RESUMEN

Meteorological conditions play an important role in seasonal ozone variations. In order to understand the meteorological formation mechanisms of ozone pollution in Guangdong Province in winter, the moderate ozone pollution process in Guangdong Province from January 3-6, 2022 was selected for comparison with that in autumn (high pollution season) of 2015 to 2021. The research was based on ground and vertical sounding data and reanalysis data of air quality and meteorological elements. Eight cities exceeded the standard during the pollution process, among which Zhaoqing showed moderate pollution (219 µg·m-3) on January 4th. The average O3-8h concentration in Guangdong Province was 123 µg·m-3, which was 21% higher than that in historical autumn. However, its scope of pollution was less than the historical pollution processes in autumn. Lower wind speed, longer sunshine hours, and airflow reflux effect under local circulation were the most important ground-level meteorological conditions for the ozone pollution process, whereas lower temperature might be an important reason for its small scope of pollution. Vertical detection analysis showed that the combined effect of low-level temperature inversion layer, stronger downdraft, and lower wind speed kept the NO2 concentrations at a high level, and further induced a higher increment of ozone concentration in the morning (34.2 µg·m-3 higher than that in the non-pollution period). The downward transport of ozone in the residual layer aggravated the ozone pollution on January 4th. The accumulation of ozone and its precursors was an important factor of the aggravation of ozone pollution the following day. Airflow trajectory analysis revealed that the ground-level ozone was affected by horizontal transport and vertical mixing of pollutants. Potential sources at 10 m were mainly distributed in the coastal areas from Fujian to Guangdong, and the scope of potential sources was concentrated in Guangdong as the height increased. Airflow backward trajectories at different heights during the ozone process passed over the areas of potential sources in Guangdong, which indicated that the local emissions in Guangdong had a greater impact on the ozone pollution process.

OBJECTIVE: To compare accuracy of anterior cervical pedicle screws between assist of rapid prototyping 3D guide plate and free-hand insertion, and evaluate the safety of two methods. METHODS: Eight adult cervical cadaver specimens after formaldehyde immersion, including 4 males and 4 females, aged 32 to 65(40.3±5.6) years old. After X-ray examination to exclude bone damage and deformity, 4 of them (3D guide plate group) randomly selected were for CT scan to obtain DICOM format data, and the data was imported into Mimics software for model, designed the ideal entry point and nail path for anterior cervicaltranspedicular screw (ATPS). After obtaining the personalized guide plate of the nail channel, it was exported as STL data, and the individual guide plate was printed by rapid prototyping and 3D printing technology. In turn, with the assistance of 3D guide plates, one-to-one personalized ATPS screws were placed on the four lower cervical cadaver specimens. Another 4 (free-hand group) lower cervical cadaver specimens were implanted with ATPS screws using free-hand technique. All specimens were performed CT thin-layer scanning and three-dimensional reconstruction after operation. The Tomasino method was used to evaluate the safety of the screws on the CT cross-sectional and sagittal images, to determine whether there was a cortical puncture of the lower and inner edges of the pedicle. According to the CT rating results, gradeⅠandⅡwere safe, and grade Ⅲ- Ⅴ were dangerous.And the accuracy of screws was recorded and analyzed between two groups. RESULTS: Two screws were inserted in each segment from C3 to C7 in 8 adult cadavers. A total of 80 screws were inserted, 40 in the 3D guide plate group, and 40 in the free-hand group. The Tomasino screw rating method was used to evaluate the safety of screw, 21 screws were gradeⅠ, 14 screws were gradeⅡ, 3 screws were grade Ⅲ, 1 screw was grade Ⅳ, 2 screws were grade Ⅴ in 3D guide plate group, while 14 screws were gradeⅠ, 8 screws were gradeⅡ, 8 screws were grade Ⅲ, 6 screws were grade Ⅳ, 2 screws were grade Ⅴ in free-hand group. The safety rate of 3D guide plate group was 87.5%, and 55.0% of the free hand group (χ2=8.7, P=0.003). CONCLUSION: The 3D printing rapid prototyping guide plate assisted insertion of the anterior cervical pedicle screw can significantly improve the accuracy and safety, and provide a theoretical basis for further clinical application.

The high content of tetracycline in municipal sludge may result in vermicomposting products carrying a high abundance of antibiotic resistance genes, thus lowering the utilization value of the vermicompost. Hence, this study aimed to reveal the underlying effects of tetracycline concentrations on the resistance genes involved in vermicomposting systems for sludge recycling. For this purpose, fresh sludge substrates with different concentrations of tetracycline (100, 500, and 1000 mg·kg-1) were vermicomposted for 60 days using Eisenia foetida. In parallel, sludge treatment without the addition of the tetracycline was used as a control. During the experiment, changes in bacterial communities, tetracycline resistance genes (tetC, tetG, tetM, tetW, and tetX), and class 1 integron (intI1) were detected using high-throughput sequencing and qualitative PCR, respectively. The results showed that the addition of tetracycline reduced the abundance of Proteobacteria but increased the abundance of Bacteroidetes in the sludge vermicompost. Furthermore, the concentrations of tetracycline had a significant negative correlation with Shannon and Pielou indexes of bacterial diversity. In addition, tetracycline increased intI1 genes and tetracycline resistance genes in the sludge by 4.25 times and 4.7-186.9 times, respectively. Moreover, there was a significantly positive correlation between the abundance of tetM genes and tetracycline concentration. This study suggests that higher concentration of tetracycline in sludge can modify the microbial community structure of vermicompost, enhancing the abundance of antibiotic resistance genes and their associated dissemination risks.

BACKGROUND: Osteoarthritis (OA) is a major health problem in the increasingly elderly population. Therefore, it is crucial to prevent and treat OA at an early stage. The present study investigated whether pamidronate disodium (PAM), a bone-loss inhibitor, can significantly prevent or reverse the progression of early anterior cruciate ligament transection (ACLT)-induced OA. Whether therapeutic intervention is associated with regulation of the expression of osteoprotegerin (OPG), receptor activator of nuclear factor-κB ligand (RANKL), metalloproteinase-9 (MMP-9) or Toll-like receptor-4 (TLR-4) in cartilage and/or subchondral bone was also investigated. METHODS: 60 New Zealand rabbits were randomized into four groups: Sham-operated (n = 20); ACLT (n = 20); short-term treatment with PAM (PAM-S, n = 10) and long-term treatment with PAM (PAM-L, n = 10). For cartilage and subchondral bone testing, rabbits from Sham and ACLT groups were harvested at 2, 4, 6, and 14 weeks. Rabbits were given PAM from the 4th week after ACLT operation in PAM-S and PAM-L group, and were harvested at 6 and 14 weeks, respectively. Trabecular characteristics and cartilage changes were detected using Micro-CT, safranin O and rapid green staining, respectively. Immunohistochemical staining for OPG and RANKL were also performed. OPG, RANKL, MMP-9 and TLR-4 expression was evaluated by western blot analysis. RESULTS: Micro-CT and histology analyses indicated that PAM treatment for 2 or 10 weeks could completely prevent or reverse osteoarthritic subchondral bone loss and cartilage surface erosion. Immunohistochemistry and western blot analysis indicated that expression of OPG and RANKL increased, although RANKL expression increased more significantly than that of OPG. Therefore the ratio of OPG to RANKL was lower in the ACLT group. However, the ratio of OPG to RANKL in the PAM group was significantly higher than that in the ACLT group. Additionally, expression of MMP-9 and TLR-4 were upregulated in the ACLT group and downregulated in the PAM treated groups. CONCLUSIONS: PAM can significantly inhibit and even reverse early osteoarthritic subchondral bone loss, thus alleviating the process of cartilaginous degeneration. The mechanisms involved may be associated with the upregulation of OPG expression, and downregulation of RANKL, MMP-9 and TLR-4 expression.
