RESUMEN

An attempt was made to understand the 'floral bud distortion' (FBD), an unexplored disorder prevailing in soybean. Cytological behaviour of floral reproductive organs and in silico characterization of differentially expressed transcript-derived fragments (TDFs) in symptomatic and asymptomatic soybean plants were carried out. Pollens in asymptomatic plants do not have defects in number, size, shape and function. However, in symptomatic plant, pollens were found nonviable, abnormal in shape and with reduced germination ability. Here, we employed a computational approach, exploring invaluable resources. The tissue-specific transcript profile of symptomatic and asymptomatic sources was compared to determine differentially expressed TDFs associated with FBD to improve its basic understanding. A total of 60 decamer primers produced 197 scorable amplicons, ranged 162-1130 bp, of which 171 were monomorphic and 26 were differentially regulated. Reproducible TDFs were sequenced and characterized for their homology analysis, annotation, protein-protein interaction, subcellular localization and their physical mapping. Homology-based annotation of TDFs in soybean revealed presence of two characterized and seven uncharacterized hits. Annotation of characterized sequences showed presence of genes, namely auxin response factor 9 (ARF9) and forkhead-associated (FHA) domain, which are directly involved in plant development through various pathways, such as hormonal regulation, plant morphology, embryogenesis and DNA repair.

RESUMEN

The nature of gene action and of maternal influence governing cottonseed oil attributes were determined with four lines, two each with high and low seed-oil percentage. For this purpose, P1, P2, F0, F1, F2 and alternative sets of BC1 and BC2 generations were analysed in six cross-combinations and their reciprocals. Marginal extents of heterosis for seed-oil percentage were noticeable in F1, with inbreeding depression in F2. Data from reciprocal backcrosses provided evidence in favour of maternal rather than cytoplasmic effects of seed-oil development. Relatively higher extents of heterosis, sizeable inbreeding depression and reciprocally unequal F2 averages were characteristic of the seed index trait, which often showed a reversal of effects from F1 to F2. Reverse reciprocal backcrosses exhibited some differences, including greater resemblance between the types, (A/B)A and (B/A)A, in addition to variable dose effects in seed index. Thus, the differences between F1 seed index values were not due to cytoplasmic influence. Positive heterotic effects for seed-oil index, especially among the backcrosses, ranged between 16.08% and 47.29% over midparent averages. Genetic component estimates from analysis of similar sets of crosses differing only in reciprocal backcrosses, and also from sets of reciprocal crosses between any two parental combinations, were inconsistent. Scaling tests detected presence of epistasis within and between a majority of cross-combinations. Despite reciprocal differences, additive gene effects for seed-oil percentage were significant in 7 out of 24 crosses, representing high x low, low x high and low x low seed-oil parents. Those were, however, accompanied by significant dominance effects of higher order. In crosses involving low seed-oil percentage parents SA1060 and SA229, all six components were detected significant, with opposite effects of dominance and dominance x dominance epistatic components. Significant additive components were also detected for seed index and seed-oil index in 7 and 5 out of 24 crosses, respectively. In the inheritance of seed index and seed-oil index, dominance effects were more important. Epistatic components of additive x additive, and to a lesser extent, those of dominant x dominant were found significant.

The role of 19 structural, developmental and biochemical traits in relation to specific adaptation was analysed in a set of 17 diverse lines with quantified adaptation, representing contemporary cultivars and land races of chickpea (Cicer arietinum L.), using multivariate analysis. Significant varietal variation was observed for most characters, particularly for the activity of the enzyme nitrate reductase (NR) and protein content in the plant. The distance analysis (D(2)-statistic) revealed that seed size and pod number and their associated attributes were important forces of divergence. The additional forces of divergence were NR activity at the flower initiation stage, yield components such as number of primary and secondary branches, and other features such as plant habit and duration of flowering. The principal component analysis revealed some similarities and also differences from the distance analysis. Leaf size, days to flower initiation, seed size and, to some extent, NR activity at flower initiation stage, were important in the first vector. Developmental traits such as chlorophyll depth, NR activity at the pod initiation and grain filling stages, and the percent protein content in the plant at flower initiation were important in the second vector. In general, the clustering pattern was not related to the geographical origin, seed colour, size of regression coefficient for yield, or deviation from linearity. The importance of the developmental and biochemical attributes in the divergence of cultivated chickpea, such as days to flower initiation, duration of flowering, NR activity and the rates of protein accumulation in developing seeds, and in adaptation, suggests the critical role of these attributes. NR activity at the flower initiation stage would appear to have a major role in the domestication of this crop and its intra-specific differentiation, as an increased seed size could not have been possible without better nutrient uptake and utilization.
