Variability in sea ice conditions, combined with strong couplings to the atmosphere and the ocean, lead to a broad range of complex sea ice dynamics. More in-situ measurements are needed to better identify the phenomena and mechanisms that govern sea ice growth, drift, and breakup. To this end, we have gathered a dataset of in-situ observations of sea ice drift and waves in ice. A total of 15 deployments were performed over a period of 5 years in both the Arctic and Antarctic, involving 72 instruments. These provide both GPS drift tracks, and measurements of waves in ice. The data can, in turn, be used for tuning sea ice drift models, investigating waves damping by sea ice, and helping calibrate other sea ice measurement techniques, such as satellite based observations.

Records of the ice pressures on the joggle skirt of the coal quay in Spitsbergen were performed over winter seasons in 2013 and 2015. The ice thickness below the quay was above 2 m. Ice temperature over the ice thickness and water pressure at the sea bed were measured synchronously. It was discovered that sea water migrates through the ice confined inside the joggle skirt under the influence of tidal changes of the water pressure below the ice. It leads to the formation of floods on the ice surface during high tide. The ice surface becomes dry during low tide. Ice temperature and ice pressure on the joggle skirt are changed according to the semidiurnal cycle. Spectral analysis and correlation analysis are performed to analyse the loads caused by thermal expansion of the ice. A thermo-mechanical model of ice based on elastic-plastic rheology with thermal effect is used in numerical simulations of the observed phenomena. Numerical simulations are performed with finite element software.This article is part of the theme issue 'Modelling of sea-ice phenomena'.

A model of the thermo-elastic behaviour of saline ice is formulated, and model solutions describing thermo-elastic waves (TEW) propagating into a half-space of the ice are investigated. The model is based on a proposal that saline ice is a matrix, which encompasses both closed brine pockets and permeable channels filled with brine. Experiments on the thermal expansion of saline ice samples, and on TEW in saline ice, have been performed in the cold laboratories of the University Centre in Svalbard and in University College London. The experimental data are compared with theoretical conclusions. The experimental data support our hypothesis that the brine in saline ice is divided between closed pockets and open, permeable channels.This article is part of the themed issue 'Microdynamics of ice'.

A compact Raman lidar system for remote sensing of sea and drifting ice was developed at the Wave Research Center at the Prokhorov General Physics Institute of the Russian Academy of Sciences. The developed system is based on a diode-pumped solid-state YVO(4):Nd laser combined with a compact spectrograph equipped with a gated detector. The system exhibits high sensitivity and can be used for mapping or depth profiling of different parameters within many oceanographic problems. Light weight (∼20 kg) and low power consumption (300 W) make it possible to install the device on any vehicle, including unmanned aircraft or submarine systems. The Raman lidar presented was used for study and analysis of the different influence of the open sea and glaciers on water properties in Svalbard fjords. Temperature, phytoplankton, and dissolved organic matter distributions in the seawater were studied in the Ice Fjord, Van Mijen Fjord, and Rinders Fjord. Drifting ice and seawater in the Rinders Fjord were characterized by the Raman spectroscopy and fluorescence. It was found that the Paula Glacier strongly influences the water temperature and chlorophyll distributions in the Van Mijen Fjord and Rinders Fjord. Possible applications of compact lidar systems for express monitoring of seawater in places with high concentrations of floating ice or near cold streams in the Arctic Ocean are discussed.

Alpha-fetoprotein (AFP) is a biological drug candidate of high medicinal potential in the treatment of autoimmune diseases, cancer, and regenerative medicine. Large-scale production of recombinant human alpha-fetoprotein (rhAFP) is desirable for structural and functional studies and applied research. In this study we cloned and expressed in the secreted form wild-type glycosylated human rhAFP and non-glycosylated mutant rhAFP(0) (N233S) in the yeast strain Saccharomyces cerevisiae with multiple chromosome-integrated synthetic human AFP genes. RhAFP and rhAFP(0) were successfully produced and purified from the culture liquids active naturally folded proteins. Elimination of the glycosylation by mutation reduced rhAFP(0) secretion about threefold as compared to the wild-type protein showing critical role of the N-linked glycan for heterologous protein folding and secretion. Structural similarity of rhAFP and rhAFP(0) with natural embryonic eAFP was confirmed by circular dichroism technique. Functional tests demonstrated similar type of tumor suppressive and immunosuppressive activity for both recombinant species rhAFP and rhAFP(0) as compared to natural eAFP. It was documented that both types of biological activities attributed to rhAFP and rhAFP(0) are due to the fast induction of apoptosis in tumor cells and mitogen-activated lymphocytes. Despite the fact that rhAFP and rhAFP(0) demonstrated slightly less effective tumor suppressive activity as compared to eAFP but rhAFP(0) had produced statistically notable increase in its ability to induce inhibition of in vitro lymphocyte proliferation as compared to the glycosylated rhAFP and eAFP. We conclude that N-linked glycosylation of rhAFP is required for efficient folding and secretion. However the presence of N-linked sugar moiety was shown to be unimportant for tumor suppressive activity but was critically important for its immunoregulative activity which demonstrates that different molecular mechanisms are involved in these two types of biological functional activities attributed to AFP.
