RESUMEN

Agricultural managed aquifer recharge (Ag-MAR, on-farm recharge), where farmland is flooded with excess surface water to intentionally recharge groundwater, has received increasing attention by policy makers and researchers in recent years. However, there remain concerns about the potential for Ag-MAR to exacerbate nitrate (NO3-) contamination of groundwater, and additional risks, such as greenhouse gas emissions and crop tolerance to prolonged flooding. Here, we conducted a large-scale, replicated winter groundwater recharge experiment to quantify the effect of Ag-MAR on soil N biogeochemical transformations, potential NO3- leaching to groundwater, soil physico-chemical conditions, and crop yield. The field experiment was conducted in two grapevine vineyards in the Central Valley of California, which were each flooded for 2 weeks and 4 weeks, respectively, with 1.31 and 1.32 m3 m-2 of water. Hydrologic, geochemical, and microbial results indicate that NO3- leaching from the first 1 m of the vadose zone was the dominant N loss pathway during flooding. Based on pore water sample and N2O emission data, denitrification played a lesser role in decreasing NO3- in the root zone but prolonged anoxic conditions resulted in a significant 29 % yield decrease in the 4-week flooded vineyard. The results from this research, combined with data from previous studies, are summarized in a new conceptual model for integrated water-N dynamics under Ag-MAR. The proposed model can be used to determine best Ag-MAR management practices.

Two studies were conducted to determine how methyl jasmonate (MeJA), jasmonic acid (JA), and 1-aminocyclopropane-1-carboxylic acid (ACC) affect grape berry abscission in the initial days after treatment. The overarching goal was to determine whether JA, with or without ACC, may hold the potential to sufficiently reduce fruit detachment force (FDF) and increase the proportion of berries with dry stem scars while minimizing preharvest abscission, effects that could be useful in the production of stemless table grapes. On Thompson Seedless grapes, JA was at least as effective as MeJA for stimulating berry abscission based on reduced fruit detachment force (FDF) and yielding detached berries with dry stem scars. Further, since previous studies showed that ACC improved MeJA-induced grape abscission, we tested ACC effects on JA activity. We found that JA rapidly induced preharvest berry abscission, confirming previous results. ACC alone did not induce preharvest berry abscission, but ACC improved the effectiveness of JA on reducing FDF and increasing dry stem scar development. These studies also demonstrated that JA-induced abscission occurs within the first day after treatment. Commercial use of JA plus ACC as an abscission agent requires that FDF sufficiently declines, and the incidence of dry stem scars increases, prior to a significant increase in fruit abscission. However, the rapid progression of fruit abscission may require harvest either within 24 and 48 h after treatment or the use of a passive catch system.

Grapevine productivity, and berry and wine flavonoid concentration, depend on the interactions of cultivar, environment, and applied cultural practices. We characterized the effects that mechanical leaf removal and irrigation treatments had on the flavonoid concentration of 'Merlot' (Vitis vinifera, L.) grape berries and wines in a hot climate over two growing seasons with contrasting precipitation patterns. Leaves were removed by machine, either at prebloom (PBLR), or at post-fruit-set (PFLR), or not removed (control) and irrigation was either applied as sustained deficit irrigation (SDI) at 0.8 of crop evapotranspiration (ETc) from budbreak to fruit set, or regulated deficit irrigation (RDI) at 0.8 ETc from bud break to fruit set, 0.5 ETc from fruit set to veraison, and 0.8 ETc from veraison to harvest, of ETc In 2014, PFLR reduced the leaf area index (LAI) compared to control. The RDI decreased season-long leaf water potential (ΨInt) compared to SDI. However, in 2015, none of the treatments affected LAI or ΨInt. In 2014, berry flavonoid concentrations were reduced by PBLR as well as SDI. SDI increased the flavonoid concentrations in wine, and PFLR increased some wine flavonols in one season. No factor affected the concentrations of wine proanthocyanidins or mean degree of polymerization. Thus, mechanical PFLR and RDI may increase berry flavonoid accumulation without yield reduction, in red wine grapes cultivars grown in hot climates when precipitation after bud break is lacking. However, spring precipitation may influence the effectiveness of these practices as evidenced by this work in a changing climate.

Hybrid (Vitis vinifera ×Vitis labrusca) table grape cultivars grown in the subtropics often fail to accumulate sufficient anthocyanins to achieve good uniform berry color. Growers of V. vinifera table grapes in temperate regions generally use ethephon and, more recently, (S)-cis-abscisic acid (S-ABA) to overcome this problem. The objective of this study was to determine if S-ABA applications at different timings and concentrations have an effect on anthocyanin regulatory and biosynthetic genes, pigment accumulation, and berry color of the Selection 21 cultivar, a new V. vinifera ×V. labrusca hybrid seedless grape that presents lack of red color when grown in subtropical areas. Applications of S-ABA 400 mg/L resulted in a higher accumulation of total anthocyanins and of the individual anthocyaninsanthocyanins: delphinidin-3-glucoside, cyanidin-3-glucoside, peonidin-3-glucoside, and malvidin-3-glucoside in the berry skin and improved the color attributes of the berries. Treatment with two applications at 7 days after véraison (DAV) and 21 DAV of S-ABA 400 mg/L resulted in a higher accumulation of total anthocyanins in the skin of berries and increased the gene expression of CHI, F3H, DFR, and UFGT and of the VvMYBA1 and VvMYBA2 transcription factors in the seedless grape cultivar.

Some plant growth regulators, including ethephon, can stimulate abscission of mature grape berries. The stimulation of grape berry abscission reduces fruit detachment force (FDF) and promotes the development of a dry stem scar, both of which could facilitate the production of high quality stemless fresh-cut table grapes. The objective of this research was to determine how two potential abscission treatments, 1445 and 2890 mg/L ethephon, affected FDF, pre-harvest abscission, fruit quality, and ethephon residue of Thompson Seedless and Crimson Seedless grapes. Both ethephon treatments strongly induced abscission of Thompson Seedless berries causing >90% pre-harvest abscission. Lower ethephon rates, a shorter post-harvest interval, or berry retention systems such as nets, would be needed to prevent excessive pre-harvest losses. The treatments also slightly affected Thompson Seedless berry skin color, with treated fruit being darker, less uniform in color, and with a more yellow hue than non-treated fruit. Ethephon residues on Thompson Seedless grapes treated with the lower concentration of ethephon were below legal limits at harvest. Ethephon treatments also promoted abscission of Crimson Seedless berries, but pre-harvest abscission was much lower (≅49%) in Crimson Seedless compared to Thompson Seedless. Treated fruits were slightly darker than non-treated fruits, but ethephon did not affect SSC, acidity, or firmness of Crimson Seedless, and ethephon residues were below legal limits.

Several species of Aspergillus section Nigri, including potential mycotoxin producers, are common residents of grape vineyards, but the relative population size of individual species throughout the growing season is difficult to determine using traditional isolation and identification methods. Using a quantitative droplet digital PCR (ddPCR) method in combination with dilution plating, total Aspergillus section Nigri populations and relative proportions of A. niger, A. welwitschiae, A. carbonarius, and A. tubingensis were measured from vineyard samples without the need for identifying individual fungal isolates. Grapes were sampled from two raisin vineyards (vineyards A and B) at berry set, veraison, harvest, and raisin stages in two consecutive years. Plate counts showed that the total population of Aspergillus section Nigri present on the fruit increased from berry set to raisin and became a larger component of the total recovered fungal population in both vineyards in both years. Results from ddPCR analysis showed that the relative proportion of A. carbonarius among the four species assayed increased later in the season (harvest and raisin) in comparison to earlier in the season (berry set and veraison). Total fungal and Aspergillus section Nigri plate counts were not significantly different between vineyards in either year. However, vineyard A generally showed higher proportions of A. carbonarius in harvest and raisin samples than vineyard B. This coincided with higher incidence and levels of ochratoxin A in vineyard A harvest and raisin fruit than in vineyard B fruit. This work demonstrates that this ddPCR method is a useful tool for culture-independent monitoring of populations of mycotoxigenic Aspergillus species during grape and raisin production.
