Cyanidin-3-O-glucoside (C3G), a natural antioxidant, plays multiple physiological or pathological roles in maintaining human health; thereby, designing advanced sensors to achieve specific recognition and high-sensitivity detection of C3G is significant. Herein, an imprinted-type electrochemiluminescence (ECL) sensing platform was developed using core-shell Ru@SiO2-CMIPs, which were prepared by covalent organic framework (COF)-based molecularly imprinted polymers (CMIPs) embedded in luminescent Ru@SiO2 cores. The C3G-imprinted COF shell not only helps generate a steady-enhanced ECL signal, but also enables specific recognition of C3G. When C3G is bound to Ru@SiO2-CMIPs with abundant imprinted cavities, resonance energy transfer (RET) behavior is triggered, resulting in a quenched ECL response. The constructed Ru@SiO2-CMIPs nanoprobes exhibit ultra-high sensitivity, absolute specificity, and an ultra-low detection limit (0.15 pg mL-1) for analyzing C3G in food matrices. This study provides a means to construct an efficient and reliable molecular imprinting-based ECL sensor for food analysis.

Piperlonguminine (PLG) is a major alkaloid found in Piper longum fruits. It has been shown to possess a variety of biological activities, including anti-tumor, anti-hyperlipidemic, anti-renal fibrosis and anti-inflammatory properties. Previous studies have reported that PLG inhibits various CYP450 enzymes. The main objective of this study was to identify reactive metabolites of PLG in vitro and assess its ability to inhibit CYP450. In rat and human liver microsomal incubation systems exposed to PLG, two oxidized metabolites (M1 and M2) were detected. Additionally, in microsomes where N-acetylcysteine was used as a trapping agent, N-acetylcysteine conjugates (M3, M4, M5 and M6) of four isomeric O-quinone-derived reactive metabolites were found. The formation of metabolites was dependent on NADPH. Inhibition and recombinant CYP450 enzyme incubation experiments showed that CYP3A4 was the primary enzyme responsible for the metabolic activation of PLG. This study characterized the O-dealkylated metabolite (M1) through chemical synthesis. The IC50 shift assay showed time-dependent inhibition of CYP3A4, 2C9, 2E1, 2C8 and 2D6 by PLG. This research contributes to the understanding of PLG-induced enzyme inhibition and bioactivation.

Introduction: Variation in plant nitrogen uptake rate and substrate preference is complicated due to the combined influence of abiotic and biotic factors. For the same species of tree across different ages, the interactions between root structural traits, nitrogen uptake rate, and soil environment have not been fully characterized, a situation that constrains our understanding of underground resource strategies employed by trees at different ages. Methods: In the present study, we examined the nitrogen uptake rate, mycorrhiza, morphology, architecture, and chemistry of the roots of Larix principis-rupprechtii in a chronosequence (aged 18, 27, 37, 46, and 57 years) in the Saihanba Mechanical Forest Farm in Northern China. Results: L. principis-rupprechtii preferred to absorb ammonium, followed in order by glycine and nitrate. The ammonium uptake rate of L. principis-rupprechtii significantly decreased (aged 18-37 years) and then increased (aged 46-57 years) with tree age. The glycine, nitrate, and total nitrogen uptake rates decreased with tree age. The root resource acquisition strategy appeared to shift from an acquisitive strategy to a conservative strategy associated with increasing tree age. Discussion: Along the root-mycorrhizal collaboration gradient, the younger L. principis-rupprechtii relied more on their own root morphology and physiology to acquire resources, adopting a "do it yourself" strategy comprising increasing the specific root length, the specific root area, and the N uptake rate (nitrate and glycine). Conversely, older trees depended more on mycorrhizal partners to acquire nitrogen resources, an "outsourcing" strategy. The results contribute to our understanding of underground resource-use strategies of plants and the nitrogen cycle in forest ecosystems according to stand age.

PURPOSE: This study is a large-scale screening supported by government departments to investigate the prevalence of scoliosis in Shijiazhuang. METHODS: In this cross-sectional study, all students underwent body posture evaluation and the Adam's forward flexure test to examine the back, and also underwent radiographic evaluation when scoliosis was suspected. RESULTS: In total, 181,935 students participated in the research, of which 289 students were diagnosed with scoliosis, with a prevalence of 2.5%, significantly higher in female (3.01%) than in male children (1.63%) (P < 0.001). Most of the patients (79.5%) had a Cobb angle from 10° to 19°.The mean Cobb angle of scoliosis was positively associated with age. The prevalence of a single curve (75.4%) was significantly higher than double curves (23.2%) and triple curves (1.38%). The percentage of scoliosis in which the thoracolumbar segment was involved (46.4%) was significantly higher than that of the single thoracic scoliosis (31.8%) and single lumbar scoliosis (21.8%). CONCLUSIONS: The prevalence of adolescent idiopathic scoliosis in XXX was high (2.50%). It is necessary to strengthen the education of adolescent scoliosis, and improve the awareness and attention of social to scoliosis. LEVEL OF EVIDENCE: IV.

Broadband spintronic terahertz (THz) radiation can be efficiently generated by spin-to-charge current conversion in a ferromagnetic/nonmagnetic heterostructure. There had been many studies on realizing the enhancement or the modulation of spintronic terahertz waves. However, reported devices so far focus on implementing certain specific modulation methods, either related to the spintronic stacks or related to the metamaterial structures. In this study, a set of femtosecond laser-driven versatile spintronic terahertz devices are proposed by integrating meta-antenna structures with W/CoFeB/Pt nanolayer stacks. These monolithic integrated devices exhibit spintronic terahertz wave emission, spectral modulation, and polarization manipulation simultaneously. The terahertz pulses are generated within the ferromagnetic heterostructure interfaces and transmitted along the metallic structures, leading to the modulation of the spintronic terahertz waves. Results have shown that the center-frequency shift is up to 140 GHz and the value of ellipticity can reach 0.6, demonstrating a set of integrated and efficient spintronic terahertz devices to modulate the emitted wave. In addition, compared with the slotline antenna, the maximum peak value of the bandpass band is enhanced up to 1.63 times by carefully designing the metamaterial structure. The spintronic meta-antenna array proposed here paves an integrated way for the manipulation of spintronic terahertz optoelectronic devices.

Because the accurate and rapid detection of antibiotics and pH plays an important role in biological systems and environmental fields, developing suitable and efficient sensors that can simultaneously detect antibiotics and pH has become important. In this work, we successfully designed and synthesized two new one-dimensional coordination polymers based on the mixed ligands L [N,N'-bis(4-methylpyridin-4-yl)-2,6-naphthalene dicarboxylamide] and H2CPG [3-(4-chlorophenyl)glutaric acid], [M(L)(HCPG)2(H2O)2] (M = Co for CP 1, and M = Ni for CP 2), which were structurally characterized by single-crystal X-ray diffraction, infrared spectroscopy, powder X-ray diffraction, and thermogravimetric analysis. CP 1 and CP 2 can be used as ultraversatile fluorescent sensors, which can sense erythromycin (ERY) and oxacillin (OXC) by turn-on fluorescent enhancement and detect furaltadone (FTD) via the turn-off fluorescent quenching effect, separately. The concentration ranges of different analytes sensed by CPs 1 and 2 were 0-0.046 and 0-0.069 mM for ERY, 0-0.04 and 0-0.028 mM for OXC, and 0-0.155 and 0-0.019 mM for FTD, respectively. Moreover, CP 2 can effectively sense pH, in both a wide pH range and the fine physiological range. The sensors have a rapid luminescence response, good recyclability, and excellent fluorescence stability. More importantly, they not only represent the first example of detecting ERY or OXC based on fluorescent CPs but also are the very rare ultraversatile fluorescent sensors. The fluorescent sensing mechanism for antibiotics and pH was discussed in detail.

Objective: Insulin resistance (IR) plays a key role in gestational diabetes mellitus (GDM) pathogenesis. The antiaging protein klotho has been proven to be closely related to IR. The purpose of this study was to investigate the effect of klotho on IR in GDM trophoblast cells. Methods: The GDM cell model of HTR-8/SVneo cells was induced by high glucose (HG). Plasmid transfection was used to mediate the overexpression or silencing of klotho. The effects of klotho on cell viability, IR, and the IGF-1/PI3K pathways were observed by RT-qPCR, western blot, Cell Counting Kit-8 detection, glucose uptake assay, and immunofluorescence detection. Results: Klotho expression was up-regulated in HG-induced cells. Overexpression of klotho could reduce the cell viability, insulin signaling molecules (INSR-α, INSR-ß, IRS1, IRS2, and GLUT4), and glucose uptake in HTR-8/SVneo cells of the HG group. In addition, the overexpression of klotho inhibited the levels of IGF-1, IGF-1R/p-IGF-1R, and the phosphorylation and activation of the signal transduction molecules PI3K/Akt/mTOR. On the contrary, klotho deletions could reverse these changes of HTR-8/SVneo cells induced by HG. Conclusion. In a word, the results of this study showed that the regulation of klotho played an important role in the IR of trophoblast cells induced by HG, which was mediated at least in part by the IGF-1/PI3K/Akt/mTOR pathway.

In comparison with traditional space infrared spectroscopy technology, the interference signals of a large focal plane array (FPA) can be used to obtain spectra over a larger area range and rapidly achieve large-scale coverage of hyperspectral remote sensing. However, the low signal-to-noise ratio of the interference signals limits the application of spectral data, especially when atmospheric detection occurs in the long-wavelength infrared (LWIR) band. In this paper, we construct an LWIR hyperspectral system of a Fourier transform spectrometer composed of a HgCdTe photovoltaic IR FPA and a Michelson interferometer. The LWIR interference signals are obtained by a high-frequency oversampling technique. We use the Kalman filter (KF) and its improved weighted adaptive Kalman filter (WAKF) to reduce the noise of multiple measured data of each pixel. The effect of overshoot and ringing artifacts on the objective signals is reduced by the WAKF. The applicability is studied by the interference signals from the different sampling frequencies and different pixels. The effectiveness is also verified by comparing the spectra of denoised interferograms with the reference spectrum. The experimental results show that the WAKF algorithm has excellent noise suppression, and the standard deviation of the interferogram can be reduced by 39.50% compared with that of KF. The WAKF is more advantageous in improving the signal-to-noise ratio of the interferogram and spectra. The results indicate that our system can be applied to atmospheric vertical detection and hyperspectral remote sensing over large area ranges because our denoised technique is suitable for large LWIR FPA.

Anastrozole (ANA), is an inhibitor of non-steroidal aromatase, widely employed for the treatment of breast cancer. However, ANA-associated liver injury cases have been documented in the application of the drug.The major purposes of the present study were to identify the structure of reactive metabolites derived from ANA and to study related metabolic pathways of ANA.We found ANA itself is an electrophilic species reactive to GSH. ANA can be metabolised to ANA-N+-glucuronide (1) catalysed by UGT1A4. An ANA GSH conjugate (2) was detected in bile and livers of rats treated with ANA. UGT1A4 participated in the phase II metabolic pathway.This work allowed us to better understand the mechanisms of the hepatotoxicity of ANA and provided new avenue to define the possible role of metabolic activation in hepatotoxicity.

Mushroom galectins are promising anticancer agents for their low IC50 values against cancer cells in vitro. In this study, two Coprinopsis cinerea galectins, CGL1 and CGL2, were heterologously expressed, and their biochemistry properties and anticancer effects were evaluated. The purified galectins were thermostable at neutral pH conditions. They both existed as tetramers and shared a high affinity towards lactose. CGL1 and CGL2 strongly inhibited the cell viability of many cancer cell lines, including three colorectal cancer cells, in a dose-dependent manner by inducing mitochondria-mediated caspase-dependent apoptosis. Furthermore, CGL1 exhibited higher apoptosis-inducing ability and cytotoxicity than CGL2. In vivo cell viability experiments based on two xenograft mouse models showed that CGL1 had a more substantial inhibitory effect than CGL2 on HCT116 tumor growth (p < 0.0001), whereas only CGL1 inhibited DLD1 tumor growth (p < 0.01). This is the first study to evaluate the anti-colorectal cancer effect of mushroom lectins in vivo, and our results showed that CGL1 is a potent agent for colorectal cancer treatment.
