RESUMEN

This study aimed to determine the expressions of HSP27, HSP60, HSP70, and HSP90 in rat ovaries during the oestrous cycle, pregnancy, and lactation. In follicle cells, HSP27 and HSP70 expression was not observed. HSP60 in oocytes was higher in the early stages of follicular development but decreased and disappeared as the follicle grew. HSP60 in granulosa and theca cells increased with follicle development and decreased with atresia. HSP90 in follicle cells did not change during follicle development or atresia. The expression of HSPs in interstitial cells was higher in the proestrus and estrus phases of the estrous cycle. The expression of HSPs in these cells was higher on day 5 of pregnancy, decreased on day 10, and decreased further on days 15 and 20. The expression of HSPs, which decreased in the second half of pregnancy, increased again on the first day of lactation. The expression of HSPs then decreased on day 5 of lactation and further decreased on days 10 and 20. HSP60 and HSP90 were positive in new and old corpus luteums (CLs) and their expression did not change during luteal development or regression. HSP27 and HSP70 were absent in new CLs. HSP27 was positive in old CLs and showed the same staining pattern during luteal regression. HSP70 expression was determined in old cyclic CLs during the oestrous cycle and pregnancy and decreased with luteal regression. HSP70 expression in old pregnancy CLs during lactation was very weak compared to the oestrous cycle and pregnancy. In conclusion, HSP60 and HSP90 may participate in folliculogenesis, luteal development, and steroidogenesis in luteal cells, and HSP27, HSP60, HSP70, and HSP90 may be effective in luteal regression and steroidogenesis in interstitial cells. HSP27 and HSP70 may be used as markers to identify old CLs in rats.

The present study was undertaken to evaluate the efficacy of 2 graded levels (0.03 and 0.05% of diet) of a monoglyceride blend containing butyric, caprylic, and capric acids in broilers' diet for optimizing gut structure and animal growth performance. For this purpose, a total of 210, one-day-old male Ross 308 broiler chicks were randomly allocated to 3 experimental treatments using 7 replicates each and 10 birds/replicate. The treatment groups involved supplementation of blend of short and medium chain fatty acids at the level of 0, 0.03, and 0.05% of the diet for 42 d. The incorporation of mixes of monoglycerides into broilers' diet linearly improved BWG between d 0 and 21 (P = 0.034). At the end of trial, however, no significant changes were observed in performance indexes (BWG, FI, FCR). Jejunal morphometric parameters (villus height, crypt depth, and their ratio) remained unaltered with the monoglyceride supplementation on d 21. The results further showed that monoglycerides supplementation increased the goblet cell counts along the jejunal villi (P = 0.034) and crypt regions (P = 0.022), as well as it effectively modulated the mRNA abundances of tight junction protein (ZO-1, P = 0.033) and nutrient transporters (SGLT, PePT1; P = 0.005, 0.023, respectively) in the jejunum. Moreover, the downregulation in mRNA abundance of TNFα (P = 0.030) was observed with the monoglyceride supplementation. The SCFAs analysis of cecal contents showed no notable differences with monoglyceride blend supplementation when compared to the unsupplemented group. Collectively, high goblet cell numbers in the jejunum along with downregulation of the mRNA abundances of pro-inflammatory cytokines, upregulation of tight junction proteins, and nutrient transporters showed favorable responses of low doses of monoglycerides blend in broiler feeding. Further studies should be conducted in different rearing conditions to examine the effectiveness of such low levels of a monoglyceride blend in the modulation of gut structure, its functionality and animal performance.

Due to consumers' concerns about broiler welfare, slow-growing broilers (SGB) have also been used in production. Fast-growing broilers (FGB) reach slaughter weight earlier the slow growing. This study was aimed to determine the sex-related changes in fattening performance, dermatitis, intestinal histomorphology, and serum IgG level of SGB and FGB under the intensive system during the 10 weeks. A total of 336 one-day-old broilers were distributed into four equal groups (FGB-female, FGB-male, SGB-female, and SGB-male) consisting of a 2 × 2 factorial design. The main factors were genotype (FGB and SGB) and sex (female and male). Each treatment had 6 duplicates consisting of 14 broilers. They were placed on the litter pens. The experiment lasted 10 weeks. The body weight, body weight gain, feed consumption, and feed-to-gain ratio were determined weekly during the experiment. One broiler close to the average body weight from each pen (6 broilers in the group) has been slaughtered each week and intestinal histomorphology was determined. Additionally, IgG levels in the blood and dermatitis were determined. While the period in which the body weight gain was the highest in the FGB was up to 6 week of age, this period lasted until the 7 week of age in the SGB (P < 0.001). It has been determined that FGB have the best feed efficiency ability during both 6 weeks of fattening period and 10 weeks of fattening period (P < 0.001). The difference between the sex in terms of the feed efficiency appeared after the age of 4 weeks and this difference continued until the age of 9 weeks (P < 0.01). It was determined that villus height, crypt depth, and the ratio of villus height to crypt depth in the jejunum region were higher in FGB from the age of 1 week than in SGB (P < 0.05). It was observed that the incidence of dermatitis increased in the FGB (P < 0.001). It has been determined that the genotype (P < 0.01) and period (P < 0.001) were effective according to the IgG level. These results indicate that 7 and 8 weeks can be considered important periods in terms of economic efficiency in SGB and FGB reared for 10 weeks under the same management systems and it was observed that both genotypes would be more susceptible to diseases at the age of 6 weeks.

This study aimed to examine the teat characteristics in relation to the animal temperament during milking in the Anatolian buffaloes using ultrasonographic, histomorphological and immunohistochemical methods. The teat canal length (TCL), teat wall thickness (TWT), teat cisternal diameter (TCD), teat diameter (TD), teat length (TL), and teat circumference (TC) values in docile (n = 5) and nervous (n = 7) buffaloes were measured at the 0th, 3rd and 6th minute of stimulation. In additional experiments, comparative histomorphology and immunohistochemical examinations of buffalo (n = 7) and cow teats (n = 8) were performed. It was determined that post-stimulation mean TCL values were significantly higher in nervous buffaloes than those of teats in docile buffaloes (p < .05). A significant positive correlation between TCD and TD, TL and TC in both docile and nervous buffaloes was noted (p < .05). Unlike nervous buffaloes where only 3/14 teat canals were open by 3rd minute of milking stimulation, almost all (9/10) teat canals were observed opened in docile buffaloes. There were fewer muscle but more collagen bundles in buffalo teats compared with cow teats. It seems that temperament of animal during milking effects the milking efficiency, and in nervous buffaloes, probably the stimulation alone may not be sufficient for opening of the teat canal and hence achieve complete milking.

The aim of this study was to determine the serum concentrations, ovarian presence and expression of anti-Müllerian hormone (AMH) in pre-pubertal, bitches with signs of ovarian remnant syndrome (ORS) and intact bitches. In addition, we aimed to verify the suitability of serum AMH concentrations for diagnostic purposes in sterilized bitches and/or in suspected cases of ORS in the field of veterinary medicine. For this purpose, 36 healthy female dogs divided into six groups: proestrus, oestrus, dioestrus, anoestrus, pre-pubertal and ORS. Serum AMH concentrations were determined by electrochemiluminescence immunoassay, and ovarian presence and distribution of AMH was confirmed by immunohistochemical and qPCR techniques. According to the results of qPCR, while the expression values of AMH were at the highest concentrations in the proestrus and oestrus, there was a statistically significant decrease in these values at the later stages of the cycle (p < 0.05). According to hormone analysis, the serum AMH values of the ORS group had decreased significantly compared with the proestrus and oestrus (p < 0.05). Although serum AMH levels of ORS group were increased compared with anestrus and pre-pubertal groups, this increase was statistically non-significant (p > 0.05). Immunohistochemically, AMH expression was first observed in the granulosa cells of primordial follicles in folliculogenesis. Expression values were the highest in the proestrous and oestrus groups, but values from bitches in later stages of the cycle were statistically significant decrease in comparison with these groups (p < 0.05). As a result, AMH concentration and expression were found to be higher in proestrus and oestrus than in other periods (p < 0.05). In addition, the measurable level of AMH concentration in bitches with ORS is an indication that it can be used in the diagnosis of ORS.

Peyer's patches are known as the immune sensors of the intestine because of their ability to transport luminal antigens. The objective of this study was both to assess the prenatal and postnatal development of sheep ileal Peyer's patches with respect to histomorphology, distribution of CD4+ and CD8+ cells, and localization of proliferating and apoptotic cells, and to examine the morphology of M cells and expression of CK18 in follicle associated epithelium (FAE). We also hypothesized that CK18 could be a potential marker for M cell. Peyer's patches completed their histomorphological development in prenatal period and involuted in the postnatal period. The distribution of the CD4+ and CD8+ cells was similar in the last trimester of pregnancy (days 120-150) and the postnatal period, but differed in the early stages of foetal development (days 70-120). In the prenatal period, the follicular area displayed high levels of proliferation and apoptosis. We observed CK18 immunoreaction only in FAE. While M cells were devoid of microfolds in the early stages of the prenatal period, these cells acquired a prismatic shape and bore distinct apical microfolds in the late prenatal period and postnatal period. As a result, it was determined that, in sheep, the development of the ileal Peyer's patches occurred in the prenatal period, independent of exogenous antigenic stimulation, and in association with high levels of lymphopoiesis and apoptosis in the follicles. We found, for the first time, that CK18 is a novel and reliable marker for FAE in sheep ileal Peyer's patches. We suggest that CK18 positive cells in FAE are M cells.

This study evaluated the effect of intra-amniotic synbiotic inclusion and continued synbiotic supplementation in the diet on the performance, intestinal epithelium integrity, and cecal microflora of broiler chickens. In Experiment 1, 510 eggs containing viable embryos were divided into 3 groups of 170 eggs each. The first group was not injected and served as a negative control ( NC: ). The next group was injected with 0.9% NaCl and was the positive control ( PC: ). The synbiotic-injected group ( S: ) was injected with a 0.5% inulin and 1 × 106 Enterococcus faecium solution. The non-injected and synbiotic injected groups were further divided into 2 sets for Experiment 2 and the birds were offered either a basal or synbiotic supplemented diet (1% inulin and 2 × 109 E. faecium cfu/kg feed). One hundred ninety-six broiler hatchlings were randomly allocated in a 2 × 2 factorial arrangement that included an intra-amniotic treatment (non-injected or synbiotic injected) and a dietary treatment (basal or synbiotic supplemented diet). The results showed that the administration of an intra-amniotic synbiotic to embryonated eggs on d 17 of incubation did not affect the hatchability or hatching weight of the birds. However, intra-amniotic synbiotic inclusion had a positive effect on FCR at d 0 to 42 (P = 0.041) and d 22 to 42 (P = 0.036). There was no significant interaction effect on the growth performance of the birds between the intra-amniotic and dietary synbiotic treatment during different or entire experimental periods. Villus height and goblet and proliferating cell nuclear antigen ( PCNA: ) positive cell counts were positively influenced by intra-amniotic and dietary synbiotic treatments. Our results also indicated that intra-amniotic synbiotic injection followed by dietary supplementation with a synbiotic significantly increased Lactobacillus colonization and decreased coliform population in the broiler cecum. Cecal butyric acid concentration increased proportionally to the cecal Lactobacillus count with dietary synbiotic supplementation on d 42. In summary, combined intra-amniotic and dietary synbiotic treatments improved broiler intestinal integrity and increased cecal beneficial bacteria populations.

The objective of this study was to examine the effect of muscle stretching on meat quality and ultrastructure of biceps femoris (BF) from beef. After slaughter, the right side of the carcasses were suspended from the Achilles tendon (AS; n=10), while the left sides were re-hanged from the pelvic bone (PS; n=10). Meat quality was evaluated by water holding capacity (WHC), cooking loss (CL), color, shear force (SF) and sarcomere length (SL). As a result, PS had no impact on WHC, CL and color parameters. PS significantly decreased SF values by 7.5% only at 2 days postmortem. At pelvic suspended sides, SL was increased by 0.13 µm, 0.14 µm and 0.12 µm at 2, 7 and 10 days postmortem, respectively (P<0.001). Electron microscopy showed that the ultrastructure of BF have longer sarcomeres and smaller fiber diameter after PS. In conclusion, PS is a useful method for improving tenderness by stretching muscles.
