RESUMEN

It is generally accepted that microbes play a critical role in maintaining gut barrier function, making them ideal to target in order to mitigate the effects of intestinal diseases such as inflammatory bowel disease with specialist supplementations such as probiotic or postbiotic preparations. In this study, specific strains of Lactobacillus helvictus both live and inactivated and Lactobacillus plantarum inactivated were fed to zebrafish at an inclusion level of 6 × 106 cells/g in order to assess the effects on gut barrier function and protection. Taken together, our results indicate that dietary administration of pro- or postbiotics strengthens the gut barrier function and innate immunity of healthy zebrafish in a strain-specific and process-dependent way. With some differences in the response intensity, the three treatments led to increased intestinal villi length and proportion of IELs, reinforcement of the GC population and up-regulated expression of biomarkers of AMP production and tight junction zona-occludin 2a (zo-2a). In addition, LPPost had an impact on the adaptive immune response, and we hypothesized that it conferred the potential to drive Th17/ILC3 immunity, as suggested by its effect on the gene expression of il22, of different AMPs, and the expression of zo2a. Moreover, LPPost showed the potential to drive Th1/ILC1-like immunity, with a higher percentage of CD8+ cells and higher ifnγ gene expression. In summary, the use of inactivated Lactobacilli species in this study represented a promising strategy for improving barrier function and regulating the immune fate of the intestinal mucosa in a strain-specific way.

With the rising awareness of antimicrobial resistance, the development and use of functional feed additives (FFAs) as an alternative prophylactic approach to improve animal health and performance is increasing. Although the FFAs from yeasts are widely used in animal and human pharma applications already, the success of future candidates resides in linking their structural functional properties to their efficacy in vivo. Herein, this study aimed to characterise the biochemical and molecular properties of four proprietary yeast cell wall extracts from S. cerevisiae in relation to their potential effect on the intestinal immune responses when given orally. Dietary supplementation of the YCW fractions identified that the α-mannan content was a potent driver of mucus cell and intraepithelial lymphocyte hyperplasia within the intestinal mucosal tissue. Furthermore, the differences in α-mannan and ß-1,3-glucans chain lengths of each YCW fraction affected their capacity to be recognised by different PRRs. As a result, this affected the downstream signalling and shaping of the innate cytokine milieu to elicit the preferential mobilisation of effector T-helper cell subsets namely Th17, Th1, Tr1 and FoxP3+-Tregs. Together these findings demonstrate the importance of characterising the molecular and biochemical properties of YCW fractions when assessing and concluding their immune potential. Additionally, this study offers novel perspectives in the development specific YCW fractions derived from S. cerievisae for use in precision animal feeds.

Cottonseed protein concentrate is a sustainable fishmeal alternative in aquafeed. A 10-week experiment was conducted to investigate the effects of a cottonseed protein concentrate-based diet with and without multi-strain yeast fractions (MsYF) on growth, bile acid metabolism, and health in largemouth bass. Four hundred fish (54.0 ± 0.0 g) were casually distributed into 16 tanks (4 replicates/diet). Fish were fed with 4 iso-nitrogen and iso-energetic diets 3 times daily, including a fishmeal diet (FM), a soy protein concentrate-based diet (SPC; replacing 81% fishmeal protein), a cottonseed protein concentrate-based diet (CPC; replacing 81% fishmeal protein), and a CPC diet supplemented with 800 mg/kg MsYF (CPCY). Results showed that the survival of SPC was the lowest, i.e., 48%, with no apparent diet effect among other treatments; we omitted the SPC in additional analyses. Fish fed cottonseed protein concentrate-based diets showed lower growth than FM (P < 0.05). Fish fed CPC showed the highest nuclear dense hepatic phenotypes ratio (50%), followed by CPCY (33%) and FM (17%). Further, dietary CPC increased hepatic total cholesterol and triglyceride levels with concurrently increased cholesterol synthesis but decreased triglyceride synthesis-associated transcription levels (P < 0.05). Furthermore, dietary CPC increased bile acid synthesis but decreased bile acid transport-associated transcription levels (P < 0.05), and then induced an increment of plasma cholic acid and hepatic chenodeoxycholic acid content and the decrement of genus Romboustia (P < 0.05). Regarding the effect of MsYF, fish fed CPCY reduced hepatic lipid accumulation and total plasma bile acid content (P < 0.05) compared to CPC, suggesting an improvement in liver health. Also, dietary MsYF could reverse the microbiota community structure showing a similar gut microbial composition to FM. In conclusion, 81% of fishmeal protein replaced by cottonseed protein concentrate suppressed growth and liver health, while dietary MsYF might mitigate the negative impact of a high cottonseed protein concentrate level diet on liver functions via gut microbiota regulation.

Probiotics are increasingly documented to confer health and performance benefits across farmed animals. The aim of this study was to assess the effect of a constant daily intake of the single-strain probiotic Pedicococcus acidilactici MA18/5M (4 × 108 CFU.day-1.kg-1 shrimp) fed over fixed, restricted ration sizes (1% to 6% biomass.day-1) on the nutritional performance and metabolism of adult penaeid shrimp Litopenaeus stylirostris (initial body-weight, BWi = 10.9 ± 1.8 g). The probiotic significantly increased the relative daily growth rate (RGR) across all ration size s tested (Mean-RGR of 0.45 ± 0.08 and 0.61 ± 0.07% BWi.day-1 for the control and probiotic groups, respectively) and decreased the maintenance ration (Rm) and the optimal ration (Ropt) by 18.6% and 11.3%, respectively. Accordingly, the probiotic group exhibited a significantly higher gross (K1) and net (K2) feed conversion efficiency with average improvement of 35% and 30%, respectively. Enhanced nutritional performances in shrimps that were fed the probiotic P. acidilactici was associated with, in particular, significantly higher α-amylase specific activity (+24.8 ± 5.5% across ration sizes) and a concentration of free-glucose and glycogen in the digestive gland at fixed ration sizes of 3% and below. This suggests that the probiotic effect might reside in a better use of dietary carbohydrates. Interestingly, P. acidilactici intake was also associated with a statistically enhanced total antioxidant status of the digestive gland and haemolymph (+24.4 ± 7.8% and +21.9 ± 8.5%, respectively; p < 0.05). As supported by knowledge in other species, enhanced carbohydrate utilization as a result of P. acidilactici intake may fuel the pentose-phosphate pathway, generating NADPH or directly enhancing OH-radicals scavenging by free glucose, in turn resulting in a decreased level of cellular oxidative stress. In conclusion, the growth-ration method documented a clear contribution of P. acidilactici MA18/5M on growth and feed efficiency of on-growing L. stylirostris that were fed fixed restricted rations under ideal laboratory conditions. The effect of the probiotic on α-amylase activity and carbohydrate metabolism and its link to the shrimp's antioxidant status raises interesting prospects to optimize dietary formulations and helping to sustain the biological and economic efficiency of the penaeid shrimp-farming industry.

Identifying vital nodes in networks exhibiting a community structure is a fundamental issue. Indeed, community structure is one of the main properties of real-world networks. Recent works have shown that community-aware centrality measures compare favorably with classical measures agnostic about this ubiquitous property. Nonetheless, there is no clear consensus about how they relate and in which situation it is better to use a classical or a community-aware centrality measure. To this end, in this paper, we perform an extensive investigation to get a better understanding of the relationship between classical and community-aware centrality measures reported in the literature. Experiments use artificial networks with controlled community structure properties and a large sample of real-world networks originating from various domains. Results indicate that the stronger the community structure, the more appropriate the community-aware centrality measures. Furthermore, variations of the degree and community size distribution parameters do not affect the results. Finally, network transitivity and community structure strength are the most significant drivers controlling the interactions between classical and community-aware centrality measures.

This study was conducted to evaluate the mucosal immune responses of rainbow trout when supplementing an experimental formulated feed with multi-strain yeast fraction product (Saccharomyces cerevisiae and Cyberlindnera jardinii). In total, 360 fish (initial BW 23.1 ± 0.2 g) were randomly allotted into three dietary treatments in an 8-week feeding trial. The dietary treatments included basal diet (control) and control + 1.5 g/kg multi-strain yeast fraction product (MsYF) fed continuously and pulsed every two weeks between control and MsYF diet. No negative effects on growth performance of feeding the MsYF supplemented diet were observed. SGR and FCR averaged 2.30 ± 0.03%/day and 1.03 ± 0.03, respectively, across experimental groups. Muscularis thickness in the anterior intestine after 8 weeks of feeding was significantly elevated by 44.3% in fish fed the MsYF continuously, and by 14.4% in fish fed the MsYF pulsed (P < 0.02). Significant elevations in goblet cell density in the anterior and posterior (>50% increase) intestine were observed after 8 weeks of feeding the MsYF supplemented diet (P< 0.03). In contrast, lamina propria width was significantly lower in fish fed the experimental diets (>10% reduction). The gene expression analysis of the intestine revealed significant elevations in expression of tlr2, il1r1, irak4, and tollip2 after 4 weeks of feeding the MsYF. Significant elevations in effector cytokines tnfα, il10 and tgfß were observed after 4 weeks of feeding the MsYF regime. After 8 weeks significant elevations in the gene expression levels of il1ß, ifnγ, and il12 were observed in fish fed the MsYF. Likewise, the expression of the transcription factor gata3 was significantly elevated (P<0.01). Supplementation of the multi-strain yeast fraction product positively modulates the intestinal mucosal response of rainbow trout through interaction with toll-like receptor two signalling pathway and potential for increased capacity of delivery of antigens to the underlying mucosal associated lymphoid tissue.

This trial tested the use of lactic acid bacteria (LAB) on pikeperch (Sander lucioperca) larvae during their first feeding. The trial included the use of two probiotic treatments and one control (no probiotics). Pikeperch larvae were exposed to LAB as follows: (1) the live feed (Treatment 1, live feed) or (2) via the live feed and the larval rearing water (Treatment 2, probiotic). Significant differences were found between the treatments in terms of total length (TL), myomere height (MH), overall survival, and the tolerance to a high salinity challenge. Larvae exposed to LAB via both the live feed and the rearing water had a significantly higher overall survival rate (85%) than the other two treatments at 21 dph. When both treatments were subjected to high salinity rates (18 parts per thousand (ppt)), both treatments exposed to LAB demonstrated higher survival rates than the control treatment (28% and 40% survival rate at 180 min for the live feed and probiotic treatments, respectively, as compared with a 100% mortality rate at 150 min for the control). At the same time, larvae exposed to the probiotic treatment had a significantly higher TL as compared to the control after 12 and 21 days post hatch (dph) (probiotic 7.13 ± 0.21 and 11.71 ± 1.1 mm, control 5.86 and 10.79 mm at 12 and 21 dph, respectively). The results suggest that the use of LAB in both the live feed and the rearing water has a positive effect on pikeperch larval quality by strengthening their resilience to stress conditions, as well as improving the growth and survival rates.

Biomedical information systems (BIS) require consideration of three types of variability: data variability induced by new high throughput technologies, schema or model variability induced by large scale studies or new fields of research, and knowledge variability resulting from new discoveries. Beyond data heterogeneity, managing variabilities in the context of BIS requires extensible and dynamic integration process. In this paper, we focus on data and schema variabilities and we propose an integration framework based on ontologies, master data, and semantic annotations. The framework addresses issues related to: 1) collaborative work through a dynamic integration process; 2) variability among studies using an annotation mechanism; and 3) quality control over data and semantic annotations. Our approach relies on two levels of knowledge: BIS-related knowledge is modeled using an application ontology coupled with UML models that allow controlling data completeness and consistency, and domain knowledge is described by a domain ontology, which ensures data coherence. A system build with the eClims framework has been implemented and evaluated in the context of a proteomic platform.

Influence in Twitter has become recently a hot research topic, since this micro-blogging service is widely used to share and disseminate information. Some users are more able than others to influence and persuade peers. Thus, studying most influential users leads to reach a large-scale information diffusion area, something very useful in marketing or political campaigns. In this study, we propose a new approach for multi-level influence assessment on multi-relational networks, such as Twitter. We define a social graph to model the relationships between users as a multiplex graph where users are represented by nodes, and links model the different relations between them (e.g., retweets, mentions, and replies). We explore how relations between nodes in this graph could reveal about the influence degree and propose a generic computational model to assess influence degree of a certain node. This is based on the conjunctive combination rule from the belief functions theory to combine different types of relations. We experiment the proposed method on a large amount of data gathered from Twitter during the European Elections 2014 and deduce top influential candidates. The results show that our model is flexible enough to to consider multiple interactions combination according to social scientists needs or requirements and that the numerical results of the belief theory are accurate. We also evaluate the approach over the CLEF RepLab 2014 data set and show that our approach leads to quite interesting results.

BACKGROUND: Cleaner-fish (wrasse, Labridae) are increasingly deployed within the Atlantic salmon (Salmo salar L.) industry as a biological control against sea-lice (Lepeophtheirus salmonis Krøyer). Two tank-based trials were performed to test the effect of farmed ballan wrasse (Labrus bergylta Ascanius) body mass and supplementary feeding on the delousing of Atlantic salmon post-smolts with an initial infection level of ∼12 lice salmon(-1) and a ∼5% wrasse:salmon ratio. RESULTS: Sea-louse levels below 0.5 lice salmon(-1) were obtained within 84 h, and preferential preying upon larger motile stages was found. The wrasse body mass and the availability of fresh, opened blue mussels (Mytilus edulis L.) did not significantly affect delousing efficiency. The functional predator response was linear, showing no minimum prey density threshold for sea-louse foraging and no satiation plateau, in spite of the high consumption rates measured. Sea-louse infection levels declined following a one-phase exponential decay model, with a standardised decline time constant of 0.8-1.3% h(-1) for each wrasse stocked per 100 salmon. CONCLUSIONS: Farmed ballan wrasse are confirmed as highly effective therapeutic and preventive biological controls against sea-lice. The study supports the current minimum hatchery size target (10 mm total length) and the use of supplementary feeding to sustain the wrasse stocks in operation. The functional predator response and the standardised decline time constant of sea-louse abundance are proposed as useful indicators of delousing efficiency.

BACKGROUND: Infections of the ciliate protozoan Ichthyophthirius multifiliis Fouquet, 1876, can cause heavy mortalities in freshwater aquaculture systems. Following the ban of malachite green on fish for human consumption, currently available chemical therapies are only partially efficacious even when repeatedly applied. There is, therefore, an urgent necessity to identify more efficient and environmentally friendly control strategies. In the present study, the use of a substrate algae feeder leopard pleco, Glyptoperichthys gibbiceps, as a biological agent to control I. multifiliis infections on blue tilapia, Oreochromis aureus, reared in a warm-water, experimental tank-based system is explored. RESULTS: The free-swimming protomont stage of I. multifiliis demonstrated selective settlement behaviour towards biofilm-covered substrates. Following a controlled exposure, the number of I. multifiliis trophonts establishing on naive blue tilapia, O. aureus, was 4.9 ± 0.4 when reared with G. gibbiceps, significantly lower than when reared in isolation (13.3 ± 0.9; ANOVA, P < 0.001). CONCLUSION: This study demonstrates for the first time the potential of G. gibbiceps as a biological control to reduce I. multifiliis on commercially valuable fish stocks in warm-water systems. This likely originated from the grazing activity of G. gibbiceps on the biofilm layer upon which the multiplicative stage of I. multifiliis was shown preferentially to settle. Therapeutic strategies against pathogenic protozoan species like I. multifiliis could greatly benefit from incorporating, where appropriate, a biological control targeting the external multiplicative life stages of the parasite, potentially reducing the current dependence on chemical interventions.

The external coloration of fish is a key driver in consumer buying decisions and is typically altered during sexual maturation in salmonids. Farmed Atlantic salmon ( Salmo salar L.) exhibiting distinct phenotypes from the typical silver and nuptial coloration were described in terms of sexual development, flesh quality, and skin pigment profiles. Reconditioning of skin coloration during storage was also tested (CIE[1976]L*a*b*) with the overall view to optimize quality management. The intermediary phenotype never reflected significant deteriorations of flesh quality. It originated from a lack of purine pigments (guanine and hypoxanthine), revealing the carotenoid compounds dominated by the yellow-orange beta-carotene. The resulting distinctive lightness and yellowness were reduced by direct ice contact at a post-mortem stage. Storage conditions can be optimized to improve and standardize the coloration of whole-fish, yielding superior flesh quality parameters. This would facilitate product quality grading during primary processing and also increase product acceptance and attractiveness.
