RESUMEN

The combined contamination of terrestrial environments by metal(loid)s (MEs) and microplastics (MPs) is a major environmental issue. Once MPs enter soils, they can interact with MEs and modify their environmental availability, environmental bioavailability, and potential toxic effects on biota. Although research efforts have been made to describe the underlying mechanisms driving MP and ME interactions, the effects of MPs on ME bioavailability in terrestrial Mollusca have not yet been documented. To fill this gap, we exposed the terrestrial snail Cantareus aspersus to different combinations of polystyrene (PS) and arsenic (As), cadmium (Cd), or mercury (Hg) concentrations. Using kinetic approaches, we then assessed the variations in the environmental availability of As, Cd or Hg after three weeks of equilibration and in the environmental bioavailability of As, Cd or Hg to snails after four weeks of exposure. We showed that while environmental availability was influenced by the total ME concentration, the effects of PS were limited. Although an increase in As availability was observed for the highest exposure concentrations at the beginning of the experiment, the soil ageing processes led to rapid adsorption in the soil regardless of the PS particle concentration. Concerning transfers to snail, ME bioaccumulation was ME concentration-dependent but not modified by the PS concentration in the soils. Nevertheless, the kinetic approaches evidenced an increase in As (2- to 2.6-fold) and Cd (1.6-fold), but not Hg, environmental bioavailability or excretion (2.3- to 3.6-fold for As, 1.8-fold for Cd) at low PS concentrations. However, these impacts were no longer observable at the highest PS exposure concentrations because of the increase in the bioaccessibility of MEs in the snail digestive tract. The generalization of such hormetic responses and the identification of the precise mechanisms involved necessitate further research to deepen our understanding of the MP-mediated behaviour of MEs in co-occurring scenarios.

Cadmium (Cd) and lead (Pb) are known to enhance immune cell damages and to decrease cellular immunity, promoting higher susceptibility to infectious diseases. Selenium (Se) is an essential element involved in immunity and reactive oxygen species scavenging. This study aimed at evaluating how Cd and Pb and low nutritional (Se) quality modulate immune response to a bacterial lipopolysaccharide (LPS) challenge in wood mice (Apodemus sylvaticus). Mice were trapped near a former smelter in northern France in sites of High or Low contamination. Individuals were challenged immediately after capture or after five days of captivity, fed a standard or a Se-deficient diet. Immune response was measured with leukocyte count and plasma concentration of TNF-α, a pro-inflammatory cytokine. Faecal and plasma corticosterone (CORT), a stress-hormone involved in anti-inflammatory processes, was measured to assess potential endocrine mechanisms. Higher hepatic Se and lower faecal CORT were measured in free-ranging wood mice from High site. LPS-challenged individuals from High site showed steeper decrease of circulating leukocytes of all types, higher TNF-α concentrations, and a significant increase of CORT, compared to individuals from Low site. Challenged captive animals fed standard food exhibited similar patterns (decrease of leukocytes, increase of CORT, and detectable levels of TNF-α), with individuals from lowly contaminated site having higher immune responses than their counterparts from highly polluted site. Animals fed Se-deficient food exhibited lymphocytes decrease, no CORT variation, and average levels of TNF-α. These results suggest (i) a higher inflammatory response to immune challenge in free-ranging animals highly exposed to Cd and Pb, (ii) a faster recovery of inflammatory response in animals lowly exposed to pollution when fed standard food than more exposed individuals, and (iii) a functional role of Se in the inflammatory response. The role of Se and mechanisms underlying the relationship between glucocorticoid and cytokine remain to be elucidated.

The Réunion harrier is an endemic raptor on Réunion Island. Several threats endanger its population, poisoning by rodenticides being considered as the main one currently. No information is available on its exposure to other chemicals notably trace metal elements such as lead (Pb), mercury (Hg), and cadmium (Cd). The Réunion harrier is still a victim of poaching nowadays. When shooting is not lethal, animals may carry embedded shot in their body and thus be exposed to toxic level of Pb as demonstrated for other raptors. Moreover, recent monitoring suggests a decrease of its breeding success over time. It is known that Hg and Cd could impair reproduction and disturb embryo development in birds. The aim is to measure metal concentrations in the tissues of 30 carcasses of harrier collected from 2016 to 2021. Lead was analyzed in the liver and humerus, while Hg and Cd were measured in livers. Concentrations were compared to toxicological reference values. Overall, the Réunion harrier was not exposed to toxic levels of Pb or Cd. For Hg, 53% of the individuals have residues higher than the threshold compatible with oxidative stress, and 13% have liver concentrations above those compatible with reproduction impairment. A positive correlation was found between the proportion of urban habitat in a 55 km2 area centered on the location where the harrier was found and the concentration of Hg in the liver. We conclude that Hg exposure could be a threat for the Réunion harrier population and recommend monitoring the exposure of the most sensitive stages, i.e., embryos and nestlings, to this metal with non-invasive methods.

Environmental risk assessment of contaminated soils requires bioindicators that allow the assessment of bioavailability and toxicity of chemicals. Although many bioassays can determine the ecotoxicity of soil samples in the laboratory, few are available and standardized for on-site application. Bioassays based on specific threshold values that assess the in situ and ex situ bioavailability and risk of metal(loid)s and polycyclic aromatic hydrocarbons (PAHs) in soils to the land snail Cantareus aspersus have never been simultaneously applied to the same soils. The aims of this study were to compare the results provided by in situ and ex situ bioassays and to determine their respective importance for environmental risk assessment. The feasibility and reproducibility of the in situ bioassay were assessed using an international ring test. This study used five plots located at a former industrial site and six laboratories participated in the ring test. The results revealed the impact of environmental parameters on the bioavailability of metal(loid)s and PAHs to snails exposed in the field to structured soils and vegetation compared to those exposed under laboratory conditions to soil collected from the same field site (excavated soils). The risk coefficients were generally higher ex situ than in situ, with some exceptions (mainly due to Cd and Mo), which might be explained by the in situ contribution of plants and humus layer as sources of exposure of snails to contaminants and by climatic parameters. The ring test showed good agreement among laboratories, which determined the same levels of risk in most of the plots. Comparison of the bioavailability to land snails and the subsequent risk estimated in situ or ex situ highlighted the complementarity between both approaches in the environmental risk assessment of contaminated soils, namely, to guide decisions on the fate and future use of the sites (e.g., excavation, embankments, and land restoration). RESUMEN

We experimentally demonstrate the generation of a short-wave infrared supercontinuum in an uncladded silicon nitride (Si3N4) waveguide with extreme polarization sensitivity at the pumping wavelength of 2.1 µm. The air-clad waveguide is specifically designed to yield anomalous dispersion regime for transverse electric (TE) mode excitation and all-normal-dispersion (ANDi) at near-infrared wavelengths for the transverse magnetic (TM) mode. Dispersion engineering of the polarization modes allows for switching via simple adjustment of the input polarization state from an octave-spanning soliton fission-driven supercontinuum with fine spectral structure to a flat and smooth ANDi supercontinuum dominated by a self-phase modulation mechanism (SPM). Such a polarization sensitive supercontinuum source offers versatile applications such as broadband on-chip sensing to pulse compression and few-cycle pulse generation. Our experimental results are in very good agreement with numerical simulations.

We demonstrate computational spectral-domain ghost imaging by encoding complementary Fourier patterns directly onto the spectrum of a superluminescent laser diode using a programmable spectral filter. Spectral encoding before the object enables uniform spectral illumination across the beam profile, removing the need for light collection optics and yielding increased signal-to-noise ratio. In addition, the use of complementary Fourier patterns allows reduction of deleterious of parasitic light effects. As a proof-of-concept, we measure the wavelength-dependent transmission of a Michelson interferometer and a wavelength-division multiplexer. Our results open new perspectives for remote broadband spectral measurements.

To date, no study has linked the environmental and the ecogenotoxicological bioavailability of contaminants to land snails. Yet, understanding the specific ecotoxicological mechanisms from bioaccumulation to genotoxicity is necessary e.g., to build an adverse outcome pathway relevant to risk assessment. Consequently, the aim of our study is to look for relationships between accumulated concentrations of As, Cd and Hg in sub-adult snails and ecotoxicological effects at the individual (survival and growth) and molecular (genomic stability) levels. This study combines random amplified polymorphic DNA (RAPD) coupled with high-resolution capillary electrophoresis system (HRS) and micronucleus (MN) assay on haemocytes to consider various types of cytogenomic damage, such as chromosomal aberrations, breakages, adducts and mutations. The results showed alteration of the individual endpoints at higher accumulation quotients (AQs) that reflect the excess of transfers to snails, especially with decreased survival for As. In addition, genotoxic effects were observed with an increased occurrence of MN in haemocytes for the three meta(loid)s considered (R2 from 0.57 to 0.61 as a function of the meta(loid)s). No concentration-dependent decrease in genome stability was highlighted by RAPD-HRS in snails exposed to As and Cd but not Hg. Our results demonstrate the complementarity of the RAPD-HRS and the MN assay for understanding the different genotoxic mechanisms of the three metal(loid)s studied in land snails. They show a way to better assess environmental risks of contaminated soils by associating ecotoxicity, genotoxicity and bioaccumulation assays (ISO 24032), i.e., ecogenotoxicological bioavailability. Convergences highlighted here between the bioaccumulation of metal(loid)s in viscera and genotoxic effects in haemocytes constitute a way to better assess the bioavailability of contaminants in soils to the land snail and the subsequent environmental risk.

We demonstrate experimentally ghost optical coherence tomography using a broadband incoherent supercontinuum light source with shot-to-shot random spectral fluctuations. The technique is based on ghost imaging in the spectral domain where the object is the spectral interference pattern generated from an optical coherence tomography interferometer in which a physical sample is placed. The axial profile of the sample is obtained from the Fourier transform of the correlation between the spectrally resolved intensity fluctuations of the supercontinuum and the integrated signal measured at the output of the interferometer. The results are in excellent agreement with measurements obtained from a conventional optical coherence tomography system.

Ghost imaging is a technique that generates high-resolution images by correlating the intensity of two light beams, neither of which independently contains useful information about the shape of the object. Ghost imaging has been demonstrated in both the spatial and temporal domains, using incoherent classical light sources or entangled photon pairs. Here we exploit the recent progress in ultrafast real-time measurement techniques to demonstrate ultrafast, scan-free, ghost imaging in the frequency domain using a continuous spectrum from an incoherent supercontinuum light source with random spectral fluctuations. We demonstrate the application of this technique to broadband spectroscopic measurements of methane absorption performed with sub-nanometer resolution. Our results offer novel perspectives for remote sensing in low light conditions, or in spectral regions where sensitive detectors are lacking.

We demonstrate cantilever-enhanced photoacoustic spectroscopy in the mid-infrared using a supercontinuum source. The approach is broadband and allows for higher photoacoustic signal intensity and an enhanced signal-to-noise ratio as compared to systems employing conventional black body radiation sources. Using this technique, we perform spectroscopic measurements of the full ro-vibrational band structure of water vapor at 1900 nm and methane at 3300 nm with relative signal enhancement factors of 70 and 19, respectively, when compared to measurements that use the black body radiation source. Our results offer a novel perspective for photoacoustic detection opening the door to sensitive broadband analyzers in the mid-infrared spectral region.
