RESUMEN

Lung adenocarcinoma (LUAD) is a common malignant tumor. Identification of biomarkers and understanding their potential functions will facilitate the treatment and diagnosis in LUAD patients. The yellow module (cor = 0.31, P = 2e-6) was selected as the core module based on weighted gene co-expression network analysis (WGCNA) by integrating RNA-seq data and tumor stage. Two upregulated genes (PLAU and GREM1) in yellow module were identified to be biomarkers. Kaplan-Meier curve analysis displayed that high expression levels of them had a poor overall survival (OS). And, their high expression levels revealed higher tumor stage and relapse possibility in LUAD patients, and could be a prognostic parameter. Both biomarkers showed similar immune cell expression profiles in low- and high-expression groups. Strongly positive correlation between both biomarkers and biomarkers of tumor-infiltrating lymphocytes were also clarified in TCGA-LUAD cohort. Importantly, single gene GSEA showed that transcriptional mis-regulation in cancer and microRNAs in cancer were enriched in LUAD patients. Therefore, a miRNA-mRNA-transcription factors (TFs) co-expression regulatory networks was constructed for each biomarker, various miRNAs and TFs were related to PLAU and GREM1. Among which, 6 downstream TFs were overlapped genes for both biomarkers. Notably, 2 of these TFs (FOXF1 and TFAP2A) exhibited significantly abnormal expression levels. Among which, FOXF1 was downregulated and TFAP2A was upregulated in TCGA-LUAD cohort. Both TFs showed a significantly positive correlation with the expression level of PLAU. In conclusion, we identified 2 biomarkers related to immune response and achieved a good accuracy in predicting OS in patients with LUAD.

BACKGROUND: The main challenge in personalized treatment of breast cancer (BC) is how to integrate massive amounts of computing resources and data. This study aimed to identify a novel molecular target that might be effective for BC prognosis and for targeted therapy by using network-based multidisciplinary approaches. METHODS: Differentially expressed genes (DEGs) were first identified based on ESTIMATE analysis. A risk model in the TCGA-BRCA cohort was constructed using the risk score of six DEGs and validated in external and clinical in-house cohorts. Subsequently, independent prognostic factors in the internal and external cohorts were evaluated. Cell viability CCK-8 and wound healing assays were performed after PTGES3 siRNA was transiently transfected into the BC cell lines. Drug prediction and molecular docking between PTGES3 and drugs were further analyzed. Cell viability and PTGES3 expression in two BC cell lines after drug treatment were also investigated. RESULTS: A novel six-gene signature (including APOOL, BNIP3, F2RL2, HINT3, PTGES3 and RTN3) was used to establish a prognostic risk stratification model. The risk score was an independent prognostic factor that was more accurate than clinicopathological risk factors alone in predicting overall survival (OS) in BC patients. A high risk score favored tumor stage/grade but not OS. PTGES3 had the highest hazard ratio among the six genes in the signature, and its mRNA and protein levels significantly increased in BC cell lines. PTGES3 knockdown significantly inhibited BC cell proliferation and migration. Three drugs (gedunin, genistein and diethylstilbestrol) were confirmed to target PTGES3, and genistein and diethylstilbestrol demonstrated stronger binding affinities than did gedunin. Genistein and diethylstilbestrol significantly inhibited BC cell proliferation and reduced the protein and mRNA levels of PTGES3. CONCLUSIONS: PTGES3 was found to be a novel drug target in a robust six-gene prognostic signature that may serve as a potential therapeutic strategy for BC.

RESUMEN

ABO blood group antigens exhibit alternative phenotypes and genetically derived structures that are located on the red cell surface. The role of ABO blood group in cancer biology has been intensely reported by several studies, and it is now widely recognized that ABO antigens are associated with the risk and prognosis of several types of tumors, namely gastric cancer and pancreatic cancer. However, there have been contentious limited issues with the association between the ABO blood group and lymphoma. In this narrative review, based on literature data, we discuss the role of ABO blood group in the risk and prognosis of lymphoma and summarize the current knowledge of the underlying pathogenic mechanisms of the association. The possible association of ABO blood group with racial disparities and pathological classification in lymphoma patients is also discussed.

Polylactic acid (PLA)/poly(ε-caprolactone) (PCL) blends have exhibited good shape memory properties and degradable characteristics in various 4D printing fields such as biomedicine, flexible electronics, and soft robotics, where the service temperature fluctuates easily by environment temperature and polymer characteristics. In this work, printed PLA/PCL 4D samples with different infill densities were prepared by material extrusion printing of pre-extruded filaments and characterized under different temperatures. The results show that the microstructures of printed samples are not influenced by printing process and have similar unique orientation as that of filaments. The thermal properties are stable and show obvious phase transition temperatures, while the mechanical properties decrease slightly in low temperature region and then decrease rapidly when temperature is over 60 °C. The increase in infill density can further improve the storage modulus more than 40% and have no significant influence on the thermal properties. The printed samples also exhibit good shape memory performances with fast recovery speeds less than 22 s. Furthermore, a two-step model is provided to predict the effective modulus of printed PLA/PCL samples and agrees well with experimental data. The results prove that temperature and infill density have different influences on the thermal, mechanical and shape memory properties of PLA/PCL blends.

RESUMEN

Industrial wastewater containing large amounts of organic pollutants is a severe threat to the environment and human health. Thus, the rapid detection and removal of these pollutants from wastewater are essential to protect public health and the ecological environment. In this study, a multifunctional and reusable surface-enhanced Raman scattering (SERS) substrate by growing Ag nanoparticles (NPs) on ZnO nanorods (NRs) was produced for detecting and degrading Rhodamine B (RhB) dye. The ZnO/Ag substrate exhibited excellent sensitivity, and the limit of detection (LOD) for RhB was as low as 10-11 M. Furthermore, the SERS substrate could efficiently degrade RhB, with a degradation efficiency of nearly 100% within 150 min. Moreover, it retained good SERS activity after multiple repeated uses. The interaction between Ag NPs, ZnO, and RhB was further investigated, and the mechanism of SERS and photocatalysis was proposed. The as-prepared ZnO/Ag composite structure could be highly applicable as a multifunctional SERS substrate for the rapid detection and photocatalytic degradation of trace amounts of organic pollutants in water.

Amylose content is a crucial physicochemical property responsible for the eating and cooking quality of rice (Oryza sativa L.) grain and is mainly controlled by the Waxy (Wx) gene. Previous studies have identified several Dull genes that modulate the expression of the Wxb allele in japonica rice by affecting the splicing efficiency of the Wxb pre-mRNA. Here, we uncover dual roles for a novel Dull gene in pre-mRNA splicing and microRNA processing. We isolated the dull mutant, du13, with a dull endosperm and low amylose content. Map-based cloning showed that Du13 encodes a C2 H2 zinc-finger protein. Du13 coordinates with the nuclear cap-binding complex to regulate the splicing of Wxb transcripts in rice endosperm. Moreover, Du13 also regulates alternative splicing of other protein-coding transcripts and affects the biogenesis of a subset of microRNAs. Our results reveal an evolutionarily conserved link between pre-mRNA splicing and microRNA biogenesis in rice endosperm. Our findings also provide new insights into the functions of Dull genes in rice and expand our knowledge of microRNA biogenesis in monocots.

Thermal conductive materials with reliable and high performances such as thermal interface materials are crucial for rapid heat transferring in thermal management. In this work, carbon fiber fabric and graphene reinforced segmented polyurethane composites (CFF-G/SPU) were proposed and prepared to obtain superior thermal, mechanical and electrical properties using the hot-pressing method. The composites exhibit excellent tensile strength and can withstand a tensile force of at least 350 N without breaking. The results show that, comparing with the SPU material, the thermal conductivity is increased by 28% for the CFF-G/SPU composite, while the in-plane electrical conductivity is increased by 8 orders of magnitude to 175 S·m-1. The application of CFF-G/SPU composite as a winding thermal interface material with electric-driven self-heating effect presents good performances of fluidity and interface wettability. The composite has great advantages in phase transition and filling the interfacial gap in the short time of few seconds under the condition of electrical field, with the interface temperature difference between two layers significantly reduced.

Autographa californica multiple nucleopolyhedrovirus (AcMNPV) orf75 (ac75) is a highly conserved gene that is essential for AcMNPV propagation. However, the key domains or residues of the AC75 protein that play a role in viral propagation have not been identified. In this study, sequence alignment revealed that residues Phe-54 and Gln-81 of AC75 were highly conserved among alphabaculoviruses and betabaculoviurses. Thus, Phe-54 and Gln-81 AC75 mutation bacmids were constructed. We found that Gln-81 was not required for viral propagation, whereas mutating Phe-54 reduced budded virus production by 10-fold and impaired occlusion body formation when compared with that of the wild-type AcMNPV. Electron microscopy observations showed that the Phe-54 mutation affected polyhedrin assembly and also occlusion-derived virus embedding, whereas western blot analysis revealed that mutating Phe-54 reduced the amount of AC75 but did not affect the localization of AC75 in infected cells. A protein stability assay showed that the Phe-54 mutation affected AC75 stability. Taken together, Phe-54 was identified as an important residue of AC75, and ac75 is a pivotal gene in budding virus production and occlusion body formation.

Autographa californica multiple nucleopolyhedrovirus (AcMNPV) orf13 (ac13) is a conserved gene in all sequenced alphabaculoviruses. However, its function in the viral life cycle remains unknown. In this study, we found that ac13 was a late gene and that the encoded protein, bearing a putative nuclear localization signal motif, colocalized with the nuclear lamina. Deletion of ac13 did not affect viral genome replication, nucleocapsid assembly or occlusion body (OB) formation, but reduced virion budding from infected cells by approximately 400-fold compared with the wild-type virus. Deletion of ac13 substantially impaired the egress of nucleocapsids from the nucleus to the cytoplasm, while the OB morphogenesis was unaffected. Taken together, our results indicated that ac13 was required for efficient nuclear egress of nucleocapsids during virion budding, but was dispensable for OB formation.

Massive fields in the primordial Universe function as standard clocks and imprint clock signals in the density perturbations that directly record the scale factor of the primordial Universe as a function of time, a(t). A measurement of such signals would identify the specific scenario of the primordial Universe in a model-independent fashion. In this Letter, we introduce a new mechanism through which quantum fluctuations of massive fields function as standard clocks. The clock signals appear as scale-dependent oscillatory signals in the power spectrum of alternative scenarios to inflation.

KEY MESSAGE: FLO15encodes a plastidic glyoxalase I protein, OsGLYI7, which affects compound starch granule formation and starch synthesis in rice endosperm. Starch synthesis in rice (Oryza sativa) endosperm is a sophisticated process, and its underlying molecular machinery still remains to be elucidated. Here, we identified and characterized two allelic rice floury endosperm 15 (flo15) mutants, both with a white-core endosperm. The flo15 grains were characterized by defects in compound starch granule development, along with decreased starch content. Map-based cloning of the flo15 mutants identified mutations in OsGLYI7, which encodes a glyoxalase I (GLYI) involved in methylglyoxal (MG) detoxification. The mutations of FLO15/OsGLYI7 resulted in increased MG content in flo15 developing endosperms. FLO15/OsGLYI7 localizes to the plastids, and the in vitro GLYI activity derived from flo15 was significantly decreased relative to the wild type. Moreover, the expression of starch synthesis-related genes was obviously altered in the flo15 mutants. These findings suggest that FLO15 plays an important role in compound starch granule formation and starch synthesis in rice endosperm.

We show that the shape of the inflationary landscape potential may be constrained by analyzing cosmological data. The quantum fluctuations of fields orthogonal to the inflationary trajectory may have probed the structure of the local landscape potential, inducing non-Gaussianity (NG) in the primordial distribution of the curvature perturbations responsible for the cosmic microwave background (CMB) anisotropies and our Universe's large-scale structure. The resulting type of NG (tomographic NG) is determined by the shape of the landscape potential, and it cannot be fully characterized by 3- or 4-point correlation functions. Here we deduce an expression for the profile of this probability distribution function in terms of the landscape potential, and we show how this can be inverted in order to reconstruct the potential with the help of CMB observations. While current observations do not allow us to infer a significant level of tomographic NG, future surveys may improve the possibility of constraining this class of primordial signatures.

Pyruvate kinase (PK) is a key enzyme in glycolysis and carbon metabolism. Here, we isolated a rice (Oryza sativa) mutant, w59, with a white-core floury endosperm. Map-based cloning of w59 identified a mutation in OsPKpα1, which encodes a plastidic isoform of PK (PKp). OsPKpα1 localizes to the amyloplast stroma in the developing endosperm, and the mutation of OsPKpα1 in w59 decreases the plastidic PK activity, resulting in dramatic changes to the lipid biosynthesis in seeds. The w59 grains were also characterized by a marked decrease in starch content. Consistent with a decrease in number and size of the w59 amyloplasts, large empty spaces were observed in the central region of the w59 endosperm, at the early grain-filling stage. Moreover, a phylogenetic analysis revealed four potential rice isoforms of OsPKp. We validated the in vitro PK activity of these OsPKps through reconstituting active PKp complexes derived from inactive individual OsPKps, revealing the heteromeric structure of rice PKps, which was further confirmed using a protein-protein interaction analysis. These findings suggest a functional connection between lipid and starch synthesis in rice endosperm amyloplasts.

The inflationary universe can be viewed as a "cosmological collider" with an energy of the Hubble scale, producing very massive particles and recording their characteristic signals in primordial non-Gaussianities. To utilize this collider to explore any new physics at very high scales, it is a prerequisite to understand the background signals from the particle physics standard model. In this Letter we describe the standard model background of the cosmological collider.

ADP-glucose pyrophosphorylase (AGPase) controls a rate-limiting step in the starch biosynthetic pathway in higher plants. Here we isolated a shrunken rice mutant w24. Map-based cloning identified OsAGPL2, a large subunit of the cytosolic AGPase in rice endosperm, as the gene responsible for the w24 mutation. In addition to severe inhibition of starch synthesis and significant accumulation of sugar, the w24 endosperm showed obvious defects in compound granule formation and storage protein synthesis. The defect in OsAGPL2 enhanced the expression levels of the AGPase family. Meanwhile, the elevated activities of starch phosphorylase 1 and sucrose synthase in the w24 endosperm might possibly partly account for the residual starch content in the mutant seeds. Moreover, the expression of OsAGPL2 and its counterpart, OsAGPS2b, was highly coordinated in rice endosperm. Yeast two-hybrid and BiFC assays verified direct interactions between OsAGPL2 and OsAGPS2b as well as OsAGPL1 and OsAGPS1, supporting the model for spatiotemporal complex formation of AGPase isoforms in rice endosperm. Besides, our data provided no evidence for the self-binding of OsAGPS2b, implying that OsAGPS2b might not interact to form higher molecular mass aggregates in the absence of OsAGPL2. Therefore, the molecular mechanism of rice AGPase assembly might differ from that of Arabidopsis.

Raman spectroscopy gas detection technology which uses a single wavelength laser detection of gas samples without contact and loss is suitable for dissolved gas detection in oil-immersed transformers. Combined with the features of Raman spectral lines, the analysis of the detection mechanism in Raman spectroscopy technology has been made. Raman spectral lines have been presented in the expression of the convolution of Lorentz function and Gaussian function, which shows preferable Raman spectrum peak linear outline basic characteristics. In this paper, the basic features of the peak height, the peak position and the half peak width, are the main targets of peak searching, and the fundamental purpose of this paper is to obtain qualitative and quantitative analysis of the sample. Therefore, the experimental data of Raman spectroscopy is designed based on the comparison method to realize the automatic peak seeking model to achieve the detection target. Therefore, according to the automatic peak searching model based on the comparison method is designed to achieve the detection target. The simulation results of using peak searching model in the Voigt linear model show that spectrum peak height and peak position are in conformity with the theory in the test experiment. Based on the establishment of Raman spectroscopy detection platform aiming at the dissolved gas in transformer oil, the analysis of the experimental data show that the actual values of the half peak height and width are (8.7,11.5)(cm-1)in the Voigt linear model with deviations. Setting the value as 10.257 cm-1 to modify the parameter, then compared with the research results, it shows that the modified Voigt linear model and the peak searching model have better adaptability and practicability. Combined with the gas detection in the experimental platform in the experimental platform of Raman spectroscopy,detection of seven kinds of transformer fault characteristic gas and analysis of peak searching have been completed effectively. In terms of methane gas, the linear relationship among the unit gas content, Raman characteristic peak intensity and the area has been obtained, which has laid a foundation for the quantitative analysis of the dissolved gas in transformer oil.
