6% (95% CI 13-56.8) were detected by ELISA, 13.9% (95% CI 4.5-23.2) by the qPCR and 7.9-11.2% (95% CI 0.08-22.3) by both tests simultaneously. The range of results is due to data obtained from literature with different specifications for test quality for ELISA. Among eight farms with females shedding C. burnetii, three farms (37.5%) could also be identified by preputial swabs from breeding sires. This indicates less reliability of preputial swabs if used as a single diagnostic tool to detect C. burnetii infection at the herd-level.

Pardosa pseudoannulata (Araneae: Lycosidae), as an important predator of crop pests, has served as a strong driver for ecological regulation of pests. Cadmium (Cd) is a toxic heavy metal widely distributed in the soil in China, which not only seriously pollutes the ecological environment, but also poses a great threat to the survival of organisms. Palpal bulbs are the genital organs of male spiders, playing an important role in reproductive physiology. However, the effects of long-term Cd stress on the genital organ of the primary pest predator were poorly understood. Therefore, we investigated the Cd effect on the male palpal organ of P. pseudoannulata at morphological and gene expression levels. The results showed that no obvious difference in the morphology between the Cd-treated and control groups was observed, but cell adhesion was affected at molecular level. Transcriptome sequencing analysis revealed that under long-term Cd stress, the biological processes including cell-cell adhesion via plasma-membrane adhesion molecules, cell-cell adhesion, and homophilic cell adhesion via plasma membrane adhesion molecules were the top three differentially expressed terms (p-adj < 0.001), and 51 unigenes were annotated into cadherin-related proteins, such as protocadherin, cadherin-87A, and cadherin-96Ca, among which, 18 unigenes were significantly upregulated under the Cd stress. Our outcomes indicate that the differentially expressed genes involved in cell adhesion may explain the negative effects of Cd stress on the spider genital organ, and the comprehensive transcriptome dataset will also provide a profound molecular information of the genital organ of P. pseudoannulata.

Body dysmorphic disorder (BDD) is a serious psychiatric condition that affects approximately 2.5% of the U.S. adult population. BDD occurs when a person develops a debilitating focus and sense of distress about a minor or imagined physical flaw. There is growing awareness that in some men with BDD, the physical preoccupation centers on the appearance of their genitals. This review explores existing research on genital manifestations of BDD in men. PubMed and PsychInfo searches with the key word's genital manifestations of BDD were conducted, which yielded a list of 48 unique articles. Articles that were included in the present review were published in English and focused on men with a formal diagnosis of BDD from a psychiatrist, psychologist, or structured clinical interview. Only five articles met these criteria, and all were conducted by the same research group. The articles are reviewed here, along with thoughts about directions for future research and implications for treatment.

The contribution of veterinary growth promoters (VGP) to the environmental burden of endocrine-disrupting chemicals (EDCs) is largely unknown. At cattle feedlots, the excrement of cattle may contain VGPs, which can contaminate aquatic systems and pose reproductive health risks. The study identifies VGPs used at cattle feedlots in South Africa and confirms associated estrogenic activity in feedlot runoff water. Using a rat model, we investigate the potential reproductive health effects and thyroid function of an environmentally relevant mixture of VGPs. Collected water samples had low levels of selected VGPs, and estrogenic activity was detected in the T47D-KBluc bioassay. Rats exposed to VGP had significant adverse effects on male reproductive health, including shortened anogenital distance, lowered sperm counts, disorganized seminiferous tubules, and thyroid parameters. In conclusion, VGP can contribute to complex environmental EDC mixtures and may adversely affect the reproductive and thyroid health of both humans and wildlife. The varied topography of individual cattle feedlots will govern the rate and extent of effluent runoff, thus continuous monitoring of VGPs in aquatic systems surrounding cattle feedlots is necessary.

Objective: To study the effects of genistein (GEN) on reproductive system in prepubertal male rats. Methods: Thirty SPF-rated male SD rats were randomly divided into control group (Con group), low-dose group (G1 group) and high-dose group (G2 group), with 10 rats in each group. Corn oil, 150 mg/kg and 300 mg/kg GEN dissolved in corn oil of equal volume were respectively administered every day and weighed the next day. After 6 weeks, the rats were sacrificed, and the testis, epididymis and prostate were dissected, and organ coefficients were calculated. Histopathological changes of testis was observed. The number of sperm was counted and the rate of sperm malformation was calculated. The concentrations of serum testosterone and estradiol were detected by radioimmunoassay. The protein phosphatase 2, regulatory subunit B, gamma (PPP2R2C) protein expression in testicular tissue was detected by immunofluorescence assay. The mRNA and protein expression levels of PPP2R2C and cyclin dependent protein kinases 2 (CDK2) in rat testis were detected by real-time quantitative fluorescence PCR (RT-qPCR) and Western blot, respectively. The protein phosphatase 2A (PP2A) activity in testicular tissue was detected by immunoprecipitation. Results: There were no statistically significant differences in body mass, sperm number, serum estradiol and PP2A enzyme activity among the groups ( P>0.05). The pathological structure of testicular in G2 group was disordered. Sperm abnormality rate in G1 and G2 groups was higher than that in Con group ( P<0.05). Serum testosterone concentration in G2 group was lower than that in Con group ( P<0.05). The expression of PPP2R2C and CDK2 in G2 group was higher than that in Con group ( P<0.05), but the protein level was lower than that in Con group ( P<0.05). PPP2R2C protein was expressed in testicular tissue in each group. Conclusion: Long-term exposure to high dose (300 mg/kg) GEN during prepuberty may cause adverse effects on reproductive function in adult male rats. Further investigation is needed to determine whether PPP2R2C-PP2A-CDK2 phosphorylation pathway affects reproductive system in rats.

In the present study, we investigated the effects of Kigelia africana (Lam.) Benth. fruits ethanolic extract in prepubertal male rats, to evaluate the influence of the extract on the reproductive system and on pubertal development. Experiments were conducted using the rodent pubertal male assay. The plant extract, analyzed by TLC, HPLC-PDA and HPLC-ESI-MS, was administered orally at doses of 200, 400 and 800 mg/kg b.w. from post-natal date 21 to post-natal day 53. Age at puberty onset, body growth, development of sexual organs exposure to plant extract or positive control were examined. Results obtained indicate that Kigelia extract, at all doses tested, significantly anticipates puberty and increases body growth and sexual organs development. These effects appears to be due to stimulation of the secretion of androgenic hormones by the compounds found in its extract and scientifically support some of its traditional uses in disorders of the male reproductive system.

Ultrasonography can provide information about the integrity of organs; however, rarely is applied to the reproductive organ evaluation of bulls. The objective of the present study was to characterize and compare values for variables and ultrasonographic characteristics of the testes, epididymis and accessory sex glands, as well as spectral Doppler indices of the testicular and internal iliac arteries, between peri- and post-pubertal Nelore and Caracu bulls. Nelore (n = 203) and Caracu (n = 79) bulls were assigned by age class: peri-pubertal (12-15 months) and post-pubertal (> 22 months). Data were analyzed using SAS's PROC MIXED procedure (P < 0.05). The biometric variables of the testes and cauda epididymis differed between peri- and post-pubertal Nelore and Caracu bulls. There was a difference between breeds for the vesicular glands, ampulla of vas deferens, disseminate portion of the prostate, and craniocaudal dimension of the bulbourethral glands. Echogenicity of the testicular parenchyma differed between breeds and age classes. The pulsatility and resistive indices of the testicular arteries differed between Nelore and Caracu bulls. The biometric and ultrasonographic characteristics of the testes, epididymis and accessory sex glands, as well as of the arterial indices in bulls are affected by genetic group and age class, and when assessed there is useful information regarding the progression of sexual maturation.

A gynandromorph of Culex sitiens Wiedemann (Diptera: Culicidae) was attracted to a human during a mosquito surveillance programme conducted in Kuala Lipis, Pahang, Malaysia on July 20, 2019. Gynandromorphism was observed in antennae, maxillary palps, legs and wings of the specimen, with distinct male characters on the left and female characters on the right, though the left maxillary palp is slightly shorter than the proboscis of a typical male. The abdomen, however, displays well-developed male genitalia. This study represents the first report of oblique gynandromorphism in Cx. sitiens, one of the vectors of Japanese encephalitis in Asia.

In most cases, male sexual differentiation occurs with SRY gene mediation. However, exceptional genotypes have been identified, as shown in this paper. This was a male adult patient seen at the Servicio de Paternidades, Instituto de Genética, Universidad Nacional de Colombia. The following procedures were carried out: Amelogenin gene and short tandem repeat analyses using human identification commercial kits, conventional karyotype, SRY fluorescent in situ hybridization, PCR analysis for Y chromosome microdeletions, clinical evaluation, and genetic counseling. We present an adult male with unambiguous genitalia, karyotype 46,XX, and an SRY negative and ZFY positive molecular profile. The diagnosis of nonsyndromic 46,XX testicular disorder of sex development (DSD) -a rare genetic condition- was established. Only 20 % of similarly diagnosed patients are SRY negative and exhibit diverse molecular profiles. Until now, available evidence seems to indicate that, even in the absence of SRY, the ZFY factor is involved in male sexual differentiation.

Chronic Wasting Disease (CWD) is a prion disease affecting several cervid species. Among them, white-tailed deer (WTD) are of relevance due to their value in farming and game hunting. The exact events involved in CWD transmission in captive and wild animals are still unclear. An unexplored mechanism of CWD spread involves transmissions through germplasm, such as semen. Surprisingly, the presence and load of CWD prions in semen and male sexual tissues from WTD has not been explored. Here, we described the detection of CWD prions in semen and sexual tissues of WTD bucks utilizing the Protein Misfolding Cyclic Amplification (PMCA) technology. Samples were obtained post-mortem from farmed pre-clinical, CWD positive WTD bucks possessing polymorphisms at position 96 of the PRNP gene. Our results show that overall CWD detection in these samples had a sensitivity of 59.3%, with a specificity of 97.2%. The data indicate that the presence of CWD prions in male sexual organs and fluids is prevalent in late stage, pre-clinical, CWD-infected WTD (80%-100% of the animals depending on the sample type analyzed). Our findings reveal the presence of CWD prions in semen and sexual tissues of prion infected WTD bucks. Future studies will be necessary to determine whether sexual contact and/or artificial inseminations are plausible means of CWD transmission in susceptible animal species.

RESUMO

Seven new species of Neotrichia Morton 1905 from Brazil are described: Neotrichia caboca sp. nov., Neotrichia capitiana sp. nov., Neotrichia cruviana sp. nov., Neotrichia damurida sp. nov., Neotrichia macuxi sp. nov., Neotrichia makunaima sp. nov., and Neotrichia matula sp. nov. Illustrations of male genitalia are provided with each description. New country records for Brazil of two additional species, Neotrichia tauricornis Malicky 1980 and Neotrichia parabullata Harris Armitage 2015 are also provided.

RESUMO

Nine species of Sycacantha Diakonoff, 1959 are recorded from China. Among them, three are described as new: S. typicusivalva, sp. nov., S. camerata, sp. nov., and S. decursiva, sp. nov. Two new combinations are proposed based on DNA barcodes and characters of the male genitalia: S. diserta (Meyrick, 1909), comb. nov., and Phaecasiophora obtundana (Kuznetzov, 1988), comb. nov. Sycacantha complicitana (Walker, 1863) and S. catharia Diakonoff, 1973 are newly recorded from China. Photographs of adults and genitalia of the new species and new combinations are provided, and a key to the species based on genitalia is given.
