RESUMEN

We report on cyclotron frequency measurements on trapped 206,207Pb+ ions by means of the non-destructive Fourier-transform ion-cyclotron-resonance technique at room temperature. In a proof-of-principle experiment using a quartz crystal instead of a coil as a resonator, we have alternately carried out cyclotron frequency measurements for 206Pb+ and 207Pb+ with the sideband coupling method to obtain 21 cyclotron-frequency ratios with a statistical uncertainty of 6 × 10-7. The mean frequency ratio R¯ deviates by about 2σ from the value deduced from the masses reported in the latest Atomic Mass Evaluation. We anticipate that this shift is due to the ion-ion interaction between the simultaneously trapped ions (≈100) and will decrease to a negligible level once we reach single-ion sensitivity. The compactness of such a crystal makes this approach promising for direct Penning-trap mass measurements on heavy and superheavy elements.

Single-ion sensitivity is obtained in precision Penning-trap experiments devoted to light (anti)particles or ions with low mass-to-charge ratios, by adding an inductance coil to an amplifier connected to the trap, both operated at 4 K. However, single-ion sensitivity has not been reached on heavy singly or doubly charged ions. In this publication, we present a new system to reach this point, based on the use of a quartz crystal as an inductance, together with a newly developed broad-band (BB) amplifier. We detect the reduced-cyclotron frequency of 40Ca+ ions stored in a 7-tesla open-ring Penning trap. By comparing the detected electric signal obtained with the BB amplifier and the fluorescence signal obtained by collecting the photons emitted by a trapped ion cloud, we show a detection limit below 110 ions. Adding the crystal, the electrical signal increases by a factor of about 30 at room temperature, which combined with the measured equivalent resistance and voltage noise, proves the feasibility of the system to reach single-ion sensitivity at 4 K.

Natural IgA antibodies are abundantly produced in vivo to protect serosal surfaces from invading infectious organisms. However, the immunotherapeutic potential of IgA has hardly been explored, although there is evidence that recombinant IgA antibodies may broaden the armentarium to combat certain infectious or malignant diseases. One of the limitations for exploring IgA's therapeutic activity has been the difficulty to obtain enough recombinant material with desired specificity for in vivo studies. Here, we describe the production and purification of monomeric recombinant IgA1 and IgA2 antibodies under serum-free conditions. For antibody production, suspension adapted CHO-K1 cells and a glutamine synthetase selection vector were used, which resulted in specific production rates of up to 2.2 pg/cell/day. Purities of >95% of monomeric antibodies were obtained by a combination of affinity chromatography-using an anti-kappa-light chain matrix-and size exclusion chromatography. Purified antibodies displayed the expected biochemical characteristics and were functionally fully active. Importantly, all required reagents and methods are commercially available and not dependent on the specificity of the desired antibody. In addition, all employed technologies and methodologies are similar to those used for the production of therapeutic IgG antibodies - thus allowing further up-scaling and streamlining according to existing antibody production technologies. In conclusion, the described methodology may assist in the development of recombinant IgA antibodies for therapeutic applications.

Buerger's disease (BD) is an entity with distinct clinical and pathologic aspects, including etiologic factors, pathogenesis, histology, clinical presentation, course, and prognosis. Comprehensive angiographic studies support the definition of the disease's evolution, provide information about distribution patterns (i.e., bilateral, upper and lower extremities, involvement of trunk arteries), aid in choosing "hot spots" for biopsies, and determine preoperative technical considerations in selection of bypass surgery candidates. In a series of 44 patients with BD, we endeavored to record the frequency of distribution and progression of various stages of disease as well as document the angiographic signs that are commonly accepted as typical of or specific to BD. Characteristic angiographic patterns of the disease are: corkscrew-shaped collateral vessels, cutoff type vascular occlusions, vessels with smooth-lined lumina in non-involved arteries and discontinuous, segmental involvement of affected arteries and veins. Results of pharmacoangiographic functional studies of circulatory disorders appear to cast doubt on the supposed specificity of phenomena such as standing-wave pattern and vasospasm in BD.
