OBJECTIVES: This study aimed to compare the antimicrobial action, cytotoxicity, cleaning ability, and erosion of dentine of hypochlorous acid (HClO) obtained from an electrolytic device at two different concentrations (Dentaqua) and three concentrations of sodium hypochlorite (NaOCl). METHODS: Microbiological test-The root canals of sixty single-rooted extracted human teeth were inoculated with Enterococcus faecalis and divided into 6 groups (n = 10), according to decontamination protocol: DW (control); 1% NaOCl; 2.5% NaOCl; 5.25% NaOCl; 250 ppm HClO and 500 ppm HClO. The colony-forming units were counted to evaluate the decontamination potential of each group, calculating the reduction in bacterial percentage. Cytotoxicity test-Cytotoxicity was evaluated after inoculation of the same tested protocols in fibroblastic cells for 3 min, calculating the cell viability percentages. Specifical statistical analysis was performed (α = 5%). Cleaning ability and erosion-Fifty-six single-rooted bovine lower incisors were divided into seven groups of 8 roots each, being the test groups 1% NaOCl; 2.5% NaOCl; 5,25% NaOCl; 250 ppm HClO and 500 ppm HClO, and a negative and positive control. Negative control was not contaminated, and the other groups were inoculated with Enterococcus faecalis. SEM images were ranked as from the cleanest to the least clean. Erosion was also assessed, being ranked from the least to the most eroded dentine. RESULTS: The highest bacterial reduction was observed in experimental groups, with no statistical differences between them (p > 0.05). The highest number of viable cells was observed in control group, followed by 250 ppm HClO and 500 ppm HClO groups, with statistical differences between them (p < 0.05). 1% NaOCl; 2.5% NaOCl; 5.25% NaOCl and 500 ppm HClO displayed the cleanest areas. All sodium hypochlorite groups displayed erosion with higher ranks with greater concentration, while hypochlorous acid did not display any erosion regardless the concentration. CONCLUSIONS: It is possible to conclude that HClO obtained from an electrolytic device presented high antimicrobial activity and low cytotoxicity in both tested concentrations. 500 ppm HClO did not display erosion and showed great cleaning ability. CLINICAL RELEVANCE: The use of 500 ppm hypochlorous acid may reduce unfavorable behavior of sodium hypochlorite whilst maintaining its antimicrobial action.

The aims of this study were to evaluate the physical and chemical properties, cytotoxicity and dentinal tubule penetration of a new calcium silicate-based root canal dressing. For pH and calcium ion release evaluation (1, 24, 72 and 168 h) were used a pH meter and colorimetric spectrophotometer, respectively. Radiopacity evaluation followed the ISO 6876:2012. Cytotoxicity was evaluated by the percentage of cell viability using MTT assay. Illustrative images of dentinal tubule penetration were obtained using confocal laser scanning microscopy (CLSM). Data from pH and calcium ion release were statistically analyzed by two-way analysis of variance and Tukey test. Radiopacity was analyzed using the Student t-test. The statistical tests for cytotoxicity results were the one-way analysis of variance and Tukey test. Both materials showed alkaline pH in all experimental times. The pH values for calcium hydroxide paste were higher than bioceramic paste at 1, 24, and 72 h (p<0.05). The calcium ion release of bioceramic was lower than the calcium hydroxide paste only at 24 h (p<0.05). The bioceramic was more radiopaque than the calcium hydroxide paste (p<0.05). Bioceramic paste presented a dose and time-dependent cytotoxic effect after MTT assay. CLSM images showed absence of tubule penetration for both pastes. The new calcium silicate-based canal dressing presented alkaline pH, high calcium release, and acceptable radiopacity. Bio C Temp showed a dose and time-dependent cytotoxic and absence of dentinal tubule penetration.

Resumo Os objetivos deste estudo foram avaliar as propriedades físicas e químicas, citototoxidade e penetração tubular de uma nova medicação à base de silicato de cálcio. Para o teste de pH, e liberação de íons cálcio (1, 24, 72 e 168 h) foi usado medidor de pH e espectofotômetro colorimétrico, respectivamente. Avaliação da radiopacidade, seguiu a ISO 6876:2012). A citotoxicidade foi avaliada pela porcentagem de células viáveis usando o ensaio MTT. Imagens ilustrativas de penetração tubular foram obtidas usando microscopia confocal de varredura a laser (CLSM). Os dados de pH e liberação de cálcio foram analisados através do teste de Análise de Variância de duas vias e teste de Tukey. A radiopacidade foi avaliada usando o teste T de Student. Para a citotoxicidade foi empregada a Análise de Variância de uma via e teste de Tukey. Ambos os materiais apresentaram pH alcalino em todos os tempos experimentais. Os valores de pH da pasta de hidróxido de cálcio foram superiores à pasta biocerâmica em 1, 24 e 72 h (p<0,05). A liberação de cálcio da pasta biocerâmica foi inferior à pasta de hidróxido de cálcio apenas em 24 h (p<0,05). Bio-C Temp foi mais radiopaco que o Ultracal XS (p<0,05). A pasta biocerâmica apresentou efeito citotóxico dependente da dose e do tempo de exposição. Imagens de CLSM mostraram ausência de penetração intratubular para ambas as pastas. A nova medicação à base de silicato de cálcio apresentou pH alcalino, alta liberação de cálcio e boa radiopacidade. Bio C Temp apresentou um efeito citotóxico dependente da dose e do tempo de exposição e ausência de penetração tubular.

Bovine leukemia virus (BLV) is a member of Retroviridae family, genus Deltaretrovirus, and the main viral agent responsible for economic loses in dairy herds. Some studies have been carried out about BLV genotypes, and at least seven genotypes were found out in samples of different regions of the world. The objective of this study was to identify BLV samples from seropositive dairy cattle in Santa Catarina state, Brazil, using molecular techniques. Blood samples were collected (454) from dairy cattle from 31 different farms, and serology using agar gel immunodiffusion test (AGID) was performed. After that, 191 seropositive samples were submitted to DNA extraction, and in 77 samples the polymerase chain reaction (PCR) for amplification of a 440 bp fragment of the env gene was performed. Nineteen DNA samples were subjected to restriction fragment length polymorphism (RFLP) analysis by digestion of the PCR fragment by five restriction endonucleases - BamHI, HaeIII, Tru9I, TaqI, and MwoI. It was found 42% seropositive animals (191/454) and 68% positives of the farms (21/31). The PCR showed 80.5% (62/77) of animals positive. The RFLP analysis identified five different genotypes dispersed by Santa Catarina state, with the highest prevalence for genotype X (47.4%). Overall, our results identified the viral genotypes present in dairy cattle and the prevalence of new variants in representative farms from Santa Catarina state.(AU)


O bovine leukemia virus (BLV) é um membro da família Retroviridae, gênero Deltaretrovirus, e o principal agente viral causador de perdas econômicas em rebanhos leiteiros. Diversos estudos têm sido feitos sobre os genótipos de BLV, e foram encontrados pelo menos sete em amostras de diferentes partes do mundo. O objetivo deste estudo foi realizar a caracterização molecular de amostras de BLV de bovinos leiteiros soropositivos no estado de Santa Catarina. Foram coletadas 454 amostras de sangue de bovinos de 31 propriedades, e fez-se inicialmente a sorologia por meio do teste de imunodifusão em gel de ágar. Após a sorologia, 191 amostras soropositivas foram então submetidas à extração de DNA, e em 77 amostras se realizou a reação da polimerase em cadeia (PCR), para a amplificação de um fragmento de 440 pb do gene env. Dezenove amostras foram submetidas à análise do polimorfismo dos fragmentos de restrição por digestão do fragmento da PCR por cinco enzimas de restrição: BamHI, HaeIII, Tru9I, TaqI e MwoI. Os resultados obtidos na sorologia apontaram 42% de animais soropositivos (191/454) e 68% de propriedades positivas (21/31). Na PCR, 80,52% (62/77) dos animais apresentaram-se positivos. A análise do polimorfismo dos fragmentos de restrição identificou cinco genótipos circulantes no estado, e a maior prevalência foi observada no genótipo X (47,4%). Este estudo permite-nos conhecer alguns dos genótipos virais presentes em bovinos leiteiros do estado de Santa Catarina, bem como identificar a existência de novas variantes e sua prevalência atual, e os resultados são úteis para futuros estudos epidemiológicos.(AU)

São descritas as manifestações clínicas, patológicas, microbiológicos e sorológicos da enfermidade natural causada pelo Vírus Respiratório Sincicial Bovino (BRSV) em uma criação extensiva de bovinos de corte no Rio Grande do Sul. Clinicamente havia tosse crônica e dispnéia intensa frente a exercícios físicos mínimos em dois animais. Os dois foram sacrificados e necropsiados. As alterações macroscópicas eram pulmonares com enfisema alveolar disseminado, focos de atelectasia e espessamento dos septos interlobulares. A imunofluorescência para BRSV em corte de pulmão congelado foi positiva em ambos os casos, sendo negativa para Parainfluenza-3 (PI-3), Diarréia Vírica Bovina (BVDV) e Rinotraqueíte Infecciosa Bovina (BHV). Foi isolado BRSV em cultivo celular de MDBK a partir de um dos animais necropsiados. Nenhuma associação foi detectada através de elisa para detecção de antígeno LPS gênero específico de Chlamydia psittaci no tecido pulmonar. O exame histopatológico evidenciou células sinciciais, enfisema crônico, hipertrofia da camada muscular peribronquiolar e metaplasia escamosa do epitélio bronquial e bronquiolar. O exame sorológico para BRSV evidenciou 79% de soropositivos em uma primeira amostragem na qual havia animais jovens e alguns com tosse. O segundo exame sorológico 6 meses após, proveniente de animais de diferentes faixas etárias, resultou em 17,3% de soropositivos. Este é o primeiro relato de doença causada por BRSV no Brasil
