RESUMEN

Two optimal variants of growth medium with different content of growth proteins obtained from cattle and porcine blood sera are recommended for culturing lymphoblastoid cells. Use of growth proteins helps optimize the conditions of cell culturing by ruling out the toxic effect and Mycoplasma contamination and ensures the standardization of virological experiments due to the absence of gamma-globulin fraction.

RESUMEN

The authors recommend to replace nonstandard native animal sera by growth-stimulating proteins isolated from the sera of cattle, northern deer, pigs, and sheep by fractionation with polyethylene glycol aqueous solutions (mol. m 6000 dalton). Growth-stimulating proteins are conducive to active proliferation of primary and continuous cell cultures at various methods of culturing. Growth-stimulating proteins were for the first time added to nutrient media during culturing of mammalian cells on microcarriers.

RESUMEN

Collase, an enzymatic preparation made from crab hepatopancreas, was used as a dissociative reagent in preparation of primary cell cultures and for detachment of continuous cells from the substrate during reinoculation. Collase was found to increase viable cell harvest by 2 to 2.5 times in comparison with trypsin and had a less detrimental effect on the cells which retained their proliferative activity, morphology, productivity, and sensitivity to viruses.

RESUMEN

Rapid methods for determining the hemolytic activity in 10% suspension of human red cells and the cytotoxicity in human embryo fibroblast and HeLa cell cultures were used to assess the biological characteristics of some experimental composite filling materials and their foreign analogs. The results permit assessing the impact on the biological characteristics of individual components, of the entire composition, of the purity of the initial ingredients and their polymerization ability, this being valuable for the development of materials with the optimal characteristics.

RESUMEN

The possibility of practical use of an increase in the level of antibody to a future causative agent of epidemic from minimal to the "critical" level in the human population against the background of a relative epidemic-free state in a town as a prepotent of an epidemic increase of influenza incidence was studied. During 4 spring and 3 autumn seasons (1978--1981) titres of antihemagglutinins to influenza A (H3N2) and B viruses were determined in 200--250 blood serum specimens from residents of Moscow selected with similar age distribution for each season against a working reference human serum. From the analysis of the antibody dynamics influenza A (H3N2) epidemic in 1979 and influenza B epidemic in 1980 were timely prognosed.

RESUMEN

The principal possibility of using beta-propiolactone-inactivated and lyophilized antigens of influenza A and B viruses and of mouse immune ascitic fluids to them for the determination of the antigenic specificity of neuraminidase has first been established. A simpler and easier method requiring no expensive reagents: inhibition of influenza virus release from erythrocytes was tested. This method is as specific as the labour-consuming and difficult test of neuraminidase activity inhibition, and allows a rapid and accurate identification of the type of neuraminidase of influenza viruses of various origins to be made.

RESUMEN

The results of the observations show that after active circulation of influenza A (H1N1), A (H3N2), and B viruses the degree of immune response (a rise in antihemagglutinin titres to the causative agent of an epidemic) differed significantly in subjects with different blood groups of the ABO(H) system. After active circulation of influenza A(H1N1) and A(H3N2) viruses, rises in the antibody titres to the etiological factor of the epidemic was more marked in subjects with blood groups O and A than in those with blood group B. After an influenza A(H3N2) epidemic a rise in the titres to this virus in subjects with blood group B occurred a season later and did not reach those high levels observed in subjects with blood groups O and A. A reverse picture was observed during increasing morbidity and epidemic of influenza B virus. A rise in the antibody titres to this virus in subjects with blood group B significantly different from that among subjects with the other two blood groups occurred earlier, in the pre-epidemic season (the spring of 1980) and persisted till the end of the epidemic season. This time difference in the onset of the antibody titre rises appears to indicate a higher genetic sensitivity of subjects with blood group O followed by blood group A to influenza A viruses and, first of all, their increased sensitivity to A(H3N2) virus, as well as higher susceptibility to influenza B virus in subjects with blood group B.

The analysis of immune and hyperimmune rabbit sera to influenza A/USSR/0249/75. A/Victoria/3/75, and A/Tokyo/1/75 viruses showed the H3 antigenic determinant common for hemagglutinins of influenza viruses of the Hong Kong variety to be poorly manifest in viruses isolated in 1975 in contrast to type A viruses isolated in previous years. Antibody to the H3 antigen is usually absent in immune sera and appears only in hyperimmune sera after reimmunization of the animals. The quantitative decrease of the H3 antigen in viruses isolated in 1975 was also clearly detected in tests of adsorption of anti-H3 monoreceptor sera.
