A series of flavanols were synthesized to assess their biological activity against human non-small cell lung cancer cells (A549). Among the sixteen synthesized compounds, it was observed that compounds 6k (3.14 ± 0.29 µM) and 6l (0.46 ± 0.02 µM) exhibited higher potency compared to 5-fluorouracil (5-Fu, 4.98 ± 0.41 µM), a clinical anticancer drug which was used as a positive control. Moreover, compound 6l (4'-bromoflavonol) markedly induced apoptosis of A549 cells through the mitochondrial- and caspase-3-dependent pathways. Consequently, compound 6l might be developed as a candidate for treating or preventing lung cancer.

Introduction: Postoperative pain and complications pose significant challenges following a hemorrhoidectomy. Attaining effective anesthesia with minimal complications is crucial. The ideal anesthesia method for ambulatory hemorrhoidectomy remains uncertain. This study aimed to investigate whether the combination of general anesthesia plus local infiltration (GAL) is associated with lower complications and reduced pain compared to spinal anesthesia (SA) in the context of hemorrhoidectomy. Methods: This retrospective single-center cohort study, conducted in a tertiary medical center in East Asia, evaluated excisional hemorrhoidectomies performed between January 1, 2017, and March 31, 2023, utilizing GAL or SA. Data on the six most common complications-pain, constipation, acute urine retention (AUR), bleeding, nausea, and headache-were extracted from medical records. A total of 550 hemorrhoidectomies were included: 220 in the GAL group and 330 in the SA group. Patient characteristics were comparable between the two groups. Results: The AUR rate was significantly lower in the GAL group compared to the SA group (15.5% vs. 32.1%, P < 0.001). Although the proportion of pain scores ≥4 did not differ significantly between the GAL and SA groups (36.2% vs. 39.8%, P = 0.429), the pain score curve indicated a stable trend. Overall, the GAL group exhibited a lower rate of adverse effects (56.9% vs. 67.4%, P = 0.023). There were no significant differences in the rates of other complications and emergency department readmission between the GAL and SA groups. Discussion: GAL emerges as a favorable choice for anesthesia in hemorrhoidectomy, demonstrating a lower incidence of urine retention and a prolonged analgesic effect in multiple hemorrhoidectomies. These findings support the conclusion that GAL represents an optimal anesthetic method for enhancing the postoperative experience in patients undergoing hemorrhoidectomy.

This study modified a passive sampling technique similar to the US EPA Method 325 A/B method but extended to include more toxic volatile organic compounds (VOCs) under varied climate conditions to enhance field applicability. A mixing chamber was built to determine uptake rates (Us) for the target compounds. It was found that the Us of 27 air toxics previously reported in the literature agreed reasonably well with our findings within 18%, thus proving the chamber's integrity. To broaden the compound coverage, both Carbopack X and Carboxen 569 were studied for a suite of toxic VOCs to meet stringent quality control (QC) criteria of correlation coefficients (R-square), method detection limits (MDL), back diffusion (BD), storage stability, as well as a wide range of climate conditions in temperature and humidity. After excluding the species that failed to pass any of the QC criteria, Carbopack X was found to fit 50 air toxics, whereas Carboxen 569 held 37. After excluding the overlapped species, 61 toxic VOCs can be determined with robust Us for a broad range of climate conditions when the two sorbents are used in pairs. A one-week field measurement was conducted to compare with the online thermal desorption gas chromatography-mass spectrometry (TD-GC-MS) with hourly data resolution. The field passive sampling showed comparable results to the means of the online hourly measurements, despite the high variability of selected target compounds, such as toluene from 0.3 ppbv as the 5th percentile to the maximum of about 80 ppbv. Passive sampling clearly demonstrated the ability to smooth out concentration variability and thus the time-averaging strength of toxic VOCs, revealing its ideal role as an exposure monitor over time. The passive sampling method can be more desired than active sampling or online methods when the aim is simply the knowledge of prolonged time-averaged concentrations.

Emotion recognition ability is the basis of interpersonal communication and detection of brain alterations. Existing tools for assessing emotion recognition ability are mostly single modality, paper-and-pencil test format, and using only Western stimuli. However, various modalities and cultural factors greatly influence emotion recognition ability. We aimed to develop a multi-modality emotion recognition mobile application (MMER app). A total of 169 healthy adults were recruited as participants. The MMER app's materials were extracted from a published database, and tablets were used as the interface. The Rasch, factor analysis, and related psychometric analyses were performed. The Cronbach alpha was 0.94, and the test-retest reliability was 0.85. Factor analyses identified three factors. In addition, an adjusted score formula was provided for clinical use. The MMER app has good psychometric properties, and its further possible applications and investigations are discussed.

MicroRNA-132 (miR-132) has been demonstrated to affect multiple neuronal functions, including dendritic growth and spinogenesis in cultured neurons and brain slices, as well as learning behavior of animals. However, its role in acquisition of temporal-associated memory remains unclear. In this study, we demonstrated that the mature miR-132 level in mouse hippocampus was significantly increased at 30 min after trace fear conditioning, a type of temporal-associated learning, and returned to baseline values in 2 h. We then knocked down miR-132 expression in vivo by infusing a lentivector expressing anti-miR-132 hairpin RNA into the third ventricle near the anterior hippocampi such RNA diffused laterally to both hippocampal formations, later confirmed by histological analysis. This approach successfully reduced hippocampal miR-132 expression in both naïve and trace fear conditioned groups, and impaired acquisition of trace fear memory in mice. To our knowledge, this result is the first demonstration of change in temporal learning behavior by reducing microRNA (miRNA) level specifically in the hippocampal region.

This study established a simultaneous screening method based on solid-phase extraction and liquid chromatography-tandem mass spectrometry (LC-MS-MS) for the detection of 23 stimulants and 23 diuretics in human urine. An electrospray ionization source and multiple reaction monitoring were used for data acquisition. All stimulants and diuretics were separated in less than 12.52 min. The limits of detection were in the range of 25-500 ng/mL for stimulants and 25-125 ng/mL for diuretics. To evaluate the performance of this method, urine samples were collected from 1627 athletes in Taiwan, and 7 positive samples were found. This LC-MS-MS method not only meets the minimum required performance limits set by the World Anti-Doping Agency but also provides a fast way to analyze the authentic urine samples in doping control laboratories.
