RESUMEN

The aim of this study was to evaluate the in vitro activity of Syzygium aromaticum essential oil (SAEO) and its main constituent eugenol (EG) against adult fleas and their action in the maturation of eggs into adults of Ctenocephalides felis felis. In order to evaluate the pulicidal activity, 10 adult fleas were exposed to a filter paper impregnated with SAEO and EG at increasing concentrations of 0.047; 0.094; 0.188; 0.375; 0.750; 1.50; 3.00; 6.00; 12.00 and 24.00 µg cm-2. Flea mortality was evaluated 24 and 48 h after exposure. In order to evaluate the inhibition of the maturation of eggs into adults, 10 eggs were exposed to filter paper impregnated with SAEO and EG at the same concentrations used in the pulicidal test, and the evaluation was performed 30 days after incubation. Untreated repetitions were maintained in both studies (control group). The lethal concentration 50 (LC50) of pulicidal activity to SAEO was 5.70 µg cm-2 in 24 h and 3.91 µg cm-2 in 48 h. The LC90 was 16.10 µg cm-2 and 15.80 µg cm-2 in 24 and 48 h, respectively. The LC50 of inhibition of the maturation of eggs into adults was 0.30 µg cm-2 and the LC90 3.44 µg cm-2. The LC50 of pulicidal activity to EG was 2.40 µg cm-2 in 24 h and 1.40 µg cm-2 in 48 h; the LC90 was 8.10 µg cm-2 and 3.70 µg cm-2 in 24 h and 48 h, respectively. The LC50 of inhibition of the maturation of eggs into adults was 0.10 µg cm-2 and the LC90 0.68 µg cm-2. Based on the results obtained, it is possible to conclude that the both SAEO and EG have in vitro pulicidal activity as well as acting as inhibitors of the maturation of eggs into adults of the flea C. felis felis.

Essential oils (EOs) are considered a new class of ecological products aimed at the control of insects for industrial and domestic use; however, there still is a lack of studies involving the control of fleas. Ctenocephalides felis felis, the most observed parasite in dogs and cats, is associated with several diseases. The aim of this study was to evaluate the in vitro activity, the establishment of LC50 and toxicity of EOs from Alpinia zerumbet (Pers.) B. L. Burtt & R. M. Sm, Cinnamomum spp., Laurus nobilis L., Mentha spicata L., Ocimum gratissimum L. and Cymbopogon nardus (L.) Rendle against immature stages and adults of C. felis felis. Bioassay results suggest that the method of evaluation was able to perform a pre-screening of the activity of several EOs, including the discriminatory evaluation of flea stages by their LC50. Ocimum gratissimum EO was the most effective in the in vitro assays against all flea stages, presenting adulticide (LC50 = 5.85 µg cm-2), ovicidal (LC50 = 1.79 µg cm-2) and larvicidal (LC50 = 1.21 µg cm-2) mortality at low doses. It also presented an excellent profile in a toxicological eukaryotic model. These findings may support studies involving the development of non-toxic products for the control of fleas in dogs and cats.

Necrotizing soft tissue infections (NSTI) are diseases that can affect the skin, subcutaneous tissue, superficial fascia, and occasionally, deep fascia, muscle tissue and adipose tissue. They present states of necrosis, which can lead to systemic toxicity and high rates of mortality. This paper describes the occurrence of a NSTI in a six year-old female beagle. The animal presented pain, areas of edema, erythema, loss of sensitivity and crepitations, in the right flank, from the dorso-lumbar area to the mammary gland. In the bacterial culture, Staphylococcus pseudintermedius and Group B Streptococcus, were isolated. The biopsy revealed necrotizing cellulites. Treatment was administered with ceftriaxone (50 mg/kg IV 12/12h), metronidazole (25 mg/kg 12/12h), and tramadol clorhidrate (4 mg/kg IV 8/8h). Wound treatment included extensive cleanings, every twelve hours, with warm diluted iodopovidone (0,01%) and compressive bandages. The diagnosis and early treatment of this disease allows a better prognosis for affected animals.

O objetivo do trabalho foi avaliar através de teste in vivo, a eficácia e o período residual de proteção do fipronil 10% "top spot" em cães infestados com diferentes cargas parasitárias de Ctenocephalides felis felis. Foram utilizados 24 cães da raça Beagle, compondo seis animais por grupo. Os cães foram divididos em quatro grupos. Os cães dos grupos controles I e II não receberam tratamento, enquanto que os cães dos grupos tratados I e II receberam tratamento com formulação de fipronil 10% "top spot". Os cães dos grupos controle I e tratado I foram infestados com 100 pulgas adultas não alimentadas, e os cães dos grupos controle II e tratado II foram infestados com 300 pulgas adultas não alimentadas. As infestações foram realizadas nos dias, -2, +5, +12, +19, +26, +33 e +40 e nos dias +2, +7, +14, +21, +28, +35 e +42 foi realizada retirada mecânica e contagem de pulgas para avaliação. As eficácias pulguicidas, para o grupo tratado I, nos dias +2, +7, +14, +21, +28, +35 e +42, foram respectivamente 99,36%; 99,73%; 99,48%; 99,74%; 99,75%; 95,06% e 67,62%. As eficácias pulguicidas, para o grupo tratado II, avaliadas nos mesmos dias, foram respectivamente 100%; 100%; 100%; 100%; 99,91%; 95,60% e 68,55%. O fipronil mostrou-se eficaz na eliminação das pulgas em cães até o dia +35. A análise estatística comparativa entre as médias de pulgas vivas, entre os grupos controle I e tratado I, demonstrou que ocorreu diferença significativa (p≤0,05) para os desafios em todos os dias experimentais, após o tratamento. Os grupos controle II e tratado II também apresentaram diferença significativa (p≤0,05) para os desafios em todos os dias experimentais, após o tratamento. A análise estatística entre os grupos tratados I e II demonstrou que não ocorreu diferença significativa (p≥0,05) para os desafios em todos os dias experimentais. O desafio foi encerrado no dia +42 já que a eficácia do fipronil nos grupos tratados I e II foram inferiores 70%. O produto em teste mostrou-se eficaz na eliminação das pulgas em cães até o dia + 35, não apresentando mais efeito residual de proteção quando os animais foram reinfestados. Não houve diferença significativa nos níveis de eficácia entre os grupos infestados com 100 e 300 exemplares adultos de C. felis felis ao longo do período experimental.(AU)

Efficacy for treated group I on days +2, +7, +14, +21, +28, +35 and +42 was respectively 99.36%, 99.73%, 99.48%, 99.00%, 74.00%, 99.75%, 95.06% and 67.62%. The efficacy for the treated group II, evaluated on the same days, was respectively 100%, 100%, 100%, 100%, 99.91, 95.60% and 68.55%. Fipronil was effective in eliminating fleas on dogs until day +35. The statistical comparisons of the mean living fleas between control I group and treated I one showed significant differences (p≤0.05) for the challenges on all experimental days after treatment. Control II group and treated II one also showed significant differences (p≤0.05) for the challenges on all experimental days after treatment. Statistical analysis between treated group I and II showed no significant difference (p≥0.05) between the challenges on all experimental days. The challenges were finished on day 42, when the effectiveness of fipronil in treated groups I and II were lower than 70%.
