RESUMEN

Several MYB transcription factors are known to play important roles in plant resistance to environmental stressors. However, the mechanism governing the involvement of MYBs in regulating tobacco mosaic virus (TMV) resistance in plants is still unclear. In this study, we found that not only is Nicotiana benthamiana MYB4-like involved in defence against TMV, but also that the ethylene pathway participates in MYB4L-mediated resistance. Transcription of NbMYB4L was up-regulated in N. benthamiana infected with TMV. Silencing of NbMYB4L led to intensified TMV replication, whereas overexpression of NbMYB4L induced significant resistance to TMV. Transcription of NbMYB4L was greater in 1-aminocyclopropanecarboxylic acid (ACC, ethylene precursor)-pretreated plants but lower when the ethylene signalling pathway was blocked during TMV infection. Gene expression analysis showed that the transcription of NbMYB4L was largely suppressed in ETHYLENE INSENSITIVE 3-like 1(EIL1)-silenced plants. The results of electrophoretic mobility shift assay and chromatin immunoprecipitation-quantitative PCR (ChIP-qPCR) experiments indicated that NbEIL1 could directly bind to two specific regions of the NbMYB4L promoter. Furthermore, a luciferase assay revealed that NbEIL1 significantly induced the reporter activity of the MYB4L promoter in N. benthamiana. These results point to NbEIL1 functioning as a positive regulator of NbMYB4L transcription in N. benthamiana against TMV. Collectively, our work reveals that EIL1 and MYB4L constitute a coherent feed-forward loop involved in the robust regulation of resistance to TMV in N. benthamiana.

PURPOSE: To explore the effect of enamel matrix proteins(EMPs) on osteogenesis and adipogenesis of stem cells from human exfoliated deciduous teeth SHED), and explore its molecular mechanism. METHODS: SHEDs were used to detect the expression of its surface antigens CD73, CD146, CD34 and CD45 by flow cytometry. SHED was induced by OB osteogenic induction liquid, and then the osteogenic differentiation ability was measured by alizarin red staining. SHEDs were divided into 4 groups, NC group had invalid sequence shRNA interfered with SHED, EMPs group had invalid sequence shRNA interfered with SHED. Then 100 µg/L EMPs was used to interfere with SHED. In miR-32 inhibitor group, miR-32 shRNA plasmid was used to interfere with SHED; while in EMPs+miR-32 inhibitor group, 100 µg/L EMP was used to intervene SHED after silencing miR-32. QPCR was used to detect the expression of miR-32, dentin sialophosphoprotein (DSPP), dentin matrix protein 1, DMP-1, peroxisome proliferators-activated receptor γ (PPARγ) and CCAAT enhancer binding protein α (C/EBPα) gene expression; Western blot was used to detect the expression of DSPP, DMP-1, PPARγ and C/EBPα protein expression; Alizarin red staining was used to detect SHED osteogenic capacity; Oil red O staining was used to detect adipogenetic capacity of SHED. RESULTS: The results of flow cytometry showed that SHED had positive expression of CD146 and CD73, and negative expression of CD34 and CD45, which was consistent with the characteristics of stem cell surface markers. Alizarin red staining and oil red O staining showed mineralized nodules and oil droplets increased significantly, consistent with the multi-directional differentiation characteristics of stem cells. Compared with NC group, the expression of miR-32 gene in EMPs group was significantly increased(P<0.05), and the expression of miR-32 in miR-32 inhibitor group and EMPs+miR-32 inhibitor group was significantly decreased(P<0.05). Compared with NC group, the expression of DSPP and DMP-1, the number of mineralized nodules in EMPs group were significantly increased(P<0.05), the expression of PPARγ and C/EBPa and the number of lipid droplets were significantly decreased (P<0.05), while the result of miR-32 inhibitor group was the opposite (P<0.05). Compared with miR-32 inhibitor group, there was no significant difference in the expression of DSPP, DMP-1, PPARγ and C/EBPα, number of mineralized nodules and oil droplets in EMPs+miR-32 inhibitor group(P>0.05). Compared with EMPs group, the expression of DSPP and DMP-1 and the number of mineralized nodules in EMPs+miR-32 inhibitor group were significantly reduced(P<0.05), while the expression of PPARγ and C/EBPα and the number of lipid droplets were significantly increased(P<0.05). CONCLUSIONS: EMPs can regulate osteogenic and adipogenic differentiation of SHED by promoting the expression of miR-32.

RESUMEN

PURPOSE: To study the effect of Kangfuxin liquid combined with Garlicin Capsules in treatment of children with recurrent oral ulcer (ROU) and on immunological regulation. METHODS: This randomized controlled clinical study prospectively enrolled 204 patients with ROU who were randomly divided into 2 groups. Patients in group A received Garlicin Capsules 1/time, 3 times/d, combined with Kangfuxin liquid 10 mL to gargle 3 times/d; patients in group B only received Kangfuxin liquid 10 mL gargle 5 min, 3/d. The treatment lasted for 2 weeks. The curative effect was compared before and after treatment, including ulcer surface pain (VAS score), time of ulcer healing, and the changes of T cell subsets (CD3+, CD4+, CD8+ and CD4+/CD8+) before and after treatment were compared by SPSS 22 software package. RESULTS: Ulcer healing time (3.5±0.6) d, duration of pain (3.1±0.3)d in group A were shorter than in group B (P<0.05); treatment efficiency was 96.08% in group A, 88.24% in group B( χ2=6.264, P<0.05). The pain scores of both groups were significantly reduced after treatment, and the difference was significant between group A and group B [(1.1±0.4) vs (3.2± 0.6)] (P<0.05). The levels of CD3+, CD4+, CD4+/CD8+ in group A were significantly higher than those in group A after treatment (P<0.05), the levels of CD8+ was significant lower than in group B (P<0.05). CONSLUSIONS: Kangfuxin liquid combined with Garlicin Capsules can improve the therapeutic effect of ROU and repair of local damaged mucosa in children, increasing the immune function of children.

RESUMEN

Two new copper(II) complexes Cu(NCS)2(4-Bzpy)2 (1) and Cu(NO3)2(4-Bzpy)4 (2) (4-Bzpy=4-benzoylpyridine) have been synthesized and characterized by IR, UV, elemental analysis and X-ray crystallography. Cu(II) atom has a square planar environment for 1 and an distorted octahedral environment for 2, respectively. In solid state there are C-H⋯π interactions and C-H⋯S hydrogen bonds between adjacent molecules in complex 1. The molecule of complex 2 is further connected by multiform π-π interactions, C-H⋯π interactions and C-H⋯O hydrogen bonds to form a three-dimensional supramolecular structure. The luminescent properties of the complexes 1 and 2 were both investigated in H2O solution and in solid state at room temperature, respectively.

RESUMEN

A new inorganic-organic hybrid compound [FTMA](2)[Co(NCS)(4)] (FTMA = ferrocenylmethyltrimethylammonium cation) has been synthesized and characterized by IR, UV, elemental analysis and X-ray crystallography. Co(II) atom has a distorted tetrahedral environment with four N atoms of four NCS(-) anions. In the solid state there are C-H⋯π interactions between adjacent ferrocenyl cations, which generate one-dimensional (1-D) supramolecular chain, and C-H⋯S hydrogen bonds between [FTMA](+) cations and cobalt thiocyanate anions. The title compound shows strong purple fluorescence emission in the solid state at room temperature.

Rational self-assembly of Sb(2)O(3) and Na(2)WO(4), or (NH(4))(18)[NaSb(9)W(21)O(86)] with transition-metal ions (Mn(2+), Cu(2+), Co(2+)), in aqueous solution under controlled conditions yield a series of sandwich type complexes, namely, Na(2)H(2)[Mn(2.5)W(1.5)(H(2)O)(8)(B-beta-SbW(9)O(33))(2)]32 H(2)O (1), Na(4)H(7)[Na(3)(H(2)O)(6)Mn(3)(mu-OAc)(2)(B-alpha-SbW(9)O(33))(2)]20 H(2)O (OAc=acetate anion) (2), NaH(8)[Na(2)Cu(4)Cl(B-alpha-SbW(9)O(33))(2)]21 H(2)O (3), Na(8)K[Na(2)K(H(2)O)(2){Co(H(2)O)}(3)(B-alpha-SbW(9)O(33))(2)] 10 H(2)O (4), and Na(5)H[{Co(H(2)O)(2)}(3)W(H(2)O)(2)(B-beta-SbW(9)O(33))(2)]11.5 H(2)O (5). These structures are determined by using the X-ray diffraction technique and further characterized by obtaining IR spectra and performing elemental analysis. Structure analysis reveals that polyoxoanions in 1 and 5 comprise of two [B-beta-SbW(9)O(33)](9-) building units, whereas 2, 3, and 4 consist of two isomerous [B-alpha-SbW(9)O(33)](9-) building blocks, which are all linked by different transition-metal ions (Mn(2+), Cu(2+), or Co(2+)) with different quantitative nuclearity. It should be noted that compound 2 represents the first one-dimensional sinusoidal chain based on sandwich like tungstoantimonate building blocks through the carboxylate-bridging ligands. Additionally, 3 is constructed from sandwiched anions [Na(2)Cu(4)Cl(B-alpha-SbW(9)O(33))(2)](9-) linked to each other to form an infinitely extended 2D network, whereas 5 shows an interesting 3D framework built up from offset sandwich type polyoxoanion [{Co(H(2)O)(2)}(3)W(H(2)O)(2)(B-beta-SbW(9)O(33))(2)](6-) linked by Co(2+) and Na(+) ions. EPR studies performed at 110 K and room temperature reveal that the metal cations (Mn(2+), Cu(2+), Co(2+)) reside in a square-pyramidal geometry in 2, 3, and 4. The magnetic behavior of 1-4 suggests the presence of weak antiferromagnetic coupling interactions between magnetic metal centers with the exchange integral J=-0.552 cm(-1) in 2.

The title complex, [Co(CO(3))(C(10)H(8)N(2))(2)]Br·3H(2)O, is isostructural with the chloride analogue. The six-coordinated octahedral [Co(2,2'-bipy)(2)CO(3)](+) cation (2,2'-bipy is 2,2'-bipyrid-yl), bromide ion and water mol-ecules are linked together via O-H⋯Br and O-H⋯O hydrogen bonds, generating a one-dimensional chain.

The title compound, bis[tris(2,2'-bipyridine)iron(II)] tetraaquatetra-mu4-oxo-pentacosa-mu2-oxo-undecaoxoiron(III)sodium(I)dodecatungsten(VI) dihydrate, [Fe(C10H8N2)3]2[NaFeW12O40(H2O)4].2H2O, consists of a dodecatungstoferrate(III) framework grafted on to an [Na(H2O)4]+ cation, two complex [Fe(2,2'-bipy)3]2+ cations (2,2'-bipy is 2,2'-bipyridine) and two uncoordinated water molecules per formula unit.
