RESUMEN

Nanosized spherical polyelectrolyte brushes (SPBs) are ideal candidates for the preparation of nanometal catalysts, protein separation, and medical diagnostics. Until now, SPBs have been synthesized by photo-emulsion polymerization in a batch reactor, which remains challenging to scale up. This paper reports a successful continuous preparation of SPBs by photo-emulsion polymerization in a self-made microreactor. The effects of residence time, monomer concentration, and feed ratios on the conversion of monomers and SPB structures are systematically investigated by dynamic lighting scattering and transmission electron microscopy. Poly(acrylic acid) (PAA) SPBs obtained in a microreactor exhibiting a narrow size distribution with a short reaction time are very effective in inhibiting the calcium carbonate scale and are comparable to those produced in a batch reactor. This work confirms the feasibility of continuous preparation and scaled-up production of SPBs.

Geocell is widely used in the treatment of poor roadbed, which can restrain soil laterally and improve the stability of soil. In cold area engineering, a change in temperature can influence the mechanical properties of geocell of different materials. To study the mechanical response of geocell at low temperatures, three types of geocell strips commonly used in engineering, namely the polyethylene (HDPE), polypropylene (PP), and polyester (PET), were studied via the uniaxial tensile test at the ambient temperatures of -5 °C, -20 °C, and -35 °C, respectively. Meanwhile, the tensile strength, fracture mode, and temperature sensitivity of geocell specimens were compared. It is concluded that: (1) at low temperatures, the tensile strengths of HDPE and PET geocell strips are significantly improved, while that of the PP geocell strip is less sensitive to the temperature. (2) The PP geocell is subject to a brittle failure at all ambient temperatures. The PET geocell strip experiences a hard-ductile failure at normal temperatures of -5 °C and -20 °C. While in the tensile test at -35 °C, it is prone to brittle failure and hard-ductile failure. The HDPE geocell strip suffers from ductile failure at all ambient temperatures. (3) At low temperatures, overall, the tensile properties of the PET geocell strip is better than those of the PP and HDPE geocell strips.

BACKGROUND AND AIM: The high-mobility group box 1 (HMGB1) protein plays an important role in a lot of biological behaviors, including DNA damage repair, gene transcription, cell replication, and cell death, and its expression is higher in many solid tumors tissues than in their adjacent normal tissues, and it is always involved in tumor proliferation, metastasis, therapeutic tolerance, and poor prognosis. However, HMGB1 in proliferation and radioresistance of esophageal squamous cell carcinoma (ESCC) remains poorly understood. In this study, the effect of HMGB1 on proliferation, cell death, DNA damage repair and radioresistance, and its underlying mechanism was investigated in human ESCC. METHODS: The immunohistochemistry scores of tumor and adjacent normal tissues in ESCC tissue microarray were analyzed. Stable HMGB1 knockdown cell lines were constructed using Kyse150 and Kyse450 cells. Cell viability, radioresistance, apoptosis, autophagy, and DNA damage were determined using CCK-8, 5-ethynyl-2'-deoxyuridine, clonogenic survival assay, immunofluorescence, flow cytometry, and western blot assays. RESULTS: Differential analyses showed that the expression of HMGB1 in esophageal cancer tissue was significantly higher than that in adjacent normal tissues. The downregulation of HMGB1 could effectively inhibit proliferation, increase radiosensitivity, impair DNA damage repair abilities, reduce autophagy, and increase apoptosis rates in ESCC cells after irradiation. CONCLUSIONS: HMGB1 is expected to be a potential target for ESCC therapy and radiosensitization.

Endocrine therapy and radiotherapy are the main treatments for luminal A breast cancer. However, drug and radiotherapy resistance could occur during long-term treatment, leading to local recurrence and distant metastasis. Some studies have found that drug resistance might be related to human epidermal growth factor receptor-3 (HER3) overexpression. However, whether HER3 plays a role in radiotherapy resistance is unknown. The purpose of this study is to elucidate the effect of HER3 in radiotherapy and to assess whether HER3 could be a potential target for radiosensitivity. We used retroviruses to construct stable low expression of HER3 in MCF-7 and ZR75-1cells. The CCK-8 assay was used to observe proliferation. Colony-forming assay was used to detect radiosensitivity. Flow cytometry was used to observe the cell cycle and apoptosis. Immunofluorescence assay was used to detect the number of γH2AX foci in the nucleus with or without ionizing radiation (IR). Western blot analysis was used to verify the change of relative proteins. Nude mice were used to observe tumor growth in vivo. In our study, silencing HER3 reduced cell proliferation and clone formation ability after IR, so silencing HER3 increased the sensitivity of luminal A breast cancer cells to radiotherapy. In terms of radiosensitivity mechanisms, it is suggested that the silencing of HER3 enhanced IR-induced DNA damage, reduced DNA repair, and increased apoptosis and G2 /M arrest. In addition, silencing HER3 combined with IR clearly inhibited the transplanted tumor growth in vivo. Therefore, we concluded that HER3 played a role in radiotherapy resistance. Silencing HER3 increased the radiosensitivity of luminal A breast cancer cells and HER3 could be a potential target for radiosensitivity.

INTRODUCTION: Radiotherapy is by means of ionizing radiation to kill tumor cells, inhibit and control the growth, metastasis and diffusion of tumor cells. During the last few decades, application of radiotherapy combined with chemotherapy and surgery are clinical mainstream treatments. However, little is known what radio-sensitization agents have been patented in China and what the potential drug candidates for patents are in China. Areas covered: This reviews covers research and patent literature of the last 10 years dealing with the discovery and development of novel radio-sensitization patents in China. Expert opinion: The 94 radio-sensitization patents granted from 2006 to 2015 mainly focus on six types of products. They are: traditional Chinese medicines (TCM), synthetic compounds, combinations of synthetic compounds and TCM, biological products, medical apparatus and others. In the course of tumor treatment, radiotherapy occupies an irreplaceable position. Previously believed that due to the prevalence of hypoxic cells in solid tumors, most of the tumor exist a certain degree of radiation resistance. To find effective ways to improve the sensitivity of tumor cells to radiation therapy has become a focus in scientific research and clinical treatment. So radiation sensitivity has been proposed and widely studied.

In order to understand the composition and quantitative variation of ecdysteroids in the adults and eggs of Opogona sacchari (Bojer), an invasive alien pest, we analyzed the ecdysteroid composition and titers in the adult and egg of this pest. On day 4 after eclosion, the titer of ecdysteroids in the male adult was 0.080 ng/adult, much lower than 5.978 ng/adult in the female adult. During the development of ovaries, the titer of ecdysteroids was low on the first two days, and high in the late period, with the peak (10.48 ng/ovary) appearing on day 3. During the development of eggs, the titer of ecdysteroids was about 0.010 ng/egg from day 1 to day 3, and then decreased to 0.006 ng/egg on day 4. In both adults and eggs, three main components of ecdysteroids were found by identification of HPLC/RIA. They were 20-hydroxyecdysone, 26-hydroxyecdysone, and an unidentified component.
