RESUMEN

The effects of dietary Lagnaria breviflora leaves extract (LBLE) on the growth performance, feed utilisation and haematological parameters of juveniles African catfish, Clarias gariepinus (mean = 8.45 ± 0.6 g) raised in a flow water (mean temperature 26.7°C, mean pH 7.52, mean dissolved oxygen 6.05 mg/L) were assessed in this study. Diets (400 g/kg crude protein; 4631.5 kcal/kg gross energy) supplemented with LBLE at levels of 0 (control), 0.5, 1.0, 1.5 or 2.0 g/kg diet were served to fish in triplicates (each replicate has 20 fish) until they appeared satiated for 12 weeks. Then, they were challenged with Aeromonas hydrophila infection for further 2 weeks. Fish served dietary LBLE had considerable higher weight gain (64.76 g) and specific growth rate (765.5% g/day) than the fish group fed with the control diet with a reduced feed conversion ratio (1.45) (p < 0.05). Compared to fish fed with the control diet, the villi height (146.30 µm) and width (284.35 µm) of the intestines increased greatly in a dose (LBLE in diets) dependent order. Similarly, dietary LBLE increased (p < 0.05) the packed cell volume (46.11%) and haemoglobin (17.03 g/dL), whereas 1.5 g of LBLE increased the counts of white blood cells. Glutathione S-transferase (769.43 U/mg protein), glutathione peroxidase (84.14 U/mg protein) and superoxide dismutase (433.15 U/mg protein) activities were significantly higher (p < 0.05) in fish fed with diets supplemented with LBLE compared to the control one. Additionally, dietary LBLE increased phagocytic and lysozyme activities and protected C. gariepinus against bacterial infections where the lowest death was observed in the fish fed on the diet containing 1.5 g LBLE/kg feed. These findings showed that the fish fed with LBLE/kg diet improved their immune system, antioxidant and growth performance in addition to providing protection from A. hydrophila infection with the optimum dose of 1.80 g/kg diet.

Edwardsiella tarda is one of the most common causes of fish diseases that hinder aquaculture. Oxidative stress in farm animals can induce a number of pathological disorders, production and general animal welfare. The use of exogenous dietary nonenzymatic antioxidants such as alpha-lipoic acid (ALA) can stop a pro-oxidant state and thus appears to have the potential to modulate the immune system and protect fish from bacterial infection. Thus, this study investigates the stimulatory effect of dietary ALA on growth performance, antioxidant capacity, liver enzymes, immunity and protection of African catfish, Clarias gariepinus (B.), against an infection with E. tarda. Five isonitrogenous and isocaloric diets (400 g/kg of crude protein) containing ALA at doses of 0.0 (control), 500, 1000, 1500 or 2000 mg/kg diet were served to 300 juveniles of African catfish (mean weight = 8.2 ± 0.2 g) adequately thrice per day for 12 weeks. Thereafter, 0.1 mL of E. tarda (ATCC 15947; 1.0 × 108 CFU/mL) was intraperitoneally injected into 10 fish from each tank and was monitored for 14 days. The results showed that ALA-fortified diets significantly boosted the fish growth, feed consumption and utilization and feed conversion ratio but no did not affect fish survival rate. The highest final fish weight (g), weight growth (g) and weight gain (%) were all considerably higher in fish fed with ALA-fortified diets (p < 0.05), especially from 1000 to 200 mg/kg ALA than the control group. Also, an enhanced hemato-biochemical, antioxidant and immune indices were noticed in African catfish-fed ALA-enriched diets. In a dose-dependent order, the levels of haematological indices such Ht, Hb, RBCs, WBCs and platelets were markedly increased (p < 0.05). Additionally, fish fed with ALA-based diets showed substantial (p < 0.05) declines in aspartate and alanine aminotransferase values, with the lowest values being found in the 2000 mg/kg diet while control group had highest values. Further, African catfish fed the feed fortified with 2000 mg ALA/kg diet showed the highest levels of lysozyme, respiratory burst, proteases and esterase activities (p < 0.05). Following exposure of fish to E. tarda infection, a significant reduction in the mortality was obtained in African catfish fed with ALA-based diets, especially from 1500 to 2000 mg ALA/kg diet (3.3%); while fish fed with the control diet had highest mortality (86.7%). Therefore, diets supplemented with ALA evoked fish growth performance, antioxidants and nonspecific immunity of African catfish. Also, resistance of African catfish to E. Tarda infection were raised when fed ALA-fortified diets at optimum inclusion rate of 1300 mg ALA/kg diet.

Effects of dietary Euphorbia heterophylla extract (EH) on growth performance, feed utilization and haemato-biochemical parameters in African catfish, Clarias gariepinus, juveniles were evaluated in this study. Diets fortified with EH at 0 (control), 0.5, 1.0, 1.5 or 2.0 g/kg were fed to the fish to apparent satiation for 84 days before challenging it with Aeromonas hydrophila. The weight gain, specific growth rate and protein efficiency ratio of fish fed EH-supplemented diets were significantly higher but lower feed conversion ratio (p < 0.05) than the control group. The villi height and width at the proximal, mid and the distal of the guts rose significantly with the increasing levels of EH from 0.5 to 1.5 g than the fish fed basal diet. Dietary EH enhanced (p < 0.05) the packed cell volume and haemoglobin, whereas 1.5 g EH boosted white blood cell, in relation to their counterpart in the control group. There were significant increase in the activities of glutathione-S-transferase, glutathione peroxidase and superoxide dismutase (p < 0.05) in the fish that were fed diets supplemented with EH than the control. Dietary EH also enhanced phagocytic activities, lysozyme activities and relative survival (RS) of C. gariepinus than the control group, with the highest RS obtained in fish that were fed diet containing EH at 1.5 g/kg level. These results revealed that the fish fed 1.5 g/kg dietary EH promoted growth performance, antioxidant and immune profiles, as well as protection against A. hydrophila infection.

The microbial phytase, derived from Buttiauxella gaviniae, Yersinia mollarettiv and Hafnia spp., is proven to be safe for avian and porcine feeding and promotes their overall growth performance. Here, we have evaluated microbial phytase's effects on the growth, bone mineral content, antioxidant status, immune responses and the resistance of African catfish (Clarias gariepinus) fed with high soybean meal-based diets against Aeromonas hydrophila infection. Five isonitrogenous diets (40% protein) were supplemented with different levels of microbial phytase ranging from 0 as a control to 250, 500, 750 and 1000 FTU/kg diet. African catfish (n = 300; 8.5 ± 0.3 g) were allocated in 15 50-L tanks (in triplicates) and were fed on the prepared tested diets for 12 weeks. After the end of the feeding period of 12 weeks, 10 fish from each replicate was intraperitoneally infected with A. hydrophila (0.5 × 105 CFU/ml) and monitored for 14 days. Dietary phytase levels linearly and quadratically improved the growth performance of African catfish and stimulated feed intake. Bone levels of calcium, phosphorus, magnesium and zinc were also positively modulated in phytase-fed fish, especially at 750-1000 FTU/kg diet. Similarly, counts of red and white blood cells as well as haemoglobin, packed cells volume, platelets, lymphocytes and heterocytes were significantly modulated in all fish fed with phytase-supplemented diets. Higher levels of serum total protein, albumin and globulin were also observed in fish fed with a 750-1000 FTU/kg diet of phytase. Conversely, aspartate and alanine aminotransferase activities were lower in fish fed with a 750-1000 FTU/kg diet of phytase, compared to those fed the control diet. Moreover, antioxidant enzymes (superoxide dismutase, catalase and glutathione peroxidase), reduced glutathione levels and immune responses (lysozyme, respiratory burst, protease and alkaline phosphatase activities) biomarkers were linearly and quadratically elevated, while malondialdehyde values were linearly and quadratically decreased in fish groups fed with phytase-based diets. After administering A. hydrophila, 60.0% of the fish fed the control diet perished, while no mortalities were observed in fish fed with 750-1000 FTU/kg diets. Taken together, the current study reveals that dietary phytase could improve the growth performance, blood profile, bone mineralization, antioxidant activities, immunity and overall protection of African catfish against A. hydrophila infection. Dietary phytase may be efficiently used in the feeding of African catfish to enhance their overall performance and mitigate health conditions with optimum level of 900 FTU/kg diet.

In the current study, growth performance, biochemical constituents of muscle, activities of enzymes in the haemolymph, and expressions of immune-related genes were evaluated in the giant freshwater prawns Macrobrachium rosenbergii fed diets supplemented with aqueous garlic (Allium sativum) extract at 0, 5, 10 and 20 g/kg (w/w) for 60 days. At the end of the feeding trial, weight gain and specific growth rate were significantly improved in garlic-fed prawn groups compared with the control (p < 0.05). Moreover, feed conversion ratio was significantly lower in the garlic-fed groups than in the control (p < 0.05). Activities of catalase, superoxide dismutase and glutathione peroxidase in the hepatopancreas, activities of alanine aminotransferase, aspartate aminotransferase and levels of albumin and total protein in the hemolymph were significantly increased in the garlic treatments (p < 0.05). Furthermore, garlic supplemented diets improved muscle biochemical profiles, particularly contents of crude protein and total ash and upregulations of immune deficiency and heat shock proteins (HSP70) gene expression (p < 0.05). Therefore, garlic has positive effects on growth performance and physio-biochemical responses of M. rosenbergii, and thus, it can be used as an additive for stress resistance and as a growth promoter in sustainable aquaculture.

This study evaluated the effect of Mitracarpus scaber leaves extract (MSLE) as a dietary supplement on growth performance, physiological, antioxidants, and mucosal immune profiles of North African catfish, Clarias gariepinus, and resistance to Edwardsiella tarda infection. Four isonitrogenous diets containing 0, 2, 4, or 6 MSLE g/kg were fed to North African catfish fingerlings (n = 240; mean weight = 5.80 g) for 56 days. After the feeding trial, 10 fish from each experimental tank were intraperitoneally injected with E. tarda (ATCC 15,947; 1.0 × 108 CFU/mL) while clinical signs and mortalities were observed for 14 days. The results show that North African catfish fed dietary MSLE had better growth performance, physiological antioxidants, and mucosal immune profiles than when fed a control diet. The best growth, physiological, antioxidant, and mucosal immune response profiles were noticed in the fish fed diet containing 6 MSLE g/kg. Furthermore, fish fed 6 MSLE g/kg diet had a higher survival rate after E. tarda infection. These results indicated that supplementation of MSLE at 6 g/kg improves growth, physiological, antioxidant, and mucosal immune profiles, and promotes resistance of North African catfish.

The aim of this study was to assess the effects of different dietary selenium sources, selenium nanoparticle (nSe), and selenomethionine (SeMet) as feed additives on growth performance, hepatic enzymes' activity, biochemical, mucosal immune parameters, liver histology, and appetite-related gene transcript in goldfish (Carassius auratus). At first, goldfish juveniles (n=480; mean 4.54 g) were fed dietary selenium nanoparticle at 0, 0.3, 0.6, and 0.9 mg nSe/kg diet and SeMet at 0, 0.3, 0.6, and 0.9 mg Se/kg for 9 weeks. Growth performance was evaluated using standard procedures. Blood, skin mucus, and tissue samples (liver and intestine) were collected for biochemical, mucosal immune response, histology, and ghrelin and insulin-like growth factor-I (IGF-I) gene expression. The results showed that fish fed diets fortified with 0.6 mg nSe/kg and 0.6 mg Se/kg had a significant higher weight gain, specific growth rates (SGR), and lower feed conversion ratios (FCR) than fish fed basal diets (p<0.05). Furthermore, dietary nSe and SeMet enhanced blood biochemical profiles especially alkaline phosphatase (ALP) (p < 0.05) and mucosal immunity than the control group in goldfish. Moreover, the liver histological investigation showed that fish fed 0.9 mg of SeMet and nSe kg-1 diets had higher liver lesion scores such as karyolysis, lipidosis, and hyperemia while fish fed 0, 0.3, and 0.6 mg of SeMet and nSe kg-1 diets had small liver changes at 9 weeks. The study further established that inclusion of nSe and SeMet in the diet of goldfish greatly promoted ghrelin and IGF-1genes expressions (p <0.05). Overall, dietary nSe performs better than SeMet and basal diets. The results evoked that nSe and SeMet stimulate the growth, biochemical, and mucosal immunity in goldfish at 0.6 mg/kg.

This study investigated the effect of Lactobacillus acidophilus-fortified diets on growth performance, antioxidant profiles, and immunity-related gene expressions of common carp, Cyprinus carpio juveniles. Four isonitrogenous diets containing 0, 102, 104, or 106 cfu/mL/kg L. acidophilus were fed to 240 Cyprinus carpio juveniles (mean = 21.34 ± 1.85 g), allotted to 12 rectangular tanks in a completely randomized designed at 3% body weight for 56 days. Growth performance and nutrient utilization were evaluated using standard procedures. Intestinal villi were measured, antioxidant profiles were evaluated from blood sera, and immunity-related gene expressions were evaluated. The results revealed that fish fed dietary 106 had significantly higher weight gain, SGR, feed intake, and lower FCR. Also, villi length, width, and areas of absorption were significantly improved in a dose-dependent manner. Furthermore, innate immune profiles, superoxide dismutase, catalase, respiratory bust activity as well as transforming growth factor beta (TGFß), interleukin 8 (IL-8), and tumor necrosis factor alpha (TNF-alpha) were significantly stimulated. This study evoked that Lactobacillus acidophilus supplementation enhanced better growth performance, improved antioxidant profiles, and modulated expression of immune-related genes of common carp, Cyprinus carpio, than the fish fed control diet. Results show that fish fed fortified diets had better growth performance, improved antioxidant profiles, and modulated expression of immune-related genes.

Plant protein (PP) sources are generally used in high levels in fish diets. Mostly, PP sources are deficient in taurine; hence, there is a need for its supplementation to fish fed high PP diets. Therefore, effects of dietary taurine were examined on growth performance, feed utilization, immunity, and antioxidant parameters of African catfish, Clarias gariepinus (B.). Fish (10.3 ± 0.4 g) were fed on diets (40% crude protein) containing different taurine levels of 0 (control), 10, 20, 30, or 40 g/kg diet for 12 weeks. Fish fed a taurine-free diet (the control) with high PP sources showed poor growth as compared with these fed taurine-enriched diets where taurine stimulatory effects were observed on fish growth and feed intake. Feed conversion ratio and fish survival rate were not significantly differed among different treatments. Fish fed taurine-enriched diets showed also higher levels of serum glucose, cholesterol, total protein, albumin, globulin, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, urea, and creatinine over that fed the control diet. Furthermore, lysozyme and respiratory burst activities as well as superoxide dismutase and catalase activities were significantly elevated in fish fed taurine-enriched diets (P < 0.05) and their highest levels were observed in fish fed 30 g/kg diet. Additionally, taurine deposition in fish muscles was positively correlated with dietary taurine levels (P < 0.05). The present study concludes that taurine is a limiting factor for growth, immunity, and antioxidants responses of African catfish fed high PP-based diets and it should be incorporated in its diets with an optimum level of 20 g/kg diet.

Clove basil, Ocimum gratissimum, is a native plant to Africa and grows virtually in tropical and subtropical regions. It has good aroma and its leaves have become used as a spicy and in traditional medicine. The use of plant leaves in fish diets may deteriorate their growth because it may content anti-nutritional factors. Thus, it is better to use plants leaves extract. In the current study, clove basil leaves extract (CBLE) was administrated to African catfish, Clarias gariepinus (B.) to evaluate its effect on growth performance, physiological, antioxidants, and innate immunity variables. Fish (10.7 ±â€¯0.5 g) were fed on diets enriched with 0.0, 5, 10, or 15 g CBLE/kg diet for 12 weeks. After the feeding trial, fish were further exposed to pathogenic bacteria (Listeria monocytogenes) for 14 days. Fish performance and feed intake were significantly enhanced with increasing CBLE levels and its optimum level is found to be 12 g/kg diet. It is noticed that the dietary CBLE in African catfish diets increased significantly the intestinal villi length, villi width, and absorption area in a dose-dependent manner and fish weight was highly correlated with villi length, villi width, and absorption area (R2 = 0.91, 0.91, and 0.92, respectively). On the other side, Dietary CBLE has significant modulatory effect on hemato- and physiological variables of African catfish in a dose-dependent manner. In this regard, blood glucose and cholesterol levels decreased significantly; mean while total protein, albumin, and globulin increased significantly in fish fed high CBLE levels (10-15 g/kg diet). Furthermore, activities of aspartate aminotransferase and alanine aminotransferase, alkaline phosphatase, urea, and creatinine levels were significantly elevated with increasing dietary CBLE levels and their maximum values were detected in fish fed 15 g CBLE/kg diet. Antioxidants and immunity variables were significantly enhanced by CBLE supplementation. Additionally, fish mortality after bacterial challenge was highest in fish fed the control diet (85%) than those fed CBLE-enriched diets. The lowest fish mortality was observed in fish fed 15 g CBLE/kg diet (13.5%). This study evoked that CBLE administration enhanced the performance, feed utilization, antioxidant, and innate immunity properties of African catfish with optimum level of 12 g/kg diet. Also, its supplementation enhanced fish challenge against L. monocytogenes.
