AIM: This study aimed to develop an anti-aging nanoformulation with Curcuma heyneana extract as bioactive substance. BACKGROUND: Curcuma heyneana Valeton & Zipj extract has been proven in previous research to have antioxidant, anti-ageing, anti-inflammatory, and wound healing properties, which makes it a potential bioactive material for anti-ageing and sunscreen cosmetic products. Phytoantioxidants need to penetrate into deeper skin layers to ensure effectivity. Thus, a transdermal delivery system is needed to deliver the extract to a deeper skin layer. OBJECTIVE: The objective of the study was to compare the permeability and anti-ageing activity of liposomal and ethosomal formulations of C. heynena rhizome ethanolic extract. METHODS: In this study, C. heyneana extract was loaded into a phospholipid vesicular system in the form of liposome and ethosome formulations using the ethanolic injection method. The anti-ageing activity was assessed by analyzing the epidermal thickness, number of sunburn cells, distance between collagen fibres, and number of fibroblasts. While the histologic specimen scoring was carried out for the in vivo penetration study. RESULTS: The ethosomal formulation had been found to have better penetration ability since it was able to reach the lower dermis area compared to the liposomes, which only reached the upper dermis. The ethosomal formulation of C. heyneana extract exhibited a better anti-ageing activity based on the parameters of epidermal thickness, sunburn cell count, fibroblast count, and the distance between collagen fibres in rat skin histology. CONCLUSION: Ethosomes have been found to be a more proficient carrier system for transdermal delivery of C. heyneana extract compared to liposomes. Meanwhile, their penetration correlated with the effectivity of the formulation, suggesting that the vesicular system enhanced the penetration ability of the extract.

Background and Aim: Sulfadiazine, one of the sulfonamide group's active compounds, is widely used for therapeutic production against several diseases. Veterinary drug residues can have a significant impact on human health conditions. This study aimed to develop a prototype of rapid test devices (RTDs) for detecting sulfadiazine residues on chicken carcasses based on the color indication. Materials and Methods: Seven samples of carcasses collected from traditional breeders in Surabaya-Indonesia were prepared and tested using RTDs. This sample represents the population considering that in the last report, the use of antibiotics was more than 40%, while the ability to monitor RTDs was estimated at 100. The standard color of purple by Hex code standard color or decimal code color was used to compare the positive samples. A light-emitting diode (LED) lamp was used to observe purple color. Analysis of sulfonamides resulting from RTDs was compared using a ultraviolet-visible spectrophotometer. Results: Sulfonamides contamination levels of 50% and 100% were detected at concentrations of 0.472 µg/mL and 0.642 µg/mL, respectively. Sulfonamides contamination that was <0.395 µg/mL did not appear purple. Conclusion: The study's findings showed that RTDs can be used to detect sulfonamides residues at a limit of detection 0.5 mg/mL after a 45 min exposure to an LED operating at a wavelength of 980 nm (p < 0.05). The limitation of RTDs was not being able to monitor the presence of residues bound in fat samples. Rapid test devices can be developed for commonly monitoring devices due to the limited technology available in the market.

Background and Aim: Human health problems due as a microbial resistance or tumors and cancers because consumption of the carcasses containing residues of tetracycline are main global problems in the context of fight against antimicrobial resistance phenomena. Explanation of the sustainable development goals, particularly point 3, is well recognized that all animal products for human consumption must be safe to live a healthy life. This study aimed to design a prototype of rapid test devices (RTD) based on principles of precipitate to obtain a specific color change after the process of reactions as an indicator to determine tetracycline residues in the carcass. Materials and Methods: Five samples of tetracycline-containing poultry carcasses using artificial add the tetracycline at pharmaceutics grade were examined using a prototype of a strong reaction solution for tetracycline fixation based on the concept bonded by ion Fe(III) at atom O in position atom C-1 at the ring of tetracycline and ion N+ as the functional branch of tetracycline. RTD detection was evaluated using a yellow color presentation and an absorbance spectrometric technique at a wavelength of 273 nm. Results: The following chemicals were used to create the best-fixed tetracycline residue: HCl and H2SO4 dissolved in H2O, chromatographic grade of 0.1 N and 0.5 N of HNO3, and 1% Fe (III) Cl. The RTD had a higher limit of detection (LOD) than the ultraviolet-visible spectrophotometer. Conclusion: The results of this study revealed that RTD, as constructed in this study, can be used to detect residue at LOD 44.764 mg/mL during 120 min of exposure through a light-emitting diode at 980 nm wavelength (p<0.05). The necessity for using RTD was because of the apparent limitations of conventional devices.

Background and purpose: This study aimed to determine the potency of kebar grass ethanol extract to overcome an increase in cerebellar neuronal cell necrosis, which has an impact on decreasing motor reflex function and spatial memory of mice from lactating mothers exposed to carbofuran. Experimental approach: Forty lactating mice were divided into four groups, 10 each; including control, T1 (carbofuran 0.0125 mg/day), T2 (vitamin C 5 mg + carbofuran 0.0125 mg/day), T3 (kebar grass extract 3.375 mg + carbofuran 0.0125 mg/day). The mice were orally administered with carbofuran, vitamin C, and kebar grass extract on days 0 to 14 postnatal. On the 15th day, brains of the mice were necropsied to measure the levels of malondialdehyde (MDA), superoxide dismutase (SOD), and glutathione (GSH), H&E staining; motor reflex tests were performed on 10-day-old mice, and the mice aged 30 days were tested on their swimming and spatial memory. Findings / Results: Carbofuran caused an increase in MDA, GSH, neuronal cell necrosis, surface righting reflex, a decrease in SOD, swimming ability, and spatial memory. Kebar grass extract and vitamin C administration decreased MDA, GSH, neuron necrosis, surface righting reflex, and increased SOD, swimming ability, and spatial memory. Conclusion and implications: Exposing to carbofuran in lactating mice caused brain oxidative stress, impaired motor reflexes, and spatial memory in mice offspring. Kebar grass extract and vitamin C administration prevented brain oxidative stress and inhibited disorders in motor reflexes, and spatial memory in mice offspring. Kebar grass extract administration was more effective than vitamin C.

ETHNOPHARMACOLOGY RELEVANCE: Graptophyllum pictum (L.) Griff., known as "handeuleum" in West Java and "Daun Ungu" in Indonesia, is traditionally used to cure hemorrhoids. AIM OF THE STUDY: The purpose of this study is to prove its effectiveness scientifically using anorectal histological parameters in Croton oil-induced hemorrhoid mice. MATERIALS AND METHODS: In vivo tests were performed by observing histomorphologic changes in mice anorectal tissue induced by croton oil. In addition, in vitro assay was performed for evaluating antioxidant activity, astringency property, and hemostasis-associated activity. The antioxidant activity was measured using a DPPH radical scavenging assay. The total flavonoid and phenolic contents were also determined spectrophotometrically. RESULTS: The in vivo assay showed that the oral-topical combination use of the ethanolic extract of G. pictum leaves demonstrated significant improvement on the croton oil-induced anorectal damage better than the single application by oral or topical application. CONCLUSION: These results showed that G. pictum has potent anti hemorrhoid activity, especially for the combinational use of oral and topical administration.

Currently, there are increasing numbers of dermal and transdermal dosage forms of both natural and synthetic compounds on the market. Therefore, it is necessary to have a method that can measure the release and penetration of the compound into the skin. This chapter presents a current method for evaluating the skin penetration of a Curcuma heyneana rhizome extract in vivo using histological parameters. We also evaluate a liposome delivery system of the same extract to determine any differences in penetration due to changes in the drug delivery system.

Background: Varanus salvator is one of the reptiles being hunted by human beings for several purposes, including traditional medicine. The studies about reproductive biology aspects were limited. Aim: This study aimed to determine the morphology, histology, and histometry of V. salvator paryphasmata and hemibaculum based on Snout-Vent Length (SVL) as an indicator of sexual maturity. Methods: This study examined 18 pairs of hemipenis of V. salvator with SVL more and less than 40 cm in equal number. Paryphasmata and hemibaculum parts were observed visually and micro-sliced, then stained with Hematoxylin-Eosin (HE). The histological observation was conducted under a 40×, 100×, and 400× magnification of a light microscope. The histometry of the paryphasmata was examined using 13 Megapixels Coolpad and OptiLab Plus for microscopic pictures. The chondrocyte cell area was measured using the Optilab Plus and Image Raster three applications. Results: The sizes of glans of hemipenis, paryphasmata, and hemibaculum increased according to the increasing of SVL. The average paryphasmata row number, epidermis, and loose connective tissue thickness were not significantly different (p > 0.05). However, dense connective tissue was thicker (p < 0.05), which corresponds to SVL. Hemibaculum was composed of fibrous and hyaline cartilage characterized by chondrocyte cells. The SVL also affects (p < 0.05) the ossification of hyaline in hemipenis, while the chondrocyte cell area followed the equation -1.87E7 + 7.09E5* SVL. Conclusion: The SVL size of V. salvator affects the paryphasmata, hemibaculum, thickness of dense connective tissue of paryphasmata, and the area of chondrocyte cells.

We present a histomorphometric analysis to enable quantitative measurement of anti-aging activity of topical substances in rat skin. In this method, the measured parameters were closely related to changes that occur on the skin caused by exposure to UV light. We used this protocol to determine the anti-aging activities of an ethanolic extract of Curcuma heyneana rhizome and retinoic acid by a measurement of the changes in epidermal thickness, the number of sunburned cells, the number of fibroblasts, and the space between collagen fibers.

ETHNOPHARMACOLOGICAL RELEVANCE: Temu giring, the rhizome of Curcuma heyneana Valeton & Zipj (C. heyneana), is native to tropical regions, especially in Indonesia. It is traditionally used as a skin care, for cosmetic and body freshness, in Javanese and Balinese women, and has many other bioactivities such as antioxidant, anticancer and antiinflammatory. AIM OF THE STUDY: The purpose of this study was to determine the antiaging activity of C. heyneana to prove its traditional use. MATERIALS AND METHODS: The antioxidant activity was determined using the DPPH free radical method, and anti-aging activity was examined using in vitro assays such as tyrosinase inhibitor and collagenase inhibitor. In vivo tests were performed by observing histomorphologic changes in rat skin exposed to Ultraviolet (UV) rays. The total curcuminoid contents and chromatographic profiles were determined by Thin Layer Chromatography (TLC) - densitometry. RESULTS: In all in vitro assays, all of the extracts showed a dose-dependent manner in the final concentration range from 62.5 to 500 µg/mL for DPPH assay and 31.25 to 250 µg/mL for tyrosinase inhibition and collagenase inhibition assay. Curcuminoid (CUR), the active principle of Curcuma genus shows antioxidant, tyrosinase inhibitor and collagenase inhibitor activity greater than all C. heyneana extracts. The in vivo assay results showed that the topical application of the crude extract of C. heyneana produced significant improvement effects on the UV-induced skin structure damage. The total CUR content was correlated with the anti-aging activity of Curcuma heyneana. CONCLUSIONS: The results show that C. heyneana contains antioxidant compounds and has potent anti-aging activity, indicating that it can be used as an anti-aging drug candidate or as a phyto-cosmeceutical.

AIM: This study was to evaluate effect of honey in repairing damage of liver tissue due to energy protein malnutrition and in mobilization of endogenous stem cells. MATERIALS AND METHODS: Male mice model of degenerative liver was obtained through food fasting but still have drinking water for 5 days. It caused energy protein malnutrition and damage of liver tissue. The administration of 50% (v/v) honey was performed for 10 consecutive days, while the positive control group was fasted and not given honey and the negative control not fasted and without honey. Observations of regeneration the liver tissue based on histologically examination, observation of Hsp70 expression, and homing signal based on vascular endothelial growth factor-1 (VEGF-1) expression using immunohistochemistry technique. Observation on expression of CD34 and CD45 as the marker of auto mobilization of hematopoietic stem cells using flow cytometry technique. RESULTS: There is regeneration of the liver tissue due to protein energy malnutrition, decrease of Hsp70 expression, increase of VEGF-1 expression, and high expression of CD34 and CD45. CONCLUSION: Honey can improve the liver tissue based on: (1) Mobilization of endogenous stem cells (CD34 and CD45); (2) Hsp70 and VEGF-1 expressions as regeneration marker of improvement, and (3) regeneration histologically of liver tissue.
