RESUMEN

Vibrio parahaemolyticus is a Gram-negative, halophilic and polymorphic coccobacillus. It is world-widely distributed and has resulted in great economic losses since its first appearance. In this study, a pathogenic strain was isolated from diseased pearl gentian grouper and identified as V. parahaemolyticus based on the sequencing results of 16S rDNA gene. In order to gain a comprehensive understanding of this isolation, the whole genome sequencing was conducted. Phylogenetic analysis of the complete genomes of 16 Vibrio species showed that LF1113, ATCC17802, ATCC33787, 2210633, FORC 004, and 160807 were the most closely related. Animal experiments demonstrated that the isolated LF1113 strain was pathogenic in a fish model. This study is the first study to describe the complete genome sequence of a V. parahaemolyticus isolate, which infected pearl gentian grouper from an outbreak in a fish factory farm in Hainan. The results will expand our understanding of genetic characteristics, pathogenesis, diagnostics and disease prevention of V. parahaemolyticus, and lay the foundation for further study.

Introduction: Plectropomus leopardus, a commercially significant marine fish, is primarily found in the Western Pacific regions and along the coast of Southeast Asia. A thorough analysis of the molecular mechanisms involved in sex differentiation is crucial for gaining a comprehensive understanding of gonadal development and improving sex control breeding. However, the relevant fundamental studies of P. leopardus are relatively lacking. Methods: In this study, a genome-wide association study (GWAS) was conducted to investigate the genetic basis mechanism of sex differentiation and gonadal developmental traits in P. leopardus utilizing about 6,850,000 high-quality single-nucleotide polymorphisms (SNPs) derived from 168 individuals (including 126 females and 42 males) by the genome-wide efficient mixed-model association (GEMMA) algorithm. Results: The results of these single-trait GWASs showed that 46 SNP loci (-log10 p > 7) significantly associated with sex differentiation, and gonadal development traits were distributed in multiple different chromosomes, which suggested the analyzed traits were all complex traits under multi-locus control. A total of 1,838 potential candidate genes were obtained by considering a less-stringent threshold (-log10 p > 6) and ±100 kb regions surrounding the significant genomic loci. Moreover, 31 candidate genes were identified through a comprehensive analysis of significant GWAS peaks, gene ontology (GO) annotations, and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses, including taf7, ddx6, apoeb, sgk1, a2m, usf1, hsd3b7, dll4, xbp1, tet3, esr1, and gli3. These trait-associated genes have been shown to be involved in germline development, male sex differentiation, gonad morphogenesis, hormone receptor binding, oocyte development, male gonad development, steroidogenesis, estrogen-synthetic pathway, etc. Discussion: In the present study, multiple genomic loci of P. leopardus associated with sex differentiation and gonadal development traits were identified for the first time by using GWAS, providing a valuable resource for further research on the molecular genetic mechanism and sex control in P. leopardus. Our results also can contribute to understanding the genetic basis of the sex differentiation mechanism and gonadal development process in grouper fish.

In order to understand the strengthening and the failure mechanism of accumulative roll bonding (ARB)-processed AZ63 Mg alloy, the interfacial bonding and fracture behavior of an ARB-processed AZ63 sheet were studied through electron microscopic analysis. The correlation between the mechanical properties, the microstructure, and the ARB processing parameters of an AZ63 sheet were presented. The experimental results have demonstrated that the average grain size of AZ63 Mg alloy processed by ARB was remarkably refined from 12.8 µm to 5.7 µm when the ARB processing temperature was set to 623 K, indicating the occurrence and development of dynamic recrystallization (DRX) nucleation. With the increase in ARB passes, the microstructure obviously became uniform. However, after five passes of the ARB process at 623 K, grains with different crystallographic orientations at the interface can be rearranged to generate the coherent eutectic plane, which inhibits the further refinement of grain size. During the ARB process of the AZ63 Mg alloy, the grain refinement was controlled by twin-induced recrystallization and dynamic recrystallization. Microcracks at the bonded interface of the ARB1 sample were eliminated during the following 3~5 rolling passes at 623 K. After three passes of the ARB process at 623 K, the strength and elongation of the AZ63 Mg alloy increased from 232 MPa and 18.5% to 282 MPa and 26.3%, respectively. The tensile fracture morphology of the sample processed by three passes of ARB exhibited numerous dimples, and the slip lines caused by the cooperative deformation of refined grains can produce a network-like dimple structure, indicating that excellent ductile fracture characteristics could be obtained.

RESUMEN

The development of Alumina-Forming Austenitic (AFA) stainless steel is reviewed in this paper. As a new type of heat-resistant steel, AFA steel forms an alumina protective scale instead of chromia in a corrosive environment. This work summarizes the types of developed AFA steels and introduces the methods of composition design. Various precipitates appear in the microstructure that directly determine the performance at high temperatures. It was found that alloy elements and the heat treatment process have an important influence on precipitates. In addition, the corrosion resistance of AFA steel in different corrosive environments is systematically analyzed, and the beneficial or harmful effects of different elements on the formation of alumina protective scale are discussed. In this paper, the short-term mechanical properties, creep properties and influencing factors of AFA steel are also analyzed. This work aims to summarize the research status on this subject, analyze the current research results, and explore future research directions.

Anisotropy is the difference in the microstructure or mechanical properties of materials in different directions. Anisotropic behavior occurs in rolled sheets, and this anisotropy is very obvious in laminated composites. In this work, the influence of anisotropy on the microstructure and mechanical properties of Ti/Al laminated composites fabricated by rolling was investigated. The results show that the microstructure and mechanical properties of the Ti/Al laminated composites were obviously anisotropic. The grains in the Al layer of the composites were elongated along the rolling direction and were compressed perpendicular to the rolling direction. The grains in the Ti layer of the composites had no obvious preferential orientation and comprised mainly twins. With the rolling direction as 0°, the mechanical properties of the Ti/Al laminated composites varied greatly as the angle of the composites increased. The tensile strength, elongation and bond strength of the Ti/Al laminated composites decreased with increasing angle of the composites. In addition, the microhardness of the Ti/Al laminated composites increased with increasing angle of the composites.

In this paper, the initial values of damage parameters in the Gurson-Tvergaard-Needleman (GTN) model are determined by a microscopic test combined with empirical formulas, and the final accurate values are determined by finite element reverse calibration. The original void volume fraction (f0), the volume fraction of potential nucleated voids (fN), the critical void volume fraction (fc), the void volume fraction at the final failure (fF) of material are assigned as 0.006, 0.001, 0.03, 0.06 according to the simulation results, respectively. The hemispherical punch stretching test of commercially pure titanium (TA1) sheet is simulated by a plastic constitutive formula derived from the GTN model. The stress and strain are obtained at the last loading step before crack. The forming limit diagram (FLD) and the forming limit stress diagram (FLSD) of the TA1 sheet under plastic forming conditions are plotted, which are in good agreement with the FLD obtained by the hemispherical punch stretching test and the FLSD obtained by the conversion between stress and strain during the sheet forming process. The results show that the GTN model determined by the finite element reverse calibration method can be used to predict the forming limit of the TA1 sheet metal.

In this study, we first assembled and characterized the complete chloroplast genome of red seaweed Halymenia maculata. It is 190,431 bp in length, with an AT content of 70.56%. In total, 122 genes were identified, and they consisted of 87 protein-coding genes, 33 tRNA genes, and two rRNA genes. The chloroplast genome of H. maculata did not show an obvious quadripartite structure. A total of five microsatellites (SSRs) were identified in the genome using MISA. Phylogenetic analysis revealed that H. maculata was first clustered with Grateloupia filicina and Grateloupia taiwanensis in a monophyletic clade that provides useful data for the phylogeny and taxonomy of Rhodymeniophycidae.

The complete chloroplast genome of Caulerpa serrulata from South China Sea has been assembled and characterized for the first time. The circular chloroplast genome was 177,848 bp in length, with a GC content of 33.80%. It contained 117 genes, which included 78 protein-coding genes, 37 tRNA genes, and 2 ribosomal RNA genes. Like other species in Caulerpa, the chloroplast genome of C. serrulata did not demonstrate a typical quadripartite structure. A total of 35 microsatellites (SSRs) were identified in the genome using MISA. Phylogenetic analysis showed that C. serrulata was closer to Caulerpa cupressoides, which further clarified the phylogenetic relationships of species in Caulerpa.

The complete chloroplast (cp) genome of Caulerpa sertularioides f. longipes was first assembled and characterized using Illumina pair-end sequencing from the South China Sea. It is 133,626 bp long, with a GC content of 32.94%. In total, 92 genes were identified in the genome, and they consisted 60 protein-coding genes (PCGs), 30 tRNA genes, and 2 rRNA genes. Like other species in Caulerpa, the whole cp genome of C. sertularioides f. longipes did not demonstrate an obvious quadripartite structure. A total of 31 microsatellites (SSRs) were identified in the cp genome using MISA. A phylogenetic tree revealed that C. sertularioides f. longipes was closer to Caulerpa cupressoides, which further clarified the phylogenetic relationships of species in Caulerpa.

RESUMEN

One new flavonoid derivative flavoside A (1), one new 5-hydroxyanthranilic acid derivative crassilin (2), along with the known angucyclinone PD116740 (3) and oxachelin (4), was isolated from the EtOAc extract of the fermentation broth of the sea urchin (Anthocidaris crassispina)-derived actinobacterium, Streptomyces sp. HD01. The structures of these compounds were established on the basis of their HR-ESI-MS and NMR spectroscopic data. All of these compounds were assessed for their antibacterial activity.

Four new 5-hydroxyanthranilic acid related compounds, named anthocidins A⁻D (1⁻4), two known analogues n-lauryl 5-hydroxyanthranilate (5) and isolauryl 5-hydroxyanthranilate (6), together with benzamide (7), 3-hydroxy-4-methoxycinnamamide (8), and (3S-cis)-hexahydro-3-[(3,4-dihydroxyphenyl)methyl]pyrrolo[1,2-a]pyrazine-1,4-dione (9), were isolated from the fermentation broth of the marine-derived actinomycete, Streptomyces sp. HDa1, which was isolated from the gut of a sea urchin, Anthocidaris crassispina, collected from Hainan Island, China. The structures of these secondary metabolites were elucidated on the basis of their 1D and 2D-NMR and mass spectroscopic data, and anthocidin A was confirmed by single-crystal X-ray diffraction with Cu Kα radiation. Anthocidins A⁻D (1⁻4) feature an acetyl group substitution at the amino group and varying alkyl side chains at the carboxyl group of 5-hydroxyanthranilic acid, and compound 5 was isolated as a natural product for the first time. The cytotoxic and antibacterial activity of compounds 1⁻9 were evaluated.
