RESUMEN

Diabetes Mellitus is accompanied by chronic hyperglycemia, inflammation, and related molecular processes, which leads to diabetic neuropathy. In this work, we tested Thiadiazine-thione (TDT) synthetic derivatives TDT1 and TDT2 against streptozotocin (STZ)-induced diabetic neuropathy. Sprague Dawley's rats, SH-SY5Y neuronal and BV2 microglial cells were employed in this work, followed by behavioral, biochemical, and morphological studies utilizing RT-qPCR, ELISA, Immunoblotting, immunohistochemistry, Immunofluorescence, and in silico analyses. TDT1 and TDT2 abolished STZ-induced allodynia and hyperalgesia. Next, we examined IRS1/PI3K/AKT signaling to assess TDT1 and TDT2's impact on diabetic neuropathy. STZ downregulated IRS1, PI3K, AKT mRNA and protein expression in rat spinal cord and SH-SY5Y neuronal cells. TDT1 and TDT2 improved IRS1, PI3k, and AKT mRNA and protein expression. STZ elevated GSK3ß mRNA and protein expression in vivo and in vitro, whereas TDT1 and TDT2 mitigated it. STZ increased the expression of inflammatory mediators such as p-NF-κB, TNF-α, and COX-2 in rat spinal cord lysates. TDT1 and TDT2 co-treatment with STZ decreased inflammatory cytokine expression by ameliorating astrocytosis (revealed by increased GFAP) and microgliosis (indicated by increased Iba1). TDT1 and TDT2 reduced STZ-induced JNK, Iba1, and COX-2 upregulation in BV2 microglial cells validating our in vivo findings. In silico molecular docking and MD simulations analyses suggested that TDT1 and TDT2 have IRS binding affinity, however, both compounds had an identical binding affinity, but distinct interaction pattern with IRS protein residues. Overall, these findings demonstrate that TDT derivatives mitigated STZ-induced neuropathy through modulating the insulin and inflammatory signaling pathways.

This study examined the changing character of the last honours of those who died of COVID-19 in Kashmir and the life experiences of the families of the deceased. A semi-structured interview schedule was used to collect information from 21 participants. Using qualitative data analysis approaches, five key themes were identified vis-à-vis the impact of COVID-19 on burial rituals and customs; effects on bereaved families, shades of grief, bereavement care, community response, and coping with loss. Based on examining the pandemic-induced changes related to customs and rituals around death, the study found that the bereaved family members were in danger of marginalization, economic burdens, psychological traumas, and overall reduced quality of life. This study would be a credible addition to the existing literature on death practices as there is a shortage of research on funeral rituals during the post-pandemic period in Kashmir.

Soil pollution with heavy metals (HMs) has become a world environmental problem. This study focuses on surface soil contamination with Cr, Mn, Co, Ni, Cu, Zn, Cd, Hg, Pb, Fe, and Al, their sources, and potential hazards along the basin of River Swat, Pakistan. The average concentrations (mg/kg) of HMs were the most abundant for Al (24,730.19) followed by Fe (22,419.41) > Mn (386.78) > Zn (57.75) > Cr (38.07) > Ni (32.46) > Cu (23.43) > Pb (19.59) > Co (10.77) > Cd (3.18) > Hg (0.12). The concentrations of Cr and Mn in 5.45% each, Co in 10.90%, Zn in 27.27%, Cu in 36.36%, Ni in 41.81%, and Hg in 92.72% of the total soil samples exceeded their respective background values. The geostatistical approaches determined the distribution patterns of HM pollution along the basin, whereas the statistics of principal component analysis exposed the likely sources of HM contamination in the area. Pollution indices evaluated the overall HM distribution and pollution status in the area. Contamination factor showed a high degree of HM contamination in 82% of the total sampling sites, while the geo-accumulation index designated low to moderate contamination with Cr, Mn, Co, Ni, Cu, Zn, Hg, and Pb, and moderate to extreme contamination with Cd, Fe, and Al. The trend of ecological toxicity showed potential ups and downs along with the sites from low to considerable hazard (< 95 < PEHI < 190), whereas the human carcinogenic hazard was within the USEPA acceptable limits (1 × 10-7-1 × 10-4), but the non-carcinogenic hazard was higher than the threshold (HI > 1) for children because they are more exposed than adults.

RESUMEN

The modified surgical flap technique for placement of implant in the anterior maxilla has been described. The flap design has been used to achieve optimal soft-tissue contour for implant restoration. A modification in the surgical technique has been employed with the intention to preserve the interdental papilla, which is the key to the maintenance of hygiene and esthetics around anterior implants. This, in turn, prevents black triangles and improves soft-tissue contours. The modified surgical flap method maintains both functional and esthetic values.

AIM: To evaluate the effect of hyperbaric oxygen (HBO) therapy on the osseointegration of dental implants by resonance frequency analysis. MATERIALS AND METHODS: Six rabbits of age 2 to 2 and 1/2 years, weight approximately 2 kg were selected and tagged 1-6. For all the animals' right femur was selected as a control group (R) and left femur as test group (L). Initially, implants of dimensions 3.75×8mm (Adin Touareg) were placed in the right femur. Implant stability quotient (ISQ) values were recorded using OSSTELL ISQ at the time of surgery (R0), after one month (R1), and the end of the second month (R2). After two months of uneventful healing, implants were placed on the left femur of all the six rabbits and three were grouped as 2S (subjected to 2 HBO sessions at the weekly interval) and other three as 4S (subjected to 4 HBO sessions at weekly interval for a month). At the time of surgery (L0), end of one month (L1) and two months (L2), ISQ values were recorded and subjected to statistical analysis. The total duration of the study was 4 months from 3 March 2013 to 03 July 2013. RESULTS: The data were statistically analyzed using t-test and analysis of variance (ANOVA) F. On the comparison between the control group (R) and test groups (2S and 4S) ISQ values for test groups were more which was highly statistically significant (p <0.001). Among the two test groups 4S group has more ISQ values compared to 2S (p <0.001). CONCLUSION: This study indicated that HBO therapy has a promotive effect on the rate of osseointegration of dental implants. CLINICAL SIGNIFICANCE: Study opens new scope for further in vivo research in utilizing hyperbaric oxygen therapy (HBOT) in implant surgeries, maxillofacial trauma cases and irradiated patients to hasten or improve osseointegration.

RESUMEN

Reattachment of tooth fragment is a simple, conservative, and noninvasive procedure, and it is the most currently acceptable treatment option. This article presents management of two accidentally damaged maxillary incisors using direct composite resin restoration and fractured tooth fragment. With the advancements in adhesive dentistry, tooth fragment reattachment procedure has become simpler and clinically reliable. The present paper is a report of 3-year follow-up of coronally fractured tooth treated with a very conservative technique of tooth fragment reattachment using vertical groove preparation and reinforcement with fiber post.

INTRODUCTION: The aim of this study was to evaluate & compare the wetting ability of five saliva substitutes & distilled water on heat-polymerized acrylic resin. Contact angle of the saliva substitute on denture base can be taken as an indicator of wettability. Good wetting of heat-polymerized acrylic resin is critical for optimum retention of complete dentures. METHODS: Two hundred & forty samples of heat-polymerized acrylic resin were fabricated using conventional method. 240 samples divided into 6 groups with 40 samples in each group. Advancing & Receding contact angles were measured using Contact Angle Goniometer & DSA4 software analysis. RESULTS: Anova test was carried out to test the significance in difference of contact angle values in the six groups. The mean of advancing angle values & mean of receding angle values of all the six groups has shown statistically significant difference between the groups. The mean of angle of hysteresis values of all the six groups are statistically not significant between the groups. A multiple comparison using Bonferroni's test was carried out to verify the significance of difference between the contact angles in a pair of groups. Statistically significant difference was seen when Aqwet (Group II) was compared to Distilled water (Group I), Wet Mouth (Group III), E-Saliva (Group IV), Biotene (Group V), and Moi-Stir (Group VI). CONCLUSION: The contact angles of five saliva substitutes and distilled water were measured and compared. Group II (AQWET) has the lowest advancing and receding contact angle values and the highest angle of hysteresis on heat-polymerized acrylic resin. Based on contact angle values, Group II (AQWET) has the best wetting ability on heat-cured acrylic resins. The ability of saliva to wet the denture surface is one of the most important properties for complete denture retention in dry mouth cases.

INTRODUCTION: Xerostomia (dry mouth) is dryness of the mouth which is due to reduced salivary flow. Lack of adequate saliva causes discomfort in denture wearing patients and decreases retention of dentures. The ability of saliva to wet the tissue surface is one of the most important properties for oral comfort and retention of complete denture in dry mouth patients. AIM: This study was conducted to evaluate and compare the wetting ability of therapeutic Aloe vera saliva substitute and commercially available Aqwet saliva substitute on heat-polymerized acrylic resin. Contact angle of liquid saliva substitute was considered as an indicator of wettability. MATERIALS AND METHODS: Aloe vera liquid (Aloe vera - Group I) and Aqwet saliva substitute (Aqwet - Group II) were compared in terms of their wetting ability. Forty samples of heat-polymerized acrylic resin were fabricated and divided into two groups with 20 samples in each. Advancing, receding contact angles and angle of hysteresis were measured using contact angle goniometer and DSA4 software analysis. Mann-Whitney U test was applied for statistical analysis of the study. RESULTS: The mean advancing angle and receding angle of Group I (Aloe vera) was smaller than Group II (Aqwet). Mean angle of hysteresis of Group I (Aloe vera) was higher than Group II (Aqwet). Mann-Whitney U test revealed that there is no significant difference in contact angles between the two groups. CONCLUSION: Wetting ability of Group I (Aloe vera) saliva substitute was found to be better compared to Group II (Aqwet) on heat-polymerized acrylic resin.

BACKGROUND: Patients undergoing radiation therapy for oral squamous cell carcinoma (OSCC) have an increased risk of developing oral candidiasis. Radiation-induced hyposalivation is thought to be a major predisposing factor for it. Radiation therapy for 2-3 weeks leads to increase in the number of candidal species in the oral cavity, eventually leading to oral candidiasis. OBJECTIVE: To evaluate and correlate the prevalence of candidal species in the oral cavity of OSCC patients and to compare with radiation dosage at 3rd and 6th week following radiotherapy. MATERIALS AND METHODS: This study includes fifty patients undergoing radiation therapy for OSCC. Patients were examined at 3rd and 6th week following radiation therapy, and smear samples were obtained from lesional sites and the radiation dose was recorded. Smear samples were cultured on CHROMagar, and identification of various candidal species was done on the basis of colony color and their morphology. RESULTS: The present study isolated four types of candidal species, namely, Candida albicans, Candida glabrata, Candida tropicalis and Candida krusei. All the organisms, except C. krusei, showed a significant increase from 3rd to 6th week of radiotherapy whereas C. glabrata and C. tropicalis showed a sudden exponential increase. Total radiation dosage did not show any correlation to candidal colonization at 3rd and 6th week following radiotherapy. CONCLUSION: OSCC patients undergoing radiotherapy show an increase in candidal colonization which is independent of radiation dosage and may be related to other factors. However, such findings should be further evaluated using a larger sample size.

Parafunctional activities associated with the stomatognathic system include lip and cheek chewing, nail biting, and teeth clenching. Bruxism can be classified as awake or sleep bruxism. Patients with sleep bruxism are more likely to experience jaw pain and limitation of movement, than people who do not experience sleep bruxism. Faulty occlusion is one of the most common causes of bruxism that further leads to temporomandibular joint pain. Bruxism has been described in various ways by different authors. This article gives a review of the literature on bruxism since its first description.

OBJECTIVE: To determine the frequency of metabolic syndrome in both genders, in a limited adult type 2 diabetic population presenting to Pakistan Institute of Medical Sciences, Islamabad. METHODS: This was a cross sectional study conducted in a tertiary care teaching hospital. During the six months of study period, 106 adult type 2 diabetics were examined and evaluated for the presence of metabolic syndrome according to the ATP-III criteria. Asian standards for the waist circumference were used. RESULTS: Out of 106 patients, 91 (85.8%) had metabolic syndrome of whom 95% were females. Abdominal obesity was present in 91% females and 86% males. Low HDL levels were present in all females and 83% males. Seventy eight percent females and 63% males had elevated levels of triglycerides. Hypertension was present in 68% and 73% females and males respectively. CONCLUSION: This study showed a very high prevalence of the metabolic syndrome in type2 diabetic population. Females were more affected than males in all respects.
