RESUMEN

BACKGROUND: Ginsenoside Rg3 is a component of ginseng that protects against myocardial ischemia/reperfusion (MI/R) injury. Ferroptosis is a new form of cell death characterized by oxidative damage to phospholipids. The purpose of this study was to examine the role and of ginsenoside Rg3 in MI/R and the mechanism. METHODS: A mouse model of left anterior descending (LAD) ligation-induced myocardial ischemia/reperfusion (MI/R) injury and oxygen-glucose deprivation/reperfusion (OGD/R) were used as in vitro and in vivo models, respectively. Echocardiographic analysis, 2,3,5-triphenyltetrazolium chloride (TTC) staining and hematoxylin-eosin (H&E) staining were used to assess the cardioprotective effects of ginsenoside Rg3. Western blotting, biochemical analysis, small interfering RNA analysis and molecular docking were performed to examine the underlying mechanism. RESULTS: Ginsenoside Rg3 improved cardiac function and infarct size in mice with MI/R injury. Moreover, ginsenoside Rg3 increased the expression of the ferroptosis-related protein GPX4 and inhibited iron deposition in mice with MI/R injury. Ginsenoside Rg3 also activated the Nrf2 signaling pathway. Ginsenoside Rg3 attenuated myocardial ischemia/reperfusion-induced ferroptosis via the Nrf2 signaling pathway. Notably, ginsenoside Rg3 regulated the keap1/Nrf2 signaling pathway to attenuate OGD/R-induced ferroptosis in H9C2 cells. Taken together, ginsenoside Rg3 attenuated myocardial ischemia/reperfusion-induced ferroptosis via the keap1/Nrf2/GPX4 signaling pathway. CONCLUSIONS: Our findings demonstrated that ginsenoside Rg3 ameliorate MI/R-induced ferroptosis via the keap1/Nrf2/GPX4 signaling pathway.

We introduce a tempering approach with stochastic density functional theory (sDFT), labeled t-sDFT, which reduces the statistical errors in the estimates of observable expectation values. This is achieved by rewriting the electronic density as a sum of a "warm" component complemented by "colder" correction(s). Since the warm component is larger in magnitude but faster to evaluate, we use many more stochastic orbitals for its evaluation than for the smaller-sized colder correction(s). This results in a significant reduction in the statistical fluctuations and systematic deviation compared to sDFT for the same computational effort. We demonstrate the method's performance on large hydrogen-passivated silicon nanocrystals, finding a reduction in the systematic deviation in the energy by more than an order of magnitude, while the systematic deviation in the forces is also quenched. Similarly, the statistical fluctuations are reduced by factors of ≈4-5 for the total energy and ≈1.5-2 for the forces on the atoms. Since the embedding in t-sDFT is fully stochastic, it is possible to combine t-sDFT with other variants of sDFT such as energy-window sDFT and embedded-fragmented sDFT.

Enoyl-CoA hydratase and 3-hydroxyacyl CoA dehydrogenase (EHHADH), a member of the 3-hydroxyacyl-CoA dehydrogenase family, were previously demonstrated to be involved in the tumorigenesis of various cancer types. This study is aimed at determining of the diagnostic and prognostic value of EHHADH in osteosarcoma (OS). The overexpression of EHHADH was found both in OS and also other sarcoma types, and according to the retrospective cohort study, the EHHADH level was related to the overall survival and disease-free survival of the OS patients. Furthermore, knockdown of EHHADH under the influence of EHHADH small interfering RNA significantly suppressed the proliferation ability of the tumor cells. Moreover, EHHADH overexpressed was found in human OS tissues. In summary, the progression of OS could be enhanced by EHHADH, which may be a potential diagnostic and prognostic biomarker for OS patients.

Camera calibration is essential for various vision-based 3D metrological techniques. In this paper, a novel camera calibration method, to the best of our knowledge, combining synthetic speckle pattern and an improved gray wolf optimizer algorithm is presented. The synthetic speckle pattern serves as the calibration target. The particle swarm algorithm-based digital image correlation is employed to achieve matches among 3D control points and 2D image points; then the improved gray wolf optimizer algorithm is used to calculate the camera parameters. For verification, simulated and real tests are conducted. Through the analysis of calibration results, the proposed method performs better and is more stable than other calibration targets. Research on the influence of camera pose and optimization algorithm is conducted, showing that the improved gray wolf optimizer algorithm performs better than other benchmark algorithms. The camera parameters can be obtained through one captured image when the speckle patterns are added in the portion of the camera sensor.

Considering the multi-sources, heterogeneity and complexity of dam safety assessment, a dam safety assessment model based on interval-valued intuitionistic fuzzy set and evidence theory is proposed to perform dam safety reliability evaluations. In the proposed model, the dynamic reliability based on the supporting degree is applied to modify the data from homologous information. The interval-valued intuitionistic fuzzy set is used to describing the uncertainty and fuzziness between heterogeneous information. Evidence theory is employed to integrate the data from heterogeneous information. Finally, a multiple-arch dam undergoing structural reinforcement is taken as an example. The evaluation result before reinforcement shows that the safety degree of the dam is low and the potential risk is more likely to be located at the dam section #13. From the geological survey before reinforcement, there exist weak fracture zone and broken mud belt in the foundation of the dam section #13. The comparison between the evaluation results before and after reinforcement indicates that the dam become safer and more stable after reinforcement.
