RESUMEN

Male Wistar rats were treadmill-trained for 8 weeks using one of the two regimens: with the constant running speed or with alternating high-speed and low-speed intervals. Both training regimens led to an increase of rat aerobic capacities and to a higher citrate synthase activity in the medial head of gastrocnemius muscle. No differences between the effects of two training regimens were observed. However, in contrast to constant-speed training the interval one resulted in myocardium hypertrophy and also in less pronounced changes in diaphragm muscle, such as slow-direction shift of myosin phenotype and reduction of muscle fiber cross-sectional area. Neither of the training regimens had an effect on corticosterone and thyroid hormones levels in rat blood, whereas the interval training resulted in a higher level of testosterone. Anabolic influence of testosterone during interval aerobic training may be favorable for heart hemodynamic capacity and force characteristics of the diaphragm.

RESUMEN

Influence of corticotropin-releasing factor fragment CRF4-6 on metabolism in the rat was investigated in this study. Tripeptide (10 mkg/rat) after intracerebroventricular infusion to anesthetized animals increased metabolic rate, not affecting the respiratory ratio, induced hyperglycemia and hyperthermia. Synchronously with that heart rate and arterial pressure did also increase. Changes in energy metabolism induced by CRF4-6 closely resemble the effect of untruncated corticotropin-releasing factor molecule. Results of this study and the range of tripeptide effects described previously evidence that CRF4-6 is a physiologically active regulatory molecule, a derivative of corticotropin-releasing factor, that expands and prolongs effects of the parent molecule.

The effects of tripeptide corticoliberin fragment CRF4-6 (Pro-Pro-Ile) on blood glucose level and the rat body temperatire were investigated. Intracerebroventricularly injected CRF4-6 (6, 30, 150 nmol/head) causes a dosedependent hyperglycemia and hyperthermia in anaesthetized animals. Corticotropin releasing factor antagonist alpha-helical CRF4-6 (6.5 nmol/head) abolishes the influence of tripeptide CRF4-6 (6 nmol/head) on blood glucose level and body temperature of rats. Bilateral adrenalectomy has no effect on tripeptide-induced hyperglycemia and hyperthemia. This result indicates that hyperglycemic and hyperthermal effects of tripeptide occur independently of adrenal gland catecholamines. In addition, non-pituitary corticoliberin receptors are involved in CRF4-6 influences on blood glucose level and body temperature.

The effects of centrally administered tripeptide fragment CRF(4-6) of corticotropin-releasing factor on convulsive activity in outbred albino rats were investigated. The peptide CRF(4-6) (icv; 6, 30, 150 nmol/head) causes dose-dependent increase in total EEG power in 1-40 Hz frequency range as a reflection of tripeptide-induced various epileptiform EEG signs such as single peaks and spike trains without external convulsion. Higher doses of CRF(4-6) (icv; 150, 225, 300 nmol per animal) induce tonicoclonic seizures. Switching to convulsive activity occurs at CRF(4-6) dose of 150 nmol per animal: injection of this dose leads only to EEG paroxysmal activity under habitual conditions and induces pathological locomotor activation under stressing conditions. Thus, CRF(4-6) similarly to full-length corticotropin-releasing factor induces epileptiform activity in rats.

The effects of corticoliberin fragment CRF(4-6) (Pro-Pro-Ile) on pain sensitivity of rats in "hot plate" test were investigated. Intracerebroventricular administration of tripeptide CRF(4-6) (6, 30, 150 nmol/head) induced dose-dependent antinociception: the latency of paw lick response increased by 7.4 +/- 1.4, 10.1 +/- 1.5 and 16.7 +/- 4.2 s respectively from the basic level of 10.2 +/- 0.9 s. Duration of tripeptide antinociceptive action was 30 min (for 6 nmol) and 60 min (for 30 and 150 nmol). Pretreatment with corticotropin-releasing factor antagonist alpha-helical CRF(9-41) (6.5 nmol/head) 60 minutes before tripeptide administration completely abolished the antinociceptive effects of CRF(4-6) (6 nmol). Therefore corticoliberin receptors seem to be involved in realization of tripeptide influence on pain sensitivity. The data obtained suggest that CRF(4-6) can either directly interact with corticoliberin receptors or modulate activity of CRF-ergic neurons.

RESUMEN

Centrally administered corticotrophin-releasing factor fragment CRF(4-6) (Pro-Pro-Ile) inhibits components of male rat sexual behaviour. Administration of 1 and 2 microg of tripeptide leads to dose-dependent increase in mount, intromission and ejaculation latencies. Alterations of sexual behaviour parameters suggest suppression of both sexual motivation and sexual performance.

The effects of tripeptide corticoliberin fragment CRF(4-6) (Pro-Pro-Ile) on blood pressure and heart rate of rats were investigated. Intracerebroventricularly injected CRF(4-6) (1.5-15.0 nmol/head) increases the mean arterial pressure and heart rate in conscious and anaesthetized animals in a dose-dependent manner. Corticotropin releasing factor antagonist: alpha-helical CRF(9-41) (6.5 nmol) completely abolished the influence of tripeptide CRF(4-6) (1.5 nmol) on blood pressure and heart rate in anaesthetized rats. This result indicates that corticoliberin receptors are involved in cardiovascular effects of CRF(4-6).

RESUMEN

The approaches for high sensitive and specific determination of nonylphenol (NP) with the help of immune enzymatic (ELISA) method have been developed. The process of preparation of conjugates of NP with proteins, antiserum obtaining, purification of immunoglobulin (Ig) fractions and study of specificity of the obtained antibodies were described in detail. It was shown that the antiserum and total Ig fraction do not differ in respect of specificity and binding abilities. The developed algorithm of ELISA method fulfillment is able to provide NP revealing with the sensitivity in the range of 20-50 ng/ml and working controlled concentrations--from 20-50 to 1000 ng/ml.

The effects of tripeptide corticoliberin fragment CRF4-6 (Pro-Pro-Ile) on heart rate and behaviour of rats with simultaneous monitoring of these parameters in free-moving animals in their home cage were investigated. Intracerebroventriculary administered CRF4-6 (6, 30, 150 nmol) induces arousal effect increasing the duration of active behaviour, decreasing the duration of passive behaviour and sleep both. At the same time the tripeptide increases the heart rate during sleep, passive and exploratory activity. CRF4-6 (30, 150 nmol) also increases the heart rate in anaesthetised rats. All observed effects of the tripeptide are dose dependent. Taking into account these facts we suggest that CRF4-6 influences behaviour and heart activities independently.

Behavior of nonlinear rats and animals from Wistar and WAG/Rij (with inborn generalized absence epilepsy) strains was examined in the elevated plus-maze and the hole board. WAG/Rij rats demonstrated low exploratory behavior in both tests. In the elevated plus-maze, WAG/Rij rats were more balanced and more anxious than Wistar and nonlinear rats. Administration of ethosuximide completely eliminated spike-wave discharges but did not change behavioral interstrain differences. Since the spike-wave patterns develop in WAG/Rij at the age of 3 months, the behavior of young (2-moth-old) pups from different strains was compared and significant differences were revealed. Correlation between the genetically defined features (spike-wave discharges) and behavioral peculiarities in WAG/Rij rats is supposed.

RESUMEN

The active immunization of rabbits and white rats to antidepressant sydnophen results in the formation of antibodies binding norepinephrine, dophamine, serotonine as well peptide regulatory compounds: substance P, pynorphine, vasopressin and beta-endorphin. The immunization against endogenic antidepressant thyroliberin induces the formation of antibodies to the same biogenic amines and to the gamma-aminobutyric acid, oxytocin and leu encephalin. The data obtained are discussed in connection with some physiological and biochemical changes found earlier during immunization to antidepressants.

The influence of TRH (100 micrograms/kg) and ACTH4-7 Pro-Gly-Pro (15 micrograms/kg and 25 micrograms/kg) involved an acceleration of the spatial learning on the 12-arm radial maze and on T-maze in rats; an increase in the number of correct choice after short (5-10 min) retention interval; and affected the working and reference spatial memory in the test of reinforced place performance. The effects of the TRH and ACTH4-7 Pro-Gly-Pro did not interfere with one another.
