INTRODUCTION: Blood culture contamination remains a dilemma issue in the diagnosis of bloodstream infection. However, to date, there is no national data on blood culture contamination and the common organism isolated in Malaysia. This is a pioneer multi-centre study involving public hospitals with medical microbiologists in Malaysia to determine the blood culture contamination rate and the common organism isolated. MATERIALS AND METHODS: This retrospective cross-sectional study involved record review of all blood culture results over 9 months period from 1st January 2018 until 30th September 2018 in 27 government hospitals in Malaysia. For each positive culture result, the type of isolated organism was classified to represent true bacteraemia or contamination. RESULTS: We analysed 448,109 blood culture records from the participating hospitals. The blood culture positivity rate was 12.5% (57395 of 448109) and 25.0% (14367 of 57395) of the positive blood culture represents contamination. The national blood culture contamination rate in Malaysia was 3.2%. The contamination rate in the adult population was significantly higher than the paediatric population (3.6% vs. 2.6%; p<0.001). The blood contamination rate by institution ranged from 1.5% to 6.8%. The most frequently isolated microorganisms in the contaminated cultures were coagulase-negative staphylococci (71.0%). CONCLUSION: Blood culture contamination is a major issue that warrants priority in recognition, and interventions should be implemented to reduce the blood contamination rate in Malaysia.

OBJECTIVES: The aims of the study were to construct a new prognostic prediction model for detecting prostate cancer (PCa) patients using machine-learning (ML) techniques and to compare those models across systematic and target biopsy detection techniques. METHODS: The records of the two main hospitals in Riyadh, Saudi Arabia, were analyzed for data on diagnosed PCa from 2019 to 2023. Four ML algorithms were utilized for the prediction and classification of PCa. RESULTS: A total of 528 patients with prostate-specific antigen (PSA) greater than 3.5 ng/mL who had undergone transrectal ultrasound-guided prostate biopsy were evaluated. The total number of confirmed PCa cases was 234. Age, prostate volume, PSA, body mass index (BMI), multiparametric magnetic resonance imaging (mpMRI) score, number of regions of interest detected in MRI, and the diameter of the largest size lesion were significantly associated with PCa. Random Forest (RF) and XGBoost (XGB) (ML algorithms) accurately predicted PCa. Yet, their performance for classification and prediction of PCa was higher and more accurate for cases detected by targeted and combined biopsy (systematic and targeted together) compared to systematic biopsy alone. F1, the area under the curve (AUC), and the accuracy of XGB and RF models for targeted biopsy and combined biopsy ranged from 0.94 to 0.97 compared to the AUC of systematic biopsy for RF and XGB algorithms, respectively. CONCLUSIONS: The RF model generated and presented an excellent prediction capability for the risk of PCa detected by targeted and combined biopsy compared to systematic biopsy alone. ML models can prevent missed PCa diagnoses by serving as a screening tool.

A permanent cell line, SPB (Snubnose pompano brain) was established from Trachinotus blochii by the explant culture method. It has been sub-cultured more than 75 passages and showed optimal growth at 28°C using L-15 medium supplemented with 15% to 20% FBS. The SPB cells were cryopreserved at different passage levels for various applications. SPB cells were composed of fibroblastic and epithelial-like cells. The SPB cells were tested for mycoplasma contamination which was found to be negative. The origin of the SPB cell line from T. blochii was confirmed by amplification of the mitochondrial cytochrome oxidase I (COI) gene. The transfection efficiency of SPB cell line is 15% assessed by expression of green fluorescent protein using pEGFP-N1 plasmid. In addition, two CMV promotor plasmids pFNCPE42-DNA and pcDNAVP28 were transfected to SPB cells and it shows high expression levels of FNCP of fish nodavirus and VP28 protein of white spot syndrome virus by immunostaining. The SPB cells showed susceptibility to SJNNV and the infection was confirmed by RT-PCR, Western blot, ELISA, TCID50 and RT-qPCR. Experimental infection was carried out in T. blochii using SJNNV propagated in SPB cell line and found 100% mortality with clinical signs. The infection was confirmed by RT-PCR. The SPB cell line can be used for propagation of fish viral pathogens and production of the recombinant proteins.

Paclitaxel (PTX) is a potent anticancer drug. In the present study, PTX was loaded in poly-3-hydroxybutyrate-co-3-hydroxyvalarate (PHBV) to fabricate the PTX/PHBV (drug-loaded) nanoparticles via the nanoprecipitation method. Blank PHBV nanoparticles were also prepared. The drug-encapsulation efficiency of PTX/PHBV nanoparticles was 45±0.4%. The PTX/PHBV nanoparticles exhibited a pH-sensitive release profile and followed a quasi-Fickian diffusion mechanism. Cytotoxic properties of PHBV and PTX/PHBV nanoparticles were checked against the MCF-7 and Caco-2 cell lines. The PHBV nanoparticle did not inhibit the proliferation of MCF-7 and Caco-2 cell lines, thus depicting their non-toxic and biocompatible nature. On the other hand, the PTX/PHBV nanoparticles demonstrated 1.03-fold higher cytotoxicity and 1.61-fold enhanced apoptosis after treatment with the PTX/PHBV nanoparticles versus free PTX. In summary, the PHBV nanoparticles could be a potential candidate for the delivery of PTX for cancer treatment.

Background: The symmetry between maxillary anterior teeth and the face holds significant importance. This study assessed and analyzed the relationship between facial parameters and anterior teeth in the maxillary arch of male and female subjects. Specifically, individual width and combined width (CW) measurements of the maxillary anterior teeth were investigated. Methods: This study involved a total of 150 dentate Yemeni subjects (74 men and 76 women), whose ages ranged from 18 years old to 30 years old. A maxillary cast was created, and two digital photographs of the face of each subject were taken and analyzed. Digital calipers and AutoCAD were used to gather measurement data of the dental parameters (intercanthal distance [ICD], interpupillary distance [IPD], interalar width [IAW], intercommissural width [ICW], and bizygomatic width [BZW]) and facial parameters (profile distance). Results: Significant correlations were found for the following: IPD and width of six maxillary anterior teeth of each of the study subjects; ICD and their central incisors; and BZW and their canine width measurements. In contrast, IAW and ICW were not correlated with all tooth measurements. Linear regression findings showed that the CW measurement of the four incisors was significantly correlated with all facial parameter measurements, excluding the ICW and IAW in females and the IAW, ICW, and profile distance in males. Conclusion: The IPD and ICD of males and females may be used to determine their CW measurements. The BZW and IPD of males can be used to take precise anthropological measurements of the width of the central canines and incisors. Meanwhile, the IPD distance of females can be used to assess the central and lateral incisor widths.

Bisphenol A (BPA) is one of the most potent endocrine-disrupting chemicals (EDCs) that adversely affect aquatic organisms. The present investigation explored the effects of exposure to BPA at 0.1 and 1 mgL-1 concentrations on the fecundity of Biomphalaria alexandrina, snail's infection with Schistosoma mansoni, and histology of the ovotestis and topographical structure of S. mansoni cercariae emerged from exposed snails. The 24 h LC50 and LC90 values of BPA against B. alexandrina were 8.31 and 10.88 mgL-1 BPA, respectively. The exposure of snails to 0.1 or 1 mgL-1 BPA did not affect the snail's survival. However, these concentrations caused an increase in the reproductive rate (Ro) of infected snails. A slight decrease in egg production was observed in snails exposed to 0.1 mgL-1 BPA after being infected (infected then exposed). However, a significant increase in egg production was noted in snails exposed to 1 mgL-1 BPA after infection with S. mansoni. Histopathological investigations indicated a clear alteration in the ovotestis tissue structure of exposed and infected-exposed groups compared to the control snails. Chronic exposure to BPA caused pathological alterations in the gametogenic cells. SEM preparations of S. mansoni cercariae emerged from infected-exposed snails showed obvious body malformations. From a public health perspective, BPA pollution may negatively impact schistosomiasis transmission, as indicated by the disturbance in cercarial production and morphology. However, it has adverse effects on the reproduction and architecture of reproductive organs of exposed snails, indicating that B. alexandrina snails are sensitive to sublethal BPA exposure.

Shrimp farming is an important socioeconomic activity worldwide. Infectious myonecrosis virus (IMNV) is an important shrimp virus responsible for significant mortality (up to 70%) in Litopenaeus vannamei. We produced recombinant capsid protein (r-IMNV31) and obtained a highly specific antibody, anti-r-IMNV31, which was used in WOAH-approved ELISA and Western blot to detect IMNV. Further, anti-r-IMNV31 was employed in an indigenously developed lateral flow immunoassay (LFA) with gold nanoparticles as a visual label. Using LFA, IMNV could be detected rapidly (20 min) from tissue homogenate with high specificity, reproducibility, and sensitivity (LOD = 103 viral particles). LFA was validated with "gold standard" qRT-PCR using 60 samples with high sensitivity (100%), specificity (86%). A Cohen's kappa coefficient of 0.86 suggested "good agreement" between LFA and qRT-PCR. With a shelf-life of ~ 1 year at ambient temperature, the use of LFA in the on-site detection of IMNV by shrimp farmers will be a reality.

An aggregation-induced emission (AIE) active Schiff base L was obtained by reacting pyridoxal and 2-hydroxy-1-naphthaldehyde with p-phenylenediamine in two simple steps. The colorimetric, UV/VIS and fluorescence studies of L revealed that the yellow emissive L (λem =540 nm, λex =450 nm) in pure DMSO turned to a red-emissive L, when the poor solvent fraction (HEPES buffer, 10 mM, pH 7.4) was increased above 50 % in DMSO. The SEM and DLS results indicated the formation of self-aggregates of L that restricted the intramolecular motion and promoted the excited state intramolecular proton transfer (ESIPT) process. The cations sensing ability of the AIEgen L was explored in HEPES buffer (5 % DMSO, 10 mM, pH 7.4), where Cu2+ selectively quenched the fluorescence at 608 nm due to the chelation-enhanced fluorescence quenching (CHEQ) effect with an estimated sensitivity limit of 0.9 µM. Subsequently, the in situ formed AIEgen L-Cu2+ complex was applied for the cascade detection of glutathione (GSH), cysteine (Cys) and homocysteine (Hcy). The decomplexation of Cu2+ from the AIEgen L-Cu2+ by GSH, Cys and Hcy restored the quenched fluorescence emission of AIEgen L at 608 nm. With this Cu2+ displacement approach, the concentration of Cys, Hcy and GSH can be detected down to 2.8 µM, 3.12 µM and 2.0 µM, respectively. The practical utility of AIEgen L and AIEgen L-Cu2+ was examined by monitoring the selective analytes in real environmental and biological samples, and also applied successfully for the cell imaging applications.

Carbon nanotubes (CNTs) have been widely used in developing polymer hybrid coatings for anticorrosive application. In the present study, poly [(3,5-dimethyl-lH-pyrazole-1-yl) methyl methacrylate-co-glycidyl methacrylate] (PyM) was prepared by solution polymerization. Single-wall carbon nanotubes (SWCNT) were incorporated in the PyM by solution blending technique at different proportions. The PyM and its SWCNT (PyM-SWCNT) nanocomposites were characterized by FT-IR spectroscopy, X-Ray Diffraction, FE-SEM and HR-TEM. Different concentrations of PyM or PyM-SWCNT prepared in the present study were assessed separately for their toxicity by in vivo and in vitro assays using zebrafish embryos and gill cell line of zebrafish (DrG), respectively. The nanocomposites at the concentration of 400 µg ml-1 of PyM in 1.0% of SWCNT was found to be non-toxic and recommended for anticorrosive application whereas the nanocomposites with above 1% of SWCNT was found to be toxic. The nanocomposites with 1.5% of SWCNT delayed the hatching rate of eggs, decreased survival rate and heart beat in zebrafish embryos, and induced the morphological changes in DrG cells. Gene expression studies revealed that PyM-SWCNT with high concentration of SWCNT induced oxidative stress by activating ROS generations in zebrafish embryos and DrG cells. The immersion study of uncoated and coated with recommended concentration of PyM-SWCNT on mild steel (MS) in sea water was studied using FE-SEM and EDS, and the results showed effective corrosion protection without leaching behaviour. The nanocomposites with novel polymer in the present study may be used in the industry for anticorrosive purpose.

Partial anomalous pulmonary venous return (PAPVR) is an uncommon type of congenital heart disease occurring due to abnormal drainage of one or more, but not all the pulmonary veins to the systemic veins or directly to the right atrium. The PAPVR might have single (to the systemic veins) or dual drainage (to the systemic as well as left atrium). Management depends on the shunt impact on the heart and lungs, and it is usually surgical correction. In case of PAPVR with dual drainage, there is a new trend of percutaneous occlusion of the vertical vein with dual drainage anatomy, so that the blood is obliged to flow to the left atrium as in normal hearts. The scope of this manuscript is to highlight the availability of this alternative option and to present our experience and outcome in 6 PAPVR patients with dual drainage treated using this percutaneous approach.

K-ion batteries (KIBs) are promising for large-scale electrical energy storage owing to the abundant resources and the electrochemical specificity of potassium. Among the positive electrode materials for KIBs, vanadium-based polyanionic materials are interesting because of their high working voltage and good structural stability which dictates the cycle life. In this study, a potassium vanadium oxide phosphate, K6(VO)2(V2O3)2(PO4)4(P2O7), has been investigated as a 4 V class positive electrode material for non-aqueous KIBs. The material is synthesized through pyrolysis of a single metal-organic molecular precursor, K2[(VOHPO4)2(C2O4)] at 500 °C in air. The material demonstrates a reversible extraction/insertion of 2.7 mol of potassium from/into the structure at a discharge voltage of ∼4.03 V vs. K. Operando and ex situ powder X-ray diffraction analyses reveal that the material undergoes reversible K extraction/insertion during charge/discharge via a two-phase reaction mechanism. Despite the extraction/insertion of large potassium ions, the material demonstrates an insignificant volume change of ∼1.2% during charge/discharge resulting in excellent cycling stability without capacity degradation over 100 cycles in a highly concentrated electrolyte cell. Robustness of the polyanionic framework is proved from identical XRD patterns of the pristine and cycled electrodes (after 100 cycles).

Effluent produced during the electroplating process can contain high concentrations of heavy metals that can enter the environment and induce toxicity to aquatic organisms. Relatively high concentrations of zinc (Zn) and mercury (Hg) have been detected in treated electroplating industrial effluent (TEPIE), though the cytotoxic potential of these compounds has not been well assessed in fish gills. A novel cell line, Danio rerio gill (DrG), were exposed to TEPIE and concentrations of Zn, Hg, and Zn + Hg previously measured in treated effluent to evaluate the use of the DrG cell line following exposure to environmental pollutants. Several cytotoxic assays were employed to assess the effect of TEPIE, Zn, and Hg on this cell line. The percent cell viability was significantly reduced in a concentration-dependent manner following exposure to TEPIE, Zn, Hg, and Zn + Hg (p < 0.05) for 24 h, with additional morphological changes observed in exposure treatments relative to controls. Additionally, there was a significant induction of DNA damage detected in all exposure treatments determined through comet assay tail length. An increase in intracellular ROS generation was also observed in cells exposed to TEPIE, Zn, Hg, and Zn + Hg, corresponding to dose-dependent increases in apoptosis. Our study confirmed that TEPIE and the metals present in it induced cytotoxicity in the DrG cell line, demonstrating its usefulness as a model to explore relationships between pollutants and fish gills.

The rarity of acute psychosis in typhoid fever can result in delayed and misdiagnosis of the condition. We report a case of a 20-year-old man who presented with fever and acute psychotic symptoms. This was associated with headache, dizziness, and body weakness. There were no other significant symptoms. Neurological examination revealed reduced muscle tone of bilateral lower limbs but otherwise unremarkable. The computed tomography (CT) scan of his brain showed no abnormality. Blood specimens for microbiological culture grew Salmonella Typhi. This isolate was susceptible to chloramphenicol, ampicillin, ceftriaxone, ciprofloxacin, and trimethoprim-sulfamethoxazole. He was treated with intravenous ceftriaxone for one week and responded well. He was discharged with oral ciprofloxacin for another week. The repeated blood and stool for bacterial culture yielded no growth of Salmonella Typhi.

Infectious myonecrosis (IMN) is an important shrimp viral disease caused by infectious myonecrosis virus (IMNV). Based on previous reports, an attempt was made to propagate IMNV in apparently healthy C6/36 subclone of Aedes albopictus cell line. The confirmatory assays such as RT-PCR, real-time PCR and bioassay revealed that C6/36 cells were found to be susceptible to IMNV and these cells could be used easily for isolation and propagation of IMNV. The results of real-time PCR assay showed that a lower CT value of 22.25 in IMNV-infected cells was obtained on 10 day post-infection (d p.i.), whereas the higher CT value of 35.21 was obtained in IMNV-infected cells on 2 d p.i. There is no significant difference between CT values of IMNV production in vitro using C6/36 cell line and in vivo using shrimp. The IMNV propagated in C6/36 cells is capable of infecting shrimp and caused 100% mortality in shrimp. Clinical signs observed in shrimp injected with IMNV propagated in C6/36 cell line were found to be similar to naturally infected shrimp.

Pulmonary hydatid cyst (PHC) in pregnancy is a very rare pathology. We report here a case of ruptured PHC during pregnancy in a 26-year old Syrian (refugee) woman who presented with complaints of productive cough with metallic taste and dyspnoea. PHC was suspected due to her clinical and radiological findings. Interestingly, the sputum examination confirmed the diagnosis as numerous protoscoleces were present. Serology for Echinococcus infection revealed positive at high titre. Early diagnosis and prompt treatment by providing care improves the patient outcome. Parasitological examination of the respiratory specimen in suspected ruptured PHC is desirable as a valuable detection tool.

BACKGROUND: Despite decades of research, neurodegenerative disorders like Parkinson's disease remain a leading cause of disability worldwide, due to the insufficient reduction of disease burden by available medications. Recently, the benefits of dietary supplements like co-enzyme Q10 in neurodegenerative diseases have been reported. ; Aim: The protective effects of supplemental co-enzyme Q10 (CQ10) and possible additive benefits of CQ 10/Levodopa-Carbidopa (LD) in Chlorpromazine (CPZ)-induced Parkinsonism-like changes in mice were investigated. ; Methods: Male mice were assigned to ten groups of 30 mice each. Groups included: Vehicle control (fed Standard Diet (SD), and given intraperitoneal {ip} plus oral saline), LD group (fed SD, and given ip saline plus oral LD), two groups fed CQ10-supplemented diet (at 60 and 120 mg/kg of feed), and given ip plus oral saline, CPZ group (fed SD, and given ip CPZ plus oral saline), CPZ/LD group (fed SD, and given ip CPZ plus oral LD), two groups fed CQ10-supplemented diet (at 60 and 120 mg/kg of feed) and given ip CPZ plus oral saline, and another two groups fed CQ10-supplemented diet (at 60 and 120 mg/kg of feed) and given ip CPZ plus oral LD. The total duration of study was 21 days, and treatments were administered daily. Bodyweight and food intake were measured weekly, while neurobehavioural and biochemical tests were assessed at the end of the experimental period. ; Results: CQ10-supplementation was protective against CPZ-induced parkinsonism-like changes including, reduction in mortality, the reversal of retardation of open-field behaviours and reduction of catalepsy, increase in dopamine levels and decreased oxidative stress. CQ10 also showed significant improvements in these parameters when co-administered with LD. CQ10 (in groups administered CPZ/CQ10 60) showed greater benefit over LD on anxiety-related behaviours and also had additive benefits on working-memory. ; Conclusion: Dietary CQ10-supplementation was associated with demonstrable benefits in CPZinduced Parkinsonism-like changes in mice.

The spread of different types of cancer has been on a rise in the recent century. The use of chemical medications develops drug resistance and causes serious side effects. Eucalyptus camaldulensis (E. camaldulensis) is one of the most famous herbal remedies considered owing to its anti-inflammatory effect and boosting the intimate immune system; moreover, it has demonstrated some anti-proliferative effects on cancerous cell lines. The current study assessed the cytotoxic activity of alcoholic extract of Eucalyptus camaldulensis (E. camaldulensis) at different concentrations of 6.125, 12.5, 25, 50, and 100 µg/mL on breast cancer cell line MCF-7. The 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS)) analyses were employed to study the antioxidant efficiency of ethanolic extract of Eucalyptus camaldulensis. The results of Fourier transmission infra-red analysis and Phytochemical screening pointed to the presence of many active compounds in this extract, such as Tannins, Saponins, Phenolic compounds, Reducing sugar, Terpenoids, Steroids, Glucosides, Alkaloids, and Flavonoids. Furthermore, the results demonstrated that this extract significantly inhibited the growth of the MCF-7 cell line in a concentration-dependent manner, as compared to the control, and the cytotoxic activity of this extract elevated with an increase in the concentration. The results pointed out that E. camaldulensis can be considered a particularly valuable source of effective anti-proliferative and cytotoxic agents. The experimental findings demonstrated that E. camaldulensis extract possessed significant antioxidant efficiency and anti-proliferative effects on cancerous cell lines.

@#The rarity of acute psychosis in typhoid fever can result in delayed and misdiagnosis of the condition. We report a case of a 20-year-old man who presented with fever and acute psychotic symptoms. This was associated with headache, dizziness, and body weakness. There were no other significant symptoms. Neurological examination revealed reduced muscle tone of bilateral lower limbs but otherwise unremarkable. The computed tomography (CT) scan of his brain showed no abnormality. Blood specimens for microbiological culture grew Salmonella Typhi. This isolate was susceptible to chloramphenicol, ampicillin, ceftriaxone, ciprofloxacin, and trimethoprim-sulfamethoxazole. He was treated with intravenous ceftriaxone for one week and responded well. He was discharged with oral ciprofloxacin for another week. The repeated blood and stool for bacterial culture yielded no growth of Salmonella Typhi.
