INTRODUCTION: Despite advancements in implantable cardioverter-defibrillator (ICD) technology, sudden cardiac death (SCD) remains a persistent public health concern. Chagas disease (ChD), prevalent in Brazil, is associated with increased ventricular tachycardia (VT) and ventricular fibrillation (VF) events and SCD compared to other cardiomyopathies. METHODS: This retrospective observational study included patients who received ICDs between October 2007 and December 2018. The study aims to assess whether mortality and VT/VF events decreased in patients who received ICDs during different time periods (2007-2010, 2011-2014, and 2015-2018). Additionally, it seeks to compare the prognosis of ChD patients with non-ChD patients. Time periods were chosen based on the establishment of the Arrhythmia Service in 2011. The primary outcome was overall mortality, assessed across the entire sample and the three periods. Secondary outcomes included VT/VF events and the combined outcome of death or VT/VF. RESULTS: Of the 885 patients included, 31% had ChD. Among them, 28% died, 14% had VT/VF events, and 37% experienced death and/or VT/VF. Analysis revealed that period 3 (2015-2018) was associated with better death-free survival (p = .007). ChD was the only variable associated with a higher rate of VT/VF events (p < .001) and the combined outcome (p = .009). CONCLUSION: Mortality and combined outcome rates decreased gradually for ICD patients during the periods 2011-2014 and 2015-2018 compared to the initial period (2007-2010). ChD was associated with higher VT/VF events in ICD patients, only in the first two periods.

Ticks are blood-feeding arachnids that are known to transmit various pathogenic microorganisms to their hosts. During blood feeding, ticks activate their metabolism and immune system to efficiently utilise nutrients from the host's blood and complete the feeding process. In contrast to insects, in which the fat body is known to be a central organ that controls essential metabolic processes and immune defense mechanisms, the function of the fat body in tick physiology is still relatively unexplored. To fill this gap, we sought to uncover the repertoire of genes expressed in the fat body associated with trachea (FB/Tr) by analyzing the transcriptome of individual, partially fed (previtellogenic) Ixodes ricinus females. The resulting catalog of individual mRNA sequences reveals a broad repertoire of transcripts encoding proteins involved in nutrient storage and distribution, as well as components of the tick immune system. To gain a detailed insight into the secretory products of FB/Tr specifically involved in inter-tissue transport and humoral immunity, the transcriptomic data were complemented with the proteome of soluble proteins in the hemolymph of partially fed female ticks. Among these proteins, the hemolipoglyco-carrier proteins were predominant. When comparing immune peptides and proteins from the fat body with those produced by hemocytes, we found that the fat body serves as a unique producer of certain immune components. Finally, time-resolved transcriptional regulation of selected immune transcripts from the FB/Tr was examined in response to experimental challenges with model microbes and analyzed by RT-qPCR. Overall, our data show that the fat body of ticks, similar to insects, is an important metabolic tissue that also plays a remarkable role in immune defense against invading microbes. These findings improve our understanding of tick biology and its impact on the transmission of tick-borne pathogens.

The objective was to evaluate the effects of bovine leukemia virus (BLV) infection, as determined by BLV seropositivity and proviral load, on 305-d milk, fat, and protein production of dairy cows. A cross-sectional study was conducted among 1,712 cows from 9 dairy herds in Alberta, Canada. The BLV status was assessed using an antibody ELISA, whereas BLV proviral load in BLV-seropositive cattle was determined with quantitative PCR. Dairy Herd Improvement 305-d milk, fat, and protein production data were obtained for all enrolled cattle. Differences in these milk end points were assessed in 2 ways: first, by categorizing cows based on BLV serostatus (i.e., BLV positive or negative), and second, by categorizing based on BLV proviral load (i.e., BLV negative, low proviral load [LPL] BLV positive, and high proviral load [HPL] BLV positive). A mixed-effect multivariable linear regression model was used to assess differences in milk parameters. We found that BLV positivity, adjusted for parity and natural log-transformed somatic cell count (SCC), was not associated with reduction in 305-d milk, fat, or protein production. However, significant reductions in 305-d milk, fat, and protein yield occurred in HPL cows, but not in LPL cows, compared with BLV-negative cows, when adjusted for parity number and natural log-transformed SCC. In summary, BLV proviral load may predict effects of BLV infection on milk, fat, and protein production.

Tick saliva injected into the vertebrate host contains bioactive anti-proteolytic proteins from the cystatin family; however, the molecular basis of their unusual biochemical and physiological properties, distinct from those of host homologs, is unknown. Here, we present Ricistatin, a novel secreted cystatin identified in the salivary gland transcriptome of Ixodes ricinus ticks. Recombinant Ricistatin inhibited host-derived cysteine cathepsins and preferentially targeted endopeptidases, while having only limited impact on proteolysis driven by exopeptidases. Determination of the crystal structure of Ricistatin in complex with a cysteine cathepsin together with characterization of structural determinants in the Ricistatin binding site explained its restricted specificity. Furthermore, Ricistatin was potently immunosuppressive and anti-inflammatory, reducing levels of pro-inflammatory cytokines IL-6, IL-1ß, and TNF-α and nitric oxide in macrophages; IL-2 and IL-9 levels in Th9 cells; and OVA antigen-induced CD4+ T cell proliferation and neutrophil migration. This work highlights the immunotherapeutic potential of Ricistatin and, for the first time, provides structural insights into the unique narrow selectivity of tick salivary cystatins determining their bioactivity.

O suporte do supervisor à família conceitua-se como comportamentos dos supervisores voltados a apoiar os papéis familiares dos funcionários. O presente estudo objetivou obter evidências de validade da Medida de Comportamento Suporte do Supervisor à Família, nas versões abrangente e reduzida, em amostras brasileiras. Participaram desta pesquisa 503 trabalhadores, de ambos os sexos. As análises fatoriais confirmatórias evidenciaram que a versão abrangente ficou composta por 14 itens, divididos em quatro fatores de primeira ordem (emocional, instrumental, modelo de papéis e gestão criativa) e um fator de segunda ordem. A versão reduzida unifatorial compôs-se por quatro itens. Ambas as versões apresentaram bons índices de consistência interna. Os instrumentos correlacionaram-se positivamente com o enriquecimento trabalho-família, suporte do supervisor e satisfação no trabalho, e negativamente com o conflito trabalho-família e intenção de rotatividade. Concluiu-se que os instrumentos apresentaram propriedades psicométricas que recomendam seu uso em investigações futuras.(AU)


The concept of a family-supportive supervisor refers to supervisors' behaviors aimed at supporting employees' family roles. This study aimed to obtain evidence of validity of the Measure of Family Supportive Supervisor Behaviors, both in its comprehensive and abbreviated versions, using Brazilian samples. A total of 503 workers of both sexes participated in this research. Confirmatory factor analyses showed that the comprehensive version consisted of 14 items, grouped into four first-order factors (emotional, instrumental, role model and creative management), all contributing to a second-order factor. The unifactorial reduced version consisted of four items. Both versions showed good internal consistency indices. The instruments exhibited positive correlations with work-family enrichment, supervisor support, and job satisfaction while demonstrating negative correlations with work-family conflict and turnover intention. It was concluded that the instruments presented psychometric properties that recommend their use in future research.(AU)


El apoyo del supervisor a la familia se define a las acciones de los supervisores dirigidos para respaldar los roles familiares de los empleados. El presente estudio tuvo como objetivo obtener evidencias de validez de la Medida del Comportamiento de Apoyo del Supervisor a la Familia, en las versiones extensa y breve, en muestras brasileñas. En esta investigación participaron 503 trabajadores de ambos sexos. Los análisis factoriales confirmatorios revelaron que la versión extensa quedó compuesta por 14 ítems, divididos en cuatro factores de primer orden (emocional, instrumental, modelo a seguir y gestión creativa) y un factor de segundo orden. La versión breve unifactorial constaba de cuatro ítems. Ambas versiones mostraron buenos índices de consistencia interna. Los instrumentos se correlacionaron positivamente con el enriquecimiento trabajo-familia, el apoyo del supervisor y la satisfacción laboral, y negativamente con el conflicto trabajo-familia y la intención de rotación. Se concluye que los instrumentos presentan propiedades psicométricas que respaldan su uso en futuras investigaciones.(AU)

Acute pancreatitis (AP) is a serious inflammatory disorder and still lacks effective therapy globally. In this study, a novel Ranacyclin peptide, Ranacin, was identified from the skin of Pelophylax nigromaculatus frog. Ranacin adopted a compact ß-hairpin conformation with a disulfide bond (Cys5-Cys15). Ranacin was also demonstrated effectively to inhibit trypsin and have anticoagulant and antioxidant activities in vitro. Furthermore, the severity of pancreatitis was significantly alleviated in l-Arg-induced AP mice after treatment with Ranacin. In addition, structure-activity studies of Ranacin analogues confirmed that the sequences outside the trypsin inhibitory loop (TIL), especially at the C-terminal side, might be closely associated with the efficacy of its trypsin inhibitory activity. In conclusion, our data suggest that Ranacin can improve pancreatic injury in mice with severe AP through its multi-activity. Therefore, Ranacin is considered a potential drug candidate in AP therapy.

BACKGROUND: Implant-based submuscular breast reconstruction (SBR) can be performed with the aid of acellular dermal matrices (ADM) for implant coverage on their inferolateral pole, aiming at providing a biological interface for hiding the implant and therefore reducing the risk of complications. The purpose of this study is to assess the long-term post-operative outcomes obtained using the SBR-specific Native® ADM (DECO med s.r.l., Marcon, Venice, Italy). METHODS: All cases of Native®-assisted immediate SBR performed at our institution between October 2016 and March 2020 were retrospectively analysed. Demographic and surgical data were collected, and post-operative outcomes, including minor and major complications, were evaluated. Particular attention was paid to complications emerging before and after patient discharge. Dependence analyses were performed to uncover statistically significant relationships between risk factors and reconstructive outcomes. RESULTS: Data on 100 patients were collected, for a total of 128 breasts. The mean age of the cohort was 49.5 years, the mean BMI was 23.4 kg/m2, and the mean follow-up was 24 months. Out of this, 14.1% of patients received pre-operative radiotherapy, while 16.4% underwent post-mastectomy radiotherapy. Breasts appeared to develop short-term minor complications more likely during hospitalisation (11.7% vs. 7.8%), while short-term major complications occurred more often after discharge (7.8% vs. 15.6%). The most frequent long-term complications were capsular contracture and contour defects (both 9.4%). Risk factors that showed a statistically significant relationship with complications were pre- and post-mastectomy radiotherapy and post-operative chemotherapy. CONCLUSIONS: The retrospective analysis showed results in line with clinical outcomes reported in the literature for the same reconstructive technique. The use of Native® ADM in SBR is safe and effective in the long term.

The hematophagy behavior has evolved independently several times within the Arthropoda phylum. Interestingly, the process of acquiring a blood meal in ticks is considerably distinct from that observed in other blood-feeding arthropods. Instead of taking seconds to minutes to complete a blood meal, an adult female Ixodes scapularis tick can remain attached to its host for numerous days. During this extended feeding period, the tick undergoes drastic morphological changes. It is well established that the tick midgut plays a pivotal role not only in blood meal digestion but also in pathogen acquisition and transmission. However, our understanding of the underlying molecular mechanisms involved in these events remains limited. To expedite tick research, we conducted a comprehensive longitudinal RNA-sequencing of the tick midgut before, during, and after feeding. By collecting ticks in different feeding stages (unfed, slow feeding, rapid feeding, and early post-detached), we obtained a comprehensive overview of the transcripts present in each stage and the dynamic transcriptional changes that occur between them. This provides valuable insights into tick physiology. Additionally, through unsupervised clustering, we identified transcripts with similar patterns and stage-specific sequences. These findings serve as a foundation for selecting targets in the development of anti-tick control strategies and facilitate a better understanding of how blood feeding and pathogen infection impact tick physiology.

Objective: to evaluate the results of performance indicators 6 (proportion of people with hypertension, with consultation and blood pressure checked in the semester) and 7 (proportion of people with diabetes mellitus, with consultation and glycated hemoglobin requested in the semester) of the Previne Brasil Program in the Massif of Baturité, located in Ceará in the period from 2020 to the year 2023. Methods: Cross-sectional study with a quantitative approach Results: The Maciço de Baturité region has 136 primary health care units in active status and registered in the National Register of Health Establishments (CNES), with a total of 99 Family Health teams (eSF), displaying a coverage indicator of 100% eSF. However, the performance indicators of the Previne Brasil program showed inadequate results. Indicator 6 did not reach the target of 50% coverage in the years 2020 to 2022 and indicator 7 reached the target only in 2021, however it shows a downward trend in 2022. Conclusion: the coverage of the indicators is unsatisfactory, in view of the predominance of quarters where the targets established for the indicators are not achieved. However, the study reveals that the coverage rate of Family Health teams is adequate in the thirteen municipalities evaluated. Thus, the research expresses the need to review the profile of care for patients with chronic non-communicable diseases.


Objetivo: avaliar os resultados dos indicadores de desempenho 6 (proporção de pessoas com hipertensão, com consulta e pressão arterial aferida no semestre) e 7 (proporção de pessoas com diabetes mellitus, com consulta e hemoglobina glicada solicitada no semestre) do Programa Previne Brasil no Maciço de Baturité, situado no Ceará no período de 2020 ao ano de 2023. Métodos: Estudo transversal de abordagem quantitativa Resultados: A região do Maciço de Baturité possui 136 unidades de atenção primária à saúde em situação ativa e registradas no Cadastro Nacional de Estabelecimentos de Saúde (CNES), com o total 99 equipes de Saúde da Família (eSF), exibindo um indicador de cobertura de eSF de 100%. Contudo, os indicadores de desempenho do programa Previne Brasil apresentaram resultados inadequados. O indicador 6 não alcançou a meta de 50% de cobertura nos anos de 2020 a 2022 e o indicador 7 atingiu a meta somente no ano de 2021, todavia apresenta uma tendência de queda em 2022.  Conclusão: a cobertura dos indicadores é insatisfatória, em observância ao predomínio de quadrimestres onde as metas estabelecidas para os indicadores não são alcançadas. No entanto, o estudo revela que a taxa de cobertura de equipes de Saúde da Família encontra-se adequada nos treze municípios avaliados. Assim, a pesquisa expressa a necessidade de revisão no perfil de atendimento a pacientes com doenças crônicas não transmissíveis.

BACKGROUND: The tick Amblyomma sculptum is the major vector of Rickettsia rickettsii, the causative agent of the highly lethal Brazilian spotted fever. It has been shown that R. rickettsii inhibits apoptosis in both human endothelial cells and tick cells. Apoptosis is regulated by different factors, among which inhibitors of apoptosis proteins (IAPs) play a central role. In the study reported here, we selected an IAP of A. sculptum that has not yet been characterized to assess its role in cell death and to determine the effects of its gene silencing on tick fitness and R. rickettsii infection. METHODS: An A. sculptum cell line (IBU/ASE-16) was treated with specific double-stranded RNA (dsRNA) for either IAP (dsIAP) or green fluorescent protein (dsGFP; as a control). The activity of caspase-3 and the exposure of phosphatidylserine were determined in both groups. In addition, unfed adult ticks, infected or not infected with R. rickettsii, were treated with either dsIAP or dsGFP and allowed to feed on noninfected rabbits. In parallel, noninfected ticks were allowed to feed on an R. rickettsii-infected rabbit. Ticks (infected or not with R. rickettsii) that remained unfed were used as a control. RESULTS: Caspase-3 activity and the externalization of phosphatidylserine were significantly higher in IBU/ASE-16 cells treated with dsIAP than in those treated with dsGFP. The mortality rates of ticks in the dsIAP group were much higher than those in the dsGFP group when they were allowed to feed on rabbits, independent of the presence of R. rickettsii. Conversely, lower mortality rates were recorded in unfed ticks. CONCLUSIONS: Our results show that IAP negatively regulates apoptosis in A. sculptum cells. Moreover, IAP-silenced ticks experienced higher mortality rates following the acquisition of a blood meal, suggesting that feeding may trigger the activation of apoptosis in the absence of this physiological regulator. These findings indicate that IAP is a potential antigen for an anti-tick vaccine.

Sexual assault perpetrated by older adults is still an underreported crime and is not fully understood. To describe a series of cases of sexual assault by Brazilian older adults and discuss the associated factors based on the literature. This is a cross-sectional descriptive analysis of five cases of older adults accused of sexual assault in the Forensic Psychiatry sector of the Medical Legal Institute (Coroner's Office) in the City of Belo Horizonte'. This review focuses on five cases involving male defendants with a mean age of 59.75 years; two with previous psychiatric treatment, and one with a previous criminal record. Their victims were prepubescent children, four of them female. Psychiatric conditions related to forensic diagnosis included dementia, schizophrenia, alcohol-related substance use disorder, paraphilia, and one without a psychiatric diagnosis. The final forensic report on all five cases found that two of the older adults diagnosed with a mental disorder in two were considered to have a mental illness that completely impaired the cognitive and volitional domains; two other adults were diagnosed with mental health disturbance that partially impaired these domains; and the last adult was diagnosed and found without mental health impairment. While the incidence of sexual crimes committed by older adults is low, the absolute number of cases has expected to rise due to population aging. Assessing the factors related to this type of crime is therefore an initial step to understanding and formulating preventive strategies.

Abstract Job crafting is characterized by worker's proactive behavior aiming to give meaning and significance to their work relations, allowing the individual to assume a leading role by acting as an agent of change. The study aimed to gather evidence of validity for the Job Crafting Hierarchical Scale in Brazil. Study sample consisted of 675 Brazilian workers of both genders, aged between 18 and 82 years. Confirmatory Factor Analysis pointed out that the 18-item model, divided into four reflective factors (increase in resources, increase in challenging demands, reduction in demands, and optimization of demands) and one formative factor (job crafting) presented the best adjustment indexes. Results showed a positive correlation with work engagement and a negative correlation with work exhaustion. In conclusion, the instrument presented psychometric properties that recommend its use in future investigations on job crafting in the Brazilian context.


Resumo O job crafting é caracterizado pelo comportamento proativo do trabalhador que objetiva dar sentindo e significância a sua relação laboral, possibilitando que o indivíduo assuma um papel de protagonista ao atuar como um agente da mudança. O estudo objetivou reunir evidências de validade da Escala Hierárquica de Job Crafting, no contexto brasileiro. A amostra constituiu-se por 675 trabalhadores brasileiros, de ambos os sexos, com idades variando entre 18 e 82 anos. A Análise Fatorial Confirmatória apontou que o modelo de 18 itens, divididos em quatro fatores reflexivos (aumento de recursos, aumento de demandas desafiadoras, redução de demandas e otimização de demandas) e um fator formativo (job crafting) foi o que apresentou os melhores índices de ajuste. Os resultados demonstraram correlação positiva com engajamento no trabalho e negativa com a exaustão no trabalho. Concluiu-se que o instrumento apresentou propriedades psicométricas que recomendam seu uso em futuras investigações sobre o job crafting no contexto brasileiro.


Resumen Job crafting se caracteriza por el comportamiento proactivo del trabajador que pretende dar sentido y trascendencia a su relación laboral, posibilitándole al individuo asumir un rol protagónico actuando como un agente del cambio. Este estudio tuvo como objetivo recopilar evidencias de validez de la Escala Jerárquica de Job Crafting, en el contexto brasileño. La muestra estuvo compuesta por 675 trabajadores brasileños, de ambos sexos, con edades entre 18 y 82 años. El Análisis Factorial Confirmatorio señaló que el modelo de 18 ítems, divididos en cuatro factores reflexivos (aumento de recursos, aumento de demandas desafiantes, reducción de demandas y optimización de demandas) y un factor formativo (job crafting), fue el que presentó mejores índices de ajuste. Los resultados mostraron una correlación positiva con el compromiso en el trabajo y negativa con el agotamiento en el trabajo. Se concluyó que el instrumento presentó propiedades psicométricas que recomiendan su uso en futuras investigaciones sobre job crafting en el contexto brasileño.

BACKGROUND: Chronic Chagas Cardiomyopathy (CCC) usually develops between 10 and 20 years after the first parasitic infection and is one of the leading causes of end-stage heart failure in Latin America. Despite the great inter-individual variability in CCC susceptibility (only 30% of infected individuals ever present CCC), there are no known predictors for disease development in those chronically infected. METHODOLOGY/PRINCIPAL FINDINGS: We describe a new susceptibility locus for CCC through a GWAS analysis in the SaMi-Trop cohort, a population-based study conducted in a Chagas endemic region from Brazil. This locus was also associated with CCC in the REDS II Study. The newly identified locus (rs34238187, OR 0.73, p-value 2.03 x 10-9) spans a haplotype of approximately 30Kb on chromosome 18 (chr18: 5028302-5057621) and is also associated with 80 different traits, most of them blood protein traits significantly enriched for immune-related biological pathways. Hi-C data show that the newly associated locus is able to interact with chromatin sites as far as 10Mb on chromosome 18 in a number of different cell types and tissues. Finally, we were able to confirm, at the tissue transcriptional level, the immune-associated blood protein signature using a multi-tissue differential gene expression and enrichment analysis. CONCLUSIONS/SIGNIFICANCE: We suggest that the newly identified locus impacts CCC risk among T cruzi infected individuals through the modulation of a downstream transcriptional and protein signature associated with host-parasite immune response. Functional characterization of the novel risk locus is warranted.

Adherence to secondary prevention measures after acute coronary syndrome (ACS) is essential to prevent disease recurrence. In Brazil, the Brazilian Unified Health System (SUS, in Portuguese), and the private healthcare system (PHCS) coexist. We aimed to evaluate the adherence to secondary prevention in patients with ACS who were assisted by either SUS or PHCS. In this longitudinal prospective study, patients with ACS were admitted to the four cardiological reference hospitals of Sergipe, three of which assisted PHCS users, and one, SUS users. We analyzed the two patient care models with multiple logistic regression models for adherence to physical activity, pharmacotherapy, and smoking cessation. We enrolled 581 volunteers in this study: 44.1 % from SUS and 55.9 % from PHCS. PHCS users showed greater adherence to pharmacotherapy at both 30 and 180 (p = 0.001) days after ACS with better results in all classes of medications (p < 0.05) than SUS users did. They also showed better adherence to physical activity (p = 0.047). There was no distinction between the groups regarding smoking cessation. The secondary prevention measures after ACS were more effective in PHCS users than in SUS users due to better adherence, especially to pharmacotherapy and regular physical activity.

Ticks are blood-feeding arthropods that use the components of their salivary glands to counter the host's hemostatic, inflammatory, and immune responses. The tick midgut also plays a crucial role in hematophagy. It is responsible for managing blood meals (storage and digestion) and protecting against host immunity and pathogen infections. Previous transcriptomic studies revealed the complexity of tick sialomes (salivary gland transcriptomes) and mialomes (midgut transcriptomes) which encode for protease inhibitors, lipocalins (histamine-binding proteins), disintegrins, enzymes, and several other tick-specific proteins. Several studies have demonstrated that mammalian hosts acquire tick resistance against repeated tick bites. Consequently, there is an urgent need to uncover how tick sialomes and mialomes respond to resistant hosts, as they may serve to develop novel tick control strategies and applications. Here, we mimicked natural repeated tick bites in a laboratory setting and analyzed gene expression dynamics in the salivary glands and midguts of adult female ticks. Rabbits were subjected to a primary (feeding on a naive host) and a secondary infestation of the same host (we re-exposed the hosts but to other ticks). We used single salivary glands and midguts dissected from individual siblings adult pathogen-free female Ixodes ricinus to reduce genetic variability between individual ticks. The comprehensive analysis of 88 obtained RNA-seq data sets allows us to provide high-quality annotated sialomes and mialomes from individual ticks. Comparisons between fed/unfed, timepoints, and exposures yielded as many as 3000 putative differentially expressed genes (DEG). Interestingly, when classifying the exposure DEGs by means of a clustering approach we observed that the majority of these genes show increased expression at early feeding time-points in the mid-gut of re-exposed ticks. The existence of clearly defined groups of genes with highly similar responses to re-exposure suggests the existence of molecular swiches. In silico functional analysis shows that these early feeding reexposure response genes form a dense interaction network at protein level being related to virtually all aspects of gene expression regulation and glycosylation. Lipophorin is a major hemolymph lipoprotein found in insects with a molecular native mass of 700 kDa. In mosquitoes, two different types of apolipoproteins are characterized, apolipophorin-I (ApoLp-I, ~250 kDa) and apolipophorin-II (ApoLp-II, ~80 kDa). This concentration depends on the stage of development and the age of the insects. Lipophorins are best studied in mosquitoes of the genus Aedes and Anopheles. In this study, we analyze the lipophorin sequence and show the lipophorin purification of the Culex quinquefasciatus and the transcriptional profile of the lipophorin gene in different life cycle stages. Similar amino acid composition and molecular weights are founded in three mosquitoes species lipophorins amino acid sequence. The two subunits of purified lipophorin (Apo I and Apo II) showed molecular masses of approximately 248 and 93 kDa, like that found in other mosquitoes. A gradual increase in the lipophorin expression gene was obtained during the previtellogenic period and after feeding we obtained peak expression at 24 h after feeding. With our results, we conclude that C. quinquefasciatus protein sequence has the same characteristics as those observed in other mosquitoes and that the expression of its apolipophorins is induced by blood feeding.

(1) Background: Quality of life (QOL) is used as a health indicator to assess the effectiveness and impact of therapies in certain groups of patients. This study aimed to analyze the QOL of patients with acute coronary syndrome (ACS) who received medical treatment by a public or private health care system. (2) Methods: This observational, prospective, longitudinal study was carried out in four referral hospitals providing cardiology services in Sergipe, Brazil. QoL was evaluated using the Medical Outcomes Study 36-Item Short-Form Health Survey. The volunteers were divided into two groups (public or private health care group) according to the type of health care provided. Multiple linear regression models were used to evaluate QoL at 180 days after ACS. (3) Results: A total of 581 patients were eligible, including 44.1% and 55.9% for public and private health care, respectively. At 180 days after ACS, the public health care group had lower QoL scores for all domains (functional capacity, physical aspects, pain, general health status, vitality, social condition, emotional profile, and health) (p < 0.05) than the private group. The highest QoL level was associated with male sex (p < 0.05) and adherence to physical activity (p ≤ 0.003) for all assessed domains. (4) Conclusions: This shows that social factors and health status disparities influence QoL after ACS in Sergipe.

Dietary restriction (DR) reduces adiposity and improves metabolism in patients with one or more symptoms of metabolic syndrome. Nonetheless, it remains elusive whether the benefits of DR in humans are mediated by calorie or nutrient restriction. This study was conducted to determine whether isocaloric dietary protein restriction is sufficient to confer the beneficial effects of dietary restriction in patients with metabolic syndrome. We performed a prospective, randomized controlled dietary intervention under constant nutritional and medical supervision. Twenty-one individuals diagnosed with metabolic syndrome were randomly assigned for caloric restriction (CR; n = 11, diet of 5941 ± 686 KJ per day) or isocaloric dietary protein restriction (PR; n = 10, diet of 8409 ± 2360 KJ per day) and followed for 27 days. Like CR, PR promoted weight loss due to a reduction in adiposity, which was associated with reductions in blood glucose, lipid levels, and blood pressure. More strikingly, both CR and PR improved insulin sensitivity by 62.3% and 93.2%, respectively, after treatment. Fecal microbiome diversity was not affected by the interventions. Adipose tissue bulk RNA-Seq data revealed minor changes elicited by the interventions. After PR, terms related to leukocyte proliferation were enriched among the upregulated genes. Protein restriction is sufficient to confer almost the same clinical outcomes as calorie restriction without the need for a reduction in calorie intake. The isocaloric characteristic of the PR intervention makes this approach a more attractive and less drastic dietary strategy in clinical settings and has more significant potential to be used as adjuvant therapy for people with metabolic syndrome.

BACKGROUND: Digital dermatitis (DD) is a multifactorial infectious disease affecting the skin on feet of cattle causing erosion and inflammation above the heel bulbs. Some cases of DD cause lameness and significantly impact animal welfare and productivity. While DD has emerged as a concern for the beef industry, key information regarding early detection and its impact on cattle behaviour is lacking. The primary objective of this study was to determine if an established DD experimental model for dairy calves could be used to induce DD lesions in beef calves. A secondary objective was to describe changes in behaviour and pain associated with induction of DD lesions. Eight beef calves acquired from a single cow-calf operator were enrolled in the study. Upon enrolment, calves were evaluated and determined to be free of foot lesions. Within the experimental environment, calves were housed in individual pens and assigned to two groups (mock-inoculated and inoculated). Both hind feet of each calf were enrolled. Within calf, inoculation protocol was consistent, and a 28-day experimental protocol was employed. Two days prior to inoculation, both hind feet of each calf were abraded (area above the heel bulbs and below the dewclaws), moistened, and wrapped to facilitate an anaerobic condition. Feet were inoculated with macerated DD lesion material or mock inoculum and remained wrapped until clinical signs of DD or protocol endpoint. RESULTS: After a period of 14 to 18 days post inoculation, three of five inoculated calves developed clinical signs (lameness), and upon close inspection, DD lesions were present on at least one hind foot. Two of five inoculated calves did not develop lesions within 28 days. Zero of three mock-inoculated calves developed DD. Treponema spp. were detected by quantitative polymerase chain reaction from biopsies of induced lesions. Measurements of behaviour prior to disease induction were numerically different between DD affected and mock-inoculated calves. CONCLUSIONS: An experimental infection model established for dairy cattle was used to successfully induce acute DD lesions in three of five inoculated beef calves. This model can provide a framework to study intervention protocols and to evaluate the impact of DD on behaviour and pain.

OBJECTIVE: To analyze the effects of electrolysis, through a medium frequency current, associated to aerobic physical activity in the body composition of young women. METHODS: The study was composed of 34 sedentary women (24.35 ± 4.43 years, 71.30 ± 7.08 kg, 1.61 ± 0.06 m, 27.31 ± 1.67 kg/m2) which were evaluated for their anthropometric measures and body composition. The volunteers were randomly assigned to two group: Electrolyphysis plus Aerobic Exercise (gEEA): 17 volunteers were submitted to the application, for 60 min , of the Aussie current, followed by aerobic physical activity (77% of HRmax) on the trampoline for 40 min, through video-lessons of Jump; and Aerobic Exercise group (gEA): 17 volunteers performed only physical activity following the same parameters mentioned above. Each group performed its protocols twice weekly, for 5 weeks, totaling 10 sessions. For the data analysis, measures repeated ANOVA was performed to compare the means of the variables analyzed before and after the treatment protocols using the SPSS - 21.0 software, adopting a p ≤ 0.05. RESULTS: Although gEEA decreased suprailiac skinfold (p = 0.04), abdominal skinfold (p = 0.03) and circumference at umbilical scar (p = 0.02) in an intragroup analysis, these means differences in anthropometric measures were not important between-groups (p > 0.05). Furthermore, there were no effect of treatment on body composition (p > 0.05). CONCLUSION: To this studied condition, our results suggested that application of medium frequency electrolysis did not enhance the losses on anthropometric measures and body composition.
