RESUMEN

We recently reported that restoring the CYP27A1-27hydroxycholesterol axis had antitumor properties. Thus, we sought to determine the mechanism by which 27HC exerts its anti-prostate cancer effects. As cholesterol is a major component of membrane microdomains known as lipid rafts, which localize receptors and facilitate cellular signaling, we hypothesized 27HC would impair lipid rafts, using the IL6-JAK-STAT3 axis as a model given its prominent role in prostate cancer. As revealed by single molecule imaging of DU145 prostate cancer cells, 27HC treatment significantly reduced detected cholesterol density on the plasma membranes. Further, 27HC treatment of constitutively active STAT3 DU145 prostate cancer cells reduced STAT3 activation and slowed tumor growth in vitro and in vivo. 27HC also blocked IL6-mediated STAT3 phosphorylation in nonconstitutively active STAT3 cells. Mechanistically, 27HC reduced STAT3 homodimerization, nuclear translocation, and decreased STAT3 DNA occupancy at target gene promoters. Combined treatment with 27HC and STAT3 targeting molecules had additive and synergistic effects on proliferation and migration, respectively. Hallmark IL6-JAK-STAT gene signatures positively correlated with CYP27A1 gene expression in a large set of human metastatic castrate-resistant prostate cancers and in an aggressive prostate cancer subtype. This suggests STAT3 activation may be a resistance mechanism for aggressive prostate cancers that retain CYP27A1 expression. In summary, our study establishes a key mechanism by which 27HC inhibits prostate cancer by disrupting lipid rafts and blocking STAT3 activation. IMPLICATIONS: Collectively, these data show that modulation of intracellular cholesterol by 27HC can inhibit IL6-JAK-STAT signaling and may synergize with STAT3-targeted compounds.

The atrophy of skeletal muscles in patients with Krabbe disease is a major debilitating manifestation that worsens their quality of life and limits the clinical efficacy of current therapies. The pathogenic mechanism triggering muscle wasting is unknown. This study examined structural, functional, and metabolic changes conducive to muscle degeneration in Krabbe disease using the murine (twitcher mouse) and canine [globoid cell leukodystrophy (GLD) dog] models. Muscle degeneration, denervation, neuromuscular [neuromuscular junction (NMJ)] abnormalities, and axonal death were investigated using the reporter transgenic twitcher-Thy1.1-yellow fluorescent protein mouse. We found that mutant muscles had significant numbers of smaller-sized muscle fibers, without signs of regeneration. Muscle growth was slow and weak in twitcher mice, with decreased maximum force. The NMJ had significant levels of activated caspase-3 but limited denervation. Mutant NMJ showed reduced surface areas and lower volumes of presynaptic terminals, with depressed nerve control, increased miniature endplate potential (MEPP) amplitude, decreased MEPP frequency, and increased rise and decay rate constants. Twitcher and GLD dog muscles had significant capacity to store psychosine, the neurotoxin that accumulates in Krabbe disease. Mechanistically, muscle defects involved the inactivation of the Akt pathway and activation of the proteasome pathway. Our work indicates that muscular dysfunction in Krabbe disease is compounded by a pathogenic mechanism involving at least the failure of NMJ function, activation of proteosome degradation, and a reduction of the Akt pathway. Akt, which is key for muscle function, may constitute a novel target to complement in therapies for Krabbe disease.

Despite lacking jaws and substantial rigid support for feeding muscles, hagfishes can forcefully grasp and ingest chunks of flesh from their prey. When feeding, bilaterally folding dental plates protrude from the mouth, then forcefully retract. This cyclic protraction and retraction occurs in the anterior region of the hagfish feeding apparatus (HFA) and involves both a cartilaginous skeleton and a complex array of muscles that act as a hydrostat. We recorded motor patterns from the largest muscles in the HFA in six specimens of Myxine glutinosa: the deep protractor muscle (DPM), clavatus muscle (CM), perpendicularis muscle (PM), and tubulatus muscle (TM). Individuals normally used four gape cycles to ingest food and four gape cycles to intraorally transport food. We measured burst duration from each muscle and the onsets of kinematic events and the onsets of CM, PM, and TM bursts relative to the onset of the DPM. The DPM fired during protraction, while the CM, PM and TM fired during retraction. Our study corroborates our anatomical predictions about DPM and CM function. Activation of the circumferentially and vertically oriented fibers of the TM and PM stiffens the origin of the CM, allowing it to forcefully retract the dental plates. The progressive decrease in retractor muscle activity during gape cycles following ingestion suggests a reliance on passive properties of the musculoskeletal system for retraction.

Abstract: A total of 175 spotted snapper Lutjanus guttatus were collected monthly in the Los Cobanos and Puerto La Libertad, El Salvador, from January to December 2000 to determine its feeding habits. The fishes studied ranged 9.8 - 58.0 cm in total length. Were collected using push-net and hook and line. This snapper is a bentonic opportunistic carnivorous predator. The total biomass of the stomach contents was 260.5 g. Crustaceans (Squillidae, Portunidae, Dynomenidae. Penaeidae, Sicyoniidae, Callianassidae), were the most abundant group: they accounted for 50.4% of the total biomass. Numerically, Portunus asper was the most abundant prey. Ontogenic differences were observed in the diet. In juveniles (16 cm TL). at any time of the year, the most frequent and abundant components were crustaceans and in adults were crustaceans, fishes and mollusks. The relative importance of different components of the diet was assessed with two indexes that combine. in different ways. percentage frequency of occurrence, percentage number and percentage weight of prey categories. The commercial use this resource and the absence of management strategies are discussed.
