Photothermal therapy (PTT) has attracted much attention due to its less invasive, controllable and highly effective nature. However, PTT also suffers from intrinsic cancer resistance mediated by cell survival pathways. These survival pathways are regulated by a variety of proteins, among which heat shock protein (HSP) triggers thermotolerance and protects tumor cells from hyperthermia-induced apoptosis. Confronted by this challenge, we propose and validate here a novel MXene-based HSP-inhibited mild photothermal platform, which significantly enhances the sensitivity of tumor cells to heat-induced stress and thus improves the PPT efficacy. The Ti3C2@Qu nanocomposites are constructed by utilizing the high photothermal conversion ability of Ti3C2 nanosheets in combination with quercetin (Qu) as an inhibitor of HSP70. Qu molecules are loaded onto the nanoplatform in a pH-sensitive controlled release manner. The acidic environment of the tumor causes the burst-release of Qu molecules, which deplete the level of heat shock protein 70 (HSP70) in tumor cells and leave the tumor cells out from the protection of the heat-resistant survival pathway in advance, thus sensitizing the hyperthermia efficacy. The nanostructure, photothermal properties, pH-responsive controlled release, synergistic photothermal ablation of tumor cells in vitro and in vivo, and hyperthermia effect on subcellular structures of the Ti3C2@Qu nanocomposites were systematically investigated.

Background and objectives: To compare the efficacy and safety of topical interventions used for recurrent aphthous stomatitis. Materials and Methods: This network meta-analysis was conducted in accordance with the PRISMA statement. We searched four electronic databases, PubMed, Web of Science (WOS), Cochrane Central Register of Controlled Trials and Embase, for randomized controlled trials reporting efficacy and safety data on topical interventions for recurrent aphthous stomatitis. We performed a quality evaluation using a methodology based on the Cochrane Handbook. Two authors independently extracted data on healing effect, size reduction effect, symptom reduction effect, recurrence and safety assessment. Network meta-analysis was then performed using ADDIS and RevMan. Results: A total of 72 trials (5272 subjects) involving 29 topical interventions were included. Honey, lnsulin liposome gel, laser, amlexanox, glycyrrhiza and triamcinolone had better efficacy performance. Probiotics and chlorhexidine helped to prolong ulcer intervals and reduce recurrence. Doxycycline and penicillin had a high risk of adverse events. Hematologic evaluation showed no preference. The rank possibility of size-reducing effect and symptom-reducing effect supported the short-term effect of laser and the long-term effect of probiotics. Conclusions: We recommend the use of laser as a short-term intervention during the exacerbation phase of RAS and probiotics as a long-term intervention during the exacerbation and remission phases of RAS.

A cost-effective and highly reproducible capillary-based surface-enhanced Raman scattering (SERS) platform for sensitive, portable detection and identification of fentanyl is presented. Through encapsulating gold trisoctahedra (Au TOH) in the capillary tube for the first time, the SERS platform was constructed by combining the superior SERS properties of Au TOH and the advantages of capillaries in SERS signal amplification, facile sample extraction, and portable trace analysis. The effects of the size and density of Au TOH on the SERS performance were investigated by experiments and simulations, which showed that the maximum SERS enhancement was obtained for Au TOH with the size of 75 nm when particle density reached 74.54 counts/µm2. The proposed SERS platform possesses good reproducibility with a relative standard deviation (RSD) of less than 5%. As a demonstration, the platform was applied to detect fentanyl spiked in aqueous solution and serum samples with a limit of detection (LOD) as low as 1.86 and 40.63 ng/mL, respectively. We also validated the feasibility of the designed platform for accurate identification of trace fentanyl adulterated in heroin at mass concentration down to 0.1% (10 ng in 10 µg total). Overall, this work advances more explorations on capillary-based SERS platform to benefit portable trace analysis.

In the insect immune system, nodules are known to be a product of the cellular response against microorganisms and may be a preferential target for melanization. However, the mechanism of nodule-preferential melanization remains to be explored. In this study, we identified several mechanisms of nodule-preferential melanization by analyzing congregation and the activation of several factors involved in the prophenoloxidase (proPO)-activating system in the silkworm, Bombyx mori. Microorganism-binding assays revealed that B. mori larval plasma have an effective invading microorganism-surveillance network consisting of at least six pattern-recognition receptors (PRRs). We also found that a hemolymph serine proteinase, BmHP14, can bind to Saccharomyces cerevisiae. Pull-down assays showed that PRR C-type lectins form protein complexes with serine proteinase homologs, BmSPH1 and BmSPH2, which leads to the activated forms of BmSPH1 and BmSPH2 being gathered on microorganisms and trapped in nodules. Immunostaining analysis revealed that most factors in the proPO-activating system and some factors in the triggering system for antimicrobial peptide production exist in the granules of hemocytes which can gather in nodules. Western blot analysis showed that factors in the proPO-activating system are congregated in formed nodules by their concentration in plasma and aggregating hemocytes.

Nodules consisting of hemocytes and trapped microorganisms are important targets for melanization, which is best known in the insect immune system. We investigated factors functioning in nodule melanization and the mechanism by which these factors congregate in the nodule. BmHP21, BmSPH1 and BmSPH2, Bombyx mori orthologs of Manduca sexta serine protease HP21, serine protease homologs (SPH1 and SPH2), and a prophenoloxidase, BmPO1 were observed as inactive forms in the plasma, but as putatively active forms in the nodule. Production of prophenoloxidase-activating proteinases, BmPAP1 and BmPAP3/PPAE and BmPO1 were confirmed in hemocytes. BmSPH1 and BmSPH2 were observed on trapped bacterial cells in the nodule and were isolated from the surface of bacterial cells incubated with plasma. BmSPH1 and BmSPH2 were found in plasma in complex with a pattern recognition receptor, BmLBP. These data suggest that melanization-regulating factors congregate in nodules through a combination of microorganism-dependent and hemocyte-dependent routes.
