High-fat diet (HFD)-induced obesity results in bone loss associated with an imbalanced gut microbiota and altered immune status. Probiotics are live microorganisms that are beneficial to the host and are important in maintaining bone health and gut homeostasis. In this study, the probiotic Lactobacillus coryniformis subsp. torquens (T3L) was isolated from traditional yak milk cheese produced in Lhasa and showed distinct acid and bile salt resistance as potential probiotics. Our data indicated that T3L not only reversed HFD-induced gut dysbiosis, as indicated by decreased Firmicutes-to-Bacteroidetes ratios but also reduced bone loss. The anti-obesity, microbiome-modulating, and bone-protective effects were transmissible via horizontal faeces transfer from T3L-treated mice to HFD-fed mice. The protective effects of T3L on bone mass were associated with regulatory T (Treg) cell-mediated inhibition of osteoclast differentiation. Our data indicate that T3L is a regulator of the gut microbiota and bone homeostasis in an animal model.

In order to discuss the necessity of repeated renal arteriography in the treatment of severe bleeding after percutaneous nephrolithotomy, this study retrospectively analyzed the clinical data of patients with severe bleeding after percutaneous nephrolithotomy in the Department of Urology Surgery of the First Hospital of China Medical University from August 2010 to July 2020, summarily analyzing treatments, outcomes and follow-up results of 27 patients who were treated by renal arteriography more than twice. Of these 27 patients who underwent repeated renal arteriography, 23 of them were treated by two times, 4 by three times, all of whom were diagnosed as renal vascular injury. And 15 of them were diagnosed as pseudoaneurysm, 4 of them renal arteriovenous fistula, and 8 of them pseudoaneurysm combined with renal arteriovenous fistula. After clear diagnosis, all these patients were performed with renal artery embolization, after which the symptoms of hematuria and lumbar discomfort were relieved or disappeared immediately. These patients were followed up from 6 months to 5 years, without corresponding symptoms recurring and with the renal function equivalent to that before embolization. The results showed that repeated renal arteriography was of great significance in the treatment of patients with severe bleeding after percutaneous nephrolithotomy, helping to clarify the cause of bleeding and giving appropriate and timely treatment.

Objective: To explore the effects and potential molecular mechanism of age on the stiffness and the fibrotic phenotype of fibroblasts (Fbs) of human hypertrophic scar. Methods: The experimental research method was used. From January to June 2020, the surgically removed hypertrophic scar tissue of 10 scar patients (4 males and 6 females) and residual full-thickness normal skin tissue of 10 cases (5 males and 5 females, aged 7-41 years) were collected after operation in Department of Burns and Plastic Surgery of the Fourth Medical Center of the PLA General Hospital. The hypertrophic scar tissue of 6 patients aged (10.7±1.6) years was included into the young group and the hypertrophic scar tissue of 4 patients aged (40.0±2.2) years was included into the elderly group according to the age of patients. For the normal skin tissue and scar tissue in the two groups, hematoxylin eosin (HE) staining was performed to observe the tissue morphology, Masson staining was performed to observe the morphology and arrangement of collagen and quantify the content of collagen, and scanning electron microscope was used to observe the microscopic difference of dermal collagen fibers after the samples were freeze-dried and metal coated. The stiffness of scar tissue in the two groups was measured by atomic force microscope under the liquid phase. The scar tissue in the two groups was collected and the Fbs were isolated and cultured. The morphological differences of the Fbs were observed under the inverted phase contrast microscope, and the protein expression of paxillin was detected with cellular immunofluorescence to reflect the morphology of the Fbs. Cellular immunofluorescence was used to detect the expressions of pro-fibrosis protein α-smooth actin (α-SMA), transforming growth factor-ß1 (TGF-ß1), and type Ⅰ collagen, mechanotransduction-related protein Yes-associated protein (YAP), and the proliferation-related protein Ki67. Real-time fluorescent quantitative reverse transcription polymerase chain reaction was used to detect the mRNA expressions of pro-fibrosis genes of TGF-ß1, α-SMA, and type Ⅰ collagen, fibrosis inhibiting gene of TGF-ß3, and mechanotransduction-related genes of Rho-associated protein 1 (ROCK1) and YAP. Data were statistically analyzed with one-way analysis of variance and least significant difference t test. Results: HE staining showed that the epidermal layer of normal skin was uneven, and blood vessels and sweat glands could be seen in the dermal layer; the epidermal layer of the scar tissue in the two groups was relatively flat, and blood vessels and sweat glands were rare. Masson staining and scanning electron microscopy showed that the collagen fibers in normal skin arranged loosely and disorderly, while the collagen fibers in scar tissue of the two groups arranged densely and orderly, and the collagen fibers in scar tissue of the young group were denser than those of the elderly group. The collagen content in scar tissue of the young group and the elderly group was significantly higher than that of the normal skin tissue (t=8.02, 3.15, P<0.05 or P<0.01), and the collagen content in scar tissue of the elderly group was significantly lower than that of the young group (t=4.84, P<0.05). The dermal stiffness of scar tissue in the elderly group was (50.3±1.1) kPa, significantly higher than (35.2±0.8) kPa in the young group (t=11.43, P<0.05). There were no obvious differences in the morphology of scar Fbs in the two groups observed under inverted phase contrast microscope and by cellular immunofluorescence. The expressions of type Ⅰ collagen and TGF-ß1 in scar Fbs cytoplasm of the elderly group were significantly higher than those in the young group, while the expressions of α-SMA in scar Fbs cytoplasm were close in the two groups. The expressions of YAP in cytoplasm and nucleus of scar Fbs in the elderly group were significantly higher than those in the young group, while the expressions of Ki67 in scar Fbs nucleus of the two groups were close. The mRNA expressions of TGF-ß1 and type Ⅰ collagen in scar Fbs of the elderly group were significantly higher than those in the young group (t=2.87, 4.85, P<0.05 or P<0.01), the mRNA expression of TGF-ß3 in scar Fbs of the elderly group was significantly lower than that in the young group (t=3.36, P<0.05), and the mRNA expressions of α-SMA in scar Fbs of the two groups were close (t=1.14, P>0.05). The mRNA expressions of ROCK1 and YAP in scar Fbs of the elderly group were significantly higher than those in the young group (t=2.98, 7.60, P<0.05 or P<0.01). Conclusions: The elderly are prone to scar healing after skin injury. The molecular mechanism may be attributed to the production of extracellular matrix components with higher stiffness, which increases tissue stiffness and thereby activates the expressions of ROCK and YAP/transcriptional co-activator with PDZ-binding motif genes, promoting pro-fibrosis gene and protein expression.

Objective: To develop a fast track transfer to intensive care unit (ICU) for the perioperative high-risk elderly patients after hip fracture surgery and analyze the preliminary clinical effect of the application. Methods: From January 2014 to December 2017, before the application of postoperative fast track transfer to ICU, the clinical data of 195 elderly patients with hip fracture were included in a retrospective analysis. Among 195 hip fracture patients, 18 were transferred to ICU post operation (non-fast track group). Multivariate logistic regression analysis was applied to investigate relevant risk factors for transferring to ICU after hip fracture surgery. Based on risk factors acquired from the analysis and clinical experience, the fast track transfer to ICU for the perioperative high-risk elderly patients after hip fracture surgery was constructed according to the preliminary and experiential criteria. From January 2018 to December 2019, the clinical data of 70 patients (fast track group) who were transferred to ICU after hip fracture surgery through the fast track were collected and compared with non-fast track group. Results: Multivariate regression analysis revealed that American Society of Anesthesiologists classification(≥Ⅲ) (OR=4.260, 95%CI:1.157-15.683, P=0.029), pre-hospital stage (≥48 h) (OR=4.301, 95%CI:1.212-15.266, P=0.024), hemoglobin concentration at admission(<90 g/L) (OR=7.979, 95%CI:1.936-32.889, P=0.004), coronary heart disease as one comorbidity(OR=6.063, 95%CI:1.695-21.693, P=0.006) were independent risk factors for transferring to ICU after hip fracture surgery. There were no significant difference in gender, age, fracture type, hemoglobin concentration at admission and time of pre-hospital stage between the non-fast track group and fast track group(all P>0.05). However, the number of comorbidities in the fast track group was significantly higher than that in the non-fast track group (Z=-1.995, P=0.046). The time to surgery, postoperative hospital stay, and length of hospital stay in fast track group were all significantly less than those in non-fast track group (Z=-2.121, -2.726, -3.130, all P<0.05). Also, there were fewer medical consultations needed and fewer patients who stayed in ICU more than or equal to 2 nights in fast track group than that in non-fast track group(all P<0.05). There were no significant difference in the rate of patients who transferred from the general ward to ICU after transferring from ICU to the general ward, the proportion of patients who received more than or equal to 4 departments, operation time, hospitalization expense, mortality during hospitalization, 30-day mortality and 90-day mortality after operation between the two groups(all P>0.05). Conclusions: The fast track constructed in this study can reduce time to surgery, postoperative hospitalization stay and length of hospitalization stay for the perioperative high-risk elderly patients with hip fractures and is a specific clinical application of eras concept based on multidisciplinary team.

An integrated in-fiber coupler is proposed and demonstrated, which can excite whispering-gallery mode (WGM) in a microsphere resonator. This device is fabricated firstly by using femtosecond laser micromachining and a fusing splicing technique to create an inner air cavity with a suspended fiber core, then an open micro-channel is drilled on the top of the inner air cavity, and finally a microsphere is placed inside the air cavity, in contact with the suspended fiber core, to excite the WGM through the evanescent field. In the transmission spectrum of the device, the slopes of the two asymmetric Fano resonance lines of 41.12 dB/nm and $ - {18.46}\;{\rm dB/nm}$-18.46dB/nm, respectively, and a symmetrical Lorentz line with a quality factor of ${9.32} \times {{10}^3}$9.32×103 can be obtained simultaneously. Such an in-fiber WGM microsphere resonator has the advantages of compact structure, convenient operation, and high durability.

We propose and demonstrate parallel structured optical fiber in-line Fabry-Perot interferometers for high temperature sensing. The device consists of three Fabry-Perot cavities in parallel connection, which allows three independent fringe patterns superimposed at its output, and, as a result, a number of dominant fringe peaks/dips appear, thus enabling unambiguous measurement in a large range. The device is featured with compact size, robust structure, and excellent high temperature sustainability, which makes it promising in extreme environment monitoring.

OBJECTIVE: The importance of circular RNAs in malignant tumors increases the attention of researchers. The role of circ_0067934 in glioma remains unclear. Our study aims to uncover how circ_0067934 functions in glioma development. PATIENTS AND METHODS: Real Time-quantitative Polymerase Chain Reaction (RT-qPCR) was utilized to determine the level of circ_0067934 in glioma tissues. Circ_0067934 was knocked down in glioma cells. Cell migrated and invaded ability was detected through functional assay in vitro and in vivo. Further mechanism assays were performed to explore the potential targets of circ_0067934. RESULTS: The circ_0067934 was highly expressed in glioma tissues compared with adjacent samples. The expression of circ_0067934 was upregulated in glioma cell lines. The cell migrated and invaded ability of glioma cells was inhibited after circ_0067934 was knocked down. Besides, CSF1 expression was decreased via knockdown of circ_0067934. Furthermore, tumor metastasis was inhibited after circ_0067934 was knocked down in nude mice. CONCLUSIONS: The circ_0067934 could suppress cell migration and invasion of glioma by upregulating CSF1.

Burn rehabilitation in China started from compression therapy in the mid-1970s, which showed the dual effects of prevention and treatment of hypertrophic scars. It not only promoted functional rehabilitation but also strengthened the confidence of patients in rehabilitation treatment. Thereafter, more therapies were brought into practice, such as intra-scar injection of triamcinolone acetonide, application of plastic splints, hydrotherapy, exercises with equipment, skin care, local therapeutic massage, active and passive exercises, as well as external use of drugs for inhibiting scars and pigments. Since the beginning of the 21st century, rehabilitation therapies have been gradually increasing. Psychological rehabilitation, occupational therapy, external use of silicone gel, wax therapy and sound, light, electricity, and radiation therapy have been carried out. Many hospitals have established foundations and held summer camps for children. As far as the whole country is concerned, compared with the huge demand, we still face a number of problems such as shortage of working staff, limited working space, capital chain rupture, lack of multi-disciplinary cooperation, untimely treatment, and incomplete rehabilitation. Nowadays, with increasing knowledge of burn rehabilitation and number of practitioners, improvement of equipment and economic situation, the pace of rehabilitation has accelerated, and the overall implementation of burn rehabilitation therapy has shown great vitality. Patients with burn injury involving over 80% total burn surface area (TBSA) of total burn area or full-thickness burn over 60% TBSA were cured and recovered in different levels of hospitals nationwide, which not only reflects the superb level of burn treatment in China but also reflects the overall improvement of rehabilitation level of the country.

Objective: To analyze the type-specific prevalence of human papillomavirus (HPV) among women aged 18-45 years from the general population in Liuzhou, Guangxi Zhuang Autonomous Region. Methods: Totally, 2 300 women aged 18-45 years old were enrolled in Liuzhou,from March to July, 2013. Cervical exfoliated cells were collected for liquid based cytological and HPV DNA tests. Women were referred to colposcopy exam, based on the clinical practice guideline. Results: Overall, the prevalence rates of any HPV or oncogenic HPV appeared as 22.7% (95% CI: 21.0%-24.4%) and 17.3% (95% CI: 16.0%-19.1%), respectively in this population under study. The high-risk HPV prevalence peaked at the age groups of 18-25 and 41-45, increasing along with the severity through cytological and histological tests. Statistically significant differences between the prevalence of CIN2+ (Cervical intraepithelial neoplasia 2+) in women older than 26 years (1.7%, 95% CI: 1.0%-2.4%) and 18-25 years (1.2%, 95% CI: 0.5%-1.9%) of age, were not observed. Among samples diagnosed as CIN2+, positivity of HPV bivalent (16/18) and nine-valent (6/11/16/18/31/33/45/52/58) vaccine, related high risks on the types of HPV types appeared as 44.1% and 97.1%. Conclusions: The age-specific HPV prevalence rates in the general women aged 18-45 in Liuzhou presented as having bimodal distribution, suggesting that the disease burden of cervical diseases in women aged 26-45 years should not be ignored. Nine-valent HPV vaccine might provide more effective prevention outcomes on cervical cancer in China.

Objective: To understand the level of bactericidal antibodies against Neisseria meningitidis and their rate of protection in children aged between 3 and 23 months, in Liuzhou, in 2012. Methods: Convenience sampling was performed in Liujiang, Rong'an and Sanjiang Counties from May through August, 2012. The inclusion criteria for 603 subjects were: Children aged between 3 and 23 months; no history of meningococcal meningitis; no vaccination against Neisseria meningitidis serogroup C; more than 30 days from the last vaccination against Neisseria meningitidis serogroup A. Demographic information and immunization history of the subjects were obtained using questionnaires. Venous blood samples (2.0 ml each) were collected and levels of Neisseria meningitides antibodies determined using a Serum Bactericidal Assay (SBA). The geometric mean titer (GMT) of serum bacterial antibodies was positive when it was greater than or equal to 1∶2 and protective when greater than 1∶8. Chi-square and Fisher's exact tests were used to compare differences in the positive and protective rates of serum antibodies of Neisseria meningitidis serogroup A and Neisseria meningitidis serogroup C, among children with different demographic characteristics. Kruskal-Wallis H test was used to compare differences in the GMT of serum antibodies of Neisseria meningitidis serogroup A and Neisseria meningitidis serogroup C, among children with different demographic characteristics. Results: Of 603 subjects, 325 (53.9%) were female and 278 (46.1%) were male; 276 (45.8%), 143 (23.7%) and 184 (30.5%) subjects were administered, respectively, no treatment, 1 dose vaccine and 2 doses vaccine. The GMT of serum antibodies against group A Neisseria meningitidis was 1∶1.11, the positive rate was 7.6% (46) and the protective rate was 2.0% (12). The GMT of antibodies in children receiving 1 vaccine dose was 1∶1.16 and the protective rate was 3.5% (5), both values higher than those in children receiving 2 vaccine doses (GMT: 1∶1.2, protective rate: 3.5% (5)). However, these differences were not statistically significant (GMT: H=0.64, P=0.728; protective rate: Fisher's exact test, P= 0.080). The GMT of antibodies in children receiving 1 and 2 doses of meningococcal polysaccharide vaccines were 1∶1.12 and 1∶2.30, respectively (≤180 d). The GMT of serum antibodies for group C meningococcal vaccine was 1∶1.18 and its positive and protective rates were 14.6% (88) and 2.2% (13), respectively. Conclusion: Children aged between 3 and 23 months are susceptible to Neisseria meningitidis groups A and C. The immune effectiveness of group A meningococcal polysaccharide vaccine is relatively poor in this age group and their antibody levels decreased rapidly.

OBJECTIVE: Early diagnostics of colorectal cancer is complicated by the lack of reliable serum biomarkers. This study aimed to investigate if the serum level of activin A can be used for diagnostics of this disease. PATIENTS AND METHODS: In this study we measured the level of activin A in patients with colorectal adenocarcinoma, benign colorectal polyps, as well as in healthy subjects. RESULTS: We found that the level of activin A was significantly higher in patients with colorectal adenocarcinoma, as compared to patients with polyps and healthy controls. Furthermore, activin A levels correlated well with the stage of colorectal cancer. The level of activin A was substantially reduced in post-operative patients. Immunohistochemical staining demonstrated that high levels of activin A were present in the adenocarcinoma tissue samples but not in the non-cancerous samples. RT-PCR further confirmed that mRNA of ßA subunit of activin A is significantly over-expressed in the majority of cancerous samples. Western blotting results further demonstrated the elevated level of activin A in cancer samples. CONCLUSIONS: Taken together, the findings suggest that colorectal adenocarcinomas directly secret activin A into the blood stream. Measuring the serum level of activin A might be used as a reliable diagnostic and screening tool in clinical practice.

Activin A, a stimulator of follicle-stimulating hormone secretion from the pituitary, acts as a neurotrophic and neuroprotective factor in the central nervous system. Activin receptor-interacting protein 1 (ARIP1) has been identified as a cytoplasmic protein that interacts with the type II receptor of activin (ActRII). However, the distribution pattern and function of ARIP1 are not well characterised in the brain. In the present study, we confirmed the existence of mRNA and protein of ARIP1 in the mouse brain, and found that ARIP1 was mainly localised at the hippocampus and hypothalamus in the cerebrum, granular layers in the cerebellum (especially in Purkinje cells of the cerebellum) and choroid epithelial cells by immunohistochemical staining. Furthermore, in contrast to the significant increase of activin A mRNA, ARIP1 mRNA and protein expression decreased in the mechanically lesioned brain of the mouse. Using neuroblastoma-derived Neuro-2a cells to investigate the function of ARIP1, we found that overexpression of ARIP1 down-regulated the activin A-induced signal transduction and significantly decreased the voltage-gated Na(+) current (I(Na) ). These data indicate that ARIP1 is a key molecule for the regulation of the action of activin in neurones, and also that decreased ARIP1 expression in the lesioned brain may be beneficial to the neurotrophic and neuroprotective roles of activin A in recovery after brain injury.

OBJECTIVE: This study investigated the potential use of serum follistatin (FST) as a marker for ovarian cancer alongside serum cancer antigen-125 (CA-125). METHODS: Serum samples were collected from patients with ovarian cancer (n = 45), benign ovarian cysts (n = 40) or other cancers (n = 100) and from healthy subjects (n = 60) for the determination of FST and CA-125 levels using enzyme-linked immunosorbent assays. Expression of FST in ovarian tissue was investigated using immunohistochemical staining. RESULTS: Compared with healthy subjects and patients with benign ovarian cysts, serum FST and CA-125 levels were significantly increased in patients with ovarian cancer. Using the 95% confidence interval for the healthy subjects group as the cut-off value, tumour marker sensitivity and specificity in ovarian cancer were 53.3% and 97% for FST and 77.8% and 84% for CA-125, respectively. Tissue expression of FST protein was more pronounced in ovarian cancer than in normal ovary. CONCLUSIONS: The serum FST level was elevated in the peripheral blood of patients with ovarian cancer and has potential as a tumour marker for ovarian cancer diagnosis. It may be particularly useful when combined with CA-125 detection to reduce the number of false-positive results.
